1.STUDY ON THE REACTION OF CHITOSAN WITH CARRAGEENAN IN SOLUTION AND THE CHARACTERISTCS OF THE COMPLEX

Solutions of chitosan and carrageenan were co-reacted under various experimental conditions to form water-insoluble precipitates. DR. spectroscopic studies and the counter -ions analyses revealed that the combination of the polyions proceeded through the electrostatic interaction between the - NH+3 of chitosan and the - SO3 of carrageenan, and the mole retios of N/S in the poly-electrolyte complexes (PEC) thus prepared have large scope depending on pH, the degree of deacetyla-tion (DD) of chitosan as well as the mixin ratio of the complexing components. The observation of the PEC by scanning electron microscopy showed that there exist fibrous structures in some PEC depending on such reactive conditions as pH, DD of chitosan related with the charge densities of chitosan chains. The solubility of PEC, which is greatly affected by the DD of chitosans, and the blood compatibility have also been studied.
2.Pharmacological Study on the Function of "Qi-promoting, Blood-breaking, Stagnation-removing and Pain-relieving" of Zedoray Rhizome

Objective To discuss Zedoray Rhizome on the correlation between its traditional Chinese herbal nature(Property and Flavor,Cannel Tropism,Efficacy and so on)and its pharmacologic actions.Methods By searching for and summarizing the literatures on the nature and pharmacologic actions of Zedoray Rhizome recorded in CNKI,CBM,Wanfang and Vip Database from 1989 to 2007,analyzed the correlation between its herbal nature and its modern pharmacologic actions.Results The efficacies of"Qi-promoting Blood-breaking and stagnation-removing" are related with the modern pharmacologic actions of resisting tumor,improving factors of hemorheology,resisting inflammation and virus,and preventing acute liver injury,etc;the efficacy of "Warming middle energizer to relieve pain"means the function of improving activities of spleen,stomach and uterus in modern pharmacology;and the spleen channel tropism is related with and its pharmacologic action of enhancing immune activity.Conclusion There are some correlations between Zedoray Rhizome's traditional Chinese herbal nature and its modern pharmacologic actions.The correlations between Zedoray Rhizome's traditional Chinese herbal nature and its modern pharmacologic actions are waiting for the further improvements to provide the dependable basis for the clinical application.
3.Expression of Vascular Cell Adhesion Molecule 1 and Intercellular Adhesion Molecule 1 in Endometrium and Serum in Endometriosis

Objective:To study the expressions of Vascular Cell Adhesion Molecule 1 (VCAM-1) and Intercellular Adhesion Molecule 1 (iCAM-1) in patients with endometriosis and to explore its effect on the onset of endometriosis.Methods:VCAM-1 mRNA and ICAM-1 mRNA were detected by real-time PCR method in 15 specimens of eutopic endometrium and 15 specimens of ectopic endometrium from patients with endometriosis(EMT group)and 15 specimens of endometrium from patients without endometriosis(control group).VCAM-1 and ICAM-1 protein were detected by Western-Blot method.Soluble VCAM-1 and soluble ICAM-1 in 44 serum of patients with endometriosis(EMT group) and 28 serum of patients without endometriosis(control group) were tested by ELISA.Meanwhile the correlation of expressions of VCAM-1 and ICAM-1 were analyzed in different groups.Results:①The expressions of VCAM-1 mRNA and ICAM-1 mRNA in ectopic endometrium were significantly higher than those of eutopic endometrium and the controls(P <0.01).There was no significant different mRNA expression between eutopic group and control group(P>0.05).②The protein expressions of VCAM-1 and ICAM-1 in eutopic group were significantly higher than those of ectopic group and the control group(P <0.01).The protein expression of VCAM-1 in eutopic group were higher than those in the controls group(P<0.01).There was no significant different protein expression of ICAM-1 between eutopic group and the controls group(P >0.05).③The concentration of sVCAM-1 and sICAM-1 in serum from patients with EMT were significantly higher than those in the control group(P<0.01).The concentration of slCAM-1 in serum from patients with EMT in stage Ⅲ-Ⅳ were significantly higher than those in stage Ⅰ-Ⅱ (P < 0.01).④There was no significant correlation in the expression of mRNA,protein and serum between VCAM 1 and ICAM-1 in ectopic group and eutopic group(P > 0.05).Conclusions:The abnormal expressions of VCAM-1 and ICAM-1 in ectopic endometrium may play an important role in the development of endometriosis.The signaling pathways and protein expression way may be different between VCAM 1 and ICAM 1 in endometriosis.
4.Sensitive surface-enhanced Raman spectroscopy detection of thiram based on hybrid substrate

