2.Effect of low concentrations of strophanthidin on isolated guinea pig failure hearts

AIM To study the effects of strophanthidin (Str) on cardiac function and Na +,K +-ATPase activity in isolated guinea pig heart failure model. METHODS Langendorff isolated heart failure models made by perfusing heart with K-H solution containing sodium pentobarbital. Eight-channel physiological recording instrument was used to determine cardiac function. Colorimetry method was used to determine cardiac sarcolemmal Na +,K +-ATPase activity. RESULTS Str increased the heart rate, left ventricular systolic pressure and the maximum rise or decline rate of left ventricular pressure in a concentration-dependent manner at 1?10 -9~1?10 -7 mol?L -1. But Str caused first a rise, then a reduction of contractility and arrhythmia accompanied by inhibition of Na +,K +-ATPase when the concentration of Str was higher than 1?10 -6 mol?L -1. Str had no obvious effect on Na +,K +-ATPase activity at 1?10 -7 mol?L -1, but increased cardiac activity at 1?10 -10~1?10 -8 mol?L -1. CONCLUSION The inotropic effect and heart toxicity of Str at higher concentration is due to inhibition of Na +,K +-ATPase, but the inotropic effect of Str at lower concentration is not the result of inhibition of Na +,K +-ATPase activity.

With the advance of medicine, nowaday most men with SCI can finish erection, ejaculation and assisted reproduction. This article reviewed the reproductive dysfunctions in men with SCI, and discussed the relevant aspects.
4.Abnormal patterns of functional connectivity between right temporal parietal junction and whole brain in children with attention deficit hyperactivity disorder

Objective To investigate the pathophysiological distinction of the functional connectivity between right temporal parietal junction(rTPJ) and whole brain in children with attention-deficit/hyperactivity disorder(ADHD) on resting-state functional magnetic resonance imaging(fMRI) and explore the neurological mechanisms of ADHD at the point of functional connectivity.Methods Resting-state functional magnetic resonance imaging scans were obtained in 30 children with ADHD from The Third Affiliated Hospital of Soochow Univcrsity and another 33 with age and gender matched controls from a normal primary school over the same period, and then,blood oxygenation level dependent(BOLD) signal was acquired to calculate the functional connectivity of rTPJ with other brain regions.Significant differences of connectivity intra-class and between groups were analyzed.Also, the correlation between the connectivity strength and the ADHD rating scales were tested.Results The pattern of functional connectivity of the rTPJ for the ADHD group was similar to that of the normal control group.Compared with the normal controls, the left superior temporal gyrus (MNI coordinates-33,6,48) , the right anterior c ingulate coltex(MNI coordinates 6,39,-3) , the right inferior parietal lobule (MNI coordinates 42,-33,30) , the left medial frontal gyrus(MNI coordinates-3,63,-18) in children with ADHD sbowed an increased resting-state functional connectivity with the whole brain ,while the right middle temporal gyrus(MNI coordinates 57,-33,-12) , the right cingulate gyrus(MNI coordinates 18,-21,36) showed a decreased resting-state functional connectivity with the whole brain.The average connection strength between the rTPJ and the whole brain showed positive correlation with the ADHD scores,while in normal controls,it showed negative.The connection strength between the rTPJ and those 6 brain areas with significant statistical difference was relevant to the ADHD scores,but they had no statistical significance.Conclusion The functional connection abnormalities between tbc rTPJ and the whole brain in children with ADHD may be relevant to the clinical symptom of attention-deficit.
5.Inhibition of valdecoxib on the proliferation of human gastric cancer cells

AIM To study the effect and mechanism of valdecoxib, a selective COX 2 inhibitor, on human gastric cancer BGC 823 cells. METHODS MTT assay and flow cytometry were used to observe the effect of valdecoxib on proliferation, apoptosis and the cell cycle distribution of BGC 823 cells. Laser confocal microscopy, transmission electron microscope and DNA fragmentation assay were further used to detect the apoptosis. The content of LDH was examined using LDH kit. RESULTS Valdecoxib in 25~400 ?mol?L -1 significantly inhibited the proliferation of BGC 823 cell in a time and dose dependent fashion, the inhibition rate of proliferation was 24 0%~92 0% after 72 h, and the rate of apoptosis was increased from (2 6?0 7)% to (7 6?1 5) %~(16 5?1 5)%. 100~400 ?mol?L -1 valdecoxib also decreased the proliferation index and the proportion of cells in the S phase, increased the proportion of cells in the G 0/G 1 phase, but had no effect on the proportion of cell in the G 2/M phase. CONCLUSION Valdecoxib inhibits human gastric cancer BGC 823 cells growth by inducing apoptosis and cell cycle arrest. The growth inhibitory effect of 400 ?mol?L -1 valdecoxib is also associated with cell necrosis.
6.The focal cerebral ischemia and reperfusion model induced by Endothelin-1 in rat

