1.Expression and role of miR-34a in bladder cancer

Objective To investigate the expression and biological function of microRNA-34a (miR-34a) in bladder cancer.Methods Forty-two cases of bladder cancer specimen,including 17 noninvasive carcinoma and 25 muscle invasive carcinoma classified by UICC 2002 TNM,or 18 low-grade and 24 high-grade classified by WHO 1973.Meanwhile,mucosa adjacent to the carcinoma was selected as normal control.The gene expression of miR-34a was determined using real-time quantitative polymerase chain reaction in 42 cases of bladder carcinoma samples.Mature mimics of miR-34a were chemically synthesized and transiently transfected-intoT24 bladder cancer cells.The effects of miR-34a on apoptosis and proliferation in T24 cells were evaluatedby flow cytometry and MTS respectively.Results 61.9% of carcinoma samples showed low expression of miR-34a,which was correlated with the malignancy and tumor size of bladder carcinoma.26.5% (11 cases) was negative and 11.6% (5 cases) showed high expression.Furthermore,upregulation of miR-34a in T24 cells contributed to cell growth and apoptosis.The apoptosis rate of T24 cells was (9.11 ± 0.41)%,which had significant difference compared with NC mimics and blank control group respectively (P < 0.01).Conclusion The relative low expression of miRNA-34a may be involved in the tumorigenesis and development of bladder carcinoma.
2.Effects of aspirin and verapamil alone or in combination on mesenteric microcirculation of rats

AimTo study the effects of aspirin (Asp) and verapamil(Ver)alone or in combination on mesenteric microcirculation of rats. MethodsAcute microcirculation disturbance(AMD) was produced by highmolecular weight dextran(Mr 480, 000) 360 mg· kg-1 iv. Arteriol and venul blood flow velocity and diameter (ABFV, VBFV, AD, VD) and blood flow state(BFS) were observed by intravital microcirculation method. Results Asp 2.5, 5 mg · kg-i, Ver 0.3, 0.6 mg · kg-1,Asp+ Ver(1 + 0.15), (2. 5+ 0. 3) mg· kg-1 iv significant increase ABFV by 11.1%, 31.3%, 18.7%,19.5%, 26.5%, 37.3%, VBFV by 12.5%, 5.7%, 2.5%, 3.7%, 30%, 34.7%,AD by 4.3%, 17.9%, 31.5%, 35%, 20%, 38.1% and VDby 2.2%,4.2%, 25%, 31.5%,5.8%,23.5%espectively, and got a raise in the number of capillaries and a marked improvement of BFS. Asp + Ver(1+0.15, 2.5+0.3) mg · kg-1 iv could reverse AMD. Conclusion Asp is superior to Ver in the increase of BFV, but Ver is superior to Asp in expansion of blood vessel. Asp in combination with Ver produces marked synergistic action and protection againist AMD.
3.Engraftment of bone marrow-derived cells after nonlethal radiation in syngeneic C57BL/6 mice

Objective To study the characteristics of cell engraftment in mice at a lower dose under nonlethal radiated condition.Methods A syngeneic C57BL/6 mouse model,transplanted with 1 × 107 bone marrow cells and exposed to 2.5 Gy whole body irradiation (WBI),was selected to study the chimerism of cells from green fluorescent protein positive (GFP +) transgenic mice.The control group was injected with GFP + cells without receiving irradiation.In addition,an allogenic transplantation model of BALB/c mice was also investigated which was infused by GFP + cells from C57BL/6 mice.The engraftment of bone marrow-derived cells (BMDCs) was detected by immunohistochemistry in bone marrow,liver,lung,small intestine and spleen.Results The transplanted bone marrow cells successfully grafted in the haematopoietic tissues from syngeneic GFP transgenic mice.The transplanted GFP+ cells were also detected in the non-haematopoietic tissues,such as the small intestine,liver,spleen and lung,after irradiation.However,a lethal dose irradiation of 8 Gy was required to establish successful chimerism in allogeneic transplantation model by infusing the bone marrow cells from C57BL/6 mice to BALB/c mice.Conclusions Bone marrow-derived cells can be successfully grafted into various recipient tissues receiving a 2.5 Gy dose of radiation in syngeneic mice,but not in allogeneic mice.This nonlethal model may help to further study the plasticity and mechanism of bone marrow-derived cells in tissue repair and regeneration after radiation injury.

