1.Let-7 and HMGA in tumors
Journal of International Oncology 2011;38(9):646-649
High mobility group protein A (HMGA) plays a critical role in the development of a variety of tumors. It's expression level will abnormally increase with the increase of the malignant degrees of astrocytoma and metastasis potential.Let-7 which is a kind of small RNA endogenous coding negatively regulates HMGA genes as a tumor suppressor. The expression of let-7 reduces while HMGA rises in many kinds of tumors was obversed. Therefore, this finding provides new references for the early diagnosis of tumor and also provides new approaches for therapy of tumor.

Chinese Journal of Applied Physiology 2014;30(4):364-367
OBJECTIVETo investigate the effects of E 4031, a sodium calcium exchanger (NCX) agonist, on the isolated cardiac function and resting Ca2+ level in myocardial cells from rats with chronic heart failure.
METHODSRats chronic heart failure model was established by abdominal aorta coarctation with; Isolated heart perfusion by Langendorff apparatus was used to detect heart function and the effects of E 4031 on haemodynamic indexes; Myocardial cells of rats in the model group were extracted quickly and co-incubated with calcium fluorescent indicator fluo3/AM and the impact of E 4031 on the fluorescence intensity in myocardial cells were evaluated by confocal microscopy.
RESULTSHeart function of rats in the model group detected by Langendorff perfusion was significantly reduced after 12 weeks, E 4031 at 10 micromol/L could improve their left ventricular developed pressure(LVDP) and systolic / diastolic maximum rate (+/- dp/dtmax). Compared with the control and sham operation groups, the resting Ca2+ fluorescence intensity of the myocardial cells of rats in model group was at a higher level and went through a process of transient rise and drop, then stably remaining at a low level after co-incubated with 10 micromol/L E 4031.
CONCLUSIONE 4031 can improve the isolated heart function of rats with chronic heart failure, which may be associated with its enhancing the activity of NCX in the myocardial cell membrane and stabilizing intracellular Ca2+ level.

4.Influence of tranilast on expression of OPN and interstitial ED1+ macrophages infiltration in rats with chronic cyclosporine A nephrotoxicity
Chinese Journal of Practical Internal Medicine 2006;0(20):-
Objective To evaluate the effect of tranilast on chronic cyclosporin A(CsA)nephrotoxicity rats and the possible mechanisms of protective effect of it.Methods Rats were on low salt diet and CsA was administered by gastric gavage at a dose of 20 mg/(kg?d)for 28 days, and tranilast was gave to these rats in a dose of 400 mg/(kg?d).The renal function and renal morphology were observed at1,2 and 4 weeks after administration of tranilast. Results CsA induced the corruption of normal condition and weight lost were markedly delayed after administration of tranilast. On histopathology tranilast attenuated tubular cells vacuolations as well as interstitial damage including interstitial ED1+ macrophages infiltration and focal interstitial fibrosis.Meanwhile, tranilast down-regulated the protein of OPN in kidney. Conclusion Tranilast can attenuate the structural changes of kidney in chronic CsA nephrotoxicity rats.Decreased CsA-related ED1+ macrophages,OPN up-regulation expression in the kidney may be related to mechanisms of protective effect of tranilast in the treatment of CsA induced chronic nephrotoxicity.
5.An experimental osteoarthritis induced by implanting composite material into subchondral bone of rabbit knee
Orthopedic Journal of China 2006;0(03):-
[Objective]To investigate the pathogenesis of osteoarthritis by increasing stiffness of subchondral bone to simulate subchondral bone sclerosis to induce the osteoarthritis.[Methods]The proportion between polymethylmethacrylate(PMMA),methylmethacrylate(MMA),hydroxyapatite(HA) and distilled water was adjusted to keep the maximum reaction temperature under 40℃.The composite material PMMA/HA was obtained and compressed on a mechanical material machine to determine its ultimate strength and stiffness, which were compared with those of subchondral bone. The subchondral bone of medial tibial plateau was scraped, where the composite material PMMA /HA was implanted into.Rabbits were sacrificed at 3,6,9 and 12 weeks, respectively. The articular cartilages of the medial tibial plateau were harvested for histological observation.Immunohistochemical analysis was adopted to record the expression and distribution of collagen type Ⅱ and MMP-1 in cartilage at protein level, which were compared with the blank and control groups.Transmission electron microscope was used to examine the ultramicroscopic changes of chondrocytes in blank and 6,12-week-postsurgery groups.[Results]With the time going, the experimental groups gradually presented degeneration, and had increased histological Mankin score.TEM demonstrated degenerative changes of chondrocytes.Collagen type Ⅱ expressed mainly in transition zone and upper deep zone, and MMP-1 expressed predominantly in the superficial and upper intermediate layers of cartilage. Both expressions steadily increased with time passing.[Conclusion]The increased stiffness of subchondral bone induced osteoarthritis in rabbits, which suggests that subcondral bone sclerosis may induce cartilage damage, and that subcondral bone sclerosis is one of the initial pathogenesis of osteoarthritis.

