1.Clinical efficacy of percutaneous kyphoplasty combined with anti-osteoporosis drugs treatment of vertebral compression fracture with osteoporosis in elderly patients

Objective To investigate the clinical effect of percutaneous kyphoplasty(PKP) combined with anti-osteoporosis drug treatment of vertebral compression fracture with osteoporosis in elderly patients.Methods Fifty-three cases of elderly patients with thoracolumbar vertebral compression fracture due to osteoporosis were selected as subjects,who were hospitalized in the Central Hospital of Sujiatun District of Shenyang from October 2010 to December 2014.They were randomly divided into the observation group (27 cases) and control group(26 cases).All patients were treated with PKP.The patients in the observation group were given the anti-osteoporosis therapy.Before and after operarion,the visual analogue scale(VAS) score was estimated and Cobb's angle of the vertebrae were measured.Postoperative complications,bone mineral density in the first half year after operation were recorded.Results VAS of treatment and of control group before operation were 7.92± 1.00 and 7.89±0.99,higher than those of one week after operation(2.68± 1.01,2.73 ± ± 1.03).Two groups of treatment and control had no significant difference(P>0.05),but before and one week after operation had significant difference(P=0.037,0.038).Cobb's angle of treatment and control group were (28.23±6.00) ° and (28.10±5.99) ° before operation,(10.90±2.99) ° and (11.09±3.00) ° one week after operation.Two groups of treatment and control had no significant difference(P>0.05),but before and one week after operation had significant difference (P =0.027,0.029).The bone mineral density in observation group was (0.79±0.12) g/cm2,higher than that in control group((0.65±0.06) g/cm2,P=0.048) in sixth months after operation.The duration of follow up after operetion were 12 months.Recurrence rate of vertebral compression fracture in control group was 15.4% (4/26),higher than that in observation group (0 (0/26)),and there was significant difference between two groups (P =0.041).Conclusion PKP combined with anti-osteoporosis treatment of vertebral compression fracture with osteoporosis can quickly relieve vertebral fracture pain and reduce the incidence of new vertebral fractures hair.The treatment with minimally invasive and orthopedic effect is worthy of clinical application.
2.Protective Effect of Taurine on Diaphragm in Diabetic Rats

[Objective] To observe protective effect of taurine, a main component of Calculus Bovis and Scorpio, on diaphragm in diabetic rats. [Methods] Twenty-four SD rats were equally randomized into normal control group, model group and taurine group. Except the normal control group, the other rats were given intraperitoneal injection of streptozotocin 50mg/kg to induce diabetic models. Taurine group was fed with 10g/L water solution of taurine, and the other two groups with water for 4 weeks. After treatment, the rats were executed and the isolated diaphragm strips were prepared. Single contraction (SC) , maximum titanic tension, contraction time, half relaxation time, force-frequency curve and fatigue index (FI) of the diaphragm strips were examined. The contents of blood sugar, superoxide dismustase (SOD) and malondialdehyde (MDA) in the diaphragm were detected and the diaphragm ultrastructure was also observed. 3.Effects of early peritoneal puncture and catheter drainage on severe acute pancreatitis

ObjectiveTo evaluate the effect on organ function by early peritoneal puncture and catheter drainage for intra-abdominal hypertension during the course of severe acute pancreatitis (SAP).MethodsA retrospective study was conducted on 405 patients of severe acute pancreatitis with IAH at Department of Pancreatic Surgery,Wuhan Union Hospital from January 2001 to September 2010.In the early course of SAP,137 cases did not receive peritoneal puncture and drainage decompression,268 cases were treated with peritoneal puncture and drainage decompression.ResultsThe differences of the scores of UBP and APACHE Ⅱ were statistically significant between the puncture group and the non-puncture group (P < 0.05).The occurrence of ARDS,ARF and MODS in the group of puncture were significantly lower than those of the non-puncture group (14% vs.27%,21% vs.32%,9% vs.14%,respectively,all P <0.05 ).At the same time,the differences between rate of early laparotomy and rate of early mortality in the two groups were statistically significant (4% vs.12%,3% vs.6%,respectively,all P <0.05).However,there was no significant difference in the occurrence time of negative balance in the two groups (5.9 ±2.9 vs.5.9 ± 2.4,P > 0.05 ).ConclusionsEarly abdominal puncture and drainage reduce the abdominal pressure of patients with SAP and IAH.It also reduces the occurrence of organ failure,the rate of early laparotomy and the rate of early mortality in SAP patients.
4.Negative Modulation of NO for Diaphragmatic Contractile Reduction Induced by Sepsis and Restraint Position

