1.Establishment of a sandwich ELISA method for measuring soluble IL-2 receptor

Two kinds of mouse monoclonal antibodies which recognize distinct epitopes of the IL-2 receptor (a chain) were used to establish a sandwich ELISA for measuring soluble IL-2 receptors. The results indicated that this method is sensitive and specific for detecting soluble IL-2 receptors of cell culture supernatant and serum from both healthy donor and patients with leukemia. Therefore this sandwich ELISA method is useful for basic and clinic immunology research.
2.Studies on the requirements for growth of Epstein-Barr Virus thransformed human lymphoblastoid B cells at low cell density

We obtained human lymphoblastoid B cells secreting human antibodies IgM and IgG to HFRS virus by Epstein-Barr virus(EBV)transfomation of PBMC.Conditioned media from the supernatant of a human fibroblast cell CRL1506 culture and irradiated CRL1506 cells as feeder cells could markedly promote the growth and antibody produotion of the human B lymphoblastoid cells at low cell density.It was shown that conditioned media contained a high level of interleukin 6(IL-6) .These results would be very useful for human monoclonal antibody technique including improving human B lymphoblastoid cell growth at low cell density and enhancing cloning efficiency andantibody production.

To study the changes in vestibular function in Parkinson's disease patients, and its significance, eletronystagmography (ENG) was used to examine 54 patients with Parkinson's disease, and also 20 hea thy elderly subjects. The results of vestibular function tests including sensory organization test, nystagmus test, cold caloric test and positional test of these two groups were recorded for statisticed analysis. It was found that the rate of vestibular dysfunction in Parkinson's disease patients were 81 4%, and that of the controls 30%. There was significant difference between the two groups ( P

5.Single nucleotide polymorphism in studying complex diseases susceptibility gene

Most diseases are complex genetic traits caused by multiple genetic and environmental components. It has been proposed that common genetic variations, mainly single nucleotide polymorphisms (SNPs), influence the susceptibility to complex diseases. We have conducted an extensive review on the characters of SNPs, the related website information, and the genotyping methods of SNPs such as direct sequencing, SnaPshot, Taqman, real time quantitative (kinetic) PCR with allele specific amplification, denaturing high performance liquid chromatography, and OLA/PCR.The strategies for studying the relation between SNPs and complex diseases susceptible genes were also reviewed.
6.Study on quality standard for Jianweishu Granule

Objective: To establish the assay methods for paeoniflorin and baicalin in Jianweishu Granules (Radix Scutellariae, Radix Paeoniae Alba, Radix Astragali seu Hedysari, Radix Notoginseng, etc.). Methods: Radix Astragali, Radix Notoginseng; Radix Paeoniae Alba and Herba Dendrobii in Jianweishu Granule were identified by TLC. The paeoniflorin and baicalin in Jianweishu Granule were determined by HPLC on shim pack CLC ODS C 18 Column (6.0mm?150mm,10?m); using methanol water phosphoric acid (47∶53∶0.2) and methanol water (30∶70) as mobile phases, respectively; the flow rate were at 1.0 mL?min -1 ; the detection wavelength were 280nm and 230nm, respectively; the column temperature were at room temperature and 40?C respectively. Results: The average recoveries are 98.94% and 98.67%. RSD are 2.31% ( n =5) and 1.36% ( n =5). Conclusion: The methods are simple, sensitive, accurate and reproducible.
7.Effects of domestic recombinant adenovirus p53 injection combined with cisplatin on growth and apoptosis of ovarian carcinoma cell lines

Objective:To evaluate the effects of domestic recombinant adenovirus p53 injection (rAd-p53,Gendicine) combined with cisplatin (DDP) on growth and apoptosis of ovarian carcinoma cell lines. Methods:MTT assay, flow cytometry, and Western blotting were used to compare the effects of rAd-p53, DDP and their combination on the growth, cell cycle, apoptotic rate, and p53 protein expression of ovarian carcinoma cell lines. Results:rAd-p53 inhibited ovarian carcinoma cells proliferation in dosE- and timE-dependent manners. After combined treatment, the growth inhibition rate was significantly increased (P<0.01). There was no significant diffE-rence in the inhibition potency induced by combined treatment with different sequences (P>0.05). Combined treatment significantly arrested ovarian carcinoma cell lines in G0/G1 phase at 72 h, the cell number in S phase was significantly decreased and the apototic rate was significantly increased (P<0.01). High expression of p53 protein was detected in ovarian carcinoma cell lines at 72 h after administeration of rAd-p53. Conclusion:rAd-p53 tranfects the exogenous widE-type p53 into the genome group of human ovarian tumor cells, induces p53 protein expression, thereby arresting tumor cells at G0/G1 phase, inhibiting tumor growth, and inducing apoptosis.

9.Risk factor analysis on early postoperative complications after pancreaticoduodenectomy

ObjectiveTo investigate the risk factors of early postoperative complications after pancreaticoduodenectomy.MethodsThe clinical data of 159 patients undergoing pancreaticoduodenectomy from March 2005 to October 2010 was analyzed retrospectively.The related factors of early postoperative complications were analyzed.ResultsThe incidence rate of early postoperative complications was 30.8% (49/159).Univariate analysis showed that preoperative fasting blood glucose level,total serum bilirubin level,serum albumin level,high density lipoproteins level and intraoperative bleeding were closely correlated with early postoperative complications in patients undergoing pancreaticoduodenectomy (P < 0.05),but gender,age,preoperative jaundice,history of diabetes mellitus,operation time and whether portal vein reconstruction were no significantly associated with early postoperative complications in patients undergoing pancreaticoduodenectomy (P > 0.05).Multivariate analysis showed that preoperative fasting blood glucose level( ≥6.1 mmol/L),total serum bilirubin level ( ≥ 171 μ mol/L) and serum albumin level( < 35 g/L) were the important independent risk factors of early postoperative complications in patients undergoing pancreaticoduodenectomy.ConclusionThe risk factors of pancreaticoduodenectomy should be fully recognized and dealt with promptly.
10.The change of lung lymph after endotoxin-induced lung injury

The change of lung lymph after endotoxin-induced lung injury was studied using a sheep model with chronic lung lymph fistula. After endotoxin injection Ppa、PMV、 Q_L、CL_p、L_A/P_A、and Kf all increased markedly. The ratios of effects of pressure and permeability on Q_L were 1:0.53 and 1:2.31 during phase I and phase H respectively. The results indicated that the formation of pulmonary edema after injury related mainly to the increases of PMV and Kf and to the decrease of effective colloid osmotic pressure. The PMV was the main factor of formating edema during phase I and the permeability was that during phase Ⅱ.
