1.Detection of Rpsl Gene Mutation of Mycobacterium Tuberculosis by PCR-SSCP Silver Staining

Objective:To study the value of detection of rpsl gene mutation in streptomy cin(SM)-resistance in clinical isolates of mycobacterium tuberculosis by PCR -SSCP,and to expect to set up a r apid detective method of rpsl gene mutation in mycobacterium tuberculosis,Method Eighty seven specimens isolated from patients were detected by PCR-SSCP silver staining,and using strain H 37Rv as a control,Results In 87 mycobacterium tuberculosis clinical isolates,the r psl DNA fragments from 16 drug-susceptible isolates had no mutation in rpsL gene ,48 of 62 sputum specimens showed rpsl gene mutation by PCR-SSCP,positive rate w as 77.4%.Conclusions The rpsL gene mutation is an important molecular mechanism of M.tuberc ulosis resistance to SM.PCR-SSCP might become a rapid detecting method of SM-r esistance of M.tuberculosis.
2.CLINICAL ANALYSIS OF 11 PATIENTS WITH OLFACTORY NEUROBLASTOMA

PURPOSETo analyze the role of radiotherapy in the treatment of olfactory neuroblastoma. METHODS From 1974 to 1994. 11 patients with olfactory neuroblastoma were treated in Xinjiang Cancer Hospital. All patients were confirmed histologically. Six cases were stage B and 5, stage C. Five patients were treated with surgery and postoperative radiotherapy (4. stage B and 1, stage C), and 6. with radiotherapy alone. The tumor dose ranged from 45~68 Gy, 2. 0 Gy/Fx. Neck irradiation with total dose of 60~65 Gy was carried out for 3 patients with cervical lymph node metastases. RESULTS Three patients had local recurrences and 2 cases, distant metastases. The actuarial overall survivals at 3 and 5 year were 55% and 36%, respectively. Three and five year survivals were 33%and 17% for the patients treated with radiotherapy alone, and 80% and 60% for the patients treated with surgery and postoperative irradiation, respectively(P= 0. 2727). CONCLUSION For the patients with stage B and C olfactory neuroblastoma, combined modality of surgery and radiotherapy should be the treatment of choice.

4.Inflammatory markers for short-term prognosis of acute cerebral infarction

Objective To investigate the potential of inflammatory markers for short-term prognosis of acute cerebral infarction.Methods A total of 272 consecutive patients with acute cerebral infarction were divided into a high hs-CRP group (hs-CRP level >3 mg/L) and a low hs-CRP group (hs-CRP level ≤3 mg/L),and their general information and medical history were collected.The Trial of Org 10172 in Acute Stroke Treatment (TOAST) stroke subtype classification was conducted and scores of the National Institutes of Health Stroke Scale (NIHSS),the Barthel index (BI) and the modified Rankin scale (mRS) were collected after admission.White blood cell count,blood glucose,blood homocysteine (Hcy) and C-reactive protein were measured within 24 hours following admission.Multivariate Logistic regression analysis was performed to identify independent risk factors for short-term prognosis of acute cerebral infarction.Results Between the high hs-CRP group and the low hs-CRP group,there were significant differences in the incidences of atrial fibrillation history,cardiogenic embolism of TOAST,blood homocysteine,blood glucose,white blood cell count,NIHSS,BI and mRS score 1,7,14 days after admission (P<0.05 for all).The hs-CRP level (OR=0.876,P<0.001,95% CI:0.817-0.917),white blood cell count (OR=1.137,P=0.029,95% CI:1.013-1.275),lipid metabolism disorders (OR=2.863,P<0.001,95% CI:1.561-5.250),and BI score (OR=1.038,P=0.047,95% CI:1.001-1.077) 1 day after stroke were independent risk factors for short-term prognosis of acute cerebral infarction.Conclusions Increased levels of the inflammatory marker hs-CRP and elevated white blood cell count may be independent risk factors for short-term prognosis of acute cerebral infarction.
6.Expression of activating transcription factor 4 in osteogenic induction of bone marrow mesenchymal stem cells

BACKGROUND:Activating transcription factor 4 is found as an activating factor that can regulate osteogenic differentiation and function, and plays a critical role in the osteogenic differentiation.
OBJECTIVE:To investigate the expression and significance of activating transcription factor 4 in the osteogenic differentiation of bone marrow mesenchymal stem cells from Sprague-Dawley rats.
METHODS:Bone marrow mesenchymal stem cells from Sprague-Dawley rats were cultured using the whole bone marrow adherence method, and ossification revulsant was added to induce passage 3 cells. cells with no osteogenic induction served as controls. RT-PCR and western blot assay were employed to dynamical y monitor expression of activating transcription factor 4.
RESULTS AND CONCLUSION:RT-PCR results showed that activating transcription factor 4 mRNA expression increased with the increasing osteogenic differentiation, and peaked at day 16. Western blot analysis showed that the protein expression of activating transcription factor 4 tended to increase with the increasing osteogenic differentiation, peaked at day 16 and stil maintained at a higher level at day 19. Compared with the uninduced cells, activating transcription factor 4 in the induced cells exhibited a higher expression at mRNA and protein levels (P<0.05). These findings indicate that activating transcription factor 4 expression is elevated during osteogenic differentiation, showing a positive correlation with osteogenic ability of bone marrow mesenchymal stem cells.
7.Effects of immediate loadings with different forces on immediate implant-bone ossification

