1.The effect of respiratory syncytial virus infection on neutrophil adherence to airway epithelial cells.

BACKGROUND: Respiratory syncytial virus (RSV) is the most common cause of lower respiratory infection in infants and young children, but the pathogenesis of RSV-induced inflammation is not well defined. MATERIAL AND METHOD: In order to examine the potential interactions between virus-infected airway epithelial cells and neutrophils, we studied the ability of neutrophils to adhere to yirus-infected airway epithelial cell monolayers by myeloperoxidase assay. Also we measured the ability of airway epithelial cells to secrete interleukin-8(IL-8) and inter-cellular adhesion molecule-1(ICAM-1) in virus-infected airway epithelial cell cultures by enzyme-linked immunosorbent assay(ELISA). The degree of IL-8 and ICAM-1 gene expression in the RSV-infected BEAS-2B cell cultures were analyzed by reverse transcription-polymerase chain reaction(RT-PCR). RESULTS: The RSV-infected BEAS-2B cell resulted in significantly enhanced level of neutrophil adherence compared to the uninfected control(p (0.001). IL-8 and ICAM-1 production significantly increased by RSV infection(p<0.05). There was a significant positive correlation between neutrophil adherence and IL-8 level(r=0.73, p=0.002), and ICAM-1 level (r=0.843, p=0.001) in RSV-infected cells. The degree of both IL-8 and ICAM-1 mRNA expression increased in the RSV-infected cells compared with the uninfected ones. CONCLUSION: These results suggest that RSV infection significantly enhances the production of IL-8 and ICAM-1 in airway epithelial cells which then results in increased neutrophil adherence.

4.Esophageal Actinomycosis after Insertion of Esophageal Stent: A Case of Surgical Experience.

Actinomycosis is an indolent, suppurative infection caused by an anaerobic gram-positive organism (usually actinomyces israelii) which usually causes infection in the face, mediastitum, lung, and abdomen. Primary esophageal actinomycosis which is not related with pulmonary or mediastinal actinomycosis, is very rare, especially in immunocompetent host. A 58-year-old woman has been suffered from dysphagia, odynophagia, and chest pain after insertion of esophageal stent in esophageal acid stricture. She underwent a esophagectomy with esophagogastrostomy for above mentioned symptoms. Pathologic diagnosis was a esophageal actinomycosis.

5.The Distribution of HLA Antigens and Haplotypes in Koreans.

BACKGROUND: The HLA system is known to be the most polymorphic genetic system in human and there are characteristic racial differences in the distribution of HLA antigens, alleles, and haplotypes. This study was performed to examine the frequency of HLA antigens, alleles and haplotypes in Koreans. METHODS: Two thousand healthy Koreans registered for unrelated bone marrow donors were subject to the study. HLA-A, B and C antigens were typed by the serological method, and HLA-DR DNA typing (low resolution) was done by PCR and reverse hybridization. HLA allele and haplotype frequencies and linkage disequilibrium values were calculated by the maximun likelihood method using the computer program of the 11th International Histocompatibility Workshop. RESULTS: HLA antigens identified in 2000 Koreans were 14 in A locus, 33 in B locus, 8 in C locus and 12 in DR locus. Alleles showing frequencies of more than 10% in decreasing order of frequency In each HLA locus were A2, A24, A33, All, B44, B62; CBL, Cw3, Cwl, Cw7, DR4, DR2, DRl3, DR8, and DR9. Among A-B, C-B, B-DR 2-locus haplotypes, A33-B44, A30-B13, Al-B37, Cwl-B54, Cw4-B62, B7-DR1, B37-DR10 showed strong positive linkage disequilibrium (Chi-square > 1000). The most common A-B-DR haplotypes in Koreans occurring at frequency of more than 2% were A33-B44-DRl3 (4.8%), A33-B58-DRl3 (3.2%), A33-B44-DR7 (2.6%), All-B62-DR4 (2.3%), A24-B7-DR1 (2.3%), and A30-Bl3-DR7 (2.1%) Comparison of the distribution of A-B-DR haplotypes among east Asian populations reveals that Koreans are most close to Japanese, but show higher degree of polymorphism in the distribution of HLA haplotypes compared to Japanese. CONCLUSIONS: The results obtained in this study can be used as basic data for Koreans in the fields of organ transplantation, disease association studies and anthropologic studies.

8.Benign Osteoblastoma: A Case Report

Osteoblastoma is a rare benign bone tumor and is characterized hitologically by an abundant number of osteoblasts and by the presence of s large amount of osteoid tissue in a highly vascular matrix. The lesion is frequently encountered in children and adolescents with most of the cases occuring between the ages of 10 and 25 years. There is significant sex prevalence (M.: F.=2: 1). Up to now there has been few reports of a benign osteoblastoma occuring in the cervical spine. This report describes a boy of a benign osteoblastoma which involved the cervical vertebral arch and spinous process, presenting with pain and limitation of motion. He was given en-bloc resection of tumor mass and confirmed by histological examination. Postoperation result was good and he goes school without any problem in about 12 months of follow up.

9.The Effects of TGF-beta2 and bFGF on the Proliferation of Retinal Pigment Epithelial Cells.

This study was undertaken to document the effect of transforming growth factor-beta2 (TGF-beta2(TGF-beta2) and basic fibroblast growth factor (bFGF) on the proliferation of pig retinal pigment epithelial cells (RPE). Whereas bFGF increased the proliferation, TGF-beta2 showed the inhibitory effect on the proliferation The inhibitory effect of TGF-beta2 disappeared in RPE subcultured with 10ng/ml of bFGF. Both TGF-beta2- and bFGF-specific antisense oligonucleotides blocked the autocrine effect of the growth factors. PLC-71 -specific antisense oligonucleotide inhibited the effect of TGF-beta2 and bFGF. Genistein inhibited the effect of TGF-beta2 and bFGF in dose-dependent man, ner. The data suggest the involvement. of in PLC-/1 and tyrosine kinase in signalling.

10.Effect of GM-CSF on Neutropenia Induced by Chemotherapy or Immunotherapy in Patients with Urologic Malignancy.

Purpose: Neutropenia is a major factor contributing to morbidity and mortality in patients undergoing chemotherapy for cancer. The efficacy and complications of GM- CSF (Granulocyte-Macrophage colony stimulating factor) on the neutropenia induced by chemotherapy or immunotherapy were evaluated in patients with a urologic malignancy. Material and Methods: Twenty-nine patients with a histologically confirmed urologic malignancy, who presented with neutropenia during either chemotherapy or immunotherapy, were given a fixed dose (400microgram) of GM-CSF subcutaneously for three to fifteen days. Twenty-nine patients with neutropenia were given GM-CSF for 50 cycles. The patients were evaluated by means of a complete blood cell count, white cell and differential counts. The mean time to response was 2.3 days. The statistical significance of the differences was calculated using a paired t test. RESULTS: GM-CSF treatment in patients with neutropenia resulted in an increase in the mean neutrophil count, the eosinophil count, and the monocyte count. Complications of GM-CSF included fever (76%), GI problems (19%), headache (14%), bone pain (14%), arthralgia (10%), and urticaria (5%). CONCLUSIONS: GM-CSF may be useful in reducing the toxicity of chemotherapy. Therefore, GM-CSF can be used to increase the effect of chemotherapy without delaying or stopping treatment as a result of neutropenia.
