1.Effect of ginsenoside-Rg3 on the expression of VEGF and TNF-α in retina with diabetic rats

AIM: To investigate the effect of ginsenoside-Rg3 on the expression of vascular endothelial growth factor (VEGF) and tumor necrosis factor-α (TNF-α) in retina with diabetic rats and its roles in preventing neovascularization in diabetes. METHODS: Sixty male Wistar rats were divided into 3 groups randomly: negative control group, diabetic control group and ginsenoside-Rg3 treatment group (5mg/kg, 0.2mg/mL) followed by establishing diabetic model. The expression of VEGF and TNF-α were measured after 8 weeks. RESULTS: There were significant differences among negative control group, diabetic control group and ginsenoside-Rg3 treatment group in the expression of VEGF and TNF-α (F=129.363, 211.992; all the P<0.01). VEGF and TNF-α expression were significantly higher in diabetic control group and ginsenoside-Rg3 treatment group than that in negative control group (P<0.01), with a significant reduction in ginsenoside-Rg3 treatment group than that in diabetic control group (P<0.01). CONCLUSION: Ginsenoside-Rg3 can down-regulate the expression of VEGF and TNF-α in retina, which may interfere in the development of diabetic retinopathy.
2.Study on the pharmacological mechanism of sodium ferulate for anti-asthmatic effect in guinea pigs.

OBJECTIVETo study the anti-asthmatic effect of sodium ferulate (SF) and its mechanism in guinea pig asthmatic model.
METHODSGuinea pigs were sensitized with ovalbumin as animal asthmatic model and treated with 3 different concentration of SF for 8 days. Levels of endothelin (ET) and nitric oxide (NO) in blood and lung tissue, and eosinophil (EOS) in blood and bronchoalveolar lavage fluid (BLAF) was counted at the end of trial.
RESULTSSF could significantly lower the ET content and increase the NO concentration in serum and lung tissue, reduce the number of EOS in blood and BLAF (P < 0.05, P < 0.01). Stronger effect was showed in the high dose group.
CONCLUSIONMechanism of anti-asthmatic action of SF might be to increase NO concentration, lower ET content, alleviate EOS infiltration.

3.Correlation of plasminogen activator and plasminogen active inhibitor expression and capillary density in IgA nephropathy.

Objective To analyze mRNA and protein expressions of plasminogen activator and plasminogen activator inhibitor-1 using microdissection and quantitative real-time PCR,and to analyse their associations with microvascular diseases in IgA nephropathy.Methods Twenty-four IgA nephropathy patients treated in Kidney Center and Kidney Laboratory of PLA from 2000 to 2004 were admitted into the study;20 renal glomeruli or 200 renal tubules and peritubular interstitial per patient were captured by Laser Microdissection System from IgA nephropathy renal biopsy slides(8 ?m),and mRNA was extracted.PA mRNA and PAI-1 mRNA levels in glomerulus and tubulointerstitial area were measured by Taqman quantitative real-time PCR.Capillaries densities in glomerulus and tubulointerstitial area were measured using immunohistological staining method and computer image analysis system.Results With the aggravation of lesions,the glomeruli and peritubular capillaries densities of IgA nephropathy decreased.The glomerular and peritubular capillaries densities were negatively associated with the level of serum creatinine(R2=0.6946 and R2=0.6271,P
4.Apoptosis of macrophage induced by E_2 at high concentration

AIM To study the effect of 17?-estradiol(E 2) on apoptosis of macrophages at certain concentration.METHODS Fluorescent microscopy and scanning eletron microscopy(ESEM) were used to detect apoptosis changes of macrophages induced by E 2 at relatively high concentration and intracellular Ca 2+ and matochontrial transmembrane potential were examined by laser scanning confocal microscopy(LSCM). RESULTS Apoptosis of macrophages was induced by high concentration of E 2 (≥1 ?mol?L -1). Intracellular Ca 2+ elevated and matochontrial transmembrane potential decreased after E 2 treatment. CONCLUSION Apoptosis of macrophages is induced by certain concentration of E 2 (≥1 ?mol?L -1) and this may be related to elevated intracellular Ca 2+ and decreased matochontrial transmembrane potential.
5.Determination of the Content of Stilbene Glycoside in Zhipingkang Misture by HPLC

OBJECTIVE:To establish an HPLC method for the determination of Stillbene glycoside (THPG) in Zhipingk-ang misture.METHODS:The chromatographic column was Hypersil C18(250 mm?4.6 mm,5 ?m).The mobile phase consisted of acetonitribl-water(25∶75) at a flow rate of 1.0 mL?min-1 with detection wavelength set at 320 nm. RESULTS: There was a good linear relationship within the concentration range of 0.5~8.0 ?g for THPG. The average recovery of THPG was 100.03%(RSD=1.07, n=6).CONCLUSION: The adoptd method is accurate, reproducible and suitable for the quality con-trol of Zhipingkang misture.

