1.INCREASED CELLULAR PROLIFERATION AND PROTECTION OF RECOMBINED HUMAN HEPATIC STIMULATOR SUBSTANCE

Objective Based on the result that a gene coding for human HSS had been identified and prokaryotically expressed, this study is aiming to investigate the proliferative and protective effect of recombinant HSS on hepatoma cells. Methods hHSS protein was produced in BL\|21 strain of E.Coli containing pET\|42a vector and purified with His\5Tag affinity chromatography. A various dosages of 80\|400??g/L were administrated into cell culture. The cell proliferation was analyzed by MTT, cell\|count and flow cytometry methods. Furthermore, cellular growth signaling as indexed by phosphorylation of mitogen\|activated protein kinase (MAPK) was determined with Western blot. In addition to cell growth, protection of hHSS on the cells against H\-2O\-2 was also observed. Results It was showed that recombinaht HSS of 400??g/L was able to promote DNA synthesis by 21\^5% as compared to non\|treated cells. After 24?h of HSS action, the cell division measured by MTT method as well as cell\|count was significantly enhanced. Meantime, it was indicated that the phosphorylation of MAPK Thr202/Tyr204 was increased in 79\^0% as compared with that of the non\|treated cells. Pretreatment of the cells with hHSS for 12?h prior to H 2O 2 injury preserved the survival of them.Conclusion It is postulated that hHSS is an active protein to stimulate liver proliferation. [
2.Research progress in regulation roles of G protein-coupled receptors in hepatocellular carcinoma

G protein- coupled receptors (GPCRs), also known as seven- transmembrane domain receptors, constitute the largest superfamily of cell surface receptors. By coupling to heterotrimeric G proteins, arrestins and other signaling molecules, GPCRs modulate diverse signal transduction pathways under physiological and pathological conditions. Recent studies have revealed crucial roles of GPCRs in tumorigenesis and development of cancer metastasis. This review summarizes roles of GPCRs, particularly the roles of those coupled to chemokines, prostaglandin, lysophosphatidic acid, endothelin, catecholamine and angiotensin in proliferation, invasion, metastasis and angiogenesis of hepatoma cells and development of hepatocellular carcinoma. The potential of GPCRs- based therapeutics being used for hepatocellular carcinoma is also highlighted.

5.Three-dimensional finite element analysis of two and three cannulated hip screws fixation for adductive femoral neck fractures

[Objective]To explore the biomechanical stability of fixation with two and three cannulated compression screws in treatment of the adductive femoral neck fractures by the three-dimensional finite element analysis(FEA).[Method]Two FEA models of two screws and three screws for adductive femoral neck facture fixations were set up to investigate the simulated loading of three different conditions.[Result]At the condition of the heel touching the ground and the middle position,there was no difference in the biomechanical stabilities of the two models.However,at the condition of the tiptoe departing from the groud,the model of three screws fixation was better in torsion resistance and stretching resistance.[Conclusion]The biomechanical stability of fixation with two screws is determined by the direction of loading.Three cannulated compression screws fixation provides better biomechanical stability.
6.Detection of hepatitis B virus pre-C region mutation in cases of chronic hepatitis B and its relationship with IFN treatment

Objective To investigate the relationship between hepatitis B virus pre-core region 1896 site mutation in cases of chronic hepatitis B and IFN-alpha treatment.Methods 200 patients with chronic hepatitis B were randomly divided into experimental group(n=100) and control group(n=100).The patients in experimental group were treated with regular liver-protective drugs and IFN-alpha,while Control group patients received only regular liver-protective drugs for 6 months.PCR-RFLP were used to detect hepatitis B virus(HBV)DNA with pre-C region 1896 site mutation in two grops pre and post treatment.Results The mutation rates before interferon treatment were 20%(20/100) and 18%(18/100) in experimental group and control group,respectly.After treatment,the effective rates in experimental group were 60%(12/20) and 50%(40/80) in Patients with Chronic Hepatitis B variant and wild strain virus Infection,respectly.The effective rate in experimental group was significantly higher than that in control group.Conclusion IFN-alpha therapy was not correlated with genomic variability of the core region.
7.EFFECT OF GINSENOSIDES ON DIFFERENTIATION OF MOUSE ASCITES RETICULOCYTE SARCOMA ARS CELLS

Our study demonstrated that ARS cells can be induced to differentiate into macrophage-like cells with changes in phenotype, nonspecific esterase activity and phagocytic function by adding ginsenosides in short-term cultures. Synthesis of DNA, mitosis and the growth of the culture cells transplanted in mouse are also inhibited in this condition. The size of cells, nuclei and nucleoli, as well as nuclear-cytoplasmic ratio of ginsenosides treated cells are diminished significantly. Microvilli of these cells are reduced in number with formation of. ruffles on the cell surface. Mitochondria are increased, their size and distribution become regular. The fact that numerous small cells, induced by ginsenosides exhibit the most conspicuous alteration mentioned above along with marked phagocytic activity indicates that they are highly differentiated macrophage-like cells. Whether the inhibition of cell growth and induction of differentiation by ginsenosides is caused through its action on the molecules regulating the gene expression of cell growth and differentiation needs further study.

9.Post-SARS osteonecrosis and anticardiolipin antibodies

[Objective]To explore the correlation between anticardiolipin antibodies and post-SARS patients with osteonecrosis,investigate the etiology of post-SARS osteonecrosis,and select the sensitive molecular symbols for early diagnosis and distinguish the high risk population.[Method]The studied subjects were divided into two groups.Sixty-two post-SARS patients with osteonecrosis was one group,and 52 matched healthy people was as normal controlled group.Empty stomach blood samples from cubital veins were collected from both groups.IgA,IgG and IgM of anticardiolipin antibodies types were examined by ELISA.[Result]Twenty-one of 62 post-SARS osteonecrosis patients were tested at least one type of anticardiolipin antibodies positive,among them,with IgA(11.33?11.209)APL,IgG(5.127?5.927)GPL,IGM(17.821?10.606)MPL.Four of 52 control group persons were detected positive,with IGA(10.702?3.126)APL,IgG(5.184?4.780)GPL,IgM(14.684?5.516)MPL.There were significant differences between two groups,while,there were no difference in prothrombin time(PT),thrombin time(TT),prothrombin activity(PTA)and international normalized ratio(INR).[Conclusion]The study indicates an increased positive incidence of anticardiolipin antibodies in post-SARS patients with osteonecrosis.This has reflected that anticardiolipin antibodies may play a role in osteonecrosis.
10.Interpretation of standards on training and evaluation of urine analyzers in grass-roots medical unit

Objective To analyze the training and evaluation standards on urine analyzers in the grass-roots medical unit.Methods The applicability,requirements,subject setup and items of the standards as well as the requirements of evaluation scoring chart were expounded in detail,and the related elementary knowledge was analyzed including the structure,working principle,installation and adjustment,operation,application,maintenance and etc of the analyzer.Combined personnel with the equipment was suggested to ensure the accuracy and high efficiency of the test result.Results The interpretation of the standards contributed to enhancing the quality of medical equipment training and the ability of practical support,and facilitated the examiner to perform annual evaluation with the scoring chart.Conclusion It is necessary to grasp accurately the connotation of the standards,to improve the quality of medical equipment training and to enhance combat support capability for the grass-roots medical unit and examiner.
