1.Effect of a selective JNK inhibitor on pregnant rats with ethinylestradiol induced intrahepatic cholestasis

Objective To evaluate the influence of SP600125,a selective c-Jun N-terminal kinase (JNK) inhibitor,on the levels of serum total bile salt (TBA) and Bsep,Ntcp expression in the hepatic tissue of rats with ethinylestradiol induced intrahepatic cholestasis.Methods Rats pregnant for 15days were administered the subcutaneous injection of 17- b -estradiol propylene ( EE ) to modulate the ICP animal models,and be SP600125 to intervene.Testing the level of serum TBA and the expression of c-Jun,Bsep,Ntcp in the hepalatic tissue.Results The average gray values of c-Jun in the group of ICP models were significantly lower than the normal control group ( 101.05 ± 5.20 vs 118.99 ± 5.95,P < 0.05 ).After the intervention of SP600125,comparing with the group of ICP models,the expression of Bsep,Ntcp in the group of SP600125 intervention were significantly higher,and this change in the high dose of SP600125 intervention group was more obvious ( low dose intervention group Bsep:0.452 ±0.031 vs 0.291 ±0.043,Ntcp:0.462 ± 0.015 vs 0.285 ± 0.021,P < 0.05 ; high dose intervention group Bsep:0.568 ± 0.038 vs 0.291 ±0.043,Ntcp:0.605±0.020 vs 0.285 ±0.021,P <0.05),while the level of TBA in the serum was significantly lower.Conclusions Treatment with SP600125 can down-regulate the level of c-Jun/AP-1,and it may participate in the lower expression of Bsep、Ntcp in the ICP rats which were induced by 17-bestradiol.

3.Extended-field intensity modulated radiation therapy and intra-cavitary brachytherapy combined with chemotherapy for stage Ⅰ b1-Ⅳ a cervical cancer with positive para-aortic lymph nodes

Objective To investigate the treatment effects and toxicities of extended-field intensity modulated radiation therapy (EF-IMRT) and intra-cavitary brachytherapy combined with chemotherapy for stage Ⅰ b1-Ⅳa cervical cancer with positive para-aortic lymph nodes.Methods A total of 46 stage Ⅰ b1-Ⅳa cervical cancer patients with positive para-aortic lymph nodes treated at Fudan University Shanghai Cancer Center between 2009 and 2011 were reviewed.Neoadjuvant,concomitant and adjuvant chemotherapy with paclitaxel and carboplatin were administrated for one cycle before radiation therapy,two cycles during radiation therapy or three cycles after radiation therapy.All patients received EF-IMRT and intra-cavitary brachytherapy.The positive lymph nodes received an additional boost dose.Results All patients received EF-IMRT to 50.4 Gy (1.8 Gy per fraction).Twenty-six patients was treated with boost dose of 6.0-8.0 Gy in 2.0 Gy per fraction to positive para-aortic lymph nodes.Thirty-seven patients received a positive paraaortic lymph nodes boost or(and) parametrial boost.All patient also received a high-dose-rate intra-cavitary brachytherapy at the point A dose of 20.0-30.0 Gy in 5.0 Gy per fraction.Total chemotherapy cycles were 189,and the average patient received 4.1 courses.Two cases (4%,2/46) experienced grade Ⅲ gastrointestinal toxicities,no patients suffered grade Ⅳ gastrointestinal toxicities.Fifteen cases (33%,15/46) experienced grade Ⅲ hematological toxicities,and 3 (7%,3/46) experienced grade Ⅳ hematological toxicities.Late grade Ⅲ-Ⅳ toxicity was seen in 3 cases (7%,3/46).The 3 year progressionfree survival rate was 46.2%,and the 3 years overall survival rate was 61.2%.Conclusion EF-IMRT and intra-cavitary brachytherapy combined with chemotherapy is safe and effective for stage Ⅰ b1-Ⅳa cervical cancer with positive para-aortic lymph nodes.
4.Role of Sonic Hedgehog signal pathway in embryogenesis and neural regeneration

