4.Stress Response of Multiple-Antibiotic-Resistance (Mar) Mutants of Escherichia coli K-12.

The survival or growth rate of multiple-antibiotic-resistance (Mar) mutants of Escherichia coli K-12 MC4100 or W3110, influenced by many stresses (4% ethanol, 3mM HzO, 0.5M NaC1, pH 2, pH 5, and heat-shock (42C)), and the effects of salicylate (SAL) induction were evaluated by growth rate. Under the stress conditions of pH 5 and heat-shock, growth rates of Mar mutants derived from MC4100 or W3110 induced by salicylate (SAL) were higher than those of wild type Escherichia coli K-12 and E. coli clinical isolates. Under the SAL induction, growth rates of wild type MC4100 or W3110 were higher than those of Mar mutants in case of 0.5M NaC1 or pH 2 stress, respectively. Without SAL induction, growth rates of wild type E. coli were higher than those of Mar mutants, in case of MC4100 grown with 4% ethanol, HO, or heat-shock stresses, in case of W3110 grown with pH 2, or heat-shock stress. There were sorne instances of cytotoxic effect of SAL to the Mar mutants, wild type E. coli, or E. coli clinical isolates under some stress conditions.

5.Antibiotic Resistance of Multiple-Antibiotic-Resistance (Mar) Mutants of Escherichia coli K-12.

Multiple-antibiotic-resistance (Mar) mutants were prepared from the wild-type Escherichia coli K-12 MC4100 or W3110, were analyzed for the antibiotic resistance by minimal inhibitory concentration (MIC), and compared with their isogenic parent strains and E. coli clinical isolates. Mutation frequency was approximately 10-7 to 10-9 MIC levels of chloramphenicol (Cm), ampicillin (Ap), tetracycline (Tc), or ciprofloxacin (Cp) antibiotics against E. coli Mar mutants were 50 ug/ml, 300 ug/ml, 200 ug/ml, or 30 ug/ml, respectively, at their highest MIC levels. Cross-resistances of each Mar mutants against Cm, Ap, Tc, Cp, erythromycin (Em), nalidixic acid (Na), and rifampin (Rf) were evaluated. Almost all of Mar mutants showed approximately 9 to 140-fold increase of MIC with contrast to the MIC of isogenic parent strains and E. coli clinical isolates, except Rf antibiotics. The induction multiplicity of Mar mutants by salicylate (SAL) was approximately the same, 2 to 10-fold, and 2 to 25-fold increase of MIC, in case of E. coli clinical isolates, Mar mutants derived from MC4100, and W3110, respectively.

6.An Immunohistochemical Study for the neu and ras Oncoprotein and Epidermal Growth Factor Receptor in the Breast Carcinoma.

To evaluate correlation between the expression of neu and ras oncoprotein and epidermal growth factor receptor on breast carcinoma and other known prognostic factors, immunohisto-chemical studies were performed. Positive reaction for neu, which appeared as brown granular deposits along cell surface and cytoplasm of the tumor cells, was significantly correlated with the histological grade but not with other prognostic factors such as tumor size, lymph node me-tastasis, local recurrence, and estrogen and progesteron receptor status. Also granular deposits of ras were noted in the carcinoma cells in the cytoplasm, while the epithelaial cells of the normal lobule and duct showed negative reation. But expression of ras was not significantly associated ras with other prognostic factors. The reaction for EGFR was mostly negative on epithelial cells of both the normal lobule and duct, and was not significantly associated with other prognostic factors. The results suggested that expression of the neu oncoprotein is significantly associated with the histological grade of breast carcinoma, while the ras and the EGFR do not show significant prognostic value.

Case 1: A 10 year old female had a "coupe de sabre" type of linear scleroderma on the right side of the frontal scalp and face and hemiatrophy of the right side of the face. Case 2: A 10 year old female had a type of scleroderma en bande on the palm and dorsum of the left hand and clubbed finger of the left third finger. Case 3: A 21 year old male had a linear type of skin atrophy and sclerosis of the right lower exteremity and mild walking disturbance.

10.Sequential Ultrastructural Change of Chorionic Villi in Human Placenta by Gestational Period.

A study was performed to observe the sequential morphological change of the human placental barrier by means of light microscopy, immunohistochemistry, scanning electron microscopy and transmission electron microscopy. The examined placentas ranged in age from 4 weeks gestation to the full-term(40 weeks). Sixty seven placental specimens were obtained immediately after delivery. With the progression of gestation, the microvilli on the surface of syncytinum tended to be fewer, shorter and blunter. The syncytiotrophoblasts were getting thinner with formation of vasculo-syncytial membrane. The cytotrophoblasts formed a continuous layer which progressively disappeared but still present in the mature villi. In view of presence of intermediate cells and remnant of desmosomes, the cytotrophoblasts appeared to form the syncytiotrophoblasts. In early pregnancy, capillary formation took place by the aggregation and differentiation of the proliferation and aggregation of endothelial cells and pericytes. Myofibroblasts in villous stroma were examined by desmin immunohistochemical staining, and detected from 19 weeks to the full-term. During last period of pregancy definitive smooth muscle cells could be demonstrated, suggesting that the presence of myofibroblasts or smooth muscle cells are closely related to the placental maturity. Scanning electron microscopy of the early placenta showed numerous syncytial sprouts representing stages in the formation of new villi, but in the late period of gestation syncytial sprouts were diminished. It is concluded that the syncytiotrophoblast is originated from the cytotrophoblast in early pregnancy as the placental barrier is formulated. Moreover, myofibroblasts and smooth muscle cells in villous stroma play important role in placental maturation.
