3.The study on microRNA-451 expression and its association with gastric cancer

Objective To investigate the relations between microRNA-451 (miR-451) expression and gastric cancer (GC).Methods Expression of miR-451 in tumor and paired non-cancerous tissues was measured by real-time quantitative polymerase chain reaction.The association of miR-451 expression with clinical pathological characteristics and prognosis was statistically analyzed;in addition,slow virus LV-mir-451 was applied to infect human gastric cancer cell line BGC-823 and MKN-45,and construct miR-451 over-expressed stable cell lines.Finally,to detect cell proliferation,migration and invasion.Results miR-451 expression decreased in GC tissues vs.pericarcinous tissues (0.010 ± 0.007 vs.0.014 ± 0.008,t =3.015,P < 0.05),and its down-regulation positively correlated with lymphatic metastasis,clinical staging and shorter overall survival.miR-451 expression in BGC-823 and MKN-45 cells did not impact on cell proliferation,but did reduce migration rates (0.863 ±0.026 vs.1.107 ±0.032,t =9.820,P<0.05;0.863 ±0.026 vs.1.060±0.054,t =4.369,P<0.05) and invasion rates (1.040 ± 0.055 vs.1.522 ± 0.001,t =5.84,P < 0.05;1.040 ± 0.055 vs.1.696 ± 0.078,t =10.16,P < 0.05) in BGC-823 cells.Conclusions miR-451 may act as a prognostic marker in GC.
4.Observation on osteoblasts responded to fluid induced shear cultured on different surfaces with scanning electron microscope.

OBJECTIVETo investigate morphological change of osteoblasts cultured on titanium plates with different microarchitecture structure when exposured to fluid shear stress.
METHODS14 dynes x cm(-2) fluid shear stress was applied on osteoblasts cultured on 3 different commercially pure titanium plates: Polished treatment (PT), sandblast (SB), sandblasting and acid-base (SB-AB) surfaces. Scanning electron microscope (SEM) was adopted to observe the morphological changes after 0.5, 4, 7.5 h time point respectively.
RESULTSMorphologically, no significant changes were observed after 0.5 h and few osteoblasts were seen after 7.5 h on all 3 type of different surfaces, and significant changes could only be observed after 4 h. Osteoblasts were elongated and rearranged along the flow way on different levels on PT surface. Shape of cells was altered, from long fusiform suspending over depressed areas into polygon stretching out many synapsises tightly attached to pits on SB-AB surface. Osteoblasts on SB surface displayed similar change as SB-AB surface, besides, some cells were elongated along the way of flow, stretching out threadlike synapsises attached to edges of pits.
CONCLUSIONMorphological change of osteoblast responding to fluid shear stress in physiological range depends on substrate microarchitecture and varies with the time of fluid shear stress application.

5.Effect of early intensive insulin therapy on immune function of aged patients with severe trauma.

This study examined the effect of intensive insulin therapy on immune function and inflammatory factors at the early phase after severe trauma. At day 1, 3, 5, 7 after admission, subsets of CD4(+) helper T lymphocytes (Th1/Th2) and human leukocyte antigen (HLA)-DR expression on CD14(+) monocytes were flow cytometrically measured. Levels of cytokines, including tumor necrosis factor-alpha (TNF-α), interleukin-6 (IL-6), interleukin-10 (IL-10) and other immunity markers, such as IgA, IgG, IgM, C3, C4 and C reaction protein (CRP) were examined in two groups. The results showed that TNF-α, IL-6 and CRP levels in the intensive insulin therapy group were significantly lower than those in the conventional therapy group, whereas IL-10 levels were substantially increased after intensive insulin therapy. C3 level at day 3, 5, 7 and C4 levels at day 5, 7 were lower in the intensive therapy group than in the conventional therapy group. Th1/Th2 ratios decreased gradually over time in both groups, and were much lower at day 3, 5, 7 in intensive therapy group. There were significant differences among day 3 to day 7 after admission in HLA-DR expression in CD14(+) monocytes. It was concluded that the intensive insulin therapy could decrease pro-inflammatory cytokines and increase anti-inflammatory cytokines in the elderly suffering from severe trauma, at the same time, with complement recovery being delayed. Moreover, intensive insulin therapy promoted immune suppression and, therefore, measures need be taken to address the issue.
6.Influence of zirconia content on translucency of zirconia-toughened alumina glass-infiltrated ceramic.

OBJECTIVEThe objective of this study is to investigate the influence of zirconia content which is 0-30.0% weight percentage of matrix on translucency of zirconia-toughened alumina glass-infiltrated ceramics.
METHODSSeven groups were divided according to different weight percentage of zirconia (0, 2.5%, 5.0%, 7.5%, 10.0%, 20.0% and 30.0%). After sintering, infiltrating and polishing, spectral transmittance was determined with spectrophotometer under D65 standard source. Contrast ratio was also tested by whiteness colorimeter.
RESULTSWith mass fraction of zirconia increasing from 0 to 30.0%, spectral transmittance reduced from 0.406% to 0.058%, while contrast ratio value increased from 0.849 +/- 0.005 to 1.015 +/- 0.006. When zirconia content was 10.0%, contrast ratio was 0.990 +/- 0.008. When it was more than 10.0%, transmission rate of the downward trend and contrast ratio of the rising trend became flat.
CONCLUSIONZirconia content has a direct impact on translucency of zirconia-toughened alumina glass-infiltrated ceramic, which is essentially opaque when zirconia content is 10.0%. When mass fraction of zirconia is more than 10.0%, the influence of zirconia content is reduced.