Chinese Journal of Thoracic and Cardiovascular Surgery 2010;26(1):8-12
Objective Background Predicting risk factors for valve replacements is important both for informed consent of patients and objective review of surgical outcomes. Development of reliable prediction rules requires large data sets with ap-propriate risk factors that are available before surgery. Methods Data were from Belling Anzhen Institute of heart, pulmonary and vascular diseases in the period of January 1993 to December 2004. 4482 heart valve replacement patients were analyzed.There. were 848 aortic valve replacements, 2202 mitral valve replacements and 1387 double valve replacements. Logistic regres-sion was used to examine the relationship between risk factors and in-hospital mortality. Results In the multivariable analysis,5 variables in the aortic model (older age, body area, NYHA class IV, creatin, CPB time) , 8 variables in the mitral model ( NYHA class Ⅳ, congestive heart failure, cardiac/thoracic ratio, FS, etiology, LVESD, CPB time, use of IABP) and 7 var-iables in the double valve model (older age, NYHA class Ⅳ, previous myocarditis, diabetes, CPB time, weight index, previ-ous percutaneous mitral balloon valvotomy ) remained independent predictors of the outcome. The mathematical models were highly significant predictors of the in-hospital mortality, and the results were in general agreement with those of others. The area uoder the receiver operating characteristic curve for the aortic model was 0. 921 [ 95% confidence interval ( CI ), 0. 874 to 0. 967 ], for the mitral model was 0. 859 ( 95% CI, 0. 813 to 0. 905 ) aod for dnuhle model was 0. 868 ( 95% CI, 0. 827 to 0.908). The goodness-of-fit statistic for the aortic model was χ~2 = 1.463, P=0.993, for the mitral model was χ~2 = 8.720,P = 0. 366 and for the double valve model was χ~2 = 8 . 134, P = 0. 420. Conclusion We print results and methods for use in day-to-day practice to calculate patient-specific in-hospital mortality after aortic and mitral valve surgery, by the logistic e-quation for each model or a simple scoring system with a look-up table for mortality rate.

Objective To assess the value of color Doppler sonographic appearance and ultrasound-guided needle biopsy on confirmation of diagnosis of atypical breast cancer. Methods 12 patients suspected to be suffering from breast cancer were examined with color Doppler sonography and ultrasound-guided needle biopsy, the results were then analyzed and evaluated. Results Atypical breast cancer had small tumors (diameter less than 1.5cm), rounded or lobular in shape, and regular margin without obvious spicule. The tumors presented hypoecho and there was no acoustic attenuation behind the tumors. The intensity of vascular signals could not be taken as significant evidence for positive diagnosis. Results of ultrasound-guided needle biopsy was helpful for confirmation of the diagnosis. Conclusions Color Doppler sonography was important for the examination of tumor of atypical of breast cancer. The cancer could be confirmed in good time by the extended sonography and ultrasound-guided needle biopsy.