In practice of forensic medicine, potential disease can be associated with fatal asphyxia in re-straint position. Research has demonstrated that nitric oxide (NO) and nitric oxide synthase (NOS) are plentifully distributed in skeletal muscle, contributing to the regulation of contractile and relaxation. In the current study, respiratory functions, indices of diaphragmatic biomechanical functions ex vivo, as well as NO levels in serum, the expressions of diaphragmatic inducible NOS (iNOS) mRNA, and the effects of L-NNA on contractility of the diaphragm were observed in sepsis induced by cecal ligation and punc-ture (CLP) under the condition of restraint position. The results showed that in the CLP12-18 h rats, respiratory dysfunctions; indices of diaphragmatic biomechanical functions (Pt, +dT/dtmax, -dT/dtmax, CT, Po, force over the full range of the force-frequency relationship and fatigue resistance ) declined progressive-ly; the NO level in serum, and iNOS mRNA expression in the diaphragm increased progressively; force increased significantly at all stimulation frequencies after L-NNA pre-incubation. Restraint position 1 h in CLP12 h rats resulted in severe respiratory dysfunctions after relative stable respiratory functions, almost all the indices of diaphragmatic biomechanical functions declined further, whereas little change took place in NO level in serum and diaphragmatic iNOS mRNA expression; and the effects of L-NNA were lack of statistical significance compared with those of CLP12 h, but differed from CLP18 h group. These results suggest that restraint position and sepsis act together in a synergistic manner to aggravate the great reduction of diaphragmatic contractility via, at least in part, the negative modulation of NO, which may contribute to the pathogenesis of positional asphyxia.
5.Effects of Soy Isoflavones on NO Content and NOS Activity in Myocardial Tissues of Diabetic Rats

Objective To observe the effects of soy isoflavones (SI) on NO content and NOS activity in myocardial tissues of diabetic rats. Methods Forty-eight SD rats were randomly divided into six groups: normal control group (NC), diabetic control group (DC), SI groups in low, moderate and high doses respectively (L-SI, M-SI, H-SI) and nilestriol group (NI). Except the NC group, the rats were given streptozotocin (STZ) 55mg?kg-1 intraperitneally (ip). From the 7th week, SI in the dosage of 30, 60, 120 mg?kg-1?d-1 was respectively given to L-SI, M-SI, H-SI groups by gastric gavage, nilestriol 0.2 mg?kg-1?per week was given to NI group,and 0.5%CMC-Na10 ml?kg-1?d-1 was given to NC and DC groups. After treatment, fasting blood sugar level, body weight, serum lactate dehydrogenase (LDH)and creatine kinase (CK) contents, and NOS activities and NO content in myocardial homogenate were measured. Objective To study the reliability and validity of Chinese version of housebound scale in Chinese elderly people in community.Methods The housebound scale was amended and translated according to the Procedure of the European organization for Research and Treatment of Cancer (EORTC) Quality of Life Group.Include two scales,which is whether is housebound and the reason of housebound.The reliability and validity of Chinese version of housebound scale were tested in a sample of 860 Chinese elderly people in two communities among 127 communities in Tangshan from October 2012 to March 2013.Reliability was evaluated using the splithalf reliability and internal consistency.Validity was assessed by content validity,discriminate validity and construct validity.Results The Cronbach’s α coefficient of the first subscales was 0.743,the split-half reliability was 0.856,the convergent validity ratio was 0.960,and the scale had good discriminate validity (P < 0.01).The Cronbach's αcoefficient of the second subscales was 0.853,the split-half reliability was 0.816,the convergent validity ratio was 0.910.Four factors body and mental factors,environmental factors,psychological and social factors,listening factors were extracted by a factor analysis,which explained 48.37% of the total variance.The loading value of each items in their respective common factor ranged from 0.42 to 0.82.Conclusion The housebound scale has better reliability and validity,which is available for the evaluation of housebound amnong elderly people.
7.Observation and analysis of the individual behavior with weaning of infant Rhesus Monkeys

