1.Expression of Endothelial Nitric Oxide Synthase and Vascular Endothelial Growth Factor in Human Malignant Melanoma

Objective To investigate the relationship between the expression of endothelial nitric oxide synthase (eNOS), vascular endothelial growth factor (VEGF) and the angiogenesis in human malignant melanoma (MM). Methods Tissue sections from 31 MM patients were examined. The protein expression of eNOS and VEGF was detected using immunohistochemistry technique and morphological quantitative analysis. Microvessel density (MVD) was also counted by immunostaining with anti-FⅧRAg antibody. Results (1)The positive staining of eNOS and VEGF was detected in 71.4% (24 cases) and 83.9% (26 cases) of MM respectively. As negative control there was no staining in the nevus pigmentosus tissue sections. (2)There was a positive correlation between the expression of eNOS and VEGF in MM. (3)The positive correlation was also found between the MVD and the expression of eNOS and VEGF in MM. The number of MVD was higher in MM than that in nevus pigmentosus tissue. (4)The lymphatic metastasis was not correlated with the expression of eNOS and VEGF. Conclusion The number of the MVD increases along with the high expression of eNOS and VEGF in MM.
2.The effects of prostaglandin E_2 on hemodynamics and cardial function in normal rabbits and cardioprotection in experimental myocardial infarction in rabbits

The effects of prostaglandin E_2 on hemodynamics and cardial function in normal rabbits and on cardioprotection in experimental acute myocardial infarction rabbits were investigated. To determine the action of prostaglandin E_2 in normal rabbits (n = 10), we measured arterial systolic pressure (ASP), arterial diastolic pressure (ADP), mean arterial pressure (MAP), heart rate (HR), left ventricular ejection time (LVET), +dp/dt max, -dp/dt max, Vc_E-50, T time after injected prostaglandin E_2 (1?g/min/kg). The results showed that ASP, ADP, MAP were decreased. HR, Vc_E-50, LVET were unchanged. To determine the effects of prostaglandin E_2 on the cardioprotection in acute myocardial infarction in rabbits (n=8), left ventricular systolic pressure (LVEP), +dp/dt max,-dp/dt max, ST of ECG, infarct size were also measured by us. The results showed that +dp/dt max, -dp/dt max were higher than control (n=9), ST lower than control, LVSP and infarct size were unchanged. Thus, prostaglandin E_2 could relax vascular smooth muscle, reduce laod of heart, decrease myocardial oxygen demands, maintain cardiac function during acute myocardial infarction.
3.STUDY ON THE PROTECTIVE IMMUNITY ACTIVATED BY ANTIGENS OF ADULT WORMS, NEWBORN LARVAE AND MUSCLE LARVAE OF TRICHINELLA SPRIALIS

AimTo compare the protective immunity against challenge infection in mice immunized by crude antigen preparations derived from adult worms, newborn larvae and musele larvae of T richinella spiralis. MethodIntestinal adult worms, muscle larvae and blood absolute eosinophil level were counted; the level of serum IgG to antigens of T. spiralis muscle larvae was detected by ELISA. ResultsAdults reduimg rate was 82.19 %, 72.31% and 42.88 % in the adult, newborn larvae and muscle larvae antigen groups respectively. Muscle larvae were reduced 68.83%. 78.19% and 51.96% respectively. The serum IgG titer in all immunized groups increased significantly, the GMRT values of adult, newborn larvae and mudscle larval antigen groups were 7.46, 5.28and 4.92times higher than that in control group respectively. Periphery blood absolute eosinophil level enhanced significantly. ConclusionThe data demonstrate that all of the adult, newborn larvae and muscle larvae antigens of T. spiralis can activate specific humoral and cellular immunity which induce protection to challenge infection in mice. Among these antigens, adult and newborn larvae antigens show better immunogenicity and it may be possible that adult antigen will provide a potential vaccine for trichinosis.
4.Murine basophils express highly in liver and regulate priming of na(i)ve CD4+ T cells to develop into Th2 cells

