1.Nicotine effects on the osseointegration of implants with different treatments

BACKGROUND:Studies have confirmed that nicotine affects the activity of osteoblasts, osteoclasts, fibroblasts and erythrocytes.
OBJECTIVE:To study the nicotine effects on osseointegration and the expression of osteoprotegerin and bone morphogenetic protein 2 after implantation of dental implants with surface treatment by sandblasting or acid etching.
METHODS:Twenty-four Sprague-Dawley rats were randomly assigned to two groups and received daily injections for 2 weeks as folows: Nicotine 2 mg/kg twice for experimental group, saline solution for control group. Then the titanium implants with surface sandblasting or acid etching were implanted into the tibiae folowed by continuous nicotine or normal saline injection. At weeks 2 and 4 after implantation, the implants and surrounding bone tissue were prepared for CT, X-ray and hematoxylin-eosin staining examinations to evaluate bone healing and expression levels of bone-related genes were measured by quantitative RT-PCR.
RESULTS AND CONCLUSION: Compared with the control groups, the degree of osseointegration and the expression of osteoprotegerin and bone morphogenetic protein 2 in the experimental groups were decreased significantly (P < 0.05), except that the expression of bone morphogenetic protein 2 in the experimental group with acid etching was not significantly reduced. In addition, the expression of bone morphogenetic protein 2 in the experimental group with acid etching was higher than that in the experimental group with sandblasting at 2 weeks after implantation (P < 0.05). The X-ray and CT show that the quantities of new generation bone and the degree of bone mineralization of the sandblasting group were significant lower than those of the acid etching group under the intervention of nicotine. Hematoxylin-eosin staining showed that the activity and quantity of osteoblasts around the implants down-regulated significantly, but acid etching-treated implants showed better outcomes than sandblasting-treated implants.
2.Adverse reactions of compound salvia drop pills and its preventive measures

Objective To learn and analyze adverse reactions of compound salvia drop pills in order to promote rational use.Methods ADR reports of Compound Salvia Drop Pills were retrieve and analyze through CNKI database from 1979 to 2013.Results Ten ADR reports were found.Adverse reactions of compound salvia drop pills were mainly gastrointestinal reaction,including cardiovascular,uropoiesis,nervous reactions.Conclusion More attention should pay to the patient who have taken compound salvia drop pills.Preventive measures should be prepared for possible adverse reactions.Suitable treatment should be carried out promptly against severe ADR.
3.The research progress of Lynch syndrome-associated endometrial cancers

Lynch syndrome.(LS) is an autosomal dominant condition caused by a mutation in the mismatch repair genes (MMR).Endometrial cancer (EC) is the most common extra-colonic cancers of LS Ⅱ type.Among the female members of LS family with the MMR gene mutation,EC has an overall lifetime risk more than 35%.LS-associated EC is more likely present as the clinical pathological characteristics of early age of onset,endometrioid adenocarcinoma,lower grade,lower uterine segment involvement,and better prognosis.LS patients with family history and early age of onset should be performed with a combined screening and genetic testing,while high-risk patients should be taken clinical monitoring methods as endometrial biopsy.Oral contraceptives mav be reasonable chemopreventive agents in the patients with LS.Although prophylactic hysterectomy with bilateral salpingo-oophorectomy is an effective strategy to prevent EC in women with LS,preoperative counseling should address the trade-offs between the reduction in the risk of cancer,and the risks and side effects of surgery.The patients need hormone replacement therapy after surgery.
4.The effect of basic periodontal therapy on blood glucose control in diabetes patient with periodontitis

Objective:To investigate the effect of basic periodontal therapy on levels of blood glucose,blood lipid,and tumor necrosis factor-alpha(TNF-?) among patients with periodontal diseases in insulin dependent diabetes patients.Methods:Forty insulin dependent diabetes patients with periodontitis were selected and divided into basic periodontal therapy group and control group.The periodontal clinic index,blood glucose,blood lipid,glycosylated hemoglobin(HbA1c),and serum TNF-? concentrations were assessed before treatment and 4 weeks,3 and 6months after basic periodontal therapy.Results:The HbA1c,TNF-?,total cholesterol(TC),tyiglycerides(TG) in basic periodontal therapy group were significant decreased after periodontal therapy,and meanwhile had significant difference compared with that in control group.Conclusion:The basic periodontal therapy can help to control the blood glucose level in insulin dependent diabetes mellitus with periodonitis.

Objective To study the dynamic changes and clinical significance of D-D, t-PA and PAI in patients with acute renal failure during the process of different methods of blood purification. Methods Thirty-seven ARF patients were divided into three groups: HD group, HDF group and HF group. Plasma D-D level, t-PA and PAI activity were determined 1 hour, 4 hours before and after treatment. Normal control group consisted of fourteen healthy people. 7.Pharmaceutical Care for a Case of Pan-drug Resistant Acinetobacter baumannii Pulmonary Infection

OBJECTIVE:To explore the role of clinical pharmacists in the treatment of pan-drug resistant Acinetobacter bau-mannii infection. METHODS:Clinical pharmacists participated in the treatment for a severe pneumonia case of pan-drug resistant A. baumannii infection. Clinical pharmacists supplied overall pharmaceutical care and suggestions with respects to initial medication scheme evaluation,pathogen judgment,therapy drug selection,ADR disposal,etc.,including anti-infective treatment of moxifloxa-cin 0.4 g,ivgtt,qd+meropenem 0.5 g,ivgtt,q8 h+linezolid 0.6 g,ivgtt,q12 h;anti-pan-drug resistant A. baumannii infection of cefoperazone sodium and sulbactam sodium 3.0 g,ivgtt,q8 h+tigecycline 50 mg,ivgtt,q12 h;liver protection of ademetionine 1, 4-Butanedisulfonate 1.0 g,ivgtt,qd+reduced glutathione 1.8 g,ivgtt,qd. RESULTS:After 25 d treatment,the patient hadn’t been fe-vered,and hemogram and hepatic function index decreased to normal value. CONCLUSIONS:Clinical pharmacist should be en-gage in anti-infective treatment and pharmaceutical care,and provide physicians reasonable medication suggestion so as to promote care rate in the clinic.
8.Correlation of Clinical Efficacy of XPD Gene Polymorphisms and Platinum-based Chemotherapy in Ad-vanced Non-small Cell Lung Cancer:A Meta-analysis

