1.Optimization of conus genomic DNA isolation methods

Conopeptides (Conotoxin) derived from tropical marine gastropod, cone snail ( Conus ) venoms have been an useful tool agent in neuro investigation and new drug source. Recently, new method of Conotoxin genes cloning from Conus genomic DNA has been established. So to get Conus genomic DNAs rapidly is the basis for diverse Conotoxin gene isolation, gene bank construction and the medical source exploitation. In this report, different DNA extractions from different tissue and organs of six cone snail species were performed in order to optimize the DNA isolation methods. The optimized brief phenol SDS method was established which fit for cone snails genomic DNA extraction.
2.Clinical study of two kinds of different invasive arterial blood pressure flushing liquid

Objective To discuss the clinical effect of normal saline flushing and heparinized saline flushing in invasive arterial blood pressure monitoring in the critical patients. Methods A total of 140 critical patients were randomly divided into the experimental group and the control group with 70 cases in each group according to the envelope method. The experimental group used normal saline for tube flushing, the control group used heparinized saline (5 U/ml) for tube flushing. The incidence of complications and coagulation index were compared between two groups. Results There was no statistically difference in the incidence of the catheter jam and local infection (P>0.05). The incidence of bleeding events was 14.3% (10/70) in the control group, which was higher than 2.9% (2/70) of the experimental group. The difference was statistically significant(χ2=4.466, P<0.05). The activated partial thromboplastin time, prothrombin time of the control group were (32.22 ± 4.22) s, (11.56 ± 2.12) s, which were longer than (29.84 ± 3.64) s, (10.66 ± 1.48) s of the experimental group. And international normalized ratio of the control group was 1.12±0.14, which was higher than 1.03±0.20 of the experimental group. The difference was statistically significant (t values were-3.573,-2.912,-3.084, P <0.05). Conclusions The normal saline flushing liquid was more safer and reliable in the invasive arterial blood pressure monitoring.
3.Nursing management in the treatment of large number of infants with urinary calculi

To explore the measures of nursing management in the treatment of large number of infants with urinary calculi. The nursing management measures included launching the preparedness and response project for sudden public health events, formulating scientific and standardized nursing management system,optimizing work flow,strengthening nurse training,focusing on the details in nursing management,implementing disinfection and isolation system seriously,and paying close attention to health education for the parents of minority infants. Scientific nursing management can ensure the treatment effectiveness and nursing safety for the infants with urinary calculi.
4.Association research of nonalcoholic fatty liver disease with waist, hip circumference and body mass index

Objective To analyze the correlation of nonalcoholic fatty liver disease (NAFLD) with waist,hip circumference and body mass index in order to explore the prevention countermeasures.Methods The datum of routine physical examination and questionnaire survey among 2 503 employees of 12 enterprises in November 2013 were collected.The indexes of height,weight,waist circumference,hip circumference,blood pressure,blood lipid and blood glucose of the subjects were measured.The grouping was according to whether the subjects suffering from NAFLID.The correlation and epidemiological characteristics between each group and the risk factors of body weight,waist,hip circumference and body mass index were analyzed.Results 2 503 subjects were collected including 490 NAFLID patients (19.57%).The body weight and body mass index of NAFLID patients were significantly lower than those of the control group.The result of BMI classification showed that the subjects of the control group were overweight while the subjects with NAFLID were obesity.The waist circumference and hip circumference of NAFLID patients were significantly larger than that of the control group.Conclusions Larger waist and hip circumference and overweight are risk factors of NAFLID.Effective intervention measures,scientific control of body weight,rational diet,the strengthening of physical exercises should be taken in order to prevent and control the development of fatty liver.
5.Conotoxin RNA isolation and its cDNA synthesis

Two methods of different RNA extractions from Conus venom tube and toxin gland of seven cone snail species were performed. ThermoScript~ TM RNase H~ - Transcriptase and AMV Transcriptase were used to synthesize cDNA separately. The cDNAs were used as templates to amplify conotoxin genes by PCR.The optimized RNA isolation and cDNA synthesis methods were obtained according to RT-PCR results, which would be the basis for cloning of new conotoxin genes.
6.The effects of experimental periodontitis on serum C reactive protein and insulin resistance in obese rats

Objective:To investigate the effects of experimental periodontitis on serum C reactive protein(CRP) and insulin resistance in obese rats.Methods:4 week old SD rats (n=35)were fed with high fat diet for 16 weeks to establish the model of obesity.According to the obesity criteria,5 rats were screened out.According to the proportion of 1:2,10 rats were randomly selected as obese control group,20 obese rats were used for the establishment of the obesity with periodontitis model(combination group) by periodontal ligation using silk thread.4 rats died after ligation.Before periodontal ligation,1 and 4 weeks after periodontal ligation,orbital venous blood were collected for the detection of fasting blood glucose and fasting insulin.Homeostasis model assessments of insulin resistance index(HOMA-IR) and beta cell function index(HOMA-β) were calculated.The level of serum CRP was examined by ELISA.Results:The level of CRP in the combination group was increased 1 week after periodontal ligation,and then gradually decreased in the following 4 weeks(F=7.773,P=0.004).HOMA-IR in the combination group were higher than that in the obese group(F=-4.691,P=0.000),and beta cell function index was significantly lower(F=3.672,P=0.002) than that in the control group 4 weeks after periodontal ligation.Conclusion:The experimental periodontitis may affect the serum level of CRP in obese rats,aggravate insulin resistance and decrease the function of beta cells in obese rats.
7.Study on bacteria contamination during the collection, processing and storage of umbilical cord blood

