1.The relationship between serum growth hormone, C-peptide, blood lipids levels and type 2 diabetes nephrophathy

Objective To investigate the relationship between serum growth hormone, C-peptide, blood lipids levels, blood glu-cose, hypertension and type 2 diabetes nephropathy. Methods 82 type 2 diabetes (including 40 without nephropathy and 42 with nephropa-thy) and 39 healthy objects were enrolled. BG,GH,C-P,HbAlc,TC,TG,LDL-C and HDL-C were detected. Results The results showed that the values of blood glucose in each time-phase and HbAlc in both type 2 diabetes groups were significantly higher than that in healthy objects. In type 2 diabetes nephropathy group, the serum levels HbAlc, FGH, TC, TG, LDL-C, systolic blood pressure and diastolic blood pressure were significantly higher than that without nephropathy and those healthy objects(P<0.01). C-P (Oh)and C-P (2h)in type 2 di-abetes nephropathy group was significantly lower than that without nephropathy and those healthy objects(P<0.01). In groups patients of type 2 diabetes nephropathy, the disease duration had significantly longer than patients without nephropathy (P<0.01). The results of cor-related analysis with Pearson showed that HbAlc was significantly positive correlated with FGH(r=0.912, P<0.01), HbAlc was signifi-cantly negatively correlated with C-P(0h)(r=-0.803, P<0.01)and HbAlc was significantly negatively correlated with C-P(2h)(r=-0.771, P<0.01). FGH was significantly positive correlated with TC (r=0.413, P<0.05). Conclusion These results suggested that type 2 diabetes nephropathy has persistent elevated levels of blood glucose and disorder of lipid metabolism. The elevated levels of FGH, hy-pertension and islet B cell secretion function deficiency are important risk factors for type 2 diabetes nephropathy.
2. The role of red nucleus in the modulation of spinal nociceptive transmission and in nociception elicited by muscle spindle afferents

Objective: To analyse the antinociceptive effect of red nucleus (RN) and its role in the antinociceptive effect of muscle spindle afferents. Methods: The single units of RN or wide dynamic range (WDR) neuron in the spinal cord dorsal horn were extracelluarly recorded. The effects of RN stimulation on nociceptive responses (C-fibers-evoked responses, C-responses) of WDR neurons were observed. The influence of muscle spindle afferents elicited by intravenous administration of succinylcholine (Sch) on the spontaneous discharge of RN neurons and on C-responses of WDR neurons were observed. The effect of muscle spindle afferents on C-responses of WDR neurons after unilateral lesions of RN was also observed. Results: Electrical stimulation of the RN produced a significantly inhibitory effect on the nociceptive responses of WDR neurons. RN neurons were excited by muscle spindle afferents. Muscle spindle afferents significantly inhibited C-response of WDR neurons and this inhibitory effect was reduced by lesions of RN. Conclusion: RN neurons have a significant antinociceptive effect and might be involved in the antinociceptive effects elicited by muscle spindle afferents.
4.Study on Quality Standard of Prostatitis Capsules

OBJECTIVE:To establish the quality standard for Prostatitis capsules. METHODS:TLC method was performed to qualitatively identify Pseudostellaria heterophylla,Acortw tatarinowii,Sargentodoxa cuneata,Rubia cordifolia and Vaccaria segeta-lis in the preparation. HPLC method was adopted to determine the content of leonurine in the preparation. The determination was performed on Agilent Eclipse XDB-C18 column with mobile phase consisted of methanol-0.025 mol/L potassium dihydrogen phos-phate(pH adjusted to 2.5)(24 : 76,V/V)with phosphoric acidat the flow rate of 1.0 mL/min. The detection wavelength was set at 277 nm,and column temperature was 30 ℃. The sample size was 10 μL. RESULTS:The TLC spots of P. heterophylla,A. tatari-nowii,S. cuneate,R. cordifolia,and V. segetalis were clear and well-separated without interference from negative control. The lin-ear range of leonurine were 4.05-81.00 μg/mL(r=0.9999). RSDs of precision,stability and reproducibility tests were all lower than 2.0%. The recovery was 98.47%-103.83%(RSD=2.04%,n=9). CONCLUSIONS:Established standard can be used for quali-ty control of Prostatitis capsules.
5.Determination of the Concentration of 5-Fluorouracil in Human Plasma With HPLC

OBJECTIVE:To establish a HPLC method for the determination of 5-fluorouracil in human plasma METHODS:The plasma samples were extracted with ethyl acetate-isopropanol(85∶15,V/V)following precipitation of plasma proteins with ammonium sulfate The solvent was evaporated to dryness under a stream of nitrogen The dry extract was dissolved in mobile phase and then injected into the Diamonsil C18 column(150mm?4 6mm,5?m)and 0 01mol/L KH2 PO4(pH5 5) was taken as mobile phase The flow rate was 1 5ml/min,UV detection wavelength was 267nm The internal standard was 5-bromouracil RESULTS:The minimal detectable drug concentration in plasma was 0 025?g/ml The calibration curve was linear over a range from 0 3?g/ml to 10?g/ml The intra-day RSD≤5 0%(n=5) and the inter-day RSD≤11 5%(n=5) CONCLUSION:The method can be used for studying the pharmacokinetics and bioavailability of 5-fluorouracil
6.Study on relationship between Hcy,hs-CRP,Cys-C and Fib with acute cerebral infarction

