3.Physiological S-phase Checkpoint
Progress in Biochemistry and Biophysics 2006;0(02):-
Cell cycle checkpoints are protective mechanism responding to DNA damages originated from externalor internal factors. When cells are exposed to genotoxic stress or when nutrition crisis occurs, cell cycleprogression is usually stopped or slowed down by cell cycle checkpoints to allow for DNA repair or for handlingthe crisis. Besides, recent studies suggest that some cell cycle checkpoint proteins are also involved in regulatingphysiological DNA replication via controlling the rate of DNA replication. Cell cycle checkpoint proteins ATR,9-1-1 complex, Chk1, Cdc25A and CDK2 may participate in this process. This kind of regulation is supposed to bevery important for ensuring accurate DNA replication and maintaining genomic stability.
4.EXPERIMENTAL STUDIES ON BOLBOSTEMMOSAPONINS USED AS INTRAVAGINAL SPERMATOCIDAL AGENTS
Journal of Xi'an Jiaotong University(Medical Sciences) 1981;0(03):-
This experiment has proved that bolbostemmosaponin and its constituents A and D have relatively powerful spermatocidal effects. They can kill all spermatozoa in a moment at the concentrations of 0.04%, 0.04% and 0.03% respectively. These spermatocidal agents mainly distroy the biomembrane system of sperm. Using the fixed gelatin membrane technique proved that all of them can strongly inhibit tne activity of a single sperm acrosin at the concentration of 0.05%.

Routine treatment group was treated with general therapy, such as gastrdavage, catharsis and so on. While hemoperfusion was added to HP treatment group besides general therapy. Results The number of cure in HP treatment group was higher than that in routine treatment group(? 2=8.17 P
7. Role of extracellular signal-regulated protein kinase in estrogen-induced proliferation of breast cancer cell line MCF-7
Academic Journal of Second Military Medical University 2010;30(4):395-399
Objective: To investigate the role of extracellular signal-regulated protein kinase (ERK) in estrogen-induced proliferation and cell cycle transformation of breast cancer cell line MCF-7 and the related mechanisms. Methods: Estrogen receptor-positive breast cancer cell line MCF-7 was used in our study. The effects of 17β-E2 on the proliferation of MCF-7 cells was investigated by MTT assay to determine the optimal concentration of 17β-E2 for the following experiment. The effect of PD98059 on 17β-E2-induced proliferation of MCF-7 cells was measured by MTT assay to determine the intermediate concentration of PD98059. The cell cycle was analyzed by flow cytometry and telomerase activity was determined by Telomerase repeat amplification protocol PCR (TRAP-PCR) silver staining. The expression of wild-type p53 and phosphorylated ERK1/2 protein was determined by Western blotting and the expression of wild-type p53 mRNA was detected by RT-PCR. Results: ERK phosphorylation inhibitor PD98059 inhibited the proliferation of MCF-7 cells treated with 17β-E2 in a time- and dose-dependent manner(P<0.01). The intermediate concentrations of PD98059 were 89.28 μmol/L for 24 h, 39.81 μmol/L for 48 h and 21.87 μmol/L for 72 h. Treatment with 20 μmol/L PD98059 for 48 h reversed the promoting effect of 17β-estradiol on the cell cycle transformation of MCF-7, increasing the number of G1 phase cells and decreasing the number of S and M phase cells (P<0.01), inhibited the enhancing effect of 17β-E2 on the telomerase activity of MCF-7 cells(P<0.05), increased the protein expression level and genetic transcription of wild-type p53 (P<0.01), and decreased the expression of p-ERK1/2 protein(P <0.01). Conclusion: ERK plays an important role in 17β-E2-induced proliferation and cell cycle transformation of breast cancer cell line MCF-7, which might be related to the changes of genetic transcription of wild type p53 and telomerase activity.
9.Effects of Shuangshen oral liquid on glucocorticoid-induced osteoporosis rats
Chinese Journal of Tissue Engineering Research 2007;0(41):-
BACKGROUND: Shuangshen oral liquid consisting of renshen and danshen is firstly used to treat glucocorticoid-induced osteoporosis. OBJECTIVE: To establish a rat osteoporosis model through glucocorticoid and to observe the preventive effects of shuangshen oral liquid (SS) on bone substance and bone mass. DESIGN, TIME AND SETTING: A completely randomized controlled grouping animal experiment was performed in the Animal Experimental Center and Department of Pharmacology, Guangdong Medical College from June to September in 2008. MATERIALS: High-dose SS consisted of 3.6 kg danshen, 5 kg renshen, and then prepared by water extraction twice, and the concentration was 870g/L. On this basis, high-dose SS was diluted three times to obtain low-dose SS with a concentration of 290 g/L. The prednisone acetate raw medicine was purchased from Zhejiang Xian Ju Pharmaceutical Co., Ltd. METHODS: Thirty-two 3-month-old male SPF rats were randomly divided into 4 groups, and every group had 8 rats. The normal control group rats were fed with distilled water 5 mL/(kg?d) by gavage. Rats in prednisone group were treated with 0.7 g/L prednisone suspension (5 mL/kg per day) by gavage. SS group rats were treated with 0.7 g/L prednisone suspension (5 mL/kg per day) by gavage and then given the high does and low does SS (5mL/kg per day) respectively by gavage 3 hours later. The rats were sacrificed by heart hemospasia 88 days later. MAIN OUTCOME MEASURES: Body mass, bone mineral content and bone hydroxyproline content per unit bone, bone mineral density in the left femur, biomechanics indexes of the right femur. RESULTS: The long-term administration of prednisone restrained the increasing of rats body mass (P
