Objective To study the effect of Bushen Tiaojing Chinese herbs on the expressions of endometrial integrin ?3 in infertile patients with polycystic ovary syndrome (PCOS) receiving ovulation induction therapies. Methods Thirty-six PCOS related infertility patients were divided into 2 groups at random, receiving different ovulation induction therapies:17 patients of group A were given clomephine citrate (CC)/human chorionic gonadotropin (hCG) therapies, and 19 patients of group B were given CC/ hCG combined with Chinese herbs therapies. 22 infertile women with fallopian tubal or male infactors were chosen as controls. The mRNA expressions of integrin ?3 in midluteal endometrium were measured by reverse transcription-polymerase chain reaction. Results Compared with controls, the expressions of endometrial integrin ?3 were weaker in group A (P0.05). Conclusion CC/hCG ovulation induction therapies may induce the decline of endometrium receptivity during midluteal phase, which Chinese herbs may promote.

Objective To analyze the characteristics of subjective visual horizontal ( SVH) and evaluate its clinical value for vestibular function in peripheral unilateral vestibular hypofunction ( UVH). Methods Eighty-five patients with UVH (study group) and 39 normal persons (control group) accepted vestibular function tests, including SVH, subjective visual vertical ( SVV ) and caloric test by video-nystagmography. The parameters of the angle of SVH and SVV, directional preponderance ( DP) and unilateral weakness(UW) of caloric test were observed. The correlation between SVH/SVV, DP, UW and the course of disease were investigated respectively. SPSS 16. 0 software was used to analyze the data. Results Reference range of SVH and SVV was from - 2° to 2° in the control group. Among the 85 patients, 46 cases(54. 1% ) and 43 cases(50. 6% ) had the abnormal values of SVH and SVV respectively, with no statistical significance( χ2 = 12. 5, P = 0. 481 ) by chi square test Fifty-five cases (64. 7% ) withabnormal DP had no statistical significance when compared with SVH and SVV respectively (χ2 values were 0. 19 and 2.86, respectively, P value were 0. 164, 0.067, respectively). In UVH, there were positive correlation between SVH, SVV and DP( r value was 0. 939, 0. 648, 0. 658, all P <0. 05) respectively, but no correlation between UW and SV H or SVV (r value was 0. 048, 0. 085, all P > 0. 05). According to the permutation and combination of the four parameters, positive or negative, three main groups could be defined [SVH(+)DP(+)UW( +), SVH( -)DP( +)UW( +), SVH( -)DP( -)UW( +); SVV( + ) DP ( +)UW( +), SVV( -)DP( +)UW( +), SVV( -)DP( -)UW( +)]. The course of disease in the three main groups was positively skewed distribution, with median of 5. 0, 10. 0, 15. 0 d and 5. 0, 9. 5, 14. 5 d respectively. By Kruskal-Wallis Test, χ2 value were 8. 80 and 6. 26, respectively( P value were 0.012, 0. 040, respectively), with statistical significance between the above three main groups. Conclusions The SVH value can evaluate the function of the otolithic. The angle of SVH and SVV are changing in the course of disease, SVH and SVV can be used as a guidance of the vestibular compensation evaluation.

Objective:To compare the prognostic value of two predictive models based on C-reactive protein(CRP)and albumin(ALB),namely the CRP to ALB ratio(CAR)and the Glasgow prognostic score(GPS),in newly diagnosed patients with diffuse large B-cell lymphoma(DLBCL).Methods:The data of newly diagnosed DLBCL patients admitted to our center from May 2014 to January 2022 were reviewed.A total of 111 patients who completed at least 4 cycles of R-CHOP or R-CHOP-like chemotherapy with detailed clinical, laboratory data and follow-up information were included. The receiver operating characteristic(ROC)curve was performed to evaluate the predictive value of pre-treatment CAR on disease progression and survival.Furthermore, the association between CAR and baseline clinical, laboratory characteristics of patients was evaluated, and progression-free survival(PFS)and overall survival(OS)were compared between different CAR and GPS subgroups. Finally, the univariate and multivariate COX propor-tional hazard regression models were used to analyze the factors affecting disease outcomes. Results:ROC curve showed that the area under the curve(AUC)of CAR predicting PFS and OS in DLBCL patients was 0.687(P=0.002)and 0.695(P=0.005),respectively,with the optimal cut-off value of 0.11 for both predicting PFS and OS.Compared with the lower CAR(<0.11)group,the higher CAR(≥0.11)group had more clinical risk factors, including age>60 years(P=0.025),ECOG score ≥2(P=0.004),Lugano stage Ⅲ-Ⅳ(P<0.001),non-germinal center B-cell-like(non-GCB)subtype(P=0.035),elevated lactate dehydrogenase(LDH)(P<0.001),extranodal involved site>1(P=0.004)and IPI score>2(P<0.001).The interim response evaluation of patients showed that the overall response rate(ORR)and complete response rate(CRR)in the lower CAR group were both significantly better than those in the higher CAR group(ORR:96.9%vs 80.0%,P=0.035;CRR:63.6%vs 32.5%,P=0.008).With a median follow-up of 24 months, patients with lower CAR had significantly longer median PFS and OS than those with higher CAR(median PFS:not reached vs 67 months,P=0.0026;median OS:not reached vs 67 months,P=0.002),while there was no statistical difference in PFS(P=0.11)and OS(P=0.11)in patients with GPS of 0,1,and 2.Multivariate Cox regression analysis indicated that only sex(male)and IPI score>2 were independent risk factors for both PFS and OS.Conclusion:CAR is significantly correlated with disease progression and survival in DLBCL patients; And compared with GPS,CAR has more advantages in predicting disease outcomes in DLBCL patients.

