Objective:To explore the essence of Radix Morindae Officinalis in strengthening bones through observing the effect of medicated serum of Radix Morindae Officinalis on cultured osteoblast in vitro. Methods:The medicated serum of Radix Morindae Officinalis was added to the osteoblasts which were separated from the cranium of 24-hours newborn SD rat,in order to determining the effect of TGF-?1 and the proliferation and secreting of alkaline phosphatase and osteocalcin of the osteoblasts. Results:The proliferation of the osteoblasts were signifi cantly stimulated by the alcohol and the n-butanol extract from the Radix Morindae Officinalis; and the n-butanol extract from the Radix Morindae Officinalis could promote differentiating OB excreting the ALP and OC;in addition, the ability of the osteoblasts expressing TGF-?1 was promoted by the n-butanol extract from the Radix Morindae Officinalis. Conclusion:The n-butanol extract from the Radix Morindae Officinalis can promote the ability of the osteoblast in expressing TGF-?1 mRNA and excreting the ALP,and further promote the growth of bone.

Objective To investigate the correlations of FcεRI-βgene polymorphisms with clinical manifestation and prognosis inwheezing infants. Methods One hundred and forty-six wheezing infants were recruited and divided into two groups by FcεRI-βdetection using Fluorescent quantitative PCR: Risk genotype group (n = 41) or normal genotype group (n = 105). The genotype distributions, clinical manifestation and asthma, and morbidity were analyzed and compared between the two groups. Results FcεR1 E237G AG/GG was more serious than FcεR1 AA in wheezing infants. (χ2 = 14.202; P = 0.003). No significant differences were found in AS morbidity between the two groups after two years follow-up (χ2 = 2.25;P = 0.13). Conclusion FcεRI-βgene polymorphisms are strongly related with infantile wheezing. Th risk genotype may be the severity of asthma but may not be the major influencing factor of asthma.

Objective To observe the inhibitory effect of pyridoxine hydrochloride (PN) combined with common chemotherapeutics on mice hepatoma cells H22 in vitro. Methods MTT assay was used to determine the effects of PN in combination with 10 different antineoplastic agents on H22 cells, and immuno-histochemistry was used to observe the distribution of PN in H22 cells and morphologic changes of the cells before and after PN treatment. Results After 24 hours incubation with 5 mmol/L PN, the treated cells expanded apparently with nucleus chipping. PN entered the tumor cell and was mainly condensed in cytoplasma and H22 cells were sensitive to PN. When administered concomitantly with chemotherapic agents, most of the combinations showed antagonistic effects while a few of the combinations were additive. For instance, doxorubicin (ADM) used in combination with PN inhibited cell proliferation with an IR value (IR=0.63) much lower than ADM alone (IR=0.71, P<0.01), and the CI value was less than 0.9, which indicated an antagonistic effect. However, PN in combination with ifosfamide (ICTX) showed additive effect (CI>0.9), and the IR value (IR=0.60) in combined group was higher than that (IR=0.40) in ICTX group (P<0.05). Conclusion PN treatment could increase the intracellular PLP level and result in growth inhibition and cell death, and combined administration of PN and ICTX might be a potential method to improve efficacy and to reduce toxic effects while a co-administration of PN and ADM should be avoided.

Chinese Journal of Digestive Endoscopy 2021;38(1):38-42
Objective:To explore the effects of endoscopic reprocessing on disinfection and its influential factors under the coronavirus disease 2019 (COVID-19) pandemic. Methods:A total of 450 endoscopes cleaned and disinfected according to Technical Specifications for Cleaning and Disinfection of Endoscopes from November 2019 to January 2020, and 450 endoscopes cleaned and disinfected according to The recommended procedure for cleaning and disinfection of gastrointestinal endoscopes during COVID-19 epidemic by Chinese Society of Digestive Endoscopology from February to April 2020 in the Second Affiliated Hospital of Chongqing Medical University were enrolled in the control group and observation group respectively by random number method. Both the control group and the observation group contained 200 gastroscopes, 200 enteroscopes and 50 ultrasound endoscopes. ATP fluorescence detection method and pour plate technique were used to evaluate the disinfection effect of endoscopes. Single factor analysis and multiple logistic regression were used to analyze the risk factors for unqualified sterilization after endoscopic reprocessing. Results:The disinfection pass rates of gastroscopes, enteroscopes and ultrasound endoscopes in the observation group were not significantly different compared with those of the control group ( P>0.05). The sterilization pass rates and ATP test pass rates of gastroscopes, enteroscopes and ultrasound endoscopes in the observation group were significantly higher than those in the control group (all P<0.05). Multivariate logistic regression analysis showed that non-strict implementation of endoscopic reprocessing ( OR=7.96, 95% CI: 4.55-22.84, P<0.001), non-standard operation ( OR=2.26, 95% CI: 1.24-5.63, P<0.001), insufficient concentration of disinfectant ( OR=5.43, 95% CI: 2.52-9.02, P<0.001), insufficient concentration ratio of multi-enzyme solution ( OR=4.38, 95% CI: 1.95-8.61, P<0.001), non-timely cleaning ( OR=2.86, 95% CI: 1.33-6.42, P<0.001), incomplete cleaning ( OR=3.75, 95% CI: 1.61-7.49, P<0.001) and improper endoscopic preservation ( OR=2.12, 95% CI: 1.36-4.12, P<0.001) were independent risk factors for unqualified sterilization after endoscopic reprocessing. Conclusion:In COVID-19 pandemic, endoscope reprocessing can significantly improve the disinfection effect of endoscopes, worthy of further clinical promotion. The failure to strictly implement the reprocessing procedure is an important factor that may lead to unqualified sterilization.
