Objective:To detect the levels of T help cell 17 (Th17)/regulatory T cell (Treg) in the peripheral blood of patients with ovarian cancer and explore their clinical significances.Methods:55 patients with ovarian cancer and 60 normal controls were enrolled.The percentage of Th17/Treg cells were detected by flow cytometry (FCM).The expression of retinoid-related orphan receptor gamma-t ( RORγt)/foxhead winged-helix box protein 3 ( Foxp3) mRNA was measured by real time PCR and the plasma levels of IL-17/TGF-β1 were detected by enzyme linked immunosorbent assay (ELISA).Results: The percentage of CD4+IL-17+/CD4+T cells,the mRNA expression of RORγt and the plasma levels of IL-17 increased significantly in patients with ovarian cancer than in normal controls (P<0.05).Compared with normal controls,the percentage of CD4+Foxp3+Treg/CD4+T cells,the mRNA expression of Foxp3 and the plasma levels of TGF-β1 increased significantly in patients with ovarian cancer ( P<0.05).Conclusion:The imbalance of Th17/Treg cells may exist in the peripheral blood of patients with ovarian cancer.Th17/Treg cells may participate the pathology of ovarian cancer.

OBJECTlVE To study the preventive effect of vitamin E(Vit E)on chronic Iiver injury in-duced by N,N-dimethyIformamide( DmF). METHODS HeaIthy maIe Kunming mice were randomIy divided into 4 groups,10 mice in each group. The mice in DmF modeI group and DmF+Vit E group were ig given DmF 0.5 g·kg-1 ,whiIe Vit E controI group and DmF+Vit E group were ig given Vit E 5 mg·kg-1 after 2 h once a day for 30 d. The activity of serum gIutamic-oxaIoacetic transaminase(GOT),gIutamic-pyruvic transaminase(GPT)and xanthine oxidase(XOD)and the content of maIondiaIdehyde(mDA) in Iiver tissue were measured with corresponding kits. The pathoIogicaI changes of Iiver tissue were ob-served with HE staining. RESULTS Compared with the normaI controI group,the activity of serum GOT,GPT and XOD of the DmF modeI group was significantIy eIevated(P﹤0.05,P﹤0.01),and the Iiver ceII nucIeus pycnosis,ceIIuIar edema,degeneration,necrosis occurred. Compared with DmF modeI group,the activity of serum GOT,GPT and XOD of DmF+Vit E group were aII significantIy reduced(P﹤0.05,P﹤0.01),the content of mDA in Iiver tissue was significantIy diminished(P﹤0.01),but histopatho-IogicaI changes of Iiver tissue were significantIy improved. CONCLUSlON Vit E has preventive effect on chronic Iiver damage induced by DmF through antioxidation.

N, N-dimethylformamide (DMF) is a kind of organic compound which is widely used in the modern industry. With the increasing utilization of DMF year by year, the interest in the research of DMF toxicity has increased dramatically. The data of animal experiments and epidemiological investigations show that DMF has harmful effect on reproductive function of human and animals. The effect of DMF on the reproductive function of animal and occupational crowd were reviewed in this paper, the research perspective was also presented.

OBJECTIVETo investigate the incidence and risk factors for hypertension among taxi drivers working different shifts.

METHODSUsing the cluster sampling method, 415 day-shift and 304 night-shift taxi drivers in Jinan, China were selected and investigated. The influencing factors for hypertension were analyzed.

RESULTSThe incidence of hypertension in all taxi drivers was 33.2%. The incidence of hypertension in night-shift drivers was significantly higher than that in day-shift drivers (37.8% vs 29.9%, P<0.05). According to multivariate analysis, the incidence of hypertension in day-shift drivers was closely related to body mass index (BMI), working years, working hours, sleep duration, rest days, diet quality, water intake, and smoking, while the incidence of hypertension in night-shift drivers was closely related to BMI, working years, working hours, part-time job, sleep duration, rest days, and drinking.

CONCLUSIONBMI, working years, and working hours are the common risk factors for hypertension in day-shift and night-shift drivers. Sleep duration and rest days are the common protective factors. The risk factors for hypertension in taxi drivers vary with different shifts.

Objective To explore the inhibitory effects of lobaplatin and cisplatin and their regulation of apoptosis-related genes in ovarian cancer cells in nude mice.Methods SKOV3 cells were implanted into nude mice.In monotherapy treatment study,the nude mice bearing human SKOV3 cells were randomly divided into control,lobaplatin,and cisplatin groups,with 7 mice in each group.The mice in each group were received corresponding treatment.The volume of tumor and the weight of nude mice were measured three times per week,respectively.Tumor inhibitory rate was calculated.The protein expressions of bax and bcl-2 were detected by flow cytometry.Results The growth inhibitory rate was 47.2％ in lobaplatin group and 42.8％ in cisplatin group,without significant difference between two groups (P ＞ 0.05).The expression of bcl-2 was decreased but the bax was increased in lobaplatin and ciaplatin groups compared to the control group.Conclusions Lobaplatin can significantly inhibit the growth of ovarian cancer cells,induce apoptosis by down-regulation of bcl-2 and up-regulation of bax.

