1.Meta Analysis of ADAM33 T1,S2 Polymorphism and the Susceptibility of Bronchial Asthma in China

Objective To investigate the correlation between ADAM33 T1, S2 gene polymorphism and Bronchial asthma risk in china. Methods We retrived the relevant published studies about ADAM33 T1, S2 gene polymorphism and bronchial asthma risk. Then we divided the population into Chinese and other Asian population. Odds ratio (OR) of Case group and control group was selected as the effect index. Stata 11.0 software was used to calculate heterogeneity test, ORs and 95%CI of two areas, and gave the forest plot and funnel plot of meta results. Results A total of 27 studies were included in this analysis,18 studies in ADAM33 T1 site were 3881 cases in case group, and 3780 cases in control group;and 14 studies in ADAM33 S2 site were 3222 cases in case group, and 3513 cases in control group. Additive model, dominant model, recessive model of ADAM33 T1 in Chinese had association with the susceptibility of bronchial asthma. The results were OR=1.488, 95% CI:1.002-2.167 in Additive model, OR=1.619, 95%CI:1.059-2.475 in dominant model;OR=2.523, 95%CI:1.910-3.333 in recessive model. Three models of ADAM33 T1 in other Asian country had no association with the susceptibility of Bronchial Asthma. Three gene model of ADAM33 S2 in Asian had no association with bronchial asthma susceptibility. Except ADAM33 T1 polymorphism in recessive model, other mode of T1, S2 had no publication bias in Chinese population. Conclusion There are association between ADAM33 T1 gene polymorphism and bronchial asthma, but ADAM33 S2 gene polymorphism and bronchial asthma have no association in Chinese population.
2.Changes of synaptic structure after long-lasting LTP induced by high and low frequency tetanus in slices of the rat visual cortex.

Synaptic ultrastructural changes after long-lasting long-term potentiation (L-LTP) induced by 2 and 100 Hz tetanus were investigated by electron microscopic and stereological approach in slices of the developing rat visual cortex (postnatal days 15~21). Both 2 and 100 Hz tetanus-induced L-LTP groups showed significant increases in synaptic interface curvature, synaptic numeric density and postsynaptic density thickness, as well as significant decreases in the cleft width, as compared with the control groups. In addition, the volume density of the active zone (AZ) was increased significantly in the 100 Hz tetanus-induced L-LTP group, but not in the 2 Hz group. The mean lateral area of individual AZ in the 100 Hz group was relatively higher than that in the 2 Hz group. These data suggest that newly formed synapses in the 100 Hz tetanus-induced L-LTP group are larger than those in the 2 Hz group and that 100 Hz tetanus might trigger reorganization or synthesis of postsynaptic cytoskeleton.

3.Different effects of 2 and 100 Hz tetanus on the expression of long-lasting long-term potentiation in rat visual cortical slices.

Long-term potentiation (LTP) can be induced by various tetanic parameters in the mammalian visual cortex. However, little researches have been done on the relationship between the expression of the long-lasting LTP (late phase LTP or L-LTP) lasting more than 3 h and the tetanic parameters. In the present study, the effects of 2 Hz and 100 Hz tetanic parameters on L-LTP of the field potentials were recorded from the layer II/III of the rat visual cortical slices in response to stimulation of the layer IV. As a result, tetanic parameters that had more than 300 pulses reliably induced L-LTP in the postnatal day 15-21 rats. Obviously different L-LTP expressions were induced by 2 Hz and 100 Hz tetani. There was no difference in L-LTP expression induced by the parameters with the same frequency and different total pulses. These data suggest that L-LTPs induced by different frequency parameters may have different induction and maintenance mechanisms; L-LTPs induced by the parameters with the same frequency may have the same mechanisms.

5.Clinical study of solitary bone plasmacytoma.

OBJECTIVETo study the clinical characteristics and the prognosis of solitary bone plasmacytoma (SBP).
METHODSFrom June 1997 to June 2002, ten SBP patients were treated in our department. Two lesions were in sacrum, two in pelvis, the other six lesions were in proximal femur, femoral diaphyseal, scapula, thoracic vertebra, proximal humus, and rib respectively. The preoperative laboratory tests were normal. Operation was performed on all patients. The pathologic diagnose was plasmacytoma and then they underwent further laboratory examination, urine Bence-Jone protein is positive in 3 patients, serous IgG value was higher than normal in 2 patients, abnormal M protein was found in electrophoresis in 2 patients. All patients received radiotherapy postoperatively.
RESULTSFour patients were excluded, whose follow-up were lessen than ten months, the other six patients's average follow-up is 28.2 months (from 18 to 48 months), one patient who developed multiple myeloma (MM) six months postoperatively received chemotherapy using M2 protocol and died 21 months after operation. The other five patients had disease-free survive and remain solitary bone lesion after the treatment of surgery and radiology.
CONCLUSIONCompared with MM, SBP patients are younger, the therapeutic results and prognosis are better. The main prognostic factors include age, the size of lesion, the axial bone lesion, persistence of myeloma protein after radiotherapy, early diagnosis and treatment, and so on.

6.Protective effect of trimetazidine on rabbit myocardium during ischemia- reperfusion.

