The early histological myocardial ischemic changes in both experimental rabbitsand forensic caseworks of sudden death were demonstrated by the hematoxylin basicfuchsin-picric acid staining method.After the ligation of the coronary artery,therewere discrete patchy fuchsin-stained fibers which in contrast with the normal fibersof light-brown color.The number of fuchsin-stained fibers increased with the pro-longation of the intervals after ischemia.The results were not affected by the post-mortem autolysis.We consider that this method can be used as a routine test ofearly myocardial ischemia in both clinico-pathology and forensic pathology.

3.Application of retinal nerve fiber layer thickness detected by OCT in the diagnosis of primary open angle glaucoma and its relationship with mean defect of visual field

AIM:To investigate the application of retinal nerve fiber layer (RNFL) thickness detected by OCT in the diagnosis of primary open angle glaucoma (POAG) and its relationship with the mean defect of visual field.METHODS:A retrospective study was carried out.Totally 158 cases (158 eyes) of patients with POAG were selected as POAG group, including 81 cases in early stage, 47 cases in middle stage and 30 cases in late stage.At the same time, 50 cases of persons without eye related diseases were selected as the control group.The upper, lower, nasal and bitamporal RNFL thickness and mean defect of visual field were detected, and the patients with POAG were followed up for 3mo.RESULTS:The upper, lower, nasal and bitamporal RNFL in POAG group were thinner than those in the control, and the mean defect of visual field was higher than that in the control group at different time (P<0.05).There was no significant difference in RNFL thickness and mean defect of visual field between patients with early POAG and the control group (P>0.05).The RNFL thickness of various parts was higher than that in patients with middle and late POAG, and the mean defect of visual field was lower than that in patients with middle and late POAG (P<0.05).There was no significant difference in RNFL thickness of various parts and mean defect of visual field in POAG group at different time (P>0.05).Pearson correlation analysis showed that the upper, lower, nasal and bitamporal RNFL thickness was negatively correlated with the mean defect of visual field (r=-0.719,-0.615,-0.681,-0.518, P<0.05).CONCLUSION:OCT can monitor the change of RNFL thickness in patients with POAG, and has a negative correlation with mean defect of visual field, which can be used for early diagnosis of POAG.
4.PID1 based connection of insulin resistance to hepatocellular carcinogenesis

OBJECTIVE To investigate the effect of phosphotyrosine interaction domain containing 1 (PID1, NYGGF4) onpromotion of IR and HCC, and explore its underlying mechanisms. METHODS Lentivirus were used to mediate the knockdown of PID1 in HFD induced IR mouse model as well as ob/ob mice. Intraperitoneal glucose and insulin tolerance were performed 4 weeks after lentivirus injection. Hydrodynamics-based transfection was applied to induce the liver specific overexpression of PID1. Flow cytometry was exerted to detect the proportion and function of immune cells.qRT-PCR and Western blot were used to detect the expression of downstream pathways of PID1. Liquid chromatography-mass spectrometry (LC-MS) and co-immunoprecipitation (Co-IP) were conducted to identify proteins interacting with PID1.Chromatin immunoprecipitation(ChIP)was operated to measure the modification of H3K4me3 of PID1 promoter.RESULTS PID1 restriction improved insulin resistance,hyperglycemia and fatty liver. Conversely, hepatic knockdown of PID1 attenuated liver xenografted tumor growth. Moreover,PID1 liver-specific protooncogenes via hydrodynamics-based transfection established a primary hepatocellular carcinoma mouse model,induced an immunosuppressive environment,with the reduction of CD3+,CD4+,CD8+T cells,retarded maturation of dendritic cells(DCs),pronounced differentiation of regulatory T cells(Tregs),and recruitment of MDSC.In addition,PID1 overexpression activated prolifer-ation related genes, promoted anti-inflammatory genes, suppressed pro-inflammatory genes, induced glycolysis and lipid metabolism genes to facilitate tumorigenesis in liver. Importantly, PID1 exerted its tumor-promoting function through binding to epidermal growth factor receptor(EGFR)and activation of downstream KRAS/ERK pathway.As such,PID1 exist trimethylation of histone H3 at lysine 4(H3K4me3) modification and IR up-regulated the expression of PID1 by activation the H3K4me3 modification. CONCLUSION PID1 is a new gene that exerts both liver cancer-promoting and insulin resistance inducing function.IR accelerates liver cancer development and progression partially dependent on the activation of PID1.
5.Causes analysis of early postoperative high intraocular pressure of angle - closure glaucoma underwent compound trabeculectomy

To analyze the causes and treatments of high intraocular pressure ( > 21mmHg ) of angle - closure glaucoma underwent compound trabeculectomy 1mo after surgery.
●METHODS: This was a retrospective study of our hospital, from March 2010 to March 2013. Thirty-four (38 eyes) of angle-closure glaucoma patients were collected, who underwent compound trabeculectomy with high intraocular pressure ( > 21mmHg) 1mo after operation. We analyzed the causes and summarized the treatments.
●RESULTS:The causes which lead to early postoperative high intraocular pressure included malignant glaucoma (9 eyes of 8 cases), blood clot and connective tissue block under the scleral flap (15 eyes of 13 cases), hyphema after surgery(5 eyes of 5 cases), sclera flap incision was incarcerated with iris tissue ( 3 eyes of 3 cases ), preoperative high intraocular pressure for a long time(5 eyes of 4 cases), 1 eye of 1 case for unknown reason. After proper treatments, intraocular pressures of all patients were bellowed 21mmHg.
● CONCLUSlON: Early postoperative high intraocular pressure of angle - closure glaucoma underwent compound trabeculectomy is caused by various factors, and the early prevention and timely treatment are key points of a successful operation.
6.Effects of methionine enkephalin on immune enhancement by reducing myeloid derived suppressor cells and reprogramming liver metabolism in colon cancer mice

