1.Antifungal susceptibility and genotypes of Candida albicans

ObjectiveTo test the susceptibilities of Candida albicans to 5 antifungal agents including methylrosanilinium chloride and to estimate their relationship with the genotypes of C.albicans.MethodsThe susceptibilitiesof 67C.albicansisolatestomethylrosaniliniumchloride, fluconazole, amphotericinB,itraconazole and ketoconazole were determined by using a broth dilution method as described in the National Committee for Clinical Laboratory Standards(NCCLS) document M27-A2.DNA was extracted from the C.albicans isolates and subjected to the amplification of 25S ribosomal DNA(rDNA) introns by PCR.The C.albicans isolates were divided into A,B and C genotypes according to the amplicons.The relationship was analyzed statistically by exact probability test between the genotypes and antifungal susceptibility of C.albicans.ResultsOf the 67 C.albicans isolates,8.96% were resistant to fluconazole,2.98% to itraconazole,1.49% to ketoconazole,and no resistance to amphotericin B was observed.The minimum inhibitory concentration (MIC) of methylrosanilinium chloride varied from 0.125 to 4 mg/L.Statistical analysis revealed no significant correlation between the genotypes and autifungal susceptibilities of C.albicans(all P > 0.05).Conclusions More attention should be given to the increasing resistance to fluconazole and itraconazole in some isolates of C.albicans as well as to the favorable antifungal activity of methylrosanilinium chloride.The antifungal resistance shows no apparent correlation to the genotypes of C.albicans.
2.Relationship between phospholipase expression and fluconazole resistance in Candida albicans

Objective To explore the relationship between antifungal resistance and expression of a virulence factor phospholipase B1 in Candida albicans. Methods Total RNA was extracted from 15 fluconazole-resistant and 15 fluconazole-sensitive Candida albicans isolates. RT-PCR was performed to detect the expression of phospholipase B1 mRNA. Extracellular and intracellular proteins secreted by Candida albicans cells were concentrated using salting-out method and extracted by P0013B RIPA lysis buffer, respectively followed by Western-blotting analysis to detect the expression of extracellular and intracellular phospholipase B1 protein. Results The relative expression of phospholipase B1 mRNA was 0.6173 ± 0.1090 in fluconazoleresistant Candida albicans isolates, significantly higher than in fluconazole-sensitive isolates (0.2653 ± 0.0935,P < 0.05). Increased expressions of extracellular and intracellular phospholipase B1 protein were noted in fluconazole-resistant Candida albicans isolates compared with fluconazole-sensitive isolates (0.4145 ± 0.2773 vs 0.2720 ± 0.2194, 0.1825 ± 0.1831 vs 0.2653 ± 0.0935, t = 2.703, 3.443, both P < 0.05). Conclusions The expressions of phospholipase B1 mRNA and protein are elevated in fluconazole-resistant Candida albicans isolates, suggesting that the expression of phospholipase B1 may be associated with the occurrence of antifungal resistance in Candida albicans.
4.The significance of the expression and distribution of axon guidance factors Netrin-1 in Hirschsprung' disease

Objective To observe the expression and the distribution of Netrin-1 in the spastic and normal segment of colon in patients with Hirschsprungs disease (HD) and investigate the relationship between Netrin-1 and development of HD.Methods Colon specimens of 36 cases with HD were selected for this study.Normal segment of colon served as the control group,while the spastic segments of colon as the lesion group.Immunohistochemical staining was applied to detecte the expression and the distribution of Netrin-1.Results Immunohistochemical staining showed that the expression of netrin-1 was positive in the intestinal muscular layers (longitudinal muscle,circular muscles),myenteric ganglias and mucosal layers in the control group.However,the expression of Netrin-1 was also positive in muscular layers and mucosal layer in the lesion group.There was no significant difference in the expression intensity of Netrin-1 between the two groups (P >0.05).Conclusion The expression of Netrin-1 in both the control group and the lesion group may indicate that netrin-1 have no significant correlation with genesis of HD.
6.Nursing strategy of primary PCI in the aged with acute coronary syndrome

7.Present situation and influencing factors analysis of the academic professional development for teachers who providing foundational courses in medical college

Objective To evaluate the status of the influence factors of foundational course teachers' academic profession development in medical college.Methods The survey includes academic promotion and academic innovation.In aspect of academic innovation,a total of 68 foundational course teachers in medical college were selected.The subjects were tested by scale of exterior influencing factors on innovative behavior,the assessment scale of innovative behavior and the organizational innovation climate scale,the scientific spirit scale of the scientific and technological talents.The impact of demographic characteristics on academic promotion was also tested.Results In aspect of academic promotion,teachers with different department and education showed statistically significant difference in titles(P<0.05).The age of teachers' promoted to professor is 40-50 years old,vice professor is 30-40 years old.Generally,it takes 6-10 years to promote to senior titles,and it takes less time for doctors.In aspect of academic innovation,scale was used in 5 points,the lowest 1 point,the highest 5 point.The mean score of the scale of exterior influencing factors on innovative behavior is mission design3.28 ±0.96,innovation investment 2.53±0.74,innovation environment 3.10±0.65.The mean score of the assessment scale of innovative behavior and the organizational innovation climate scale is organization target cognition 3.10 ± 0.74,colleagues relationship 3.44±0.82,team innovation motivation 3.34±0.83.The mean score of the scientific spirit scale of the scientific and technological talents in spiritual science dimension is 3.40±0.67 points.The projects with low scores are distributed in three dimensions of innovation investment,innovation environment and organization target cognition.Conclusions Teachers' academic promotion is related to the department and education.In aspect of academic innovation,teachers' self-evaluation of scientific spirit is higher,the evaluation of the mission design,colleagues relationship,team innovation motivation is also higher,the evaluation of the innovation environment,organization target cognition,innovation investment is lower.Material resources,research cooperation and so on especially should be improved.

9.Smac Involved in Promoting TGF-β1 Treated Mesangial Cells Apoptosis Induced by Triptolide

Objective To investigate the effects of Triptolide on apoptosis of cultured rat mesangial cells treated by TGF-β1 and the role of Smac in this process. Methods The mesangial cells were pre-treated with different concentrations of Triptolide for 24 hours, then stimulated with TGF-β1 for 24 hours. Apoptotic cells were detected by TUNEL assay. Smac transcription level was determined by Real time-PCR analyses. Smac expression level was assessed using Western blot anal-yses. Localization of Smac was shown by confocal fluorescence microscopy. Results Compared with control group, TGF-β1 inhibited apoptosis and Smac transcription and expression in rat mesangial cells. By contrast, Triptolide promoted mesangial cells apoptosis. In Triptolide groups, Smac mRNA and protein levels were up-regulated. Additionally, in normal and TGF-β1 groups Smac protein was mainly localized in mitochondriawhile in Triptolide groupit was mainly localized in cytoplasm and nucleus with increased fluorescence intensity. Conclusion Triptolide could promote the effect that TGF-β1 inhibited apop-tosis of mesangial cells, through both up-regulation the expression of Smac and stimulating it translocation from mitochon-dria to cytoplasm and nucleus.