OBJECTIVETo investigate the protective effects of trimetazidine on rabbit myocardium in ischemia and reperfusion.
METHODSFourty rabbits were divided into five groups randomly: normal control group, ischemia control group, ischemia and drug intervention group, reperfusion control group, reperfusion and drug intervention group. Ischemia lasted for 30 minutes and reperfusion was given for 30 minutes. Serum CPK, SOD activities and MDA concentrations were measured in each group and ischemia tissue ATP concentrations were also measured. Heart tissue was examined with electron microscope in each group.
RESULTS(1) Serum concentrations of MDA in ischemia and drug intervention group were significantly different from those in ischemia control group [(4.09+/-0.40 vs 4.79+/-0.92)nmol/ml, P<0.01], serum activities of CPK [(1322+/-1148 vs 1498+/-190) NU/ml], SOD[(324+/-71 vs 288+/-54)NU/ml] were not significantly different between ischemia and drug intervene group and ischemic control group (PP>0.05,respectively). (2) Serum activities of CPK [(1512+/-226 vs 1904+/-203) NU/ml], SOD[(213+/-71 vs 119+/-55) NU/ml], concentrations of MDA [(6.09+/-0.69 vs 7.43+/-0.20)nmol/ml] and concentrations of ATP[(1.401+/-0.248 vs 0.629+/-0.175) micromol/g] in ischemia heart tissue of reperfusion and drug intervention group were significantly different from those in reperfusion control group (P<0.001 - 0.01 respectively). (3) There were significant differences in electron microscopic observation between intervention group and control group.
CONCLUSIONTrimetazidine can improve cardiac mitochondrial metabolism and scavenge oxygen free radicals. Trimetazidine has cardioprotective function during ischemia and reperfusion.

7.Surgical classification and therapeutic strategy for metastases of proximal femur.

OBJECTIVETo establish a surgical classification system for metastases of proximal femur and discuss the therapeutic strategy with retrospective analysis and literature review.
METHODSThe data of 99 patients who underwent a total of 102 operations for femoral metastatic lesions from January 2003 to December 2011 was analyzed. There were 50 males and 49 females, and the median age was 56 years (range 15-87 years). The most common diagnosis was lung cancer (30 cases), followed by breast cancer (17 cases). All femoral lesions were divided into 4 types (I-IV) with different anatomic site and biomechanic characteristic. The patients with various surgical reconstruction mode and postoperative follow-up data were recorded.
RESULTThere were 65 side who received widely or marginal resection and 37 side who received intralesional resection. The patients were operated with bipolar hip prosthesis (n = 3), ordinary total hip replacement (THR) (n = 10), bipolar tumor prosthesis (n = 48), THR with tumor prosthesis (n = 8), intramedullary nailing (n = 21), and plate/screw (n = 12). The estimated survival for the 99 patients was 10.3 months. Type I, II, III and IV patients with postoperative American Society of bone and soft tissue tumors-93 rating were 86.5%, 77.3%, 81.3% and 69.1%. Patients with type IV were worse compared with the other 3 groups (t = 4.763, P = 0.031). The 10 operations were followed by complications of any kind. Complication rate of patients with type IV were 3/12, and it was significantly higher than the other 3 groups of patients (χ(2) = 4.018, P = 0.045).
CONCLUSIONSThe classifications and corresponsive surgical methods for upper femur metastases had some superiority in hinting prognosis and guiding treatment.

8.The surgical management of sacral chordoma.

OBJECTIVETo analyze the surgical treatment results and experience of sacral chordoma.
METHODSThe data of 51 cases of sacral chordoma surgically treated from July 1997 to July 2007 was retrospectively studied. The age of patients ranged from 21 to 75 years (mean 57 years), including 32 males and 19 females. Forty patients had the first surgery, while other 11 patients were referred to our hospital because of local recurrence from other hospital. Wide resection for all 17 S(3-5) tumors, and wide resection plus piece-meal excision for 34 tumors which involved S(3) above. Reconstruction was performed using pedicle screw and rod device to achieve the continuity between the lumbar spine and the pelvis. The oncologic and functional outcomes of 51 patients were reviewed.
RESULTSAfter a mean duration of follow-up of 3.5 years (range from 15 to 108 months), 5 of 51 patients died of disease. Seventeen of 40 (42.5%) patients who underwent first surgery recurred during follow-up, 18 of 28 patients (64.3%) got re-relapse after second or third surgeries. Postoperative wound complications were as high as 31.6%.
CONCLUSIONSChordoma is a biologically aggressive low-grade malignant tumor. Wide resection is a prerequisite for curative treatment of sacrococcygeal chordoma, intralesional curettage causes the risk of high local recurrence for which curative resection in a second or third procedure is more difficult to achieve. In order to preserve near normal bowel and bladder function for the tumor which involves S(3) above, wide en-bloc resection plus piece-meal excision in the cephalad is performed.

9.Morphological alteration of the hippocampal synapses in rats prenatally exposed to magnetic resonance imaging magnetic fields.

The present study investigated the effects of prenatal exposure to the magnetic resonance imaging (MRI) magnetic fields on the synaptic ultrastructure of hippocampal formation of rats at different postnatal development stages. Pregnant rats with gestation of 12-18 days were exposed to the magnetic fields used for MRI clinical applications. When the offspring were 1, 2, or 5-month-old, the synaptic morphologic parameters were measured in female offspring. In the 2-month-old MRI group, the curvature of synaptic interface, the length of active zone and the surface density per unit volume (S(v)) of active zone in the dentate gyrus (DG) decreased significantly, and the width of synaptic cleft increased in the CA1 area. In the 5-month-old MRI group, the width of synaptic cleft increased, the thickness of postsynaptic density and the curvature of synaptic interface decreased significantly in the CA1 region, and the width of synaptic cleft increased in the DG. No significant change was observed in the 1-month-old group. These results suggest that prenatal exposure to the medical magnetic fields causes synaptic ultrastructural changes. The relationship of these changes with behavioral impairments was discussed.
