OBJECTIVE To study the distributive properties and susceptibility of yeasts to six antifungal agents. METHODS To analyze the distributive properties of 264 clinical Candida spp isolates and study the susceptibility to amphotericin B,nystatin, fluconazole, ketoconazole, miconazole and clotrimazole. The susceptibility of yeasts was tested according to the National Committee for Clinical Laboratory Standards guideline(NCCLS M27-A2). RESULTS Strains of Candida albicans were the most frequent organism isolated accounted for 62.5% of all the isolates.C.tropicalis,C.glabrata,and C.parapsilosis accounted for 20.8%,12.5%,and 1.9%,the others accounted for only 2.3%.The main infected organs were lungs, urinary tract, and digestive tract;the susceptibility tests showed strains of Candida spp to nystatin, amphotericin B,and fluconazole were more active than to the other antifungal agents. The resistance to triazole antifugal agents could be shown. CONCLUSIONS We should strengthen the diagnosis of Candida spp and strengthen the surveillance on susceptibility of clinical isolates Candida spp so as to help the doctors choose the antifungal agents reasonably.

ObjectiveTo explore the imaging features of fetal tuberous sclerosis complex by ultrasonography and magnetic resonance imaging.MethodsRetrospective analysis on the imaging characteristics of the 10 cases of fetuses confirmed as tuberous sclerosis complex who were examined in Hubei Maternal and Child Healthcare Hospital in July 2013 to December 2014 by ultrasonography and MRI, which was compared with the pathological data of specimens and follow-up after birth.ResultsEighteen cases were diagnosed as fetal cardiac rhabdomyoma by ultrasonography among all of the 996 fetuses, in which lesions were located on the ventricular wall near septum or elsewhere in the heart cavity on ultrasonography. Fetal cardiac rhabdomyoma was characterized by circular, homogeneous high echo (singleton in 4 cases, multiple in 14 cases). Among them no subependymal nodule was found by ultrasonographic. Ten cases of subependymal nodule were found by magnetic resonance imaging, which were diagnosed as tuberous sclerosis complex with cardiac rhabdomyomas, including 3 cases of brain subcortical tubers. The subependymal nodules under the lateral ventricle wall showed characteristic low signal nodules on T2WI, protruding from the ependymal surface. Of 18 cases, only 4 cases of fetal cardiac rhabdomyoma were found by MRI. Nine cases of ifnally had termination of pregnancy. Two cases were conifrmed as cardiac rhabdomyoma with intracranial nodules after pathological examination, and 1 case was conifrmed as tuberous sclerosis complex after birth. ConclusionsPrenatal ultrasonography can diagnose fetal cardiac rhabdomyoma successfully, and MRI can diagnose the fetal brain nodules sensitively. Once ultrasonography finds cardiac rhabdomyoma, it may be promising to diagnose fetal tuberous sclerosis complex by ultrasonography combined with MRI.

The purpose of this study is to investigate the enantioselectivity of norgestrel (NG) transdermal permeation and the potential influence of linalool and lipids on the enantioselectivity. In vitro skin permeation studies of NG across the excised rat skins were performed with Valia-Chien diffusion cells, and the permeation samples were analyzed by enantioselective HPLC. The possible enantioselective permeation of NG across intact rat back skin and lipids extracted rat back skin and the influence of linalool were evaluated. The skin permeation rate of dl-NG was two times higher than that of l-NG when donor solutions (EtOH/H2O 2 : 8, v/v) containing l-NG or dl-NG. It may be mainly attributed to the solubility discrepancy between enantiomer and racemate. The enantioselective permeation of dl-NG across intact rat skin was observed when the donor solutions containing dl-linalool. The permeation flux of l-NG was 22% higher than that of d-NG. But interestingly, the enantioselective permeation of dl-NG disappeared under the same experimental condition except that the lipid extracted rat skin was used. Attenuated total reflection-fourier transform infrared spectroscopy analysis of stratum corneum showed that the wave number for asymmetric CH2 stretching vibrations of lipids treated with dl-linalool was greater than that of the control. The results indicated that the enantioselective permeation of NG may be contributed by the interaction between dl-linalool and lipids. More than half of lipids were composed of ceramides. The stereospecific interaction maybe existed among chiral enhancer (linalool), lipids (ceramides) and/or chiral drugs (NG).

A rapid liquid chromatography_tandem mass spectrometric ( LC_MS/MS) method was developed for the determination of aflatoxin B1(AFB1), Deoxynivalenol (DON), Zearalenone (ZEA) in artificial porcine gastrointestinal digested juices, as pigs reacted most sensitively to these mycotoxins. The formula feed was digested by artificial gastric and intestinal juices respectively, then the mycotoxins and adsorbent were added in ratio. After incubation and centrifugation, the supernatant was diluted 10_folds by injection solution and analyzed by LC_MS/MS. The 3 analytes were separated on a reversed phase C18 column using a gradient elution program of aqueous solution containing 0 . 2 mmol/L ammonium acetate and 0 . 1% formic acid methanol. Qualitative analysis was performed using multiple_reaction monitoring ( MRM ) , and quantitative analysis was by internal standard method. Under optimum conditions, the limit of quantitation ( LOQ ) of AFB1 , DON, ZEA was 1, 50, 40 and 0. 3, 50, 20 μg/L in artificial gastric and intestinal digested juices respectively, and the relative standard deviations (RSDs) were below 5. 0%. Then, the thermal stability was studied by incubating the analytes at 39 ℃±0. 5 ℃ for 1, 2, 5 and 10 h, and the results showed 3 analytes were stable under the conditions. Furthermore, the method was applied to evaluate the binding efficacy of 8 mineral benders and 5 organic adsorbents. The adsorbents demonstrated binding efficacy of 85. 1%-96. 5%, 8 . 1%-14 . 7%, 13 . 7%-30 . 0% and 7 . 4%-16 . 6%, 6 . 7%-16 . 2%, 18 . 6%-39 . 0% in gastric digested juice, and 76. 2%-93. 0%, 12. 3%-31. 3%, 0%-23. 2% and 8. 6%-13. 4%, 3. 8%-23. 5%, 24. 9%-34. 8% in intestinal digested juice for these 3 mycotoxins, respectively, with 2 kinds of adsorbents.