OBJECTIVE To investigate enhanced immune function of methionine encephalin (MENK) and its anti-tumor mechanism in CT26 colon cancer mouse model. METHODS 3×106 CT26 cells were implanted subcutaneously in BALB/c mice. Four days after, MENK was peritoneally administrated at the concentration of 20 mg·kg-1 for 14 d. The percentage of MDSCs in bone marrow, spleen, blood, tumor and liver were detected by flow cytometry. Non- esterified fatty acid (NEFA), triglycerides (TG) and total cholesterol (T-CHO) in liver homogenate were tested by a NEFA test kit, a TG test kit and a T- CHO test kit respectively. qRT- PCR and Western blot were used to measure mRNA and protein levels of inflammation-, glycometabolsim- and lipometabolsim-associated indexes in liver. RESULTS MENK decreased percentages of MDSCs in bone marrow, spleen, blood and tumor in colon cancer mice. MENK-treated mice displayed elevated ratio of CD4+T and CD8+T cells in spleen as well as increased T and B lymphocytes proliferation. Meanwhile, MENK also ameliorated liver damage reflected by lower levels of GPT and GOT in serum and reduced risks of cancer- associated index including inflammation, high lipid and high glucose. Furthermore, MENK lowered down the levels of NEFA, TG and T- CHO in liver homogenate. MENK treatment decreased expression of p- STAT3, increased expression of p-AKT, IRS1 and Glut4 at protein level as well as reduced lipogenesis-associated genes and elevated glycolysis-associated genes in liver of tumor bearing mice. Also, abated expression of genes associated with MDSCs generation (M-CSF, GM-CSF, IL-6, IL-1β) and migration (S100A9, KC) was observed within shrunken subcutaneous tumor by MENK intervention. CONCLUSION MENK has the ability to strength immune function against colon cancer by reducing MDSCs and improving liver metabolism.
7.PID1, a new tumor-promoting gene in insulin resistance mediated acceleration of hepatocellular carcinoma development and progression

8.An investigation for genetic polymorphisms of 10 STR loci on chromosome X of Chinese Hui nationality population in Ningxia region of China.

OBJECTIVETo investigate the alleles and genotypes frequency of 10 short tandem repeat (STR) loci (DXS101, DXS6789, DXS6799, DXS6804, DXS7130, DXS7132, DXS7133, DXS7423, HPRTB, DXS8378) on X chromosome of Chinese Hui nationality population.
METHODSThe study of 10 STR loci was performed by using the techniques of PCR, polyacylamide gel electrophoresis and silver staining.
RESULTSAmong unrelated Hui individuals, the allele numbers of 10 STR loci DXS101, DXS6789, DXS6799, DXS6804, DXS7130, DXS7132, DXS7133, DXS7423, HPRTB, DXS8378 were 9, 8, 4, 6, 6, 6, 4, 4, 5 or 5 respectively; the numbers of genotypes were 17, 22, 7, 14, 14, 15, 6, 7, 12 or 8 respectively. The distribution of genotypes from these 10 STR systems fitted the Hardy-Weinberg equilibrium (P> 0.05). Polymorphisms information content of 10 STR loci, except for DXS7133 (0.48) and DXS7423 (0.48), ranged from 0.54 (DXS6799) to 0.80 (DXS6789); the power of discrimination were from 0.89 (DXS7133, DXS7423) to 0.99 (DXS6789, DXS7132, DXS101).
CONCLUSIONThe loci of 10 STR on chromosome X are appropriate for individual identification, paternity testing involving a female child and for study on related disease.

9.Sequence-length polymorphism research of C-stretch in mitochondrial DNA hyper-variant region.

OBJECTIVE:
To investigate the polymorphism of C-stretch in mitochondrial DNA hyper-variant region and the values in forensic science.
METHODS:
The gene frequency were detected in such two loci of nt16180 and nt310 by using PCR-sequencing technique with two pair of primers according to the reference.
RESULTS:
The length of PCR products were 807 bp and 962 bp respectively. Seven genotypes in each locus were detected. AAAACCCCCTCCCC allele amounted to 87.72% of all nt16180 genotype, while AAAACCCCCCCCCCCCC was firstly reported in Han population. CCCCCCCCTCCCCCC allele amounted to 60.53% of all nt310 genotype. 15 haplotypes were detected among 114 samples. The GD value was 0.6309. The AAAACCCCCTCCCC-CCCCCCCCTCCCCCC haplotype was the most high frequency and amounted to 57.89%.
CONCLUSION
The obtained basic genetic data in two loci of nt16180 and nt310 of mtDNA will be useful for forensic science.

10.Validation the haplotype polymorphisms of the DXS7424-DXS101 on X-chromosome.

OBJECTIVE:
To validate the genetic characteristics and distribution of DXS7424-DXS101 on X chromosome in Han population.
METHODS:
DXS7424 and DXS101 loci were genotyped by PCR, PAGE and silvers stain methods. Their genetic parameters were analyzed by Arlequin software.
RESULTS:
There were 37 haplotypes detected in 151 Han unrelated males. The frequencies ranged from 0.0066 to 0.1391, with a GD value of 0.9453 and a DP value of 0.9389. Haplotypes 16-23 were the most common haplotypes in Han population.
CONCLUSION
Analysis of combined DXS7424 and DXS101 haplotypes appears to be a powerful means in population genetics and forensic practice for determination of identity and paternity.