Objective:To compare the efficacy and safety of flumatinib (FM)and imatinib (IM)as first-line treatment in newly-diagnosed patients with chronic myeloid leukemia in chronic phase (CML-CP ) in real world. Methods:A total of 84 newly-diagnosed CP-CML patients in our center from December 2019 to December 2022 were retrospectively analyzed.Among them,32 cases received FM as first-line treatment, and 52 cases received IM. Molecular response (MR),disease progression, survival and incidence of adverse events (AEs)were compared between the two groups. Results:At 3 months of treatment, the incidences of early molecular response (EMR ),MR2.0 and MR3.0 were 96.7%,70.0% and 20.0% in FM group, respectively, which were significantly higher than 77.1%,29. 2% and 0 in IM group (all P<0.05 ).At 6,9 and 12 months of treatment, the incidences of major molecular response (MMR)in FM group were 68.2%,85.7% and 90.0%,respectively,which were significantly higher than 22.9%,34.0% and 51.1% in IM group (all P<0.01).The median time to achieve MMR in FM group was 6(6-9)months,which was significantly shorter than 18(12-22)months in IM group (P<0.001 ).The 3-year progression-free survival rate and 3-year event-free survival rate in FM group were 100% and 68.8%,respectively,while in IM group were 98.1% and 55.8%.There were no significant differences between the two groups (P>0.05). The incidence of grade 3-4 hematologic AEs in FM group was 21 .9%,which was slightly lower than 25.0% in IM group, but the difference was not significant (P>0.05 ).Conclusion:In real clinical practice,FM as first-line treatment achieves MMR earlier than IM,and exhibits good safety profile in newly-diagnosed CML-CP patients, which potentially leads to improved long-term survival and treatment-free remission.

Prohibitin (PHB), an evolutionarily conserved mitochondrial inner membrane protein, is highly expressed in cells that require strong mitochondrial function. Recently, we demonstrated that the deletion of Phb in spermatocytes results in impaired mitochondrial function. In addition, PHB expression in the mitochondrial sheath of human sperm has a significantly negative correlation with mitochondrial reactive oxygen species levels, but a positive one with mitochondrial membrane potential and sperm motility. These results suggest that mitochondrial PHB expression plays a role in sperm motility. However, the mechanism of PHB-mediated regulation of sperm motility remains unknown. Here, we demonstrate for the first time that PHB interacts with protein kinase B (AKT) and exists in a complex with phospho-PHB (pT258) and phospho-AKT in the mitochondrial sheath of murine sperm, as determined using colocalization and coimmunoprecipitation assays. After blocking AKT activity using wortmannin (a phosphatidylinositol 3-kinase [PI3K] inhibitor), murine sperm have significantly ( P < 0.05) decreased levels of phospho-PHB (pT258) and the total and progressive motility. Furthermore, significantly ( P < 0.05) lower levels of phospho-PI3K P85 subunit α+γ (pY199 and pY467) and phospho-AKT (pS473; pT308) are found in sperm from infertile asthenospermic and oligoasthenospermic men compared with normospermic subjects, which suggest a reduced activity of the PI3K/AKT pathway in these infertile subjects. Importantly, these sperm from infertile subjects also have a significantly ( P < 0.05) lower level of phospho-PHB (pT258). Collectively, our findings suggest that the interaction of PHB with AKT in the mitochondrial sheath is critical for sperm motility, where PHB phosphorylation (pT258) level and PI3K/AKT activity are key regulatory factors.