Objective To compare the levels of murine basophils distribution in different organs and analyze the characteristics of CD4+T cells activation/differentiation regulated by basophils.Methods Firstly,the mononuclear leukocytes were isolated separately from periphery blood,liver,spleen,lung and lymph nodes and the percentages of FcεR I+CD49b+ basophils in different organs were investigated.Secondly,liver basophils were purified by immuno-microbead strategy and co-clutured with different cytokines for 24 h.Apoptosis status of basophils was detected by Annexin V straining method.Thirdly,Mice were subcutaneously implanted with a miniosmotic pump containing 5 μg IL-3.At day 7 after implantation,liver mononuclear cells were harvested and the percentages of basophils were analyzed.Finally,Th1/Th2 polarization influenced by basophils orientated cytokines was investigated when naive OT-Ⅱ CD4 T cells were stimulated with spleen dentritic cells and OVA peptides.Results The percentage of murine basophils in mononuclear leukocytes of liver was significantly higher than that of periphery blood,spleen,lung and lymph nodes under physiological condition.Both of in vitro and ex vivo experiments showed that IL-3 played an inhibitory role on apoptosis of basophils.Cytokines secreted by basophils was investigated to induce Th2 polarization and inhibit Th1 response when naive OT-Ⅱ CD4 T cells were co-cultured with spleen dentritic cells and OVA peptides.Conclusion Th2 response was enhanced and Th1 response was down-regulated by murine basophils,which implied that basophils might participate in the construction of liver specific physiological immuno-tolerant microenvironment.

6.Myasthenia gravis accompanied with thymomas: its treatments and prognosis

Objective To investigate the treatments and prognostic factors of myasthenia gravis (MG) accompanied with thymomas and evaluate the treatment effects of such cases. Methods Our study was based on 103 cases of MG with surgically proved thymoma treated in our hospital The patients were treated with steroid therapy, radiotherapy and chemotherapy We evaluated the long term effects, the survival rate and the prognostic factors. Results The overall effective rate was 78 6%.Twenty three patients obtained a result of complete remission (22 3%), 26 pharmacological remission (25 2%), and 32 improvement (31 1%) The overall survival rates were 77 8% and 48 5% at 5 and 10 years, respectively Based on the three histologic types, the survival rates at 5 and 10 years of lymphyocytic thymoma were 88 9% and 72 7% respectively, of mixed thymoma 83 3% and 58 3%, of epithelial thymoma 55 6% and 10 0% The survival rates of stage Ⅰ and Ⅱ were 89 8% and 76 2% respectively at 5 and 10 years, and of stage Ⅲ and Ⅳ were 35 7% and 0% The survival rates of patients treated with steroid and radiotherapy were 88 9% and 57 1% respectively. Conclusions The administration of steroids therapy, radiotherapy and chemotherapy after removal of thymomas may enhance the long term curative effect in patients of MG with thymoma However, patients with epithelial thymoma or Masaoka's stage Ⅲ and Ⅳ had rather poor prognosis
7.Analysis of risk factors for children with incomplete Kawasaki disease

ObjectiveTo study the risk factors for children with incomplete Kawasaki disease(IKD) to decrease the development of coronary artery lesions (CAL).MethodsAll children diagnosed as IKD from Jan.2005 to Apr.2011 in our department were reviewed retrospectively for their clinical data,laboratory values and treatment measures.ResultsEight of the children (8/36,22.2% ) were positive for CAL.The count of white blood cell( WBC),count of platelet(PLT),hematocrit and C reactive protein(CRP) level were ( 18.36 ± 4.63) × 109/L,(450.30 ± 155.40) × 109/L,( 25.63 ± 3.53 ),( 18.30 ± 3.80) mg/L and ( 13.48 ±3.27) × 109/L,(350.60 ± 56.80) × 109/L,( 33.78 ± 2.24 ),(9.70 ± 2.50) mg/L in the CAL group and non-CAL group respectively.And there were significant differences on the four indexes between CAL group and non-CAL group ( t =2.58,2.65,2.73,2.48,respectively,P < 0.05 ).Pyretolysis time of children first undergoing globulin treatment was (2.5 ± 1.5 ) d and ( 1.5 ± 1.0 ) d in children under 1-year old and those above l-year old respectively,and the difference was significant ( t =2.35,P < 0.05).ConclusionCAL should be cautiously prevented in IKD infants under l-year old with a fever lasting for over 5 days,decreased hematocrit and elevated WBC count,PLT count,Hct and CRP.Infant IKD patients are not so sensitive to intravenous gamma globulin and tend to occur CAL.They should be given an early diagnosis and timely treatment.
8.The preparation of 99Tcm-J591 and its SPECT imaging of nude mice bearing human prostate cancer