7.Investigation of the function of the glass colorant on the machinable infiltrated ceramics color.

OBJECTIVETo investigae the function of the glass colorant on the color of the machinable infiltrated ceramics(MIC).
METHODSFive kinds of glass with different colorant were infiltrated through the aluminous matrix by heating the components to 1 100 degrees C for 2 hours. The specimens surface was polished, and their thickness was 0.5 mm.
RESULTSThe refractive index of the MIC infiltration glass was 1.59691 (587.6 nm, nd) . The most different parameter of the MIC color were L*, then a*, and b* had little difference . The parameters of the color space of MIC were: L*(64.55-71.46), a*(3.35-7.38), b*(10.00-12.41), Ca*b*(11.38-13.95), ha*b*(54.07-73.00). These were almost close to the color parameters of Vita In-ceram.
CONCLUSIONThis experiment proved that the glass colorant was changed the MIC color parameters, and the main function was on L*, then a*. The ceramic color was up to the requirement of clinic.

8.Effects of enteral nutrition on uptake of amino acid and enzyme-protein synthesis of pancreatic acinar cell in acute pancreatic dogs.

OBJECTIVETo evaluate the effect of intrajejunal nutrition on uptake of amino acid and enzyme-protein synthesis in pancreatic acinar cell and subcellular fractionation and zymogen granules in dogs with acute pancreatitis.
METHODSFifteen dogs were induced acute pancreatitis by retrograde injection of 5% sodium-taurocholate into the pancreatic duct. Radioactive tracing and electron microscope were used to evaluate the change of amino acid uptake, enzyme-protein synthesis in acinar cell, subcellular fractionation, the quantitative analysis of mean zymogen granule number and mean zymogen granule area after injection L-(3)H-phenylalanine 30, 60, 120 1nd 180 min on the 7(th) day.
RESULTSThe radioactivity of L-(3)H phenylalanine uptake by pancreatic acinar cells and incorporations of L-(3)H phenylalanine into newly synthesized enzyme-protein peaked at 60 min. In enteral nutrition (EN) group it was higher that that in parenteral nutrition (PN) group (P < 0.05), and then gradually declined. The radioactivity peaked at 60 min in zymogen granule, lysosomal-mitochondria and microsomal subcellular fractionation. The latter two decreased, bat there was no significant difference (P > 0.05). The change of the mean number and mean area of zymogen granules were not significant different between the EN group and PN group (P > 0.05).
CONCLUSIONEN or PN do not stimulate pancreatic acinar uptake amino acid and enzyme-protein synthesis in acinar cell and subcellular fractionation.

9.Characterization and antibacterial effect of Ag-nHA-nTiO2/polyamide 66 nanocomposite membrane on oral bacteria.

OBJECTIVEUndried silver-hydroxyapatite-titania (Ag-nHA-nTiO2) nanoparticles slurry was used to make membrane with polyamide 66 (PA66) by co-polymerization method. The purpose of this study is to test the physical and chemical characteristics and antibacterial ability.
METHODSThe morphology, chemical components and structures of the membrane were characterized by atomic absorption spectrometer (AAS), X-ray diffraction (XRD), scanning electron microscope (SEM) and energy-dispersive X-ray analysis (EDX). Staphylococcus aureus (S. aureus), Escherichia coli (E. coli), Porphyromonas gingivalis (P. gingivalis), Fusobacterium nucleatum (F. nucleatum) and Streptococcus mutans (S. mutans) were utilized to test the antibacterial effect.
RESULTSXRD results demonstrated that the membrane have characteristic diffraction peaks of pure hydroxyapatite (HA). A homogeneous distribution of the Ca, P, Ti and Ag element in the membrane was confirmed by EDX. Both surface and section showed porous structure which was confirmed by SEM and the average hole size was 20-30 microm. The bacteria assay reflected to the antibacterial effect, 50.10% of S. aureus and 56.31% of E. coli were killed. However, 91.84% of P. gingivalis, 90.64% of F. nucleatum and 90.49% of S. mutans were killed and pictures of SEM showed obviously fewer cells on the surface.
CONCLUSIONThe nanocomposite membrane could be one of the bioactive materials with antibacterial properties for oral guided bone regeneration technique.

10.Clinical features of secondary epilepsy: a report of 68 cases

Objective To explore the clinical features of epilepsy after stroke. Methods One thousand and eighty-two patients with stroke, admitted to our hospital from June 2002 to December 2008,were chosen. The clinical data of 68 of these patients having epilepsy after stroke were collected; the clinical characteristics, seizure types and locations were summarized; the relations between epilepsy and both the stroke types and the locations of stroke lesions were analyzed. Results In patients with secondary epilepsy, the lesion of seizure mainly located in the temporal lobe with simple partial seizure as its main seizure type; patients with epilepsy secondary to cerebral hemorrhage had a higher prevalence as compared with those secondary to cerebral infarction and subarachnoid hemorrhage (P<0.05). Patients with secondary epilepsy having their lesions in the lobes had a higher prevalence as compared with those having their lesions in other locations (P<0.05) Conclusion Stroke patients having cerebral hemorrhage and lesions in the lobes are likely to have secondary epilepsy; the lesion mainly locates in the middle of the temporal lobe; simple partial seizure is common.
