Objective To explore the effect of low-dose ketamine combined with propofol anesthesia on expres?sion of glutamine receptor subunit 1 (GluR1) and 2 (GluR2) in the hippocampus of depressed rats after electroconvulsive therapy. Methods Healthy adult male Sprague-Dawley rats, weighing 200~250 g, were used in this study. Mental depres?sion was induced by chronic unpredictable mild stress. Thirty-two depressed rats were randomly divided into 4 groups (n=8): metal depression group (group A), ECT group (group B), ECT+propofol group (group C) and ECT+propofol+ket?amine group (group D). Eight normal rats served as control group. Control group received no treatment. Group A received intraperitoneal injection of normal saline 8 mL/kg plus sham ECT. Group B, C and D received ECT once a day for 7 con?secutive days following intraperitoneal injection of normal saline 8 mL/kg, propofol 80 mg/kg and propofol 80 mg/kg +ketamine 10mg/kg, respectively. Sucrose preference test and Morris water maze were performed to assess depressed be?havior and learning and memory function, respectively. RT-PCR and Western-blot assay were used to detect the expres?sion of GluR1 , GluR2 and their mRNA expression. Results After ECT, compared with control group and group A, changes of SPP in group B, C and D were obvious. The change of SPP in group D was much higher than all other groups (P<0.05). Rats in group B showed prolonged escape latency and shortened space exploration time, which were significantly different from all other groups (P<0.05). Rats in group D showed the most shortened escape latency and prolonged space exploration time (P<0.05). The expression of GluR1 was significantly increased in group B, C and D compared with group A (P<0.05). The expression of GluR2 and mRNA was significantly decreased in group B and C (P<0.05). The difference in GluR2 and mRNA expression was not significant among group A, D and control group (P>0.05). Conclusion Low-dose ketamine combined with propofol anesthesia exert effective antidepressive action and improve learning and memory function of depressed rats after electroconvulsive therapy. The beneficial effects of the ketamine combined with propofol anesthesia may be related to up-regulation expression of GluR1 and GluR2 in hippocampus.

Objective To investigate the anxiety and depresson status of pregnant women with gestationul diabetes mellitus (GDM), to explore the correlated factors and harmful result among these patients, and to provide a theoretic basis for prevention and treatment of the anxiety and depresson status of pregnant women with geststional diabetes mellitus. Methods 95 cases with GDM were investigated by using Self Rating Anxiety Scale (SAS) and Self-rating Depression Scale (SDS). Results Anxiety rate was 47.4% and depression rate was 33. 4% in patients with GDM. In the screening examination, patients with anxiety had blood glucose, fast blood glucose, and OGTT at 0 min., higher than those in patients without anxiety. And in the screening examination, patients with depression had higher levels of blood glucose, fast blood glucose, two-hour-later-after-a-meal blood glucose, OGTT at 0 min. and at 120 rain. than non-depression patients. Conclusions GDM Patients with anxiety and depresson have higher level blood glucose, and the control of their metalism is poor. It should pay a high attention to the negative mood of them and try to take effective intervention measures as early as possible to avoid the occurrence of a bad pregnant result.

Acta Academiae Medicinae Sinicae 2001;23(2):196-198
Objective To investigate the factors that influence the consistency of international normalized ra tio (INR) of prothrombin time. Methods Using the same automated blood coagulation analyzer and INR calibration plasma with two different types of prothrombin time (PT) reagent to assay the INR values of 19 controls (group 1), 12 patients who took long-term anticoagulant orally (group 2),12 patients who took it at the first time (group 3) and 8 patients who took long-tern anticoagulant togeth er with cephalosperin N (group 4). Results The two kinds of reagents gave quite different INR val ues in all four groups (P<0.01). The INR difference in group 3 (0.23±0.07, n = 12) was larger than that in group 2 (0.51±0.20, n=12) (P<0.01) .The difference in group 4 (0.61 ±0.21, n=8) was larger than that in group 2 ( P<0.01). Conclusions The factors that may influence INR consistency are the source and sensitivity of the reagents , as well as the period of the anticoagulant been taken and the other drugs taken at the same time.

