OBJECTIVETo establish human heart valve interstitial cells calcification culture model in vitro, and observe the effect of bone morphogenetic protein-2 (BMP-2) on calcification of human heart valve interstitial cells.
METHODSHuman heart valve interstitial cells were cultured in vitro, and divided into control group: cells were cultured in conventional media plus recombinant human BMP-2 treatment and experimental group: besides above treaments, calcification inducers ( recombinant human BMP-2, β-glycerophosphate, L-ascorbic acid, dexamethasone) were added to the culture media. The two group of cells were cultured for 14 days and were stained by Von Kossa, then the cell calcification was observed in this valvular interstitial cells calcification culture model in vitro. Protein expression of intercellular adhesion molecule 1 (ICAM-1), interleukin 8, BMP-2 and BMP-4 was determined by Western blot and BMP-2 secretion was measured by ELISA.
RESULTSIn the control group, the structure of human heart valve interstitial cells was clear, and the spindle and radial growth shaped cellular morphology was visible, and Von Kossa staining was negative. In the experimental group, the nuclei become darker in color, and granular sediment distribution was seen surrounding cells, and Von Kossa staining was positive, the cells were forming nodules of calcification. The protein expression of ICAM-1, interleukin 8, BMP-2 and BMP-4 in the experimental was significantly higher than that of the control group (all P < 0.05). The expression of BMP-2 in the experimental group was also significantly higher than that in control group ((92.5 ± 4.9) pg/ml vs. (22.2 ± 1.9) pg/ml, P < 0.05).
CONCLUSIONHuman BMP-2, β-glycerophosphate, L-ascorbic acid, and dexamethasone can induce human heart valve interstitial cells calcification and enhance inflammation in vitro by stimulating the secretion of BMP-2.

Objective To observe the effecacy of modified CHOP regimen in the initial treatment of non-Hodgkin' slymphoma (NHL) patients with low Karnofsky score. Methods Twenty initial treatment NHI.patients with low Karnofsky score were randomly allocated into treatment group and control group, respectively treated with modified and standard CHOP regimen. Results Of 13 cases in treatment group,12 pacients'Karnofsky score were over 60.1 was still below 60,overall response (OR) rate was 92.3 % (12/13).while in control group,3 patients' Karnofsky score were over 60,4 were still below 60.OR rate was 100 % (7/7),rate of marrow toxicity was 57.1 % (4/7).There were no significantly difference for OR rates (P =0.4515),but for improving Karnofsky score there were significantly difference (P =0.0149).Conclusion Modified CHOP regimen can improve Karnofsky score of NHL patients with low Karnofsky score, benefical for further treatment and reducing hospitalization cost.

Sjögren's syndrome is a chronic autoimmune disease with unclear etiology. From the point of etiology, Chinese medicine (CM) theory holds that pathological products like dry toxin, blood stasis are produced in the pathological process. They are both pathologic results and pathogenic factors for its further development. So pathological products are also named as second pathogenic factors. In this article, the concept of second pathogenic factors was sorted and defined. Main second pathogenic factors of Sjögren's syndrome were pinpointed, and their modern medical bases were analyzed. Authors came to a conclusion that clearing away second pathogenic factors is a key point in treating Sjögren's syndrome.

Objective To investigate the expression of activated ERK (p-ERK) and activated p38 (p-p38) in the keratinocytes of condyloma acuminata (CA) lesions. Methods Fifty cases of HPV 6/11 CA were diagnosed by in situ hybridization. The expression and distribution of p-ERK and p-p38 in CA lesions and 25 normal human skins (foreskins) were detected by immunohistochemistry technique (En Vision). Results ①The results showed that the expression of p-ERK and p-p38 in keratinocytes of CA lesions were significantly higher than those in normal epidermis (P

Importance of central laboratories in hospitals is rising sharply as they provide laboratory support to hospital development in its science researches. The paper introduced the mission and features of the central lab of the hospital, as well as its technical personnel makeup, instrumentation, and rules and regulations. Authors prove in the paper that central labs are playing a key role in hospital science research as its research base.

Objective To investigate the clinical therapeutic effect of electroacupuncture on ischemic stroke by recording the therapeutic effect score and performing transcranial Doppler (TCD) detection in electroacupuncture-treated convalescent stroke patients before and after treatment. Methods Sixty convalescent patients with ischemic stroke were randomly allocated to treatment and control groups, 30 cases each. The control group received routine medication and the treatment group, electroacupuncture in addition. In the two groups, pre- and post-treatment neurological defect scores were recorded and the clinical therapeutic effects were observed after four weeks of treatment. Pre- and post-treatment middle cerebral artery (MCA) pulsation indices (RI), resistance indices (RI) and ratios of systolic to end-diastolic (S/D) blood flow velocity detected by TCD were compared between the two groups. Results The total efficacy rate was 80.0% in the treatment group and 50.0% in the control group; there was a statistically significant difference between the two groups (P<0.01). There was a statistically significant pre-/post-treatment difference in the MESSS score in the two groups (P<0.01). There was a statistically significant post-treatment difference in the MESSS score between the treatment and control groups (P<0.05). There were statistically significant pre-/post-treatment differences in TCD indices (PI, RI and S/D) on the affected side in the two groups (P<0.05). There were statistically significant post-treatment differences in TCD indices on the affected side between the treatment and control groups (P<0.05).Conclusion Electro- acupuncture can promote rehabilitation in convalescent stroke patients.

Objective To evaluate the radiation protection of occupational hazards in a hospital Clinac 23EX medical electronic accelerator construction project so as to ensure the health and safety of the relevant people involved. Methods According to the relevant laws, regulations and standards of China, combined with the relevant materials provided by the construction unit, the radiation protection tests and comprehensive assessment of this project were carried out. Results The performance test results of the medical electron accelerator met the requinements of GB/T 19046-2013 The ambient dose equivalent rate in the workplace was between the background dose rate (0.10 μ,Sv/h) and 11.5 μSv/h,which suggested the computer room shielding met the requirements of radiation protection. The total body effective dose, the for 7 radiation workers were 0.85,1.19,1.64 mSv,respectively, which were lower than the dose management control values of the construction unit and the national standards.Radiation protection supplies and the management system of the construction unit met the national requirements. Conclusions The construction project can effectively control the radioactive occupational hazard factors in normal operation, and the radiation protection facilities have reached the completion requirements.

Plasma nesfatin-1 levels were assayed by ELISA in patients with impaired glucose regulation (IGR),type 2 diabetes mellitus( T2DM ),and healthy subjects with normal glucose tolerance( control group). The results showed that plasma nesfatin-1 levels in IGR and T2DM groups were significantly higher than that in control group [ ( 1.91 ±0.79 and 1.80±0.80 vs 1.41 ±0.58) μg/L,P<0.01 ].Plasma nesfatin-1 levels were positively correlated with body mass index(BMI),HbA1c,waist hip ratio, fasting plasma glucose, fasting insulin, and homoestasis model assessment insulin resistant index( HOMA-IR,P<0.05 or P<0.01 ).Multiple regression analysis showed that BMI and HOMA-IR were independent related factors in influencing plasma nesfatin-1 levels (both P<0.01 ).These results suggest that nesfatin-1 may partially contribute to the pathogenesis of insulin resistance and T2DM.
