3.Virus inactivation using peracetic acid-ethanol solution impacts osteogenic activity of demineralized bone matrix

BACKGROUND:At present, an increasing number of bone graft materials are inactivated using peracetic acid-ethanol solution, but there is no report on whether virus inactivation using peracetic acid-ethanol solution has effects on osteogenic induction of demineralized bone matrix. OBJECTIVE: To explore the effects of virus inactivation using peracetic acid-ethanol solution on the osteogenic activity of demineralized bone matrix. METHODS: Long bones of Sprauge-Dawley rats were selected to make demineralized bone matrix meal. A part of bone meal was placed into gelatin capsules and sterilized by60Coγ irradiation as control group; another part of bone meal was placed into gelatin capsules folowing virus inactivation using peracetic acid-ethanol solution, and then sterilized using60Coγ as experimental group. After that,40 Sprauge-Dawley rats were enroled, and gelatin capsules in the experimental and control groups were respectively implanted into the bilateral psoas muscles of the lumbar spine. At 2, 4, 6, 8 weeks after implantation, the bone grafts were removed for gross and histological observations. RESULTS AND CONCLUSION:At 8 weeks after implantation, the bone mass in the experimental group was not intact in shape with scattered particles, and under light microscope, a few of osteoblasts and new vessels were seen; the bone mass in the control group had hard texture and complete morphology, and under light microscope, mature bone trabeculae wrapped with osteoblasts and new vessels were visible and there were ful of fat cels and bone marrow cells between the bone trabeculae. The number of new microvessels in the experimental group was lower than that in the control group at different time (P < 0.001); at 8 weeks after implantation, the calcium content, alkaline phosphatase content, inorganic phosphorus content and new bone growth rate were all lower in the experimental group than the control group (P < 0.001). These findings indicate that peracetic acid-ethanol for virus inactivation has some negative effects on the osteogenic induction of demineralized bone matrix.

5.Targeting oncolytic adenovirus therapy in pancreatic cancer

Carrying out targeting an oncolytic adenovirus strategy in gene therapy of pancreatic cancer is a new direction.Currently,related research including vitro test and animal models vivo test,targeted strategies on molecular biology and gene level are carried out.Some of oncolytic adenovirus drugs have entered phase Ⅲ clinical trials.Oncolytic adenoviruses alone or in combination with other treatments,can enhanced anti-tumor effect.
6.Progress in Study on Animal Models of Polycystic Ovary Syndrome and Intervention with Traditional Chinese Medicine

Polycystic ovary syndrome (PCOS) is a disease of dysgenesia and dysmetabolism. Its clinical manifestations include hyperandrogenism, ovulation failure, hyperinsulinemia, elevated serum LH level, obesity and sterility. Histological examination of ovary reveals polycystic changes. It is difficult to carry out clinical research because of its complicated pathogenesis, pathophysiology and multiple biochemical changes and clinical manifestations. It is necessary to study PCOS using animal models. In this article, the authors introduce some methods of establishing animal models of PCOS, such as androgen administration (including testosterone propionate,dehydroepiandrosterone), combination of insulin with HCG, use of estradiol valerate,combination of progestrone with HCG, aromatic cyclase inhibitor method,and so on, and review the effects of traditional Chinese medicine on the intervention of PCOS.
7.Alteration and clinical significance of NK cell and T lymphocyte subpopulation in patients with rectal carcinoma

Objective To investigate the alteration and clinical significance of NK cell and T lymphocyte subpopulatien in patients with rectal carcinoma. Methods The numbers of NK cell and T lymphocyte in 26 healthy persons and 35 rectal carcinoma patients were detected by 3H-releasing method and FCM. Results NK cell and CD3, CD4, CD4/CD8 were decreased significantly in patients with rectal carcinoma (P<0.05), which was positively correlated with Dukes staging (P<0.01), and was inereased after redical or palliative operation (P<0.05), but NK cell and CD3, CD4, CD4/CD8 were further decreased in patients with unresectable tumor after operation (P<0.05). Conclusions The immunity of rectal patients was decreased with the development of the disease, and it will be improved after radical of palliative operation.
8.Therapeutic effect of mycophenolate mofetil on autoimmune encephalomyelitis in Wistar rats