Objective To study the binding performance of 99Tcm labeled anti-human prostatic specific membrane antigen (PSMA) monoclonal antibody J591 (99Tcm-J591) and prostate cancer cells in vitro,the biodistribution and SPECT imaging of 99Tcm-J591 in nude mice bearing human prostate cancer in vivo.Methods The monoclonal antibody J591 was labeled with 99Tcm by improved Schwarz method.Labeled antibody was purified by Sephadex G-50.The labeling efficiency and radiochemical purity were measured by paper chromatography and trichloroacetic acid method.The binding performance of J591 and prostate cancer cells was measured by flow cytometry in vitro.The nude mice bearing PSMA-positive C4-2 prostate carcinoma xenografts served as experiment group,mice bearing PSMA-negative PC3 tumors served as control group.6.2-8.5 MBq of 99Tcm-J591 (25 μg) was intravenously injected into mice.Gamma imaging was performed 2,6,12 and 24 h after injection,T/NT was calculated by ROI technique.After scanned 12 h post injection,4 mice of the experiment group and 5 mice of the control group were sacrificed and the tracer in vivo biodistribution was measured by gamma-counting,and the % ID/g was calculated.Two-sample t test was carried out to validate significant difference of %ID/g between two groups.Results The labeling efficiency and radiochemieal purity of 99Tcm-J591 were (78.9±6.2)% and (92.3±5.1)%,respectively,and the specific activity of 99Tcm-J591 was 68.7 MBq/mg.The antibody J591 and 99Tcm-J591 could strongly combine with PSMA-positive C4-2 cells in vitro,and didn't combine with PSMA-negative PC3 cells in vitro.SPECT imaging results showed that radioactive concentration was obvious in tumor 6 h post injection,the concentration scope became large and the tumor image was clear 12 h post injection.T/NT was 1.9±1.1 at 2 h,4.3±1.8 at 6 h,5.6±2.7 at 12 h,1.4±0.6 at 24 h,respectively.In the control group,no radioactivity concentration was found in tumor,and T/NT was less than 2.The biodistribution results showed that %ID/g of tumor tissue was 20.1±5.2 in the experiment group and 5.8±2.6 in the control group,and there was significant difference (t=5.37,P<0.001).No significant tracer uptake occurred in other tissues and organs between the two groups (all t< 1.98,all P>0.05).Conclusion The immunoactivity,characteristics of biodistribution and tumor targeting property of monoclonal antibody J591 show promising future and potential values in diagnosis and therapy of prostate cancer.
9.Determination of electrical stimulation pain perception and analysis of the related factors.

The electrical pain perception were determined in healthy Chinese adults and factors were assessed. The anxiety-depression state of one hundred healthy Chinese were evaluated and the general information was recorded. Then electrical pain perception (perception threshold, pain threshold, pain tolerance) were assessed by neuromuscular monitor at the same time. Further study of influence factors such as gender, age, body mass index (BMI), left and right sides, degree of anxiety and depression, etc. were carried out on the pain perception. The results showed that 95% of the reference range of electrical perception threshold, pain threshold, pain tolerance were 4-7mA, 7 26mA and 15-60mA, respectively. The bilateral pain perception in males were significantly higher than that in females. There was no difference of effect between left and right sides. Age was positive related to pain perception, degree of depression was negative related to pain perception. BMI, degree of anxiety did not show association with pain perception. Therefore, gender, age and degree of depression were significantly related to electrical pain perception in healthy Chinese adults.

10.Three years follow-up observation and analysis of caries status of primary and permanent teeth among 652 6-year- old children in Sichuan Province.

OBJECTIVEThis longitudinal study aimed to investigate the caries status of primary and permanent teeth among 6-year-old children in Sichuan Province from 2010 to 2012.
METHODSA sample that comprised 652 6-year-old children from six different elementary schools (three represented the urban areas, and the other three represented the rural areas) were examined according to a baseline, with follow-up examinations at 1, 2, and 3 years. Eruption and caries experience were re- corded using World Health Organization criteria.
RESULTSThe prevalence rates of primary tooth caries of 6-year-old children in Sichuan Province for 3 years were 74.23% (484/652), 75.61% (493/652), and 81.90% (534/652). The filling rate of the primary teeth was 5.87% (145/2,471) in 2012, with significant differences (P < 0.01) between the urban areas [10.84% (133/ 1,227)] and rural areas [0.96% (12/1,244)]. The total pit and fissure rate of the first molar was 14.11% (92/652) in 2012, with significant differences between the two areas (P < 0.01) [rural: 0.66% (2/303); urban: 25.79% (90/349)].
CONCLUSIONThe pre- valence of caries in the primary and permanent teeth of 6-year-old children was high. An increasing prevalence tendency was observed as the age increased. The prevalence of first molar caries indicated that prevention and control of dental caries should be performed as early as possible.