Objective Through investigation and training in the knowledge and skills of pre hospital emergency treatment in Xuzhou highway traffic policemen,we explored a more targeted and more effective first-aid skills training program.Methods Based on our hospital emergency center 91 highway traffic policemen were randomly assigned to 3 first aid skills training groups,group 1 received theoretical training and operational training,group 2 was given the same theoretical training and operational training after DVD player,group 3 took the same method as that of group 2,besides,situational training was given to group 3.The emergency treatment technique was evaluated post training.Results After the training,the scores of 3 groups were different from each other,group 3 had the best results compared with group 1 and group 2.Conclusions The highway traffic policemen had poor emergency treatment knowledge.They should be trained with first aid knowledge and skills as well as cardiopulmonary resuscitation training.

ObjectiveTo observe the effect and safety of arsenic trioxide (ATO) combined with all-trans retinoic acid (ATRA) in treating patients with acute promyelocytic leukemia (APL).Methods Eighty-three cases with APL treated for the first time were divided into two groups by random digits table method:observation group with 48 cases was received combination induction treatment of ATO and ATRA,control group with 35 cases was treated with combination induction treatment of ATRA and chemotherapy.The clinical effect and adverse reaction between two groups were compared.ResultsThe effective rate and early death rate were 100.0％( 48/48 ) and 0 in observation group,97.1％(34/35 ) and 2.9％( 1/35 ) in control group,which had no significant difference between two groups(P ＞ 0.05 ).The incidences of bone marrow suppression,infection,liver and kidney damage,cardiac toxicity and gastrointestinal symptoms were 8.3％ (4/48),10.4％ (5/48),12.5％ (6/48),6.2％ (3/48) and 18.8％ (9/48) in observation group,while 97.1％(34/35),65.7％(23/35),45.7％(16/35),37.1％(13/35) and 100.0％(35/35) in control group,which had significant differences between two groups (P ＜ 0.05).ConclusionCombination treatment of ATO and ATRA in APL has an obvious effect and few adverse reaction,which can be applied in clinic.

Objective To investigate the effect of sevoflurane anaesthesia on the cognitive function and phosphorylation of tau protein in hippocampal neurons in amyloid precursor protein (APP) transgenic mice.Methods Male APP gene mutation mice,weighing 18-22 g and aged 8-12 weeks,were used in this study.Forty-four APP positive mice were randomly divided into two groups:sevoflurane group (group AS,n =28) and control group (group AC,n =16).And other forty-four APP negative mice were randomly divided into two groups:sevoflurane group (group S,n =28) and control group (group C,n =16).The animals in groups S and AS inhaled 3％ sevoflurane for 4 hours.While in groups C and AC,the animals inhaled pure oxygen for 4 hours.Morris water maze was performed 24 hours after sevoflurane or pure oxygen inhalation.The phosphorylation of tau protein at Ser262 and Ser396 sites was detected by Western blotting on 1 day after pure oxygen inhalation in groups AC and C,and on 1,3 and 7 days after sevoflurane inhalation in groups AS and S.Results Compared with group C,the escape latency was significantly prolonged and the duration of staying at the original platform quadrant was shortened in groups S and AC,and the phosphorylation of tau protein at Ser262 site in group S and phosphorylation of tau protein at Ser262 and Ser396 sites in group AS were increased (P ＜ 0.05).Compared with group S,the escape latency was significantly prolonged,the duration of staying at the original platform quadrant was shortened,and the phosphorylation of tau protein at Ser262 and Ser396 sites was increased in group AS (P ＜ 0.05).Compared with group AC,the escape latency was significantly prolonged,the duration of staying at the original platform quadrant was shortened,and the phosphorylation of tau protein at Ser262 and Ser396 sites was increased in group AS (P＜0.05).Conclusion Sevoflurane anesthesia can aggravate the impairment of cognitive function in APP positive mice and the increase in the phosphorylation of tau protein at Ser262 and Ser396 sites is involved in the mechanism.

BACKGROUNDTo study the expression of catalylic subunit of DNA-dependent protein kinase (DNA-PKcs) in non-small cell lung cancer (NSCLC) and its relationship with apoptosis.

METHODSNSCLC tissues from 113 untreated patients were analyzed immnohistochemically with antibodies to DNA-PKcs, p53 and bcl-2.

RESULTSThere were expressions of DNA-PKcs, p53 and bcl-2 in NSCLC at different levels. The positive rate of DNA-PKcs, p53 and bcl-2 was 89.38%(101/113), 61.95%(70/113) and 59.29%(67/113) respectively. The expression of DNA-PKcs was significantly related with the histological types. Its expression in squamous carcinoma was significantly lower than that in adenocarcinoma and bronchioloalveolar carcinoma. The expression of DNA-PKcs increased with the increasing differentiated degree of NSCLC ( P < 0.05), but had no relationship with lymph node metastasis. There was no significant relation between the expression of p53 and the pathological type of NSCLC. A significant difference of bcl-2 expression existed in the histological types of lung cancer ( P < 0.01). Its expression in squamous carcinoma was significantly higher than that in adenocarcinoma, bronchioloalveolar carcinoma and adenosquamous carcinoma, but had no relationship with the differentiated degree of lung cancer and lymph node metastasis. The expressions were significantly related between DNA-PKcs and p53 ( P < 0.01), p53 and bcl-2 ( P < 0.05).

CONCLUSIONSThe expression of DNA-PKcs is fairly high in NSCLC. The high expression of DNA-PKcs and overexpressions of mutated p53 and bcl-2 may be important causes of radioresistance in NSCLC.