Objective To investigate the therapeutic effect of mycophenolate mofetil (MMF) on rats with experimental autoimmune encephalomyelitis (EAE).Methods The guinea pig spinal cord homogenate (GPSCH) and complete Freund's adjuvant were used as the antigen to induce the acute EAE model in Wistar rats which were verified according to clinical scoring.Totally 36 rats were randomly and equally divided into 3 groups,that is,normal control,EAE group and MMF treatment group.In 10 d after immunization,normal saline or MMF (30 mg?kg-1?d-1) were given intragastrically for 7 d to the corresponding groups.The rats were sacrificed on the 16th day and 30th day after immunization for pathological changes of the brain and spinal cord.Clinical manifestation was scored during the experiment.Results Compared with the EAE model group,MMF-treated rats had lower clinical scores and shorter disease process.Besides,the inflammatory infiltration,demyelination and axonal damage in the central nervous system were significantly improved in MMF treatment group.Conclusion MMF is an effective drug in treating EAE,and can relieve the clinical signs and ameliorate pathological lesions in the central nervous system of acute EAE rats.

Objective To investigate the distrbution of actin and the level of Ca 2+ in macrophages activated with LPS and IFN\|? and study effects of reorganization of actin and the level of Ca 2+ on migration and phagocytosis of macrophages. Methods Rat macrophages were activated with LPS and INF\|?,actin and intracellular free Ca 2+ were labeled with phallacidin and Fura\|2,the distribution of actin and the level of Ca 2+ were observed and analyzed with Ca 2+ image analysis system and confocal laser scanning microscope. Results The migrating cell and the phagocytizing cells increased in the activated macrophages.F\|actin and the level of Ca 2+ of the activated macrophages increased.There was a front and a back in the migrating cell.Lamellipodia and filopodia protruded from the front.F\|actin of the lamellipodium and filopodium was rich.There were stress fibers of F\|actin at the bottom of the migrating cells.In the migrating macrophages,the level fo Ca 2+ in the back was high than that in the front.When the migrating cell turned their growth direction,the level of Ca 2+ at the curve increased greatly,especially in its lateral region.When macrophage phagocytized the dead lymphocyte,Ca 2+ in the phagocytizing region increased. Conclusion When macrophages are activated with LPS and IFN\|?,migration and phagocytosis increase greatly.Particular morphologic changes in the distribution of F\|actin and the level of intracellular free Ca 2+ occur. 10.EFFECTS OF EXTRACELLULAR MATRIX ON LYMPHANGIOGENESIS in vitro

Objective In order to investigate effects of extracellular matrix on lymphangiogenesis. Methods Endothelial cells of canine thoracic duct were incubated in type I collagen gel and on Matrigel basement membrane matrix gel.Morphologic changes of endothelial cells and tube formation were observed under phase contrast microscope,structures of lymphaticlike channels were observed with transmission electron microscope,tube formation was quantified by measuring the length and area of tubes with image analyzer. Results Lymphatic endothelial cells on type Ⅰ collagen gel grew to confluent monolayers,no tubes were found.The cells underwent morphologic changes and reorganized into lymphaticlike tubes when the cells were covered with the top gel layer.Tubes of heparin\|treated dishes were more than that of the control dishes,tubes of dishes treated with heparin and bFGF together were more than that of dishes treated with heparin or bFGF alone.Endothelial cells on Matrigel basement membrane matrix gel migrated into the upper part of the gel and formed tubes.The tubes were thin,the cells became long.The tubes were in traction obviously.In ultrathin sections,tubes formed by lymphatic endothelial cells represented morphologic characteristics of lymphatic capillary.The lymphaticlike tubes contained matrix.Conclusion\ Endothelial cells of canine thoracic duct reorganized into lymphaticlike channels in type Ⅰ collagen gel and Matrigel basement membrane matrix gel.Extracellular matrix plays important roles in morphological changes and migration of lymphatic endothelial clells and lymphangiogenesis.\;[
