2.Expressions of interleukin-22 in sera and peripheral blood mononuclear cells of patients with pityriasis rosea
Chinese Journal of Dermatology 2014;47(10):746-747
Objective To investigate the role of interleukin-22 (IL-22) in the pathogenesis of pityriasis rosea (PR).Methods Peripheral venous blood samples were obtained from 48 patients with PR in acute stage and recovery stage and from 20 healthy human controls.Real-time fluorescence-based quantitative PCR was performed to measure the mRNA expression of IL-22 in peripheral blood mononuclear cells (PBMCs),and enzyme-linked immunosorbent assay (ELISA) to determine the level of IL-22 in sera. Results The relative expression level of IL-22 mRNA in the patients with PR in recovery stage was significantly higher than that in the healthy controls (2.89 ±0.91 vs.1.91 ± 0.60,t =3.84,P < 0.05),but lower than that in the patients in acute stage (2.89 ± 0.91 vs.4.15 ±1.36,t =3.61,P < 0.05).Similarly,the level of serum IL-22 was significantly increased in the patients with PR in recovery stage compared with the healthy controls ((21.44 ± 7.33) vs.(13.22 ± 4.26) ng/L,t =4.49,P < 0.05),and increased in the patients with PR in acute stage compared with those in recovery stage ((36.87 ± 10.63) vs.(21.44 ± 7.33) ng/L,t =5.01,P < 0.05).The level of serum IL-22 was positively correlated with the stage of PR (r =0.883,P < 0.05).Conclusions IL-22 is overexpressed in PBMCs and sera of patients with PR,implying that IL-22 is involved in the pathogenesis of PR.
3.Design of evaluation index system of scientific research performance based on the Balanced Score Card and Analytic Hierarchy Process in military medical universities
Chinese Journal of Medical Science Research Management 2015;28(3):225-230
Objective To design an evaluation index system of scientific research performance in military medical universities and determine the weights of these indexs. Methods Through literature study, the Balanced Scorecard Card (BSC) is used to preliminarily design the evaluation index system of scientific research performance in military medical universities. And then the Delphi Method is used to finalize the valuation indexs. The weights of valuation indexes at all levels are determined based on the Analytic Hierarchy Process (AHP).The comprehensive weight is determined by math-statistical method. Finally,we quantify the qualitative indicators of third-level indicators, collect the data on scientific research performance of two departments in recent two years and evaluate the applicability of the system. Results A systematic and comprehensive evaluation system which can reflect the scientific research performance in military medical universities, comprise 8 first-level indicators,22 secondlevel indicators and 50 third-level indicators. The weights of valuation indexes at all levels have been determined. The empirical study shows that in the comprehensive level of B department is higher than that of A.But each department has its strengths in the aspects of four dimensions. For example, compared to the score of B department, the score of A department is significantly lower in the aspect of traditional scientific input and output, comparable in the two aspects of customers and internal processes, slightly lower in 2013 but higher in 2014 in the aspect of learning and growth. Conclusions The evaluation index system of scientific research performance based on the BSC and AHP comprise long term and short-term indicators, financial and non-financial indicators, explicit and implicit indicators, qualitative and quantitative indicators.Through using the scientific consultation, analysis and calculation methods, the evaluation index system is proved to be valid and reliable.Compared to the traditional evaluation system of scientific research performance, the evaluation system based on the BSA can better and more comprehensively reflect the level of scientific research, innovation ability and sustainable development trend of a department in the aspcet of applicability.
4.Design of performance evaluation indexs of scientific research management based on the Balanced Scorecard in military medical universities
Chinese Journal of Medical Science Research Management 2015;28(2):157-159,163
The scientific outputs are usually used as the main measure basis of the traditional performance evaluation of scientific research in military medical universities. In this research,we design the performance evaluation indexs of scientific research management based on the Balanced Scorecard from four levels including finance, customers, internal processes and learning and growth, which can provide a new perspective and theoretical basis for improving performance evaluation system of scientific research management in military medical universities.
5.Diagnostic value of BG and GM test for invasive fungal infections in ICU patients
International Journal of Laboratory Medicine 2017;38(11):1468-1470
Objective To evaluate the early diagnostic value of 1,3-β-D glucan(BG) and galactomannan(GM) test for invasive fungal infections(IFI)in intensive care unit(ICU) patients. Methods From 2013 to 2015,the concentration of serum BG and GM were detected in 452 cases of ICU patients, including 182 cases with diagnosis of IFI,46 cases with possible diagnosis of IFI and 224 cases of non-IFI.The diagnostic performance of BG,GM and combined test were analyzed. Results The sensitivity, specificity, positive predictive value(PPV) and negative predictive value(NPV)of BG test were 75.0%,80.4%,75.8%,79.6%.Those of GM test were 25.80%,85.7%,59.5% and 58.7%.The sensitivity and specificity of BG and GM combined test were 87.4% and 94.7% respectively. Conclusion BG and GM combined test could improve the diagnostic efficiency, reduce the false positive rate and false negative rete, which might be helpful for the early diagnosis of IFI in ICU patients.

Journal of Shanghai Jiaotong University(Medical Science) 2000;20(5):439-441,449
Objective To study the relationship among p53 gene mutation, apoptosis and cellploidy as well as to explore the role of mutated p53 gene in tumorigenesis. MethodsA total of 85 speci-mens (20 normal ovarian specimens, 20 ovarian benign tumors, 20 non - metastatic carcinomas and 25metastatic carcinomas) were chosen. p53 gene mutations were detected by PCR- SSCP (single strand con-formation polymorphism). Cell ploidy, apoptosis role of carcinoma cells and cell distribution in each cellstage were detected by flow cytometry. Resultsp53 gene mutation and non- mutation of ovarian carci-noma was 60% and 40% respectively ( P >0.05). The rate of apoptosis of carcinoma cells with mutationof p53 gene (20.25%) was much lower than that without p53 mutation (41.68%, P <0.05). Aneuploidcarcinoma cells had been found in 17/23 carcinoma specimens with p53 gene mutation and 8/22 carcinomaspecimens without p53 mutation ( P < 0.05). Aneuploid carcinoma cells metastasize more easily thandiploid carcinoma cells ( P < 0.05). The rate of apoptosis was not different between aneuploid and diploidpatients. ConclusionMutation of p53 gene decreases the apoptosis rate of ovarian carcinoma and maypromote tumorigenesis. Cell ploidy had a linkage with p53 gene mutation, but not with cell apoptosis.

Journal of Shanghai Jiaotong University(Medical Science) 2009;29(11):1377-1382
Protein post-translational modifications are important ways to regulate protein function and cell behavior, and oxidative stress directly influences protein post-translational modifications. P53 protein has various post-translational modifications, which can be quickly regulated under stress conditions to activate a series of downstream target genes and facilitate the p53 function diversity. The effects of oxidative stress on p53 post-translational modifications of phosphorylation, ubiquitination, sumoylation, acetylation and methylation are introduced in this paper.
