Objective To investigate the effect of bezafibrate on serum pro-inflammatory cytokine levels and blood pressure change in patients with hypertriglyceridemia and hypertension after antihypertensive therapy with extended release nifedipine. Methods Fifty-eight patients with both hypertriglyceridemia and hypertension, which was treated with extended release nifedipine, were randomized into 2 groups: bezafibrate group (n=30) and placebo group (n=28). Changes in levels of blood lipids, high-sensitivity C-reactive protein (hs-CRP), tumor necrosis factor?(TNF-?) and interleukin-6 (IL-6) were observed following the treatment. OBJECTIVE: To establish an HPLC method for the determination of total(Ct) and free(Cf) concentrations of teicoplanin (TEIC) in plasma. METHODS: For determing the free concentration, the plasma samples were prepared by ultrafiltration. A C18 column was used, with acetonitrile -0.01 mol•L-1 sodium dihydrogen phosphate (25∶75, pH adjusted to 3.3 by phosphoric acid) as the mobile phase, the detection wavelength was set at 240 nm, the column temperature was maintained at 35℃, and the flow rate was 1.0 mL•min-1. The specificity, linearity, the lower limit of quantitation, precession, recovery and stability of the developed method were validated. RESULTS: The free concentration was linear with in 0.5-50 μg•mL-1, the lower limit of quantification was 0.5 μg•mL-1, and the method recovery rate was 94.63%-103.72%. The intra-day and inter-day precision(RSD) were all less than 4.00; the linearity was good with in the total concentration of 1.562 5-100 μg•mL-1, the lower limit of quantification was 1.562 5 μg•mL-1, the method recovery rate was 94.55%-99.59%. The intra-day and inter-day precision(RSD) were all less than 4.00. The TEIC was all stability at the conditions of maintaining at room temperature for 10 h, after freeze-thaw cycle and keeping at 4℃ for 72 h. CONCLUSION: The method is simple, accurate, and sensitive, and is suitable for the clinical determination of total and free concentrations of teicoplanin and the study of pharmacokinetics.

Serum cystatin C (Cys-C) is a cysteine protease inhibitor and has been used as an indicator of early renal function damage.The recent studies have found that Cys-C is closely related to the occurrence,development and prognosis of heart failure.This article reviews the recent research progress on the association between Cys-C and heart failure.

OBJECTIVETo explore the effect of silencing protein kinase D (PKD)-2 on Tca8113 cell proliferation, programmed cell death, and chemosensitivity.

METHODSThe stable cell lines of pkd-2 gene silencing and empty vector plasmid group were established. The proliferation and 50% inhibitory concentration (IC50) of shRNA-mediated Tca8113 chemotherapy drugs were detected through methyl thiazolyl tetrazolium assay (MTT). The programmed cell death rate and sensitivity to Tca8113 chemotherapy drugs before or after pkd-2 gene silencing were measured through flow cytometry. P-glycoprotein (P-gp) expression of pkd-2 silencing cells was identified by immunohistochemical methods.

RESULTSStable cell lines of pkd-2 gene silencing were established. Compared with parental cells, the proliferation of shRNA-mediated Tca8113 was not significantly different, but its IC50 was lower. Meanwhile, cell programmed death rate and sensitivity to chemotherapeutic drugs of shRNA-mediated Tca8113 significantly increased. Compared with wild group Tca8113, a significant decrease in P-gp expression was induced by chemotherapy drugs with shRNA-pkd-2 gene silencing.

CONCLUSIONThe pkd-2 gene of shRNA interference silencing Tca8113 promotes programmed cell death of Tca8113, reduces the IC50 of the chemotherapy drugs, and significantly improves the sensitivity of Tca8113 to chemotherapeutic drugs while reducing the expression of P-gp.

Objective To understand the acute effect of ambient air CO pollution on daily mortality of cerebrocardiovascular diseases among highly and long-term exposed residents of 65 years and over in Taiyuan,Shanxi,China.Methods The relationship of CO concentration and daily mortality of cerebrocardiovascular diseases among residents over 65 years old in Taiyuan(2003-2004) was analyzed by case-crossover design and conditional Logistic regression in SAS 9.0 Results The 48 h accumulated CO concentration had the most significant effect.As increment of CO average was 100 ?g/m~3,the corresponding OR of the effect on the total deaths of the cerebrocardiovascular diseases,the cardiac disease,the ischemic heart disease,myocardial infarction,cardiac failure,arrhythmia and stroke was 1.006,1.010,1.007,1.005,1.005,1.006 and 1.012 respectively.Under different air pollution conditions,the pollutant presented different effects on deaths of eerebrocardiovascular diseases.Conclusion The current CO pollution has caused a certain adverse effect on the cerebrocardiovascular mortality among residents of 65 years and over in Taiyuan city.More stringent measures should be taken to control the air pollution and decrease CO level,thus reduce the mortality of cerebrocardiovascular diseases.

Aim To evaluate the effect and safety of azithromycin injection produced in China and cefazolin in the treatment of respiratory tract infections.Methods 50 patients with respiratory tract infections were divided randomly into two groups. Patients in the test group recieved azithromycin in the dosage of 250 mg?d-1,qd for 5 d iv with double dosages at the first time and patients in the control group recieved cefazolin in the dosage of 2.0 g,bid,iv for 5～10 d. Results No statistical significant differences of overall clinical effect were observed between two groups .The fully recovery,effective and side reaction rates for the test group were 46.67%,90% and 3.3%,respectively,and for control group the data were 60%,95%,and 0%, respectively.Conclusion Azithromycin injection produced in China is effective and safe in the treatment of respiratory infections.

Objective Oridonin is a poorly water soluble drug with low bioavailability.The study aimed to prepared improve the dissolution rate of oridonin by preparation of oridonin solid dispersion with poloxamer. Methods The oridonin solid dispersion was prepared by dissolvent method with poloxamer188 as the carrier and determined by DSC and powder X-ray diffraction.The dissolu-tion rates in different pH dissolution mediums of pure drug, physical mixture and solid dispersion were determined by HPLC method. Results Oridonin exists as an amorphous state in the solid dispersion.Compared with the pure drug and physical mixture, the orido-nin solid dispersion was improved greatly without the pH influence on the drug release.The dissolution rate of oridonin solid dispersion in water, pH 6.8 PBS and pH 1.2 HCl were (92.6 ±4.2)%, (93.1 ±3.5)%and (94.4 ±2.9)% respectively. Conclusion Taking inpoloxamer188 as the carrier, the oridonin solid dispersion has successfully developed its dissolution rate.