BACKGROUND:Animal studies showed that functional early load or immediate load on immediate implant did not affect the prognosis of implant.
OBJECTIVE:To analyze the effect of immediate loading with different forces on the binding surface of immediate implant bone ossification.
METHODS:Six adult healthy male dogs were selected. Al teeth (three premolars) between cuspid teeth of lower mandible and the first molar were pul ed out. One implant (OSSTEM GSII) was immediately implanted, total y 36 implants. The vertical immediate loading by device was used on the implants at 24 hours after implantation. Grouped by the loading force values:0, 10, 20 N on the left three implants of each dog from front to back, 30, 40, 50 N on the contralateral three implants of each dog, at a frequency of 1 Hz for 10 minutes every day. The implant stability quotient (ISQ) was tested using resonance frequency measurement instrument (OSSTELL) at 0 day, 4, 8, and 12 weeks.
RESULTS AND CONCLUSION: With increased load forces, serum osteocalcin expression increased, and peaked on 20 N, but decreased in 30 N group, and lowest in 50 N group. At 4 weeks after immediate implantation, the ISQ values were slightly less than pre-implantation in each group, especial y the 50 N group. At 8 weeks after immediate implantation, ISQ values were increased in each group to different degrees. The increased degree of the 20 N group was maximal. At 12 weeks, a peak value was detected in each experimental group. The implants could bind to bone tissues to different degrees. The range of implant-bone interface formation was positively associated with time. Results indicated that the smal force cannot impact implant-bone ossification, but promotes it in a manner, but large force value (≥ 20 N) wil affect the stability of the implant-bone ossification.
8.Evaluation of uncertainty in colony forming units on the disinfected skin of nurses in operation room

Objective To develop a feasible method for evaluating uncertainty in colony forming units(CFU)on the disinfected skin of nurses of operation room.Methods A nurse of operation room in a hospital involved the study.Ten spots on her disinfected hand skin were selected for evaluation of uncertainty in the acquired data and analysis of reliability of the evaluation methods,according to Technical Norms for Monitoring the Disinfection and Sterilization Quality in Hospitals in Jiangsu Province and JJF1059-1999 Evaluation and Expression of Uncertainty in Measurement.Results The CFU on the disinfected skin of nurse was(4±2)cfu/cm2, which indicated that the disinfection of the nurse's hands was not qualified.Conclusion The evaluation method by our study is reliable for evaluating the uncertainty in CFU on nurse’s disinfected hand skin.It is simple and fitful for the uncertainty evaluation of hospital disinfection effect.
9.Effect of Zhibai Dihuang Pill on serum level of resistin,adiponectin,leptin in patients with hyperthyroidism

AIM:To investigate the effect and the mechanism of Zhibai Dihuang Pill(Radix Rehmanniae praeparata,Fructus Corni,Rhizoma Dioscoreae,Rhizoma Anemarrhenae,Cortex phellodendri Chinensis,etc.) on patients with hyperthyroidism. METHODS: Eighty-five hyperthyroid patients were randomly divided into two groups.The control group(40 cases) were treated with propylthiouracil,while the treatment group(45 cases) were treated with propylthiouracil and Zhibai Dihuang Pill;in the 12-week long treatment period,the heart rate,body weight,thyroid free FT_3、FT_4 and TSH and the serum level of FAA,resistin,adiponectin,leptin of patients in both groups were measured. RESULTS: After treatment,the heart rate,the serum of FT_3,FT_4 decreased and the body weight,TSH increased in both groups(P0.05).(CONCLUSION): Zhibai Dihuang Pill can improve the abnormal metabolism of sugar and fat in patients with hyperthyroidism by its actions on the serum level of FFA,resistin,adiponectin,leptin.
10.The role of interleukin-2 pathway in pathogenesis of nasal polyps.

OBJECTIVE:
To study the expression of Interleukin2 (IL-2) signaling pathway related factors and Treg cell in nasal tissue of nasal polyps, so that to investigate the possible mechanism of IL-2 signaling pathway in the progress of nasal polyps and the correlation between IL-2 pathway and Treg cell.
METHOD:
Thirty patients were enrolled for study, including the patients with nasal polyps and those with only deviated nasal septum as normal control. The nasal polyps tissue and the turbinate mucosa of the patients were collected during surgery. The expression levels of IL-2 and IL-2R were measured by means of ELISA. The level of pSTAT5 was evaluated by Western blot. We measured the level of Foxp3 mRNA in the tissue by real-time PCR, and the proportion of Treg cells was evaluated by flow cytometry. Finally. we analyzed the correlation between IL-2 pathway related factors and Treg cells in nasal polyps.
RESULT:
The expression levels of IL-2. IL-2R and pSTAT5 were significantly decreased in the nasal polyps compared with normal control (P < 0.05), and the level of Foxp3 mRNA and proportion of Treg cells in patients with nasal polyps were significantly lower than in normal control (P < 0.05). Moreover, there was positive correlation between the levels of IL-2 pathway related factors and the levels of Foxp3 mRNA and Treg cells proportion in nasal polyps.
CONCLUSION
The activated state of IL-2 signaling pathway got changed in nasal polyps tissue, the level of which was positively correlated with the expression of Treg cells, indicating that the IL-2 signaling pathway may play a crucial role in the development of nasal polyps, and the decreased level of Treg cells in nasal polyps may result from the downregulation of IL-2 signaling pathway.
