1.Efficacy of cryoablation combined with zoledronic acid sequential therapy for bone metastases pain

Objective To investigate the clinical efficacy and safety of cryoablation combined with zoledronic acid sequential therapy for bone metastases pain. Methods Twenty-four patients from our depart-ment suffering from malignant tumors with moderate to severe pain due to bone metastases were enrolled in this study. Those patients with 28 metastatic bone tumors were successfully treated with cryoablation operations under CT-guided. Three days after cryoablation they were offered zoledronic acid(4 mg added in 100 ml normal saline,dripping for more than 15 min),once 4 weeks. Pain level and life quality were respectively evaluated by NRS(Numerical rating scale)and KPS(Karnofsky performance status),before cryoablation,3 days after cryoablation,2 weeks and 12 weeks after zoledronic acid sequential therapy. Results Without serious compli-cations,all of 24 patients were successfully punctured to tumor lesions. Patients’pain scores were 7. 70 ± 0. 86 (before cryoablation),3. 29 ± 0. 95(3 d after cryoablation),2. 54 ± 0. 83(2 weeks after zoledronic acid sequential therapy)and 2. 17 ± 0. 76(12 weeks after zoledronic acid sequential therapy),with a significant sta-tistical difference between pretherapy and post-treatment(F = 530. 64,P < 0. 001);during observation period after treatment,the effective rate in pain was 91. 67% . KPS scores were 45. 83 ± 6. 54(before cryoablation), 49. 58 ± 6. 24(3 d after cryoablation),61. 67 ± 7. 01(2 weeks after zoledronic acid sequential therapy)and 78. 33 ± 8. 68(12 weeks after zoledronic acid sequential therapy),with a significant statistical difference between pretherapy and post-treatment(F = 418. 99,P < 0. 001);during observation period after treatment, the effective rate in KPS was 75. 00% . Six months after the treatment,there were 10 cases of CR,11 cases of PR,and the effective rate was 87. 50% . Conclusion Cryoablation combined with zoledronic acid may be an effective therapeutic method with good safety in the treatment of patients with bone metastases pain.
2.Risk factors for pregnancy-induced hypertension in Tangshan city

Objective To investigate the risk factors for pregnancy-induced hypertension in Tangshan city.Methods Ninety-five pregnant women with hypertension were involved in questionnaire survey with contents ineluding general states of health and histories of past illness. Results Pregnancy-induced hypertension were more seen in winter, women aged above 35 years old with blood type A, bad educational background, bad nutritional status, hypertension history for family or herself, diabetes history and ere (P<0.01 ). Conclusion There are ages, blood types, educational background, nutritional status, hypertension history, diabetes history for the risk factors for pregnancy-induced hypertension.
3.Correlation between wheezing and serum 25-hydroxy vitamin D3 levels

Objective To investigate the relationship between wheezing and serum levels of 25-hydroxyvitamin D3.Methods 130 children with lower respiratory tract infection were selected as the subjects, the children with wheezing in group A([Abstract] Objective To investigate the relationship between wheezing and serum levels of 25-hydroxyvitamin D3.Methods 130 children with lower respiratory tract infection were selected as the subjects, the children with wheezing in group A(n=70),normal pneumonia (no wheezing) were group B(n=60); The healthy children who were examined in the hospital at the same period were selected as the control group (n=60). The serum levels of 25-hydroxyvitamin D3 were measured by ELISA in the control and hospitalized children. The pathogens and allergens of the two groups were also detected. Results The levels of serum 25-hydroxyvitamin D3 in group A and group B were (56.92±16.88) nmol/L, (70.68±21.96) nmol/L, respectively, which was significantly lower than that in control group (82.69±17.63) nmol/L, ( t=8.50, 3.30,P=0.00,0.00); compared with group A( t=3.85, P=0.00). The positive rate of group A was 65.71%, which was significantly higher than that of group B (35.00%, χ2=12.20,P=0.00). The positive rate of group A was 30.00% compared with the control group (35.00%, χ2=0.36,P=0.54). The serum level of 25-hydroxyvitamin D3 was 75.57% in group A, which was significantly higher than that in group B (60.00%,χ2=14.21, P=0.00). The positive rate of virus detection was 57.14% in 98 children with serum 25-hydroxy vitamin D3 level<75 nmol/L, which was significantly higher than that in serum 25-hydroxy vitamin D3 level≥75 nmol/L(18.75%, χ2=14.25, P=0.00). The positive rate of allergens in serum 25-hydroxy vitamin D3 level <75 nmol/L was 35.71%, which was significantly higher than that in serum 25-hydroxy vitamin D3 level≥75 nmol/L(21.88%, χ2=2.11, P=0.14). Conclusion The main risk factor for children with wheezing is viral infection, while the low level of serum 25-hydroxy vitamin D3 in children increases the risk of viral infection, resulting in increased risk of wheezing in patients,so the clinical can occur through the detection of serum 25- hydroxyvitamin D3 levels to predict and intervention for children with wheezing.
4.Comparative study on the diagnostic effect of combined detection of plasma cleavage protein 9 and multi ligand proteoglycan 2 precursor methylation and four serum tumor markers for colorectal cancer