OBJECTIVE:To systematically review the relationship of clinical efficacy between XPD Lys751Gln (A/C),XPD Asp312Asn(G/A)and platinum-based chemotherapy in patients with advanced non-small cell lung cancer(NSCLC),and provide evidence-based reference for clinical treatment. METHODS:Retrieved from PubMed, Cochrane Library, EMBase, Medline, CJFD,VIP database and WanFang database,studies about the effects of XPD Lys751Gln and XPD Asp312Asn polymorphism on ef-fectiveness,clinical outcomes and adverse drug reaction of platinum-based chemotherapy in advanced NSCLC patients were collect-ed,and Meta-analysis was performed by using Rev Man 5.3 software. RESULTS:Totally 30 studies were included,involving 5 028 patients. Genetic testing showed that XPD Lys751Gln divided into mutant gene (Lys/Gln + Gln/ Gln) and wild-type gene (Lys/Lys),while XPD Asp312Asn divided into mutant gene (Asp/Asn + Asn/Asn) and wild-type gene (Asp/Asp). Results of Me-ta-analysis showed,the progression-free survival (PFS) of Lys/Gln+Gln/Gln patients with platinum in XPD Lys751Gln polymor-phism was obviously lower than Lys/Lys patients [OR=-1.12,95%CI(-1.73,-0.50),P<0.001],while there was no significant difference in the chemotherapy effectiveness and total survival period. The effective rate of Asp/Asn+Asn/Asn patients for platinum in XPD Asp312Asn polymorphism was lower than Asp/Asp patients [OR=0.80,95%CI(0.68,0.96),P=0.02],while there was no significant difference in the total survival period and PFS. Meanwhile,the incidence of Ⅲ-Ⅳ level gastrointestinal adverse reac-tions of Lys/Gln+Gln/Gln with platinum in XPD Lys751Gln polymorphism was higher than Lys/Lys patients [OR=0.43,95%CI (0.20,0.94),P=0.03],and there was no significant difference in Ⅲ-Ⅳ level blood system adverse reactions. CONCLUSIONS:XPD Lys751Gln polymorphism may be associated with PFS and Ⅲ-Ⅳ level gastrointestinal adverse reactions for advanced NSCLC patients with platinum-based chemotherapy,while XPD Asp312Asn polymorphism may have effect on platinum-based chemotherapy,both of them may be as estimate the chemotherapy effect and prognosis detection index of platinum-based chemo-therapy.

Objective To screen the amyloid protein-β(Aβ)degradation-associated genes through peripheral blood monocytes(PBMC)-extracted gene microarray analysis in patients with Alzheimer's disease(AD).Methods PBMC were isolated from blood of elderly AD patients versus age-matched healthy individuals(control).The cDNA(mRNAs)were analyzed using gene microarray.And real-time quantitative polymerase chain reaction detection and enzyme activity analysis were used to verify the primary outcome of gene chip.Results The expression of cathepsin D mRNA in peripheral blood monocytes was 104.70±15.96 in AD patients as compared with the control group 49.86±5.19,and the activity of cathepsin D was (22 620 ± 1 389) RFU in AD versus (32 210 ± 2 284) RFU in control (both P<0.05).Conclusions The results suggest that the decreased levels of cathepsin D could be the stage markers related to the pathophysiology of AD process.Based on the microarray data,we select cathepsin D genes for further study.
10.miR-200c Regulates RMP7-mediated Increases of Blood-tumor Barrier Permeability by Targeting RhoA

Objective To study the mechanism of miR?200c in regulating RMP7?induced increases of blood?tumor barrier(BTB)permeability by targeting Ras homolog gene family member A(RhoA). Methods Endogenous expression of miR?200c was detected by real?time PCR in hu?man cerebral microvascular endothelial cell line hCMEC/D3(ECs)after RMP7 treatment. miR?200c mimic and miR?200c inhibitor were transfect?ed into GECs(ECs with U87 glioma cells co?culturing),respectively. Transfection efficiency of miR?200c mimic and miR?200c inhibitor were de?termined by real?time PCR. HRP flux and TEER assays revealed BTB permeability. The protein expression level of RhoA was assessed by West?ern blotting. The distribution of RhoA was assessed by immunofluorescence microscopy. RhoA luciferase assays were performed using the Dual?Lucif?erase reporter assay system. Results RMP7 significantly induced a decrease in miR?200c expression in GECs of BTB. miR?200c mimic and miR?200c inhibitor were successfully transfected into GECs. Overexpression of miR?200c inhibited endothelial leakage and restored normal transendo?thelial electric resistance values. Simultaneously ,overexpression of miR?200c significantly reduced the protein expression level of RhoA. In addi?tion,immunofluorescence analysis revealed that the distribution of RhoA in the cytoplasm and nuclei of GECs were decreased in miR?200c mimic group. RhoA was one of the direct targets of miR?200c with the specific binding site being located at the seed sequence. The results of miR?200c si?lencing were opposite to that of the miR?200c overexpression group. Conclusion miRNA?200c regulated RMP7?induced increases in BTB perme?ability by targeting RhoA.
