OBJECTIVE:To establish a method for the contents determination of 5 coumarins in sulphur-fumigated and no sul-phur-fumigated Glehnia littoralis,and investigate the effect of sulphur fumigation on the coumarins in G. littoralis. METHODS:HPLC was performed on the column of Hypersil ODS C18 with mobile phase of acetonitrile-0.1% phosphoric acid (gradient elu-tion)at a flow rate of 1.0 ml/min,detection wavelength was 295 nm,column temperature was 30 ℃,and the volume was 20 μl. RESULTS:The linear range was 0.121 5-12.15 μg/L for psoralen,0.389-38.9 μg/L for xanthotoxin,0.055-5.5 μg/L for bergapten, 0.070 5-7.05 μg/L for imperatorin and 0.092 5-9.25 μg/L for isoimperatorin(r≥0.999 7);RSD of precision test was lower than 1.0,RSDs of reproducibility and stability tests were lower than 3.0;recovery was 98.01%-101.97%(RSD<3.0%,n=6). CON-CLUSIONS:The content of coumarins in G. littoralis was obviously decreased after sulphur fumigation. the method is simple and accurate,and canbe used for the quality control of sulphur-fumigated and no sulphur-fumigated G. littoralis.

OBJECTIVE:To observe the efficacy and safety of scopolamine butylbromide in the adjuvant treatment of gastric ul-cer. METHODS:The data of 160 patients with gastric ulcer were retrospectively analyzed and randomly divided into tradition group and combination group. Tradition group was orally treated with acid suppression,hemostasis and other conventional treatment. Based on the treatment of tradition group,combination group was treated with Scopolamine butylbromide for injection 20 mg by in-tramuscular injection,once a day,for continuous 7 d,continue conventional treatment after 7 d. The treatment course was 8 weeks. The clinical efficacy,different complete relief patients with abdominal pain in different time,and the recurrence rate after 1 year in 2 groups were observed,and the incidence of adverse reactions were recorded. RESULTS:The total effective rate in combination group was significantly higher than tradition group,recurrence rate was significantly lower than tradition group,different complete relief patients with abdominal pain in different time were significantly better than tradition group,the difference was statistically sig-nificant(P<0.05). There were no obvious adverse reactions in 2 groups during treatment. CONCLUSIONS:Based on the conven-tional treatment,scopolamine butylbromide can be used in the adjuvant treatment of gastric ulcer. It can obviously shorten the time of abdominal pain in patients with gastric ulcer and reduce the recurrence rate,with good safety.

Objective To investigate the effect and significance of salidroside on Eag1 expression in mouse cervical cancer xenograft.Methods The mouse subcutaneous xenograft model of cervical cancer U14 cells was established.Forty female Kunming mice were divided into 4 groups according to the random principle,the xenograft group,cisplatinum group,salidroside group and cisplatinum combined salidroside group.Then the mass size was measured by using the vemier caliper everyday.The xenograft tumor weight was measured by using the electronic scale and the tumor inhibiting rate was calculated.The immunohistochemistry method was adopted to verify that the xenograft tumor was undifferentiated squamous cell carcinoma.The tumor surrounding tissue in the xenograft group was taken as the control group.RT-PCR and Western blot were adopted to detect the expression levels of Eag1 mRNA and protein in mouse cervical cancer xenografts in each group.Results Kunming mice renograft tumor model was established by subcutaneous injection of cervical cancer U14 cells,the tumor formation rate was 100％.The tumor volume and tumor weight in the cisplatinum group,salidroside group and combined group were lower than those in the senograft group,the xenograft tumor volume and tumor weight in the combined xenograft group were significantly lower than those in the cisplatinum group and salidroside group (P＜0.05);the expressions of Eag1 mRNA and protein in the xenograft group,cisplatinum group and combined group were significantly higher than those in the control group,while which in the cisplatinum group,salidroside group and cisplatinum combined salidroside group were significantly lower than those in the xenograft group,the Eag1 expression level in the combined group was lower than that in the cisplatinum group and salidroside group,the difference was statistically significant(P＜ 0.05).Conclusion Salidroside can inhibit the growth of cervical cancer xenograft and Eag1 expression,and the effect in the combined use with cisplatinum is more significant.