Objective:To compare the difference between the combined diagnostic effect of plasma Septin9(SEPT9) and polyligand Syndecan-2(SDC2) methylation with four serum tumor markers in the diagnosis of colorectal cancer.Methods:In this study, 128 patients who were treated in the affiliated Hospital of Xuzhou Medical University from March to December in 2019 were selected for a case-control study.All the subjects were examined by gastroenteroscopy.According to the pathological results, they were divided into three groups: colorectal cancer group( n=74) and colorectal adenoma group( n=7). The patients with no abnormal or inflammatory polyps or proliferative polyps examined by gastroenteroscopy were taken as the control group( n=47). The methylation levels of SEPT9 gene and SDC2 gene were detected by Roche Lightcycler 480 II real-time fluorescence quantitative polymerase chain reaction, and the concentrations of alpha-fetoprotein, carcinoembryonic antigen, carbohydrate antigen 125 and carbohydrate antigen 199 were detected by Roche Cobas 8000 electrochemiluminescence instrument.Chi-square test was used to compare the positive rate of each marker in the three groups.Medcalc was used to draw the receiver operating characteristic curve (ROC) curve of the subjects′ working characteristic curve, and the value of each index in the diagnosis of colorectal cancer was analyzed. Results:The positive rates of SEPT9 gene and SDC2 gene methylation were 81.1%(60/74) and 67.6%(50/74) respectively in colorectal cancer group, and increased to 85.1%(63/74) after combined detection.The positive detection rates of alpha-fetoprotein, carcinoembryonic antigen, carbohydrate antigen 125 and carbohydrate antigen 199 in colorectal cancer group were 1.4%(1/74), 33.8%(25/74), 6.8%(5/74) and 13.5%(10/74), respectively.When the four tumor markers were detected together, the positive detection rates were only increased to 43.2%(32/74), except for AFP and carbohydrate antigen 125(χ 2=3.847, 2.430, all P>0.05). The differences were statistically significant (χ 2=48.230, 30.487, 43.285, 3.847, 8.788, 6.988, 8.722, all P<0.05). The area under the curve (AUC) of SEPT9 methylation, SDC2 methylation, alpha fetoprotein, carcinoembryonic antigen, carbohydrate antigen 125 and carbohydrate antigen 199 were 0.854 (0.781, 0.910), 0.795 (0.715, 0.861), 0.575 (0.485, 0.662), 0.685 (0.597, 0.764), 0.603 (0.513, 0.689) and 0.631 (0.541, 0.715), respectively.The AUC of combined detection of two DNA methylation markers was better than that of alpha fetoprotein, carcinoembryonic antigen, carbohydrate antigen 125 and carbohydrate antigen 199, and the differences were statistically significant (alpha fetoprotein: Z=4.990, P<0.001; carcinoembryonic antigen: Z=3.743, P<0.001; carbohydrate antigen 125: Z=4.951, P<0.001; carbohydrate antigen 199: Z=3.983, P<0.001). The combined detection of two kinds of gene methylation was better than the combined detection of four kinds of serum markers in the diagnosis of colorectal cancer, and the difference was statistically significant ( Z=3.334, P<0.001). Conclusion:The combined detection of SEPT9 gene and SDC2 gene methylation in plasma is more suitable for non-invasive diagnosis of colorectal cancer than the combined detection of 4 serum tumor markers.

The Szent-Gy?rgyi Prize for Progress in Cancer Research is a prestigious scientific award sponsored by the National Foundation for Cancer Research (NFCR)—a leading cancer research charitable organization in the United States that supports innovative cancer research globally with the ultimate goal to cure cancer. The coveted Szent-Gy?rgyi Prize annually honors a scientist whose seminal discovery or body of work has resulted in, or led toward, notable contribu-tions to cancer prevention, diagnosis, or treatment; and the discovery has had a high direct impact of saving people's lives. In addition, the prize promotes public awareness of the importance of basic cancer research and encourages the sustained investment needed to accelerate the translation of these research discoveries into new cancer treat-ments. In 2016, NFCR's Szent-Gy?rgyi Prize Selection Committee was unanimous in its decision to recognize an icon in human disease genetics, Dr. Mary-Claire King, for her pioneering research that demonstrated the first evidence of genetic predisposition to breast cancer. Her proof of existence of BRCA1 gene and its location has made genetic screening for breast and ovarian cancers possible, saving lives of many people who are at high risk with inherited BRCA1 mutations.
6.Study on detection value of noninvasive cerebral hemodynamics and serum nerve function indexes in patients with craniocerebral injury