Objective To prepare hybrid substrate and apply it to detect thiram with surface-enhanced Raman spectros-copy( SERS) which provides unique molecular vibration information .Methods The Au substrate was prepared by deposi-tion of gold film on the silver substrate that had a rough surface .The Au substrate was treated with amination as a linker with the silver sol before the hybrid substrate was formed .With PATP as a probe molecule ,the Raman intensity of PATP on the Au substrate and the hybrid substrate was compared ,respectively .Results and Conclusion PATP had stronger Raman intensity on hybrid substrate than on the Au , and the detection limit was 10 -9 mol/L.This method can be used for quanti-tative detection on the hybrid substrate by SERS .
5.Factors Associated with Postvitrectomy Endophthalmitis

OBJECTIVE To investigate factors associated with postvitrectomy endophthalmitis. METHODS Cases with postvitrectomy endophthalmitis in our hospital between 2003 and 2007 were collected and retrospectively reviewed. RESULTS All 14011 cases (eyes) underwent pars plana vitrectomy and postoperative endophthalmitis happened to seven cases (eyes). The incidence of infection was 0.047%. Confirmed microbiology was demonstrated in 4 of 7 cases (57.0%),in which Gram-positive cocci were found 75.0%. 71.4% Of cases with postvitrectomy endophthalmitis were older than 60. The infection was controlled in 6 cases after antibiotic treatment. One case with diabetes and culture confirmation of Gram-negative bacilli deteriorated even after the second vitrectomy,and evisceration was finally performed. CONCLUSIONS The predominant organism is Gram-positive cocci in postvitrectomy endophthalmitis. The risk factors include increasing age,presence of systemic diseases and immunodeficiency. Timely and effective antibiotic therapy is important to restore visual acuity for postvitrectomy endophthalmitis.
6.Effect of intervention model based on empowerment theory on self-efficacy and self-acceptance of patients with permanent enterostomy

Objective? To explore the effect of empowerment-based intervention model on self-efficacy and self-acceptance of patients undergoing permanent enterostomy. Methods? By purposive sampling, a total of 100 colorectal cancer patients undergoing permanent enterostomy operation in the First Affiliated Hospital of Zhengzhou University from January to December of 2018 were selected as the research objects. The 50 patients admitted from January to June of 2018 were taken as the control group while the other 50 patients admitted from July to December of 2018 were taken as the experimental group. During intervention, there were 7 patients lost in the control group and 43 valid questionnaire retrieved; in the experimental group, 2 patients were lost and 48 valid questionnaires were retrieved. The control group was given conventional nursing care while the experimental group was given the nursing intervention formulated based on the empowerment theory. Stoma Self-Efficacy Scale(SSES) and Self-Acceptance Questionnaire(SAQ) were used to assess the patients. Results? After intervention, the total score of SSES, the dimension scores of "stoma care efficacy" and "social self-efficacy"of the experimental group were higher than those of the control group, with statistical significances (P<0.05). After intervention, the total score of SAQ and the score of "self-acceptance" dimension in the experimental group were higher than those in the control group, and the differences were statistically significant (P<0.05). Conclusions? The intervention based on empowerment theory can improve the self-management efficacy and self-acceptance level of patients with permanent enterostomy. It is suggested that it should be popularized in clinical practice.
7.Application value of group psychological intervention combined with narrative nursing in the psychological adjustment of patients with colorectal cancer

