1.Effect of Transverse Position and Numbers on the Stability of the Spinal Pedicle Screw Fixation during the Pedicle Cortex Perforation.

Objective To evaluate the effect of transverse position and numbers on the stability of the spinal pedicle screw fixation during the pedicle cortex perforation. Methods The vertebral compression fracture was performed in the L1 vertebral body using the Chiba's method from 60 fresh thoracic and lumbar vertebrae samples of sheep(T13-L3),which were randomly divided into 6 groups(A,B,C,D,E,and F)using a lottery method. Bilateral pedicles of vertebral arch of T14 and L2 were inserted pedicle screws,connecting titanium rods to fix T14-L2 segments. Then the samples of groups B,C,D,E,and F were removed a quarter of right side of lateral T14 thoracic pedicle cortical,which were considered the pedicle cortex perforation model. Finally,each group was fixed on different numbers of crosslinks:group A(0 crosslink,Intact),group B(0 crosslink,NCL),group C(1 crosslink,1/2 of the rods,MCL),group D(1 crosslink,1/3 of the rods close to T14,PCL),group E(1 crosslink,2/3 of the rods away from T14,DCL)and group F(2 crosslinks,1/3 and 2/3 of the rods respectively,TCL). After all samples were subject to 10 000 times of fatigue test with biomechanics test machines,the axial compressive stiffness,range of the motion(ROM)of the 6 directions(flexion,extension,lateral bending,and axial rotation),and the maximum pullout of the screws of the T14 pedicle cortex perforation were measured and compared among these 6 groups. Results The axial compressive stiffness in groups A,C,D,E,and F was significantly higher than that in group B(all P<0.05),and group A was significantly higher than group F(P<0.05) . The maximum pullout in groups A,C,D,E,and F were significantly higher than that in group B(all P<0.05),and group A was significantly higher than group F and groups C,D,and E were significantly lower than group F(all P<0.05). ROM of flexion,extension,lateral bending,and axial rotation in groups A,B,C,D,E,and F were significantly lower than that in group B(P=0.000),and ROM of left and right axial rotation in groups C,D,and E were significantly higher than in that group F(P=0.000). Conclusions During the pedicle cortex perforation,adding of one crosslink can improve the stability of the pedicle cortex perforation,and adding of two crosslinks can approximately achieve the same stability as the pedicle screw fixation with no pedicle cortex perforation. The location of the crosslink has no obvious effect on the short segment of spinal fixation.
2.Expression and clinical significance of serum chemokines in patients with lung cancer

Objective To detect the expression levels of multiple serum chemokines including IFN-inducible T cell chemoattractant (ITAC),Fractalkine,macrophage inflammatory protein (MIP)-3α,IL-8,MIP-lα,MIP-1β in patients with lung cancer and explore their association with the clinical characteristics of lung cancer as well as the correlations among these chemokines.Methods Forty newly diagnosed patients with lung cancer and thirty healthy controls were enrolled for detection of the serum levels of 6 kinds of chemokines by Luminex technology.The correlations of clinical characteristics of lung cancer with these chemokines and the correlations among these chemokines were analyzed by SPSS 17.0 software.Results The serum levels [M (QR)] of IL-8,Fractalkine and MIP-3α in patients with lung cancer were 5.16 (4.74),128.45 (141.89),10.31 (8.88) respectively,and 2.01 (0.95),61.46 (74.81),8.08 (5.87) respectively in control group,with significant differences (Z =-4.783,P <0.001;Z =-4.046,P <0.001;Z =-3.105,P =0.002).The expression of MIP-1β in lung adenocarcinoma was significantly higher than that in squamous carcinoma [18.32 (12.27) vs.13.72 (7.31),Z =-2.212,P =0.027],and of ITAC in squamous carcinoma was significantly higher than that in small cell lung cancer [24.51 (22.48) vs.9.28 (4.85),Z =-2.460,P =0.014].The expressions of MIP-3α and Fractalkine were positively correlated in the two groups (r =0.619,P<0.001;r=0.766,P<0.001).Conclusion The expressions of IL-8,Fractalkine and MIP-3α increase significantly in lung cancer patients,and they are may play important roles in metastatic lung cancer.
3.RNase MC2 manifests antitumor effects towards human hepatocellular carcinoma

