2.Research progress of genetic engineering on medicinal plants.
China Journal of Chinese Materia Medica 2015;40(4):594-601
The application of genetic engineering technology in modern agriculture shows its outstanding role in dealing with food shortage. Traditional medicinal plant cultivation and collection have also faced with challenges, such as lack of resources, deterioration of environment, germplasm of recession and a series of problems. Genetic engineering can be used to improve the disease resistance, insect resistance, herbicides resistant ability of medicinal plant, also can improve the medicinal plant yield and increase the content of active substances in medicinal plants. Thus, the potent biotechnology can play an important role in protection and large area planting of medicinal plants. In the development of medicinal plant genetic engineering, the safety of transgenic medicinal plants should also be paid attention to. A set of scientific safety evaluation and judgment standard which is suitable for transgenic medicinal plants should be established based on the recognition of the particularity of medicinal plants.

Chinese Journal of Tissue Engineering Research 2013;(37):6620-6627
BACKGROUND:We have built the three-dimensional human skin melanoma model with human epidermal keratinocytes and MV3 melanoma cel s co-cultured on the de-epidermized dermis in vitro.
OBJECTIVE:To establish a skin model of melanoma by mixed culture of MV3 melanoma cel s and HaCaT cel s on the de-epidermized dermis in vitro.
METHODS:MV3 melanoma cel s and HaCaT cel s were mixed with different percentages and inoculated on the surface of de-epidermized dermis fol owed by a liquid culture and air-liquid culture, and then the tissue-engineered skin model was established in vitro. Routine biopsy immunohistochemical observation was performed on the constructed skin melanoma model.
RESULTS AND CONCLUSION:Hematoxylin-eosin staining showed that MV3 melanoma cel s were distributed on the surface layer of de-epidermized dermis and formed tumor masses, while the HaCaT cel s were mixed growth with tumor cel s and formed a typical epidermoid structure. Some tumor cel s infiltrated into the surface or deep of de-epidermized dermis and showed a tumor foci distribution. The CK10, CK-pan and S-100 proteins were positive for immunohistochemical staining. With the increasing of MV3:HaCaT cel percentage, CK10 and CK-pan gradual y down-moved from the surface, and changed from layer distribution to lumpy distribution, while the staining of S-100 protein was gradual y distributed layer-by-layer, and some area showed tumor-like distribution. The results show that the skin model of melanoma can be in vitro constructed successful y by mixed culture of MV3 melanoma cel s and HaCaT cel s on de-epidermized dermis.

Acta Anatomica Sinica 1955;0(03):-
The synthesis of cellular and nuclear basic proteins in the first generation of the repopulated pachytene spermatocytes (RPS) after the withdrawal of gossypol treatment and the normal control pachytene spermatocyte (CPS) were studied by the incorporation of ~3H-arginine into proteins. After treatment with gossypol for various periods of time, the synthetic activity for cellular protein in the RPS was slightly inhibited. In comparison to CPS, the inhibitory rate ranged from 12% to 26% (gossypol treatment for 3 to 12 weeks, respectively). However, the synthetic activity for nuclear basic protein in the RPS was drastically reduced in comparison to that of the CPS. The reductive rate ranged from 46% to 61% in RPS (gossypol treatment for 3 to 12 weeks, respectively).Gel electrophoretic separation of the basic protein extracted from normal pachytene cell indicated that the major basic proteins were nucleosomal linker and core proteins, i. e., histone H_1, H_(2a), H_(2b), H_3 and H_4, and with a lesser amount of spermspecific BP and X_1 proteins. In the RPS, the synthesis of sperm-specific proteins(BP and X_1) and core histones (H_(2a) and H_4) reduced markedly. Finally, the effect of gossypol on the ratio change of nuclear basic protein to DNA in RPS and its relation to nucleosomal spacing and chromatin structure are discussed.

Chinese Journal of Cancer Biotherapy 1995;0(03):-
GST/MCP-1 fusion protein was overexpressed in Escherichia coli as inculstion bodies. After isolation of the inclusion bodies, the optimum conditions of denaturation and renaturation were studied. The renatured produts were purified to be electroporetically pure by affinity chromatography on immobilised glutathione. The purified product remains biological activities and can react specifically with MCP-1 antibodies by western blot analysis. The in vitro antitumor effect showed that GST/MCP-1 could activate monocytes and lymphocytes to inhibit the growth of a lung adenocarcinoma cell line A549. In vivo, GST/MCP-1 could inhibit the tumor growth in nude mice. These results suggested that MCP-1 had antitumor effect.

Journal of Shanghai Jiaotong University(Medical Science) 2001;21(2):165-167
Objective To investigate the infiltration of inflammatory cells and the pathogenesis of nasal polyps. Methods the nasal polyps were obtained from 42 patients undergoing nasal operations. The tissues from each patient were stained with HE and Toluidine Blue for eosiophils, lymphocyte, plasma cell and mast cell and observed under the light microscope. A mean number of the above cells were counted. All data were analyzed with the Ttest. Results Eosinophils were found predominantly in 12 polyps(28.57%). Lymphocyte infiltration was shown in the other 30 polyps (71.43%). Furthermore, many mast cells with only a few eosinophils were found in the lymphocyte infiltration polyps; the number of mast cells in these polyps were much more than those in eosinophil accumulation polyps with a very significant difference(P<0.01). Conclusion The accumulation of eosinophils in the nasal polyps does not appear to be related to the presence of mast cells and allergy. The disorder of immunity may play an important role in the formation of nasal polyps.

Journal of Clinical Otorhinolaryngology Head and Neck Surgery 2015;29(14):1268-1271
OBJECTIVE:
Differences in clinical features, especially facial nerve canal leision between cholesteatoma in external auditory meatus and middle ear were compaired.
METHOD:
A retrospective clinical analysis was made. Clinical data included 125 cases of middle ear cholesteatoma with facial nerve canal leision and 28 cases of cholesteatoma occurred in external auditory canal from 2003-01-2014-08 in our hospital.
RESULT:
Clinical course of cholesteatoma in external auditory canal was 4.97 ± 7.51 years, course of middle ear cholesteatoma was 16.60 ± 14.42 years (P < 0.01). 21 cases (75%) of external auditory canal cholesteatoma were manifested as pneumatic mastoid and 110 cases (88%) of middle ear cholesteatoma were manifested as diploic mastoid respectively. 22 cases (78.6%) of facial nerve canal damage-in mastoid segment in cholesteatoma of external auditory meatus and 76 cases (60.8%) of facial nerve canal damage in tympanic segment in cholesteatoma of middle ear were observed (P < 0.01). The incidence rate of ossicular errosion in middle ear chol-esteatoma was significantly higher than that in external auditory meatus (P < 0.01). The incidence of semicircular canal defects in middle ear cholesteatoma (30.4%), was significantly higher when comparing to the incidence (10.7%) in cholesteatoma of external auditory meatus (P < 0.05).
CONCLUSION
The site of facial nerve canal lesion in middle ear cholesteatoma and cholesteatoma of external auditory meatus were different. More attention should be paid before and during operation to avoid facial nerve injury, including physical examinations, especial otologic exams, radiological reading and careful operation.
