2.Studies on Renal Histological Parameters in SD Rats and Beagles

[Objective] To investigate the renal histological parameters in SD rats and beagles and to supply evidence for renal toxicological and pathological diagnosis. [Methods] Forty SD adult rats and 20 adult beagles (half males and half females) , were respectively executed at 12 weeks and 12 months old. Then, kidney tissue was fixed in 10% neutral formalin, embedded in paraffin, made into slices at 5?m and stained by HE. The larger diameter and shorter diameter of glomerulus in the renal cortex and outer stripe of rats and beagles were determined by Spot Advanced image analysis software. The differences of the above parameters appearing in different species and genders were compared. [Results] The larger diameter was about 96 ?m, and the shorter diameter 76 ?m in the renal cortex of the beagles, and the larger was about 120?m and the shorter 94 ?m in the outer stripe of the beagles. The differences of the larger and shorter diameters were insignificant between the male and female beagles, but the larger and shorter diameters in the outer stripe were longer than those in the cortex. The larger diameter and shorter diameter were about 87 ?m and 64 ?m respectively in the renal cortex of SD rats, and the larger was about 88 ?m and the shorter 64 ?m in the outer stripe of SD rats. The differences of the larger and shorter diameters were insignificant between the male and female beagles, as well as in the outer stripe and the cortex. [Conclusion] The nephron in the outer stripe is larger than that in the cortex of beagles but no difference of the nephron exists in SD rats. The nephron in beagles is larger than that in SD rats, and there is no difference in the nephron between male and female beagles and SD rats.
3.Effects of intervening measures on postpartum depression

Objective To study the related factors of postpartum depression (PPD) and the effects of intervening measures to PPD incidence. Methods 1 597 pregnant women selected from our antenatal care clinic were investigated by using the hospital anxiety and depression questionnaire (HAD) during pregnancy and the Edinburgh postpartum depression scale (EPDS) after childbirth.All the enrolled women were randomlyd divided into control group and intervening group by the proportion of 1 to 2.Six intervening measures were used in the latter group. Results (1) There were 49 women whose HAD≥11 score (anxiety-depression mood)with 28 cases (57.1%) had got postpartum depression in the control group.In the intervening group,however,there were 94 women whose HAD≥11 score with 24 cases (25.5%) had got postpartum depression.There is a significant difference between the two groups ( P

Objective To compare the clinical effect of different materials dentures in repaired of teeth. Methods A retrospective analysis of 570 patients with framework removable partial dentures , which were divided into the Vitallium 2000 group, the pure titanium group and the Co-Cr alloy group randomly from Jan. 2010 to Dec. 2012 in our hospital. Compare the clinical effect of three groups. Results The emplacement rate (χ2=10.283,P<0.05)and the 2-week adjust rate (χ2=16.262,P<0.05)of three groups were statistical different significantly. The incidence rate of denture stomatitis(χ2=12.773,P<0.05),the masticatory efficiency(t=8.194,P<0.05)and the incidence rate of adverse reactions (χ2= 6.028,P < 0.05)of three groups were statistical different significantly. The patients′satisfaction (χ2=2.191,P>0.05)of three groups weren′t statistical different significantly. The degree of comfort(χ2= 6.827,P < 0.05),the speech satisfaction(χ2= 6.002,P < 0.05) and the chewing speech satisfaction (χ2=5.838,P<0.05)three groups were statistical different significantly. Conclusion The clinical effect of the Vitallium 2000 group is better of weight , strength and stability than other groups. The Vitallium 2000 frame removable partial denture is nontoxic , fastness and no peculiar smell , which could be ensure the aesthetics , the degree of comfort and improve the masticatory efficiency , worthy to recommend its clinical use.
6.Preparation of Graphene/Gold Nanoparticles/Glucose Oxidase Flexible Electrode Based on Poly(ethylene terephthalate) Film and Its Application in Detection of Glucose

The graphene prepared by chemical vapor deposition onto copper foils was transferred onto a poly(ethylene terephthalate) (PET) soft substrate by the aid of poly-methyl methacrylate (PMMA).The soft G/AuNPs/GOD composite electrode based on PET substrate was fabricated using a protocol in which the uniform distribution of Au nanoparticles (AuNPs) was firstly obtained by controlling the evaporation of gold sol on a graphene surface, then thioglycolic acid (TGA) was modified on the AuNPs through Au-S bond, and finally glucose oxidase (GOD) was immobilized on the surface of AuNPs through acylation reaction between TGA and the GOD.Glucose was detected in the linear range from 0.05 to 10.55 mmol/L with a linear correlation coefficient (r) of 0.9955.The detection was performed in phosphate buffer solution (pH 7) at 25℃ with a working potential of 0.6 V (vs.SCE electrode), and the detection limit was 1 μmol/L (3σ).The G/AuNPs/GOD flexible electrode based on PET substrate provided a new pathway to detect glucose in special environments using wearable equipment, which enlarged the applied field of glucose detection.
7.Effects of Maidang Rutong granule on l-dopa-induced galactozemia in rats.