Purpose To determine the effect of RNase MC2 purified from momordica charantia on cell growth of hepatocellular carcino-ma ( HCC) and its underlying mechanism. Methods MTT, colony formation and nude mice model were used to examine the activity of RNase MC2 in cell proliferation. Cell cycle analysis was done by flow cytometry. Autophagy induced by RNase MC2 treatment was observed via transmission electron microscope. Western blot was performed to detect the RNase MC2-mediated changes of proteins. Re-sults In vitro and in vivo data showed that RNase MC2 markedly inhibited HCC cell proliferation, arrested cells at G2/M phase by in-creasing expression of p53 and p21, induced autophagy via upregulating Beclin-1 and LC3-Ⅱ. Furthermore, combination of RNase MC2 and Sorafenib exerted enhanced lethal effect on HCC cells. Conclusion RNase MC2 manifests significant antitumor activities and enhances the killing effect of Sorafenib in HCC via inducing cell cycle arrest and autophagy.
4.Design, synthesis and evaluation of tacrine-methoxybenzene hybrids as cholinesterases inhibitors.

A series of tacrine-methoxybenzene hybrids (5a-5i) were designed, synthesized and evaluated as inhibitors of cholinesterases (ChEs). All the compounds had better ChEs inhibitory activities than tacrine with IC50 values at the nanomolar range. Compound 5h exhibited the strongest inhibition on acetylcholinesterase (AChE) with an IC50 value of 6.74 nmol x L(-1) and compound 5f showed the most potent inhibition on butyrylcholinesterase with IC50 value of 3.83 nmol x L(-1). Kinetic and molecular modeling studies showed that these hybrids targeted both the catalytic active site and the peripheral anionic site of AChE.
5.Genetic polymorphisms of 19 STR loci in han popu-lation from Hainan and its application in paternity testing

Objective To construct a database for the genetic polymorphism of 19 STR loci in Han population from Hainan province. To investigate the application of 19 STR loci in the paternity testing. Methods The genotypes of 462 unrelated individuals in Hainan were detected with GoldeneyeTM 20A PCR Amplification Kit. 19-STR database was acquired, analyzed and evaluated in 283 paternity testing cases. Results No deviations of allele frequency from Hardy-Weinberg equilibrium expectations were found for Chi-square test (P>0.05). Observed heterozygosity (Hobs) varied between 0.603 and 0.914, total discrimination power (TDP) of 19 STR loci was more than 0.999999999999999, cumulative probability of exclusion (CPE) for triplet cases was 0.999999994. In all 283 paternity testing cases, triplets and duos were 170 and 113 respectively; there were 36 (12.7%) excluded cases comparing to 247 confirmed cases (87.3%). 14 mutation events were observed, and all were one-step mutation. Conclusion 14 out of 19 loci showed highly polymorphic in Han population from Hainan, and 19 STR system has high cumulative probability of exclusion and can meet the needs of paternity test of the local region. But mutation should be paid special attention to.
6.Significance of Troponin-I in patients with septic shock and left ventricular diastolic dysfunction

Objective To investigatethe clinical value of troponin-I(cTnI)in patientswith septic shocka-nd left ventricular diastolic dysfunction. Methods As a retrospective analysis ,38 patients with left ventricular di-astolic dysfunction and septic shock(Sa group),as well as 20 patients with normal cardiac function(Sn group) were enrolled in this study. Moreover ,20 patients with left ventricular diastolic dysfunction and without septic shock were used as control group(Ca group). The ratio of early diastolic mitral inflow velocity to early diastolic mi-tral annulus velocity(E/e′)was measured as the evaluation index of left ventricular diastolicfunction by echocar-diography within 72 hours after admission to ICU. Level of cTnI was detected in all cases and the relationship was evaluated by E/e′. Receiver operating characteristic curve(ROC)was constructed to indicate the predictable value of left ventricular diastolic dysfunction in patients with septic shock. Results The level of cTnI was significantly elevated in both Sa group and Sn group(P<0.05),while the level of cTnI and E/e′in Sa group were significantly higher than those in Sn group(P < 0.05). cTnI was positively correlated with E/e′(r = 0.367 ,P = 0.004). The area under the curve(AUC)of cTnI was 0.834,with the cut-off value of 0.49 ng/mL(sensibility=77.6,specificity=80.7). Conclusion The level of cTnI was significantly higher in patients with septic shock. cTnI was significantly correlated to left ventricular diastolic dysfunction in patients with septic shock. cTnI ≥ 0.49 ng/mL could be an available predictor for left ventricular diastolic dysfunction in patients with septic shock.