OBJECTIVETo investigate the potential of gene therapy of rat prolactinomas mediated by adenoviral vectors with a gene encoding rat tyrosine hydroxylase.
METHODSRecombinant replication-deficient adenovirus named Ad-GFP-TH with rat TH-cDNA and control adenovirus named Ad-GFP were constructed by homologous recombination in bacterial cells. The rat pituitary prolactinoma cell line MMQ are chosen as the target cells to study the effect of gene therapy on their growth and prolactin secretion mediated by Ad-GFP-TH.
RESULTSRecombinant Ad-GFP-TH and Ad-GFP were successfully reconstructed. Transfection of MMQ cells with Ad-GFP-TH not only restrained their growth but also decreased their PRL secretion.
CONCLUSIONGene therapy may serve for a potential treatment for prolactinomas, especially invasive prolactinomas.

Journal of China Pharmaceutical University 2015;46(1):100-104
The objective of the study was to determine the potential effects of Naomaili granules on rats subjected to ischemia and reperfusion. With the exception of animals in sham group, all animals in treatment groups were subjected to ischemia and reperfusion by middle cerebral artery occlusion(MCAO)with thread technique. The ability of anti-cerebral ischemia, relieving cerebral edema and antioxidation was evaluated by the measurement of neurological deficit score, ratios of infarction area, brain water content, histopathological score and biochemical indexes related to oxidative stress in brain tissue. Significantly reduction of infarction area, histopathological score, contents of H2O2 and MDA were observed at 6 g/kg and 3 g/kg Naomaili granules compared to animals in the model group; the capacity of inhibition of superoxide anion radical(O-·2)and hydroxyl radial( ·OH)were also found to have increased. Additionally, Naomaili granules(6 g/kg)improved neurological function, reduced brain water content, and increased the level of GSH, GSH-Px and SOD compared with model group. The result suggested that Naomaili granules exhibited obvious protection against cerebral ischemia and reperfusion-induced injury due to its antioxidating effect.

Objective To understand distribution of the endemic fluorosis areas and running status of water-improving defuoridation projects in Gausu province. Methods In 2006, Gansu province endemic fluorosis areas, the content of fluoride in drinking water was measured in villages where water was not improved, running status of delluoridation projects was investigated and the content of fluoride in drinking water were determined in villages where water was improved. Dental fluorosis and skeletal fluorosis prevalence were examined in children in identified high-fluorlde villages. The fluorine content in drinking water was determined by F-ion selective electrode, dental fluorosis of children was diagnosed using Dean method, and adults skeletal fluorosis was diagnosed according to "National Standard for Clinical Diagnosis of Endemic Skeletal Fhiorosis" (GB 16396-1996). Results Water samples were examined in 1997 villages of 26 countries, among which water fluoride content was higher than 1.0 mg/L in 598 villages, accounting for 29.94%(598/1997). All 1215 water-improving and defluoridation projects had been investigated, among which 94.90%(1153/1215) of the projects were functioning well, and intermittent and abandoned projects accounted for 2.96%(36/1215) and 2.14%(26/1215). Mean fluoride of treated water of 1084 water-improving and defluoridation projects had water fluoride content ≤ 1.0 mg/L, accounted for 90.79%(1084/1194) ; mean fluoride of water from 1068 water-improving and defluoridation projects had water fuoride content ≤ 1.0 mg/L, accounting for 91.75%(1068/1164). Total 86 390 children of 8 to 12 year-old were examined, the detectable rate of dental fluorosis was 22.47%(19 414/86 390) and 142 211 adults above 16 year-old were examined, the detectable rate of skeletal fluorosis was 4.20%(5967/142 211). Conclusions Some villages yet have water fluoride content exceeding the standard. Some projects are abandoned and running badly, leading to fluoride content exceeding the standard. In a few areas, the prevalence of children dental fluorosis and adult skeletal fluorosis still exists in Gansu province, the task of prevention and control for endemic fluorosis is still arduous. We must raise the effect of prevention and treatment of this disease.