Objective To understand the characteristics and rules of behavior and preliminary study on the change of psychological state and behavior of infant rhesus monkeys after weaning by artificial feed .Methods 290 infant rhesus monkeys were born in 2012, individual behavior of infant rhesus monkeys were observed and recorded by the methods of ad libitum sampling and chronography .Results The behavior of huddling , sucking finger and sucking body of companion were observed in the first month , the behavior of huddling and sucking finger were significantly increased in the second and third month(P <0.05), sucking body of companion in second month were significantly higher than the first month (P <0.05);the new behavior pacing , sucking genitalia and mounting were observed in the second month , pacing and sucking were significantly increased in the third month ( P <0.05 ) , mounting did not have obvious change; in the third month , sucking toe behavior ware recorded and did not change with time; The proportion of huddling behavior occurred between female and female was significantly higher than opposite sex or male in the same cage .The male infant monkeys were more likely to occurred pacing behavior than female .Sucking body of companion and sucking finger behavior of male were significantly lower than the female infant monkeys ( P <0.05 ) .There was no significant difference of sucking toe behavior .Conclusions 7 behaviors were recorded during the whole stage of observation , huddling behavior , pacing behavior , sucking toe behavior , sucking finger behavior , sucking genitalia behavior , sucking body of companion behavior and mounting behavior respectively , and 7 behaviors were increased by degrees with time .Huddling behavior to reflect fear psychology;sucking behavior to represent physiological demand and defensive psychology ;pacing was insignificant behavior in the initial stage , then to reflect psychological anxiety of rhesus monkeys .Provided a way and model on assessment of psychology, praxiology, to study and research the change of psychological state and behavior of early weaning infants .
8.Outcome of allogeneic hematopoietic stem cell transplantation for relapsed/refractory acute myeloid leukemia in NR status

Objective To explore the outcome of allogeneic hematopoietic stem cell transplantation (allo-HSCT) for relapsed/refractory acute myeloid leukemia (AML) patients in nonremission (NR) status and its related risk factors.Methods Thirty-nine relapsed/refractory AML patients in NR status who received allo-HSCT between Jan.2006 and Dec.2016 were retrospectively analyzed.Major end points of study included overall survival (OS),disease free survival (DFS),and relapse rate.Results All patients achieved hematopoietic reconstitution.Median time to neutrophil and platelet engraftment was 13 (11 20) and 16 (10-58) days,respectively.Acute graft-versus-host disease (aGVHD) occurred in 25 (64.1%) patients,and 11 (31.4%) patients developed chronic GVHD.During a median follow-up period of 9.1 (1.6-93.8) months,11 (28.9%) cases survived,10(26.3%) survived without leukemia,and 21 (53.8%) relapsed.The estimated 2 year OS and DFS were 30.0% ± 8.0% and 26.7% ± 7.7%,and cumulative incidence of relapse and transplantation related mortality at 2 years was 56.63% (95% CI 37.84%-71.71%),19.7% (95% CI 8.3%-34.5%),respectively.The multivariate analysis revealed that the number of bone marrow blasts≥25% or any level of blasts in peripheral blood was significantly associated with worse OS (HR =11.91,P=0.003),DFS (HR =10.75,P =0.002) and higher rate of relapse (70.83% versus 20.22%,P =0.002).In addition,the development of grade Ⅱ-Ⅳ aGVHD also predicted an inferior OS (HR =3.18,P =0.039).Conclusion Salvage therapy with allo-HSCT can induce long-term survival in part refractory/relapsed AML patients.Decrease in the pre-transplant disease burden is the key to reduce relapse and improve survival.
9.Establishment of a method for the evaluation of emergency granulopoiesis in mouse bone marrow with EdU

Objective To label granulocytes in a state of differentiation in mouse bone marrow (BM) with EdU (5-ethynyl-2′-deoxyuridine) for further understanding the changes in granulocyte produc-tion at different stages of differentiation during inflammation. Methods C57BL/6 mice were intraperitoneal-ly (i.p.) injected with EdU and heat-inactivated Escherichia coli(HI E.coli). BM cells were harvested at different time points after HI E.coli injection and then stained with fluorescent-conjugated antibodies(Abs). Myeloblasts,promyelocytes,myelocytes, metamyelocytes and band and segmented neutrophils were identi-fied by fluorescence-activated cell sorting(FACS). The percentage of EdU-positive cells in each population was recorded. Results The percentage of EdU-positive myeloblasts in mice increased by 10.0% at 24 h af-ter intraperitoneal injection with HI E.coli,but decreased by 75.0% and 23.0% at 48 h and 72 h,respec-tively. The percentage of EdU-positive promyelocytes declined by 23.0%,54.5%,64.3% and 77.8% at 24 h,48 h,72 h and 96 h,respectively. The percentage of EdU-positive myelocytes increased by 60.0% and 10.0% at 24 h and 48 h,but decreased by 80.0% and 90.0% at 72 h and 96 h. The percentage of EdU-positive metamyelocytes increased by 50.0% at 24 h,but decreased by 33.3%,61.5% and 66.7% at 48 h,72 h and 96 h. The percentage of EdU-positive band and segmented cells increased by 14.0% at 24 h,but decreased by 50.0%, 77.8% and 88.0% at 48 h, 72 h and 96 h. Conclusion Emergency granulopoiesis occurred 24 h after the establishment of HI E.coli-induced model of acute peritonitis, which meant that the proliferation of myeloid precursor cells,especially that of myelocytes and metamyelocytes,was accelerated and resulted in increasing number of mature neutrophils immigrating to sites of inflammation.