8.DISTRIBUTION AND ORIGINS OF THE NITRERGIC PERIVASCULAR NERVES IN RAT CEREBRAL ARTERIES

Objective To observe the distribution and origins of the nitrergic perivascular nerves in rat cerebral arteries with a histochemical method for reduced nictinamide adenine dinucleotid phosphate(NADPH) diphorase activity, after resecting the membraneous structure at the ethmoidal formen. Methods Health adult SD rats were randomly distributed to three groups:sham operation group(group A), unilateral (right) operation group(group B), and bilateral operation group (group C). Both group B and C were further divided into two groups: group of resection of membraneous structure and nasociliary nerve (group BⅠ,CⅠ), and group of only removing membraneous structure (group BⅡ, CⅡ). On the seventh day after operation, the pterygopalatine ganglia and otic ganglia of group A were taken, and striped the cerebral arteries of three groups. With the method of NADPH d we investigated the positive cells in pterygopalatine ganglia, otic ganglia, and the changes of positive perivascular nerve fibers. Results A great deal of NADPH d positive cells were found in both pterygopalatine ganglia and otic ganglia. After unilateral operation, the NADPH d positive nerves in the ipsilateral middle cerebral artery(MCA) completely disappeared, but it did not change on the contralateral MCA. The positive nerves on basal artery(BA) and bilateral posterior cerebral artery(PCA), internal carotid artery(ICA), and anterior cerebral artery (ACA), obviously decreased. After bilateral operation, the positive nerve fibers in all arteries except ACA disappeared. There were no significant difference between group BⅠ and BⅡ; CⅠ and CⅡ.Conclusion There were nitrergic nerve fibers in all major cerebral arteries, and the density of positive nerve fibers in the anterior circulation of the cerebral artery was higher than that of posterior one. Different cerebral arteries have different origins of nitrergic nerve. The nitrergic nerve in BA, PCA and ICA completely originated from ipsilateral pterygopalatine ganglia; the nitrergic nerves of MCA were completely originated from ipsilateral pterygopalatine ganglion; ACA have more sources of nitrergic nerves other than bilateral pterygopalatine ganglia. Nasociliary nerve from trigeminal ganglion did not involve in the origins of nitrergic perivascular nerves in rat cerebral arteries. [
9.Projections from respiratory neurons of medulla oblongata to the motor neuron pool of phrenic nerve in rat:An HRP retrograde tracing study

ObjectiveTo observe the projections from respiratory neurons of medulla oblongata to the motor neuron pool of phrenic nerve in rat. MethodsBy using horseradish peroxidase (HRP) retrograde tracing technique after injecting HRP into phrenic nerve, retrograde labelled neurons were found in C3-C5 segment. Then, HRP was injected into the area where the phrenic motor neurons occupied, retrograde labelled neurons were found in medulla oblongata. ResultsPhrenic motor neurons locate in the C3-C5 segment ipsilaterally, occuping the intermediate portion of anterior horn and appearing typical motor neurons. Retrograde labelled neurons were found bilaterally in medulla oblongata, the neurons were located in nucleus retroambigualis (RNA), ventramedial area to nucleus retrofacialis (NRF). ConclusionThe phrenic motor neurons may receive direct projection of respiratory neuron in medulla oblongata, the projecting neurons are distributed in RNA and NRF area.
10.Main progress on studies of pharmacological activities and clinical applications of Guizhi Fuling capsule.

Guizhi Fuling capsule is a traditional Chinese medicine composed of five kinds of medicinal plants, Cinnamomi Ramulus, Poria, Moutan Cortex, Persicae Semen, and Paeoniae Radix Alba. Pharmacology studies have shown that Guizhi Fuling capsule has many activities: anti-inflammatory, analgesic, anti-tumor, regulating smooth muscle, endocrine regulation and enhancing immunity. It achieved obvious effects in the treatment of uterine fibroids, pelvic inflammatory disease, dysmenorrheal, endometriosis, ovarian cysts, breast hyperplasia and other gynecological diseases. This paper reviewed the main progress on studies of pharmacological activities and clinical applications of Guizhi Fuling capsule in recent years.