BACKGROUND:Many studies have showed that Sonic Hedgehog (Shh) signal pathway regulates the proliferation and differentiation of nerve cels, axon guidance and neural regeneration after brain injury. OBJECTIVE:To summarize the role of Shh signal pathway in embryonic development and post-natal nerve repair. METHODS:A computer-based retrieval was performed by the first author in PubMed and CNKI database to search related papers published from January 1980 to July 2015 using the keywords of “Shh signal pathway, embryogenesis, neural regeneration” in English and Chinese, respectively. Articles related to Shh signal pathway in embryonic cels differentiation, tissue development, post-natal neural regeneration, axon migration and tumor formation. A total of 38 relevant literatures were retrieved. RESULTS AND CONCLUSION: Recently, Shh signal pathway has become an issue of concern because this signal pathway is closely related with neural regeneration after brain injury. Shh is widely expressed in Hedgehog (Hh) family and plays a crucial role in embryogenesis and organogenesis. Shh is wel known to contribute to the patterning of neural tube system, regulate the proliferation and migration of progenitor cels, and control axonal growth and guidance. In addition, Shh is closely related to tumor formation. Studies have shown that Shh can reduce infarct volume and improve behavior prognosis of stroke rats.

6.EFFECTS OF CHROMIUM ON BLOOD GLUCOSE AND LIPID METABOLISM AND GENE EXPRESSION OF SKELETAL MUSCLES IN DIABETIC RATS

Objective: To study the effects of chromium on glucose and lipid metabolism and gene expression in diabetic rats.Methods: Male Wistar rats were assigned to three groups:normal control(NC), alloxan-induced diabetic control group(DM), and DM with chromium supplementation group(DM+Cr). Cr 200 ?g/(kg bw?d) was supplemented orally for 60 days. At the end of the treatment, the blood glucose, lipid and serum insulin were measured, and the changes in gene expression among three groups were studied by mRNA differential display technique.Results: Blood glucose in DM+Cr group decreased significantly than that before experiment. The levels of serum TG, TC, LDL-C and AI in DM+Cr group were lower than those of DM group, while the serum HDL-C levels were higher. Serum insulin was not improved obviously in DM+Cr group. 11 cDNA fragments larger than 400 bp expressed differences in skeletal muscles between DM+Cr group and DM group and were isolated, 4 of which expressed higher in DM+Cr group, while the rest expressed higher in DM group.Conclusion: Chromium supplementation could partially improve the disorders of glucose and lipid metabolism, and have an impact on some gene expression in diabetic rats, which may contribute to the regulating effects on its disorders of metabolism.

Objective To study of the influential factors of dynamic electrocardiogram (DCG)changes after intensity modulated radiation therapy(IMRT) for the lung cancer and breast cancer .Methods IMRT was delivered to 43 patients with lung cancer and 39 patients with breast cancer ,and they were installed DCG before and after the therapy .The associations of clinical and DVH fac‐tors with DCG changed after IMRT were analyzed .Results 32 patients have abnormal DCG after IMRT (39% ) ,the clinical data and physical factors were analyzed by Logistic regression ,gender(OR=1 .52 ,P<0 .05) and heart V40 >22% (OR=1 .15 ,P<0 .05) were the independent influencing factors for abnormal DCG .Conclusion The rate of abnormal DCG is low after IMRT for the lung cancer and breast cancer ,and gender and heart V40 were the independent influencing factors for abnormal DCG .
10.Effects of swimming in cold water on the functioning and structure of peripheral nerves in diabetic rats

Objective To observe the effects of swimming in cold water on the functioning and structure of the peripheral nerves of diabetic rats,and to compare the effects of seawater and fresh water. Methods Forty SD rats weighing ( 250 ± 20) g were randomly divided into a normal control group (A),a diabetic model group ( B ),a seawater swimming group (C) and a fresh water swimming group (D) with 10 rats in each group.The swimming training was carried on 5 times a week for 8 weeks.At the end of the 4th and 8th week of training,caudal nerve conduction velocity (CNCV) was measured.The nerve structure of the caudal nerves was observed at the end of the 8th week. Results By the 4th week,CNCV had slowed significantly in group B compared with group A,but not in groups C and group D.Compared with group B,CNCV had increased significantly in group C.There was no significant difference in CNCV between groups C and D.At the 8th week,compared with group A,CNCV had slowed in groups B and C.Compared with group B,CNCV was significantly faster in groups C and D.However,there was no significant difference between group C and group D with regard to CNCV.At the end of the 8th week demyelination was observed in the caudal nerves under a light microscope and an electron microscope in groups B,C and D,but the demyelination was milder in groups D and C. Conclusion Swimming in cold water can prevent or delay diabetic neuropathy in diabetic rats.There was no significant difference between seawater and fresh water swimming in terms of its effect on CNCV.