Objective:To explore the application value of group psychological intervention combined with narrative nursing in the psychological adjustment of patients with colorectal cancer.Methods:Totally 52 patients with colorectal cancer who were admitted into the First Affiliated Hospital of Zhengzhou University from February 2018 to June 2019 were selected by convenient sampling and divided into the control group ( n=26) and the observation group ( n=26) according to the random number table. Patients in the control group received routine care, while patients in the observation group received group psychological intervention combined with narrative nursing. Upon admission and one week after surgery, the nursing staff conducted an in-hospital survey and distributed the Hamilton Anxiety Scale (HAMA) and Hamilton Depression Scale (HAMD) , the Chinese version of the Ostomy Adjustment Inventory-20 (OAI-20) and the Perceived Derogation-Discrimination Scale (PDDS) to evaluate the patients. The negative emotions, psychosocial adjustment and stigma were compared between the two groups. Results:Before intervention, there was no statistically significant difference in the levels of negative emotions, psychosocial adjustment and stigma between the two groups ( P>0.05) . After intervention, the HAMA, HAMD and PDDS scores of the observation group were lower than those of the control group, and the OAI-20 score was higher than that of the control group, and the differences were statistically significant ( P<0.05) . Conclusions:Group psychological intervention combined with narrative nursing is of great significance in the psychological adjustment of patients with colorectal cancer, which can eliminate their stigma and ameliorate their negative emotions, and is worth promoting in clinical practice.
8. Mechanism of action for oligomeric proanthocyaniclins in pava qnat-induced acute lung injury

Objective:
The present study was designed to evaluate the protective effects of oligomeric proanthocyanidins (OPC) in mice exposed to paraquat (PQ) , and to explore the molecular mechanism.
Methods:
Four experimental groups were designed. Control group: 10 BALB/c mice were intraperitoneally injected with normal saline) . PQ group: 10 BALB/c mice were intraperitoneally injected with PQ (100 mg/kg) . PQ+OPC group: 10 BALB/c mice were administered with OPC (100 mg/kg) for 1 h before PQ (100 mg/kg) expo-sure. OPC group: 10 BALB/c mice were intraperitoneally injected with OPC (100 mg/kg) . The peripheral blood samples or lung tissue samples were collected at the designed time points for measuring the levels of oxi-dative stress indicators, the related protein levels of nuclear factor-kappa B (NF-κB) pathway and nuclear fac-tor erythroid related factor-2 (Nrf2) pathway.
Results:
Compared with the control group, the level of reactive oxygen species (ROS) , the content of malondialdehyde (MDA) in the PQ group were significantly induced, and the activity of superoxide dismutase (SOD) in the PQ group was decreased in the peripheral blood. As com-pared with the PQ group, the level of ROS and the content of MDA in the PQ+OPC group were significantly re-duced, the activity SOD in the PQ+OPC group was increased in the peripheral blood; the level of ROS and the content of MDA were also reduced in lung tissues in the PQ+OPC group. Moreover, compared with the con-trol group, the phosphorylation of IκBα and the expression of NF-κB p65 were increased in lung tissues in the PQ group. The phosphorylation of IκBα and the expression of NF-κB p65 were decreased in lung tissues in the PQ+OPC group as compared with the PQ group. In addition, compared with the control group, the expressions of HO-1 and Nrf2 were increased in lung tissues in OPC group, and these were decreased in lung tissues in PQ groups. Furthermore, the expressions of HO-1 and Nrf2 were also increased in lung tissues in PQ+OPC as com-pared with the PQ group.
Conclusion
OPC could alleviate PQ-induced systemic toxicity in mice by regulating oxidative stress via NF-κB and Nrf2 pathway.
9.Association between tumor necrosis factor-β gene polymorphisms and genetic predisposition to gastric cancer