Objective To study and investigate the detection value of noninvasive cerebral hemodynamics and serum nerve func‐tion indexes in the patients with craniocerebral injury .Methods Totally 64 patients with craniocerebral injury in our hospital from September 2013 to May 2015 were selected as the observation group ,meanwhile 64 healthy persons with the same age were selected as the control group .Then the noninvasive cerebral hemodynamics and serum nerve function indexes were compared between the two groups ,furthermore the detection results in the observation group were compared among the patients with different severity de‐grees and intracranial pressures .Results The middle cerebral artery blood flow indexes of the observation group were all higher than those of the control group ,the serum nerve function indexes were also higher than those of the control group ,and the middle cerebral artery blood flow indexes and serum nerve function indexes of the observation group had obvious differences among the pa‐tients with different severity degrees and intracranial pressures too (P< 0 .05) ,showing statistical significance .Conclusion The noninvasive cerebral hemodynamics and serum nerve function indexes have highe detection value in the patients with craniocerebral injury ,and have active clinical role for the understand of disease severity degree and intracranial pressure situation .
7.Optimization of Preparation Technology of Stachydrine in Fructus Leonuri Based on Back-propagation Neural Network and Genetic Algorithm

Objective To optimize the preparative procedure for stachydrine in Fructus Leonuri. Methods The preparation was screened by orthogonal experiment, and a mathematical model of relationship of extraction time, methanol concentration, and solid-liquid ratio with the content of stachydrine hydrochloride was established by using back-propagation (BP) neural network. And the process parameters were optimized with genetic algorithm (GA) . Results The optimum process parameters were as follows: extraction with 69% of methanol concentration and with solid-liquid ratio being 11 times for 62 min. The content of stachydrine obtained by BP neural network modeling and GA was higher than that achieved by orthogonal experiment. Conclusion The optimum preparative procedure could be achieved by combining BP modeling with GA. The model developed in this study was proved to be predictable and feasible for the optimization of process parameters of multi-dimension nonlinear system.

9.Oxidative stress-activated JNK-MAPK signaling pathway is involved in fluctuant high glucose-induced injury of hepatocytes

[ ABSTRACT] AIM:To explore the mechanisms of fluctuant high blood glucose-induced apoptosis of hepatocytes. METHODS:SD rats were randomly divided into normal control group ( N) , stable high blood glucose group ( S) , fluctu-ant high blood glucose group ( F) and insulin group ( I) .Diabetic rats were induced by intraperitoneal injection of strepto-zotocin (65 mg/kg) , and the fluctuant high blood glucose animal model was induced by intraperitoneal injection of ordinary insulin and glucose at different time points every day.The blood glucose fluctuation patterns of the animals in F group with-in 12 weeks were similar every day and no significant difference of the HbA1c concentration was observed compared with S group, indicating that the fluctuant hyperglycemia was successfully established in F group.The activity of superoxide dis-mutase ( SOD) and glutathione peroxidase ( GSH-Px) , and the content of malondialdehyde ( MDA) and nitric oxide ( NO) in the homogenate of the liver tissues were detected by colorimetry.The mRNA and protein levels of JNK, p-JNK, Bax and Bcl-2 were examined by RT-PCR and Western blot.RESULTS:After 12 weeks, the increases in the intakes of food and water, the urine output, and the abnormal liver function were observed in S group, I group and F group.Compared with N group, the MDA level was increased, the content of NO and the activity of SOD and GSH-Px were decreased, and up-regu-lation of JNK mRNA and p-JNK and Bax proteins, and down-regulation of Bcl-2 were also found in S group, I group and F group.The above effects were more obviously showed in F group.CONCLUSION:Oxidative stress activates JNK-MAPK signaling pathway, which is involved in fluctuant high glucose-induced apoptosis of hepatocytes.

OBJECTIVETo investigate the reactivity of intrapulmonary arterial rings to vasoactive substances as thromboxane A2 and endothelin-1 in patients with chronic obstructive pulmonary disease (COPD).
METHODSIntrapulmonary arterial rings isolated from patients with normal lung function and COPD were mounted in a Multi Myograph system to determine the reactivity of the intrapulmonary arterial rings to 60 mmol/L KCl, thromboxane A2 analogue U46619 and endothelin-1 before and after preconditioning with the COX synthase inhibitor indomethacin.
RESULTSThe reactivity of intrapulmonary arterial rings to U46619 and endothelin-1 was significantly decreased in patients with COPD. The reactivity to U46619 was dramatically decreased in patients with normal lung function after application of indomethacin.
CONCLUSIONThe reactivity of intrapulmonary arterial rings is significantly decreased in patients with COPD.