AIM To establish an Endothelin 1 induced focal cerebral ischemia and reperfusion model. METHODS Endothelin 1(ET 1), a potent vasoconstrictor, was injected near the middle cerebral artery to induce reduction in cerebral blood flow and ischemic neuronal damage. The changes of cerebral blood flow in striatum were characterised using hydrogen clearance technique. The neurologic scores were performed and the infarct volume was identified by TTC staining at 6 h and 24 h after ET 1 application, respectively. RESULTS ET 1 induced a dose dependent reduction of cerebral blood flow in striatum and the CBF at 10 min after ET 1 injection were the lowerest. CBF at 10 min post injection was (27 1?2 9)% in group 300 pmol, (12 7?2 1) % in group 360 pmol, (11 9?1 8)% in group 400 pmol and (9 5?1 6)% in group 500 pmol , respectively. Neurologic score showed that ET 1 could induce variable grade neurologic deficit. 7.Rifaximin in Treating Acute Infectious Diarrhea:A Clinical Study

OBJECTIVE To evaluate the clinical curative effect and safety of rifaximin in treating acute infectious(diarrhea).METHODS A multi-center randomized trial with double-blind double-analogue and parallel(-control) with positive drug was used.All the 240 chosen patients were classified into two groups.There were 120 cases in the trial group.On the first day,the patients received rifaximin 300mg tid.From the 2nd to 5th day,the patients(received) rifaximin 400 mg tid.There were 120 cases in the control group,on the first day,the patients received levofloxacin 100 mg tid;from 2nd to 5th day,they received levofloxacin 100 mg tid.The total therapeutic course was 3 to 5 days.RESULTS After 3-5 day treatment,symptoms such as ache in abdomen and diarrhea were(alleviated) or disappeared and stool examination was turned better or normally.To the trial group,the cure rate was 84.68%,the dominant effecive rate was 15.31%,and the total(effective) rate was 100.00%.The bacteria clearance rate was 100.00% in the trial group.There was no significant difference between two groups.There were no severe side effect in the two groups.CONCLUSIONS Rifaximin is an effective and safe drug for acute(infectious) diarrhea for adults.
8.Relationship between antitumor effect of valdecoxib and COX-2 expression

Objective To evaluate the inhibitory effect of valdecoxib on the growth of the cancer cell lines and involvement of COX-2 in this inhibition. Methods Western blotting and immunocytochemistry were used to detect the expression of COX-2. MTT assay was used to determine inhibitory effect of the drugs on the cell growth. The content of PGE_ 2 in cell medium was determined with PGE_ 2 ELISA kit. Results ①Clone 26 cells expressed high levels of COX-2, whereas BGC-823,HGC-27 and SK-OV-3 cell had no COX-2 expression. ②Valdecoxib inhibited the growth of BGC-823, HGC-27, SK-OV-3 and clone 26 cells, with a IC_ 50 of 110.7, 99.2, 113.3, 117.6 ?mol/L, respectively. ③The inhibitory effect of these drugs on BGC-823 and clone 26 cell was in the descending order of valdecoxib, SC-560 and indomethacin. ④PGE_ 2 did not antagonize the effect of valdecoxib, SC-560 and indomethacin on BGC-823 and clone 26 cells. ⑤The inhibitory effect of valdecoxib and indomethacin on the growth of clone 26 cells was not compatible with that on PGE_ 2 . Conclusion The inhibitory effect of valdecoxib on cell growth is not related to its effect on COX-2.
10.MtrF gene expression in Neisseria gonorrhoeae of different resistance levels

Objective To study the relationship between the expression of mtrF gene and high-level multiple resistant Neisseria gonorrhoea.Methods ① The susceptibility of 58 clinical Neisseria gonorrhoeae to 5 kinds of antibiotic agents was tested by disc diffusion method.② The minimum inhibitory concentration(MIC) of erythromycin was determined by tube dilution method.③ The expression of mtrD and mtrF gene in susceptive group,inter-mediated resistant group and high-level multiple resistant group was detected by semi-quantitative RT-PCR.Results ① There were 30 strains presenting resistance to two or more than two antimicrobial agents,which accounted for 51.7% of the 58 clinical strains.② The number of strains sensitive,intermediate and resistant to erythromycin was 7,21 and 30,respectively,and there were 17 strains with erythromycin MIC≥32.0 ?g /mL.③ Compared with that in susceptive group and inter-mediated resistant group,mtrF expression was up-regulated in high-level multiple resistant group(P