Objective To study the level changes and correlation of homocysteine (Hcy) ,high-sensitivity C-reactive protein(hs-CRP) ,cystatin C(Cys-C) and plasma fibrinogen(Fib) in the patients with acute cerebral infarction (ACI) .Methods The fasting blood samples were collected from 178 cases of ACI(ACI group) and 93 healthy individuals blood samples (control group) .The lev-els of serum Hcy ,hs-CRP and Cys-C were detected by the BECKMAN AU-680 fully automatic biochemical analyzer and plasma Fib was determined by the RAC-100 fully automatic coagulometer .Results The levels of serum Hcy ,hs-CRP ,Cys-C and plasma Fib in the ACI group were significantly increased compared with the control group (P<0 .05) .There was significantly positive correlation between Hcy with hs-CRP and Cys-C in the ACI group(r=0 .326 ,0 .361 ,P<0 .05) ,but there was no significant correlation be-tween Hcy and Fib ;there was significantly positive correlation between hs-CRP with Cys-C and Fib(r=0 .365 ,0 .421 ,P<0 .05);the same significant positive correlation also existed between Cys-C and Fib (r=0 .447 ,P<0 .05) .The positive rate of the joint de-tection of Hcy ,hs-CRP ,Cys-C and Fib was 93 .8 % ,which was obviously higher than that of the single indicator detection (P<0 .05) .Conclusion Cys-C ,hs-CRP ,Fib and Hcy participate in the occurrence and development process of ACI ,their joint detection has the important clinical significance for the prevention ,early diagnosis and treatment of ACI .
8.Analysis of P53 mutation and invasion front grading in oral squamous cell carcinomas.

We examined P53 mutation and invasion front grading (IFG) in 30 cases of oral squamous cell carcinomas (OSCCs). The association of P53 mutation and IFG scores with clinicopathological parameters was evaluated. P53 mutation existed in exon 5-8 in 15 out of the 30 OSCCs (50%). The incidence of P53 mutation was not associated with age, gender, N value and TNM stage. However, there was a significant correlation between P53 mutation and T value (P=0.046). There were no statistically significant correlations among the clinicopathological parameters and IFG. Interestingly, The IFG score in OSCCs with P53 mutation was significantly higher than that in OSCCs without P53 mutation (P<0.001). These results suggest that the high incidence of P53 mutation is a major mechanism of OSCC carcinogenesis. The presence of P53 mutation indicates the most anaplastic fields in the invasive areas of the tumors, which may predict poor prognosis for the patients.
9.Comparison of tensile properties of palmar or dorsal placement of stitches in repairing the tendons across the joints

Objective This study was designed to evaluate the biomechanical properties in tendons across the joints when Palmar or dorsal stitches were used. Methods Twenty porcine tendons were transected and then repaired in zone Ⅱand divided into two groups,using modified Kessler by placing the stitches in volar or dorsal part of tendons.The repaired tendons were placed on a joint model with flexion of 60?. An instron tensile testing machine was used to investigate 2 mm gap formation force and ultimate tensile strength. Results 2 mm gap formation force and ultimate tensile strength were both higher when dorsal placement of stitches was used. Conclusions Dorsal placement of stitches was recommended for repairing the tendons across the joints because it can offer greater tensile strength.
10.The Influence and Preventive Measures for theTest Results of Total Bile Acid about Blood Lipid Testing Reagent.

Objective To study The influence and preventive measures for the test results of total bile acid about blood Lipid testing reagent. Methods In Mindray BS -300 automatic biochemical analyzer, we detected total bile acid of the fresh mixed serum separately, then we tested the same samples after the test items of cholesterol, triglycerides, high -density lipoprotein, low - density lipoprotein. Then using fresh serum mixed with normal saline and blood lipid reagent according to 1: 1 dilution ,we measured TBA. We did cross - contamination corrected test (by the anti - cross - contamination to repeat the first step after the test) and recorded all results and had statistics analysis. Results This experiment found that reagents Ⅰ of TC, TG and the reagent Ⅱ of HDL - C , LDL - C had significant positive interference in determination of TBA results. After being corrected by anti - cross - contamination, the results returned to normal. Conclusion When using the automatic biochemical analyzer, we not only acquired the worker fououed of the equipment, and reagents, determined principle, but also pay attention to cross - contamination among reagents, and then in accordance with the equipment work characteristics to establish the appropriate measures to eliminate them, so made sure the reliability of the experimental data to provide clinic with the true objective and actual diagnosis evidence fundamentally.
