ObjectiveTo study the anti-proliferative effect and the mechanism of arotinoid Ro40-8757 on human pancreatic carcinoma cell line JF-305.MethodsMTT assay was used to evaluate the growth inhibition effect of Ro40-8757 on human pancreatic carcinoma cell line JF-305. The expression of p27 protein was measured by Western blotting, and flow cytometry for cell cycle analysis.ResultsRo40-8757 significantly inhibited the growth of JF-305 cells with a dose-, time-dependent manner. After 72 hours treatment with 10 -5mol/L Ro40-8757, the proportion of cells in G 1 phase increased from 56% to 76% while the cells in S phase decreased from 25% to 7%. Western blot analysis showed that the expression of p27 protein in cells treated with Ro40-8757 increased markedly.Conclusion Ro40-8757 inhibits the growth of pancreatic carcinoma cell line JF-305 possibly by up-regulating p27 protein expression and arresting cell cycle.

A rapid and sensitive method based on biotin-avidin mediated competitive enzyme-linked immu nosorbent assay(BA-ELISA) was established for the determination of ketamine.The optimal concentration of coated antigen and anti-ketamine monoclonal antibody were found to be 2.0 and 10.2 mg/L.The concentra tions of biotinylated goat anti-mouse IgG(Biotin-IgG) and streptavidin-horseradish peroxidase(SA-HRP) were optimized and the optimum results were found to be 0.29 and 1.0 mg/L, respectively.The linear range of the presented method was from 0.1 to 1000 μg/L, and the limit of detection was found to be 0.03 μg/L.The recoveries of ketamine spiked in human serum and urine were between 94% and 102%.Comparing the result of traditional ELISA, the present BA-ELISA method had a lower detection limit for ketamine.The experimental results indicated that the present BA-ELISA method was specific and sensitive.

Objective To investigate the imaging features of adrenal rest tumor.Methods Twelve patients of adrenal rest tumor proved by surgery or clinical diagnosis were retrospectively analyzed.Among these 12 patients,12 were examined with ultrasound,11 with MR and 1 with CT. MR and CT were performed without and with intravenous injection of contrast material.The imaging features of adrenal rest tumor were retrospectively summarized and the relevant literatures reviewed. Results The adrenal rest tumors were found in testis in 10 of the 12 patients,and in ovaries and broad ligament in the remaining two.The imaging features of the testicular adrenal rest tumor were summarized as following:all patients had bilateral testicular masses without change of the testicular contour. On ultrasonography,the lesions were hypoechoic, with some hyperechoic areas and appeared highly vascularized on Colour Doppler ultrasonography.The masses showed iso-density on plain CT,and avid enhancement on post-contrast CT images.The masses ranging in size from0.7 cm×1.0 cm×2.2 cm to 2.3 cm ×2.7 cm ×2.9 cm with uniform signal intensity,lobulated margin on MRI.They exhibited iso- or slight hyperintensity on T1 WI and hypointensity on T2WI relative to normal testicular parenchyma.The tumors showed intense enhancement on post-contrast MR images. No abnormality was detected with Colour Doppler uhrasonography and MR in 2 patients of adrenal rest tumor in ovaries and broad ligament. Conclusion Combining imaging features with the typical clinical history,the diagnosis of adrenal rest tumor could be suggested pre-operatively.

Objective To evaluate a new computer-aided technique applicable for myocardial contrast echocardiography(MCE) to quantitate automatically calibrated myocardial contrast intensity(CD and to test the feasibility of calibrated CI in assessing myocardial perfusion. To analyze the relationship on myocardial perfusion,regional contractile function and cell apoptosis in stunned myocardium. Methods According to coronary occlusion and reperfusion at different times, rabbits were divided into three groups: 15 min occlusion/30min reperfusion (group Ⅰ ),30 min occlusion / 60min reperfusion (group Ⅱ ) and 120 min occlusion / 60min reperfusion (group Ⅲ ). MCE was performed on all rabbits at baseline,occlusion and after reperfusion,and its images were analyzed by a new computer-aided technique. Myocardial calibrated CI of each segment was measured automatically by software. Percentage wall thickening (WT) of each risk segment at each stage were also measured by echocardiography. The apoptotic index(AI) in regional left myocardial dysfunction was calculated by terminal deoxynucleotidyl transferease-mediated biotinylated deoxyuridine triphosphate nick end labeling(TUNEL ). Results (1) During occlusion, WT in the areas at risk decreased to zero or negative and the calibrated CI values were significantly lower than those at baseline ( P ＜0.01 ). After reperfusion, WT in all risk segment remained depressed, but calibrated CI significantly improved in group Ⅰ and Ⅱ while those remained unchanged in group Ⅲ. (2)AI in risk myocytes were (13. 70 ± 5.48 ) %, (36.25 ± 5.55 ) % and ( 62.06 ± 6. 70 ) %, respectively, both statistically significant difference between the two groups ( P ＜0.05 or P ＜ 0.01 ). AI were negatively correlated to WT and calibrated CI ( r = - 0. 87 and r = - 0. 77, P ＜0.05). Conclusions MCE with computer-aided technique can assess quantitatively myocardial perfusion and regional contractile function. Short-term ischemiareperfusion does not cause myocardial necrosis, but it will lead to myocardial cell apoptosis and the phenomenon of myocardial stunning. Prolonged ischemia, even if given sufficient reperfusion, can lead to apoptosis and necrosis simultaneously.