ObjectiveTo evaluate bacteria contamination during collection,processing and storage of cord blood to gain insight into contamination mechanism and direct prevention.MethodsFresh cord blood was separated by hydroxyethyl starch (HES) to harvest nucleated cells.The bacteria contamination was tested by culturing 10 ml plasma-red cells with BacT/ALERT 3D-480 automatic blood culture system.Total 87 positive samples were further identified for bacteria species.Ninety six cord blood nucleated cells concentrate with bacteria positive stored in liquid nitrogen(LN2) for 6-7 years were thawed at 37 C and re-cultured for bacteria analysis.ResultsWe collected 19 062 umbilical cord blood.Among them,336 was bacteria positive ( contamination rate 1.8 % ).Eighty-seven positive samples were further investigated with facultative bacteria 58 (66.7 % ),aerobic 38(43.7% ) and anaerobic 17( 19.5% ),Gram- negative accounted for 68% while positive 32%.The most frequent bacteria were Escherichia coli ( 25.3% ),Streptacoccus intermediate ( 14.9% ) and Chromobacteria violaceum(9.2% ).Ninety-six nucleated cells concentrate with bacteria positive were cryopreserved at liquid nitrogen for researching.Of them,83 samples( 86% ) showed positive of bacteria culture after deep-low temperature storage for 6-7 years.ConclusionsBacteria contamination rate of the cord blood collection,processing and storage in 2000 ~ 2007 was 1.8%.Stored in liquid nitrogen for 6-7 years,the viability of bacteria was 86%.The aseptic procedures of cord blood collection in delivery room should be intensified.The bacteria re-culture following thawing of cord blood cells is necessary before clinical transfusion.
8.Inhibitor-based methods for detection of derepressed hyperproduction and plasmid-mediated AmpC β-lactamases

Objective To investigate the reliability of using inhibitors including Phenylboronic acid (PBA)and Fqucloxacillin(FCC)in detecting derepressed hyperproduction and plasmid-mediated AmpC B-lactamases.Methods PBA and FCC were chosen as inhibitors and double-disk potentiation method and double-disk synergy method were used to detect positive and negative control strains of AmpC β-lactamases and 107 clinical isolates for AmpC β-lactamases production.The positive control strains included E.cloacae (029M),plasmid-mediated ACT-1 type of E.coli DH5a2919,MOX-1 type of k pheumoniae,LAT-2 type of E.coil.The negative control strains included E.cloacae 029(wild-type),E.coli SHV-1,E.coli SHV-2, E.coil SHV-5,E.coli TEM-1,E.coli TEM-3,k peumoniae SHV-18 and E.coli ATCC25922.We compared the results above with the three dimensional test(3-DT)to observe the accuracy in detecting AmpC-BLA.Results 3-DT together with PBA and FCC based inhibition tests showed the 4 positive control strains and the 9 negative control strains were determined as expected.AmpC-BILA was detected in 107 clinical isolates ofEnterobacteriaceaes.The positive rate of3-DTmethod is24.3%.The positive rates ofPBA.FCC double-disk potentiation method and double-disk synergy method are 30.8%(33/107),26.2%(28/107) and 23.4%(25/107),respectively.The conjugate results in two strains of P mirabilis and one strain of K.peumoniae were positive.They were all plasmid-mediated AmpC-Bi.A.There Was a higher false positive when using PBA and FCC-based double-disk potentiation method to detect the induction type of AmpC-BLA, but the accuracy of double-disk synergy method was high.Compared with the 3-DT,the coincidence rate using PBA and FCC-based double-disk synergy method is 99.1%.Conclusions Using PBA and FCC as inhibitors in the double-disk synergy test is a accurate and reliable method to detect AmpC-BLA regardless of derepressed hyperproduction type or plasmid-mediated type.
10.A preliminary study of mechanism of periodontitis and periodontal therapy influence in insulin resistance for obesity rats

Objective To explore the mechanism of the experimental periodontitis and initial periodontal therapy on insulin resistance of obese rats associated with periodontitis.Methods Blood samples were collected from SD rats of obese group,periodontitis associated with obese group,periodontal therapy group and control group.Body index,data about insulin resistance and Observed pathological changes were calculated.Results Comparison of four groups:more severe insulin resistance and islet structural dysfunction was observed in obesity with periodontitis group than that of group with obesity alone.After periodontal therapy,inflammation and insulin resistance may be improved.Results from pathological specimens also supported this result.Conclusions The periodontitis with obesity can enhance the inflammatory state of obesity and cause the deterioration of insulin resistance.The initial periodontal treatment can relieve the inflammation of obesity with periodontitis and improve the resistance of insulin.It may be the reason of insulin resistance when obesity with periodontitis that the signaling pathways of IRS-1 and IRS-2 blocked,and PDX-1,GK,GLUT and other protein expression inhibited.