OBJECTIVETo study galactagogue effect of Maidang Rutong granule on the lactation rats.
METHODThe experiments were designed to observe the efficiency of Maidang Rutong granule on lactescence, serum prolactin, and morphology of mammary gland with rat galactozemia model established by injecting l-dopa.
RESULTMaidang Rutong granule showed significant enhancement for lactescence and the offspring's body weight. It could antagonize the decrease of serum prolactin and the atrophy of mammary gland induced by l-dopa.
CONCLUSIONMaidang Rutong granule exhibited significant galactagogue effect on the l-dopa-induced galactozemia in rats.

8.Interleukin-23 strengthens the anti-apoptotic and drug resistance of human tongue squamous cell carcinoma through the Wingless-related integration site/β-catenin pathway.

OBJECTIVEThis study aims to detect the expression level of interleukin-23 (IL-23) in tongue squamous cell carcinoma tissues and its relationship with clinical prognosis, as well as explore the anti-apoptotic and drug resistance of the tongue squamous cell line-SCC9 before and after treatment with IL-23.
METHODSThe expression of IL-23 in tumor tissues from 28 tongue cancer patients was analyzed by immunohistochemistry assay. Quantitative real-time polymerase chain reaction (qRT-PCR) was used to detect the mRNA expression of Wingless-related integration site (Wnt)1 and c-myc in SCC9 cells treated with different IL-23 concentrations. After interferencing the β-catenin with small interfering RNA (siRNA), the expression of β-catenin, B-cell lymphoma-2 (Bcl-2), ATP-binding cassette sub-family G member 2 (ABCG2), and permeability-glycoprotein (P-gp) in SCC9 was measured by Western blot analysis. The effect of IL-23 on the apoptotic resistance of SCC9 to cisplatin was examined by methyl thiazolyl tetrazolium test.
RESULTSThe expression of IL-23 in tongue cancer tissues was correlated with lymphatic metastasis, nerve invasion, and the recurrence after therapy (P<0.05). After dealing with IL-23, SCC9 showed the upregulation effect of Bcl-2, ABCG2 and P-gp expressions. IL-23 was closely related to the activation level of the Wnt pathway and significantly strengthened the resistance to cisplatin (P<0.01).
CONCLUSIONIL-23 activates the Wnt pathway in tongue squamous cell carcinoma, thereby enhancing its resistance to apoptosis and drug.

9.Isolation and purification of anaphylactic antibody from guinea pig and preliminary research on its role in passive cutaneous anaphylaxis test (PCA)

Objective To establish guinea pig model of type Ⅰ anaphylaxis, isolate the anaphy-lactic antibody(IgE and IgG) preliminarily from sera of sensitized guinea pigs, and investigate its functional characteristics. Methods Animal model was established by sensitizing guinea pigs with OVA and Al(OH)_3, level of antibody was determined by ELISA, IgE and IgG in sera were preliminarily isolated through saturated ammonium sulphate precipitate and affinity chromatograph of Protein A. A continuous passive cutaneous ana-phylaxis test (PCA) was performed by sensitizing guinea pigs individually with IgE and IgG and then challenging at different time , and the variation of blue spots in skin were observed after challenge . Results Concentration of IgE in model group and control group were 719.3750 ng/ml and 2.5250 ng/ml, the optical density of IgG in model group and control group were 0.9921 and 0.0174, the level of two antibodies in model group were significantly higher than that of control group (P<0.05). In 9 d continuous PCA test, the blue spot induced by IgE in skin lasted for 9 days and appeared the largest when challenged at day 5. The diameter of blue spot induced by IgG was the largest when challenged at day 2 and then decreased fast. Con-clusion Anaphylactic antibodies were successfully preliminarily isolated from sera of sensitizing guinea pig, both IgE and IgG play roles in type Ⅰ anaphylaxis of guinea pig, the hypersensitive reaction induced by IgG is fast and short than that induced by IgE, and IgG may become an important surrogate marker in immunotoxic-ity evaluation(type Ⅰ anaphylaxis)of vaccine.
10.Three-dimensional contrast-enhanced ultrasonic cholangiography in cadaver liver

Objective To assess the feasibility of three-dimensional contrast-enhanced ultrasonic cholangiography(3D-CEUSC) in cadaver liver.Methods The 3D-CEUSC was performed in 6 cases of cadaver liver.Image quality of 3D-CEUSC was evaluated.The visualization of branching orders,the degree of visibility and coincidence of morphous were compared with those of cholangiagraphy using fluoroscopy.Results The imaging quality of 3D-CEUSC was inferior to that of cholangiography with significant difference.The three-dimensional biliary tree structures were visualized in all 6 3D-CEUSC.The maximum visualization of branching orders in 3D-CEUSC was (3.67±0.52),which was equal to the results(4.00±0.63)by cholangiography (P=0.465).The degrees of visibility of biliary tree were equivalent with those by cholangiography in the first and second order with significant difference.The coincidence of morphous was excellent compared the images of 3D-CEUSC with direct X-ray cholangiography.Conclusion 3D-CEUSC is a new technique as a useful supplement to cholangiography in evaluation of biliary anatomy and variation before graft harvesting in LDLT.
