Objective To study the role of cytidine-phosphate-guanosine oligodeoxynucleotide(CpG ODN) on the development of Plasmodium liver stage. Methods Plasmodium yoelii BY265 18S rRNA was cloned, and the TaqMan real-time PCR was established on P.yoelii BY265 18S rRNA and mouse GAPDH as quantitative analysis model. The model was tested by the level of liver Plasmodium load with the liver cDNA in BALB/c mice infected by salivary gland sporozoites for 42 hours.Twelve BALB/c mice were randomly divided into CpG group,CpG control group and PBS control group which were injected respectively by ODN1826 30 ?g,ODN1826 control 30 ?g and 0.01 mol/L PBS 200 ?l via vena caudalis.Twenty-four hours later, each mouse was inoculated with 100 sporozoites.Mice were sacrificed in 42 hours after infection, and the liver load of Plasmodium was analyzed by TaqMan real-time PCR.Results The cloned Py BY265 18S rRNA gene showed 98% similarity to Py 17XNL.The quantitative analysis model consisted by 18S rRNA and GAPDH showed positive correlation between the level of liver Plasmodium load and the sporozoite inoculation dose to mice. The Plasmodium load in CpG ODN pre-treated mice was reduced to one fifth of the control group(0.28/1.33)(P

Objective To observe the clinical effiect and toxicity of Aidi injection and Huaier particles combined with interventional therapy for primary liver cancer patients. Methods 43 paients with primary liver cancer were randomly divided into two groups:the treatment group 22 cases(Aidi injection and Huaier particles combined with interventional therapy to the primary liver cancer)and the control group 21 cases(treated only with intervention therapy of liver cancer). The recent efficiency and toxicity of the two groups were compared. Results The effective rate of the treatment group was 59.1%,and in control group was 33.3%,and the differences was significant(P<0.01).In the treatment group, clinical benefit rate was 86.4%higher than than of the control group(71.4%),and the difference was significant(P<0.05);The treatment group significantly improved the quality of life of patients,T cell and NK cell activity were improved, and better than the control group(P<0.05).The incidence of gastrointestinal reactions and neutropenia rate in treatment group was lower than the control group(all P<0.05).Conclusion Aidi injection and Huaier Granule interventional therapy could improve the curative effect of primary liver cancer、the immune activity、quality of patients life, and the adverse effects.

Objective: To investigate the effects of normal and pathological bone marrow stromal cells(BMSC) on the proliferation and apoptosis of K562 cells in separated culture and contact culture. Methods: On 2,4,7 and 12 d after culture, the growth of K562 cells was observed under optical microscope;the apoptosis of K562 cells was detected by TUNEL. Results: the growth of K562 cells on CML BMSC in contact culture was faster than the else. The growth of K562 cells in contact culture was significantly faster than that in separated culture after 7 d(P

Objective: To investigate the effects of point thread embedding on the learning and memory of the vascular dementia(VD) model rats and the possible mechanisms. Methods: VD rat models were established by employing improved method of Pulsinelli's four-vessel occlusion, then the model rats were treated with point thread embedding.Morris maze test was used to evaluate the learning and memory of the rats.In vivo recording of long term potentiation(LTP) was performed in the dentate gyrus of hippocampus and changes in each experimental group were observed and compared after high-frequency stimulation(HFS).Results: Compared with models group, the results from the Morris tests in treated group(thread embedding group)enhanced significantly(P

Objective:To explore the diagnostic value of Caprini score for acute pulmonary embolism(APE).Methods:Totally 2 764 patients with suspected APE admitted to Yuncheng Central Hospital were enrolled from January 2012 to January 2019, and 312 patients were diagnosed APE and assigned to APE group finally.Among the patients without APE, 312 patients(control group) were matched with the patients in APE group according to age, gender, weight, blood pressure, surgical history, etc.The general clinical data of the two groups were collected, and the Caprini score of each patient was recorded. The differences between the two groups in clinical data and Caprini score were compared. The area under curves(AUC) of receiver operating characteristic(ROC) was calculated to predict the diagnostic efficacy of Caprini scale for patients with APE.Results:The Caprini score of the APE group was significantly higher than that of the control group[(5.41±2.47)points vs.(2.16±1.28)points, t=1.180, P=0.004]. The Caprini score had a favorable diagnostic efficacy for patients with suspected APE(AUC=0.915, 95% CI: 0.878-0.995, P<0.001), and when the 3.5 cutoff value of Caprini score was determined, the specificity and sensitivity were 87.76% and 95.24%, respectively, with 7.778 of positive likelihood ratio, 0.054 of negative likelihood ratio, and 0.83 of Youden index. Conclusion:Caprini score has strong diagnostic efficacy in patients with APE.

Objective To investigate the effects of endostatin combined with icotinib on chemotherapy sensitivity, tumor markers, and prognosis in patients with advanced non-small cell lung cancer(NSCLC).Methods A total of 88 patients with advanced NSCLC admitted to our hospital from January 2019 to August 2020 were selected and randomly assigned to two groups, with 44 patients in each group. The patients in both groups were treated with pemetrexed+cisplatin(PP)chemotherapy, whereas those in the control group were administered echitinib and those in the observation group was treated with endostatin combined with icotinib.Patients in the two groups were assessed for clinical efficacy, tumor markers[carbohydrate antigen(CA)50,carcinoembryonic antigen(CEA),cytokeratin 19 fragment(CYFRA)21-1],T lymphocyte subsets, microRNA expression levels(miR-34b,miR-204-5p,and miR-158-5p)and tyrosine protein excitation. We also examined janus kinase 2(JAK2)/signal transducer and activator of transcription 3(STAT3)signaling pathways, along with adverse reactions. Results We detected no significant differences between the two groups with respect the total response rate(P>0.05).Following treatment, the levels of serum CA50,CEA,and CYFRA21-1 in the observation group patients were found to be lower than those of patients in the control group(P<0.05).Similarly,the levels of CD3+,CD4+,CD4+/CD8+in observation group patients were higher than those in the control group patients, whereas the levels of CD8+were lower than those in the control patients(P<0.05).Furthermore,the expression levels of miR-34b,miR-204-5p,and miR-158-5p in the observation group patients were higher than those in the control group patients, whereas contrastingly, the levels of J AK2 and STAT3 mRNA expression were lower than those in the control group(P<0.05).In addition, the progression-free survival of the observation group patients was higher than that of control group patients(P<0.05).Conclusion The combination of endostatin and icotinib has clear therapeutic efficacy in patients with advanced NSCLC,which can con-tribute to enhancing their sensitivity to chemotherapy and immune function. We speculate that the underlying mechanisms of action may be associated with a reduction in the activity of the JAK2/STAT3 signaling pathway, which is beneficial for patient prognosis.