5.A STUDY OF ENERGY EXPENDITURE OF MEN WORKED IN THE HOT DESERT ENVIRONMENT

Energy expenditure of five young adults worked in the hot desert environment were measured by the methods of indirect calorimetry in the Wuwei Gansou in July 1989.The results indicated that the basal metabolism rate, the resting metabolism rate and the energy cost of young adults marching in the desert and Gobi (15kg load, at rate of 3.5km/h) were 2.782,3.059,14. 188, 10.305 kJ? m-2?min-1 respectively. It was shown that under the similar conditions energy expenditure marching in the desert was 38% more than in the Gobi.Ambient temperature had a greater effect on energy expenditure but the effect was various at different activities. When at 24.5-38.4℃ during marchi-ng(15kg load),1℃increase in ambient temperature was associated with anincrease of about 0.86% energy consumption. While at 25-33℃ during quiet rest the energy consumption tended to decrease with an increase of ambient temperature. But at 33-40℃ it increased with ambient temperature again. It is suggested that the energy metabolism may be more easily influenced by ambient temperature when the metabolic heat production of human body is increased.The present study recommended the estimation of energy requirement for the adult with a moderate activity in hot desert environment should be approximate 15.062MJ/d
6.Relationship between ACE gene (I/D) polymorphism and familial essential hypertension

Objective:To explore the relationship between angiotensin converting enzyme (ACE) gene insertion/dele‐tion (I/D) polymorphism and familial essential hypertension (EH) .Methods :Fluorescent in situ hybridization stai‐ning DNA sequencing test was used to detect ACE gene I/D genotype frequency and allele frequency in 46 familial EH patients (familial EH group) ,64 EH patients without family history (non‐familial EH group) and 43 healthy people (healthy control group) .Results:In healthy control group ,non -familial EH group ,familial EH group ACE gene DD genotype frequency was 11.6% ,32.3% and 37.0% ;distribution frequency of D allele was 33.7% , 52.3% and 57.6% respectively .Compared with healthy control group ,there were significant rise in ACE gene DD genotype and D allele frequency in familial EH group and non -familial EH group (P<0.05 or <0.01) ,but there were no significant difference between familial EH group and non‐familial EH group (P>0.05) .Conclusion:ACE gene DD genotype and D allele may be genetic predisposing genes of patients with essential hypertension ,and there is no significant difference in genetic constituent ratio between familial EH patients and non‐familial EH patients .
7.Effect of blood pool 18F-FDG activity on liver SUV: a calibration methodology study

Objective To identify the effect of blood pool 18F-FDG activity on liver SUV and to investigate the optimal normalization method.Methods PET/CT and common serological examination items from 1 018 subjects were retrospectively collected.Mean SUV of liver and blood were recorded as SUVmean(L) and SUVmean (B),respectively.The difference and quotient of SUVmean(L) and SUV mean (B) were calculated as SUVmean (L-B) and SUVmean (L/B),respectively.CV of SUVmean (L),SUVmean (L-B) and SUVmean(L/B) were calculated to assess their inter-individual variations.Pearson correlation analysis was used to evaluate the relationship of SUVmean(L),SUVmean(L-B),SUVmean(L/B) with SUVmean(B).Multiple linear stepwise regression was performed to identify their vulnerability to common serological examination items.Results CV of SUVmean(L/B) (15.1%) was less than that of SUmean(L) (23.2%) and SUVmean(L-B) (40.6%).Correlation between SUVmean(L) and SUVmean(B) (r =0.820,P<0.001) was more significant than that between SUVmean(L-B) and SUVmean(B) (r =0.205,P<0.001) as well as between SUVmean (L/B) and SUVmean (B) (r=-0.376,P<0.001).Blood glucose and BMI correlated with SUVmean(L) and SUVmean(L/B),but not with SUV (B).Age and HDL correlated with SUVmean(L) and SUVmean(B),but not with SUV (L/B).Fatty liver was significantly associated with SUV mean (L/B) (β =-0.047,P <0.001),but not with SUVmean(L) and SUVmean (B).Conclusions 18 F-FDG activity of blood pool affects liver SUV.SUV mean (L/B) is a simple and reliable normalization method since its inter-individual variation and vulnerability to common serological examination items are relatively lower than liver SUV.

Objective To investigate the relationship between OHSS and FSHR gene polymorphism. Methods Two hundred and two women were enrolled in this study. The FSHR gene polymorphisms at position 307 and 680 were detected. Results The distribution of the allele frequency and genotype frequency of the position 680 in FSHR gene were significantly different between women with OHSS or not. No significant differences of the position 307 in FSHR gene were observed. Conclusion Themutation of Asn680Ser in FSHR might be closely related with OHSS.