Objective:To probe the method of isolating outer hair cells (OHC) from each of four turns of the guinea-pig cochlea. Method:From eight guinea pigs the organ of Corti from each of four turns of the cochlea were dissected, and then treated using enzyme. Result:A fair amount of living OHCs from each of four turns were obtained. The length of OHCs from each of four turns were 23.81,34.50,60.48 and 71.37 μm. Conclusion:The key to success in isolating OHCs from each of four turns of the cochlea is to know very well the anatomical characteristics of each of four turns of the cochlea and be operated in accordance with normal rules.

AIM: To approach the antiobesity action and mechanisms of the daidzein derivative: LRXH609(LRX).METHODS: The body weight,Lee′s index,total weight of celiac fat tissue,food intake,serum glucose and lipids in obese rats induced by a high-fat diet were measured and the antiobesity action was tested after LRX was administered for 30 days.3T3-L1 pre-adipocytes were induced by in vitro culture,the effects of LRX on cell proliferation,lipogenesis,lipolysis were observed.RESULTS: The body weight,Lee′s index,fat tissue weight in obese rats were significantly decreased by LRX,and the concentrations of TC,FFA in serum were decreased,the proliferation of 3T3-L1 pre-adipocytes was inhibited,the activities of hormone sensitive lipase in 3T3-L1 pre-adipocytes were significantly elevated,the glycerine release from adipocytes was promoted and the concentrations of TG in adipocytes were decreased.CONCLUSION: LRX plays a role in antiobesity action and regulating blood lipid and the mechanism might be related to inhibiting proliferation and differentiation of pre-adipocytes,stimulating TG decomposition by activating hormone-sensitive lipase and decreasing the TG storage in adipocyte.

Objective To understand the performance of the basic medical security system in Guangxi for decision-making support on the development planning of the system during the 12th FiveYear Plan in Guangxi.Methods Descriptive statistics were made to analyze the annual report data of Guangxi's basic medical security system during 2009 to 2011,regarding the basic medical security for urban workers and that for urban residents,as well as those of the new rural cooperative medical system.Results The survey found that the three basic medical security systems in Guangxi have almost achieved full coverage; the compensation ratio keeps rising as the inpatient expenses of urban residents was up to 62.21％,64.96％ and 66.96％ respectively from 2009 to 2011,and that for urban residents up to 34.26％,39.96％ and 49.68％ respectively; the ratio for those covered by the new rural cooperative medical system has been maintained at 45％ for the three years; the coverage of the systems keeps expanding.Conclusion Recommend to integrate pilots of the three security systems and carry out serious diseases pilot programs; encourage reform of the payment system at institutions at or above county level; reform the risk-pooling of disease management of at primary medical institutions; strengthen the information systems of basic medical insurance system,in an effort to ensure the sound development of the basic medical insurance system.

Objective To investigate the left ventricular(LV) systolic hemodynamics and the distribution of blood flow in patients with hypertrophic cardiomyopathy(HCM) by vector flow mapping(VFM).MethodsThirty-five random non-obstructive HCM patients(case group) and forty healthy volunteers (control group) were enrolled.Peak systolic velocity (Vs),peak systolic flow (Fs) and total systolic negative flow (SQ-) of two groups were compared.Velocity vector,streamline and vortex distribution of two groups were observed.Results Vs,Fs,SQ- of the control group and Vs,Fs of the case group all showed a decreasing trend from the basal segments to the apical segment(P<0.05).SQ- of the case group lost this decreasing tendency.Vs,Fs of all segments and SQ- of the middle and apical segment of the case group were higher than those of the control group.SQ- of the basal segment of the case group were significantly lower than that of the control group(P＜0.01).Distribution of blood flow:The velocity of blood flow was faster in the case group than that in the control group and the direction was disordered.The streamlines of the case group were discontinuous,and vortexes in the case group were more than that in the control group.Conclusions VFM technology could quantitatively evaluate LV systolic hemodynamics in patients with HCM,and can clearly show the distribution of blood flow.