Objective:To investigate the association between tumor necrosis factor-β (TNF-β) gene polymorphisms and genetic predisposition to gastric cancer, and to analyze the relationship between specific genotype of TNF-β and serum levels of TNF-β.Methods:Using case control study, we selected 153 patients with gastric cancer in Fudan University Shanghai Cancer Center between September 2021 and December 2022 as the gastric cancer group, and 150 healthy individuals were chosen as the healthy control group. In the previous study, 30 peripheral blood DNA samples of gastric cancer patients and healthy controls respectively were amplified by conventional PCR, which were sequenced to identify the genotype frequencies of TNF-β polymorphic loci (rs1041981, rs2229092, rs2229094 and rs78613290); consequently, Allele-Specific Quantitative PCR was used to further detect and analyze the genotype and genotype frequencies of TNF-β polymorphic loci; serum TNF-β levels were measured by Enzyme-Linked Immunosorbent Assay (ELISA), and the relationship with specific genotypes of TNF-β was analyzed. Chi-square test and Fisher test were used to analyze the genotype distribution frequency of TNF-β polymorphic loci, and non-parametric statistics was used to analyze the differences in serum TNF-β expression levels.Results:The sequencing results showed that the genotype distribution of rs1041981 in gastric cancer group was CC 16.67% (5/30), CA 40.00% (12/30) and AA 43.33% (13/30). The genotype distribution in control group was CC 40.0% (12/30), CA 43.33% (13/30), AA 16.7% (5/30). The difference of genotype frequency between the two groups was statistically significant (χ 2=6.478, P=0.039). The genotypes of the polymorphic loci rs2229092 in both groups were AA, AG, and GG, with no statistically significant difference between the two groups (χ 2=1.888, P=0.612). The distribution frequencies of the genotypes of the polymorphic loci rs2229094 (TT and TC) and rs78613290 (GG and AG) showed no statistically significant differences between the two groups (both P>0.05). Further validation with an expanded clinical samples (153 cases in the gastric cancer group and 150 cases in the control group) found that the difference of rs1041981 genotype distribution between the gastric cancer group [CC 15.69%(24/153), CA 54.9%(84/153), AA 29.4%(45/153)] and the control group [CC 27.3%(41/150), CA 58.0%(87/150), AA 14.7%(22/150)] was significantly different (χ 2=12.366, P=0.002). Analysis of the influence of different allele frequencies on the risk of gastric cancer revealed that the odds ratio ( OR) of the A allele of rs1041981 for the risk of gastric cancer compared to the C allele was 1.701 (95% CI 1.235?2.355). Gene phenotype analysis combining the clinicopathological characteristics of gastric cancer patients found that the distribution frequency of the rs1041981 genotype was significantly different among groups of different genders, tumor invasion depths, and the lymph node metastasis, with statistically significant differences (All P>0.05). Additionally, gastric cancer patients with rs1041981 AA genotypes had higher serum TNF-β expression levels than those with CA and CC genotypes, (both P<0.05). Conclusions:The gene type frequency of the TNF-β gene polymorphic loci (rs1041981, C>A) exhibited significant differences between the gastric cancer group and the healthy control group. The presence of the A allele in rs1041981 site increased the susceptibility to gastric cancer, and patients with different gene types displayed vaning levels of serum TNF-β, among which AA genotype ranks the highest level.
10.Clinical application of percutaneous pedicle screw placement guided by ultrasound volume navigation combined with X-ray fluoroscopy: a prospective randomized controlled study.

OBJECTIVE:
To explore the feasibility and accuracy of ultrasound volume navigation (UVN) combined with X-ray fluoroscopy-guided percutaneous pedicle screw implantation through a prospective randomized controlled study.
METHODS:
Patients with thoracic and lumbar vertebral fractures scheduled for percutaneous pedicle screw fixation between January 2022 and January 2023 were enrolled. Among them, 60 patients met the selection criteria and were included in the study. There were 28 males and 32 females, with an average age of 49.5 years (range, 29-60 years). The cause of injury included 20 cases of traffic accidents, 21 cases of falls, 17 cases of slips, and 2 cases of heavy object impact. The interval from injury to hospital admission ranged from 1 to 5 days (mean, 1.57 days). The fracture located at T 12 in 15 cases, L 1 in 20 cases, L 2 in 19 cases, and L 3 in 6 cases. The study used each patient as their own control, randomly guiding pedicle screw implantation using UVN combined with X-ray fluoroscopy on one side of the vertebral body and the adjacent segment (trial group), while the other side was implanted under X-ray fluoroscopy (control group). A total of 4 screws and 2 rods were implanted in each patient. The implantation time and fluoroscopy frequency during implantation of each screw, angle deviation and distance deviation between actual and preoperative planned trajectory by imaging examination, and the occurrence of zygapophysial joint invasion were recorded.
RESULTS:
In terms of screw implantation time, fluoroscopy frequency, angle deviation, distance deviation, and incidence of zygapophysial joint invasion, the trial group showed superior results compared to the control group, and the differences were significant ( P<0.05).
CONCLUSION
UVN combined with X-ray fluoroscopy-guided percutaneous pedicle screw implantation can yreduce screw implantation time, adjust dynamically, reduce operational difficulty, and reduce radiation damage.