Objective To explore the possible functional mechanism of Er-xian Decoction and its component herbs on the expression of extrogen receptor(ER) in adrenal development rats.Methods Fifty-four development female SD rats were randomly divided into nine groups:normal control group(A),positive control group(B),Er-xian Decoction group(C),Herba Epimedii group(D),Rhizoma Curculiginis group(E),Radix Morindae Officinalis group(F),Radix Angelicae Sinensis group(G),Cortex Phellodendri group(H),Rhizoma Anemarrhenae group(I).After the rats were administrated for six days,serum were got to measure testosterone(T).Bilateral adrenal gland was weighed and embedded by paraffin.Mean optical density(MOD) were studied by the immunohistochemical method and image analyzing system.Results The adrenal coefficient was increased in group B and C,and the level of T was decreased in group C,D,E and I.ER? was mainly observed in the cytoplasm of zona glomerulosa and zona fasciculate.MOD in group C was increased while declined in group B.ER? was mainly observed in nucleus of zona glomerulosa and zona fasciculate.MOD in group C,D,E and I was less than group B.Conclusion Er-xian Decoction and its component herbs of warming Shen(Herba Epimedii,Rhizoma Curculiginis) and nourishing Yin(Rhizoma Anemarrhenae) play a phytoestrogen role by influencing the expression of ER? and ER ? in the adrenal cortex.

Popularizing the knowledge of reproductive ethics for infertile couples and foste-ring their ethics consciousness is a vital problem of normative development of human assisted reproduction technolo-gy and an effective way to solve the ethical issues of human assisted reproduction technology.

Tiredness is a physiological phenomenon,inevitably appears in certain period of exercise.If the tiredness has not been eliminated promptly,it would roll up to fatigue.It is important to analyze the generation,evaluation and recovery of fatigue,to make people come back to training,working or studying as soon as possible.

Objective To investigate indications,technique,prognosis and complications of anterior flap hemipelvectomy (hemipelvectomy using a quadriceps femoris muscle flap with superficial femoral artery)in the treatment of pelvic tumors.Methods From April 2009 to October 2010,10 patients with pelvic tumors underwent anterior flap hemipelvectomy in our department.There were 9 males and 1 female,aged from 30 to 62 years (average,46 years).There were 7 cases of chondrosarcoma,1 case of epithelioid sarcoma,1 case of pleomorphic liposarcoma and 1 case of spindle cell sarcoma.Results Nine patients were followed up for 12 to 30 months (average,21.5 months).Superficial skin necrosis occurred in 1 patient,while there was no wound infection.Local recurrence (33.3％) occurred in 3 patients,including 2 cases of chondrosarcoma and 1 case of epithelioid sarcoma.Three patients died at last,including 2 patients with local recurrence and 1 with pulmonary metastasis from liposarcoma.Two patients survived with pulmonary metastasis,and remaining 3 patients survived without recurrence and metastasis.Conclusion Anterior flap hemipelvectomy is indicated for:(1) patients with extensive soft tissue sarcoma in the buttock,and the tumor or reaction zone involve the subcutaneous tissue; (2) patients with recurrent pelvic tumors,and the conventional posterior flap can't be used due to contamination by the original incision; (3) patients whose posterior flap has poor blood supply due to radiotherapy or other factors.The prerequisite of the surgery is that the blood stream between external iliac artery and femoral artery is unobstructed; moreover the blood vessels are not in the tumor reaction zone.Compared with conventional posterior flap,the anterior flap hemipelvectomy is an easier procedure,which not only can easily cover surgical defect and obtain better surgical margin,but also has less complications.

Objective: To construct a polycistron tandem repeated Echistatin (Ecs) gene. Methods: Three Ecs genes with independent initiation and termination codon were ligated tandem through restriction enzyme sites after amplified with 3 pairs of primers using pMD18T-Ecs as template. The polycistron Ecs gene was inserted into pET30a and expressed in E.coli BL21(DE3) with IPTG induction. The expression results were identified by 18% SDS-PAGE and Western blot. Results: The expression of Ecs polycistron was accomplished with 18% expression level of total protein determined by SDS-PAGE and Western blot. Conclusion: The successful expression of Ecs polycistron provided a new method for the preparation of low molecular weight protein.