ObjectiveTo explore the influence factors of prognosis of spinal cord injury patients. MethodsFollow-up data of 121 cases with spinal cord injury was analysed with retrospective cohort study.ResultsThe highest recover rate was in the patients who accepted the special rehabilitation therapy,while that of recovering two class was in the patients who accepted the magnetic stimulation.Conclusions Special recovering cure and magnetic stimulation can surely promote recovery of spinal cord injury patients.

Objective To investigate the management of external iliac artery mycotic hemorrhage after kidney transplantation. Methods A case of external iliac artery mycotic hemorrhage after kidney transplantation managed by the use of hypogastric artery autograft was reported with review of the literature.The massive blooding occurred 3 times after kidney transplantation in a male patient 25 years of age on the 22nd,24th and 38th day after transplantation.The blooding amounted to 800 ml,2500 ml and 3800 ml respectively.The blood loss was replaced and prompt surgical exploration was carried out with the blooding site at the anastomosis sutured up on the 1st and 2nd episode of bleeding.On the 3rd occurrence of bleeding, the diseased external iliac artery segment, about 2cm in length, was resected and the gap was replaced by a 3cm long hypogastric artery autograft. Results The blood flow through the repaired external iliac artery and the blood supply to the lower extremity was adequate.Periodic hemodialysis had been restored and the patient waited for reimplantation. Conclusions External iliac artery mycotic hemorrhage after kidney transplantation is a serious and fatal complication.Simple arterial repair is usually noneffective.Resection of the diseased mycotic segment of the external iliac artery with repairing of the gap with a hypogastric artery autograft is rational,feasible and simple.The procedure is highly recommended.
9.Color Doppler sonographic evaluation in 32 patients with venous leakage in venous incompetence in erectile dysfunction

Objective To investigate the hemodynamic change of venous leakage in venogenic impotence. Methods Thirty-two patients with vasculogenic impotence were evaluated with conventional penile duplex sonography with spectral analysis and color Doppler imaging after intracavernosal injection of PGE1 to induce an erection.The color Doppler appearance of deep dorsal vein of penis,cavernous veins and bulbourethral vein wen observed and the correlativity with resistance index(RI)of cavernous artery were analyzed. Results After five minutes following intracavernous injection,the flow and diameter of the penis vein were continuous increased.The coefficient correlation r between the discharge of deep dorsal vein of penis,cavernous veins and bulbourethral vein with RI of cavernous artery were-0.55,-0.53,-0.24(P<0.05).Considering the existence of mixed venous leakage,r between the discharge of vein of penis with RI of cavernous artery was-0.88(P<0.001).Conclusions Higher qualitative ultrasound imaging could demonstrate venous leakage sensitively and assess the location and degree of venous leakage in venogenic impotence initially.
10.Bladder anatomical changes and dose variation during the course of intensity-modulated radiation therapy of cervical cancer

Objective To investigate bladder anatomical changes and dose variation in patients with cervical cancer.Methods We analyzed 20 patients,undergoing external beam radiotherapy scanning cone beam CT(CBCT)before each fraction.Bladder was contoured on each CBCT,was projected onto the planning CT and assesses anatomical changes and dose variation.Results A total 451 CBCT images,for 20 patients were collected for analysis,show more change in bladder volume and position.In 15 cases bladder volume and V45 had no significant correlation(r=0.225 -0.473,all P>0.05),4 cases shows negative correlation(r=-0.564,P<0.05;r=-0.597,P<0.01;r=-0.942,P<0.01;r=-0.816,P<0.01),1 case shows positive correlation(r=0.662,P<0.01).Have more than the criteria(V45≤50%)number is 64/451(14.2%)in whole treatment.Conclusions For most patients by filling adequacy bladder,bladder dose variation is acceptable:CTV lager for individual patients should be closely observed its regression,implementation of the offline or online calibration.
