1.miR-125b-1 gene mutation in lung cancer: relation with the clinic opathological parameters and prognosis of patients

Objective To investigate the role of miR-125b-1 gene in oncogenesis and progression of human lung cancer. Methods PCR-SSCP was used to detect miR-125b-1 gene mutation in 121 specimens from primary site of lung cancer tissues and 22 samples of nor- real adjacent lung tissues. Results There were 40 cases of miR-125b-1 gene mutation, and gene mutation was positively correlated to the status of lymph node metastasis ( r,=0.285, P<0.01 ) and clinical stage metastasis( r, =0. 241, P <0. 01 ), but no relationship between miR-125b-1 mutation and other clinical features was found. The postoperative survival time in patients with miR-125b-1 gene mutation was significantly longer than those with low expression ( P<0.05). Conclusion There exist miR-125b-1 gene mutation in lung cancer, miR- 125b-1 gene mutation probably plays a crucial role in lung carcinogenesis.

3.Protective Effect of Jiedu Huayu Decoction on Rat Hepatic Immune Injury

Objective To evaluate the protective effects of Jiedu Huayu Decoction(JHD)on hepatic immune injury in mice induced by concanavalin A(Con A) and to explore its mechanism.Methods Sixty NIH mice were randomly divided into 5 groups:normal control group(group A),model group(group B),high-dose JHD group(group C),low-dose JHD group(group D) and bifendate group(group E).Except those in group A,mice in other groups were injected with Con A(at the dose of 20 mg/kg) via the tail vein at the first day of experiment.Group C and group D were orally administrated with JHD at the dose of 72.0 g/kg and 18.0 g/kg,respectively.Group E were orally administrated with bifendate at the dose of 150 mg/kg.All the drugs were given once daily for 5 consecutive days.Four hours after last medication,mice were injected with Con A once again at the same dose of 20 mg/kg.Serum aminotransferase(ALT) activity and interleukin-6(IL-6) concentration were detected at the 8th hour after Con A administration.Hepatic histopathological examination was performed,and the expression of intercellular adhesion molecule-1(ICAM-1) in hepatic tissue was detected by immunohistochemical method.Results ALT activity and IL-6 content in group C and group D were obviously lowered,hepatic histopathological changes were alleviated,and the expression of ICAM-1 in liver tissue was attenuated compared with those in group B(P

6.Risk factors for N2 lymphatic metastasis in patients with non-small cell lung cancer

Objective To assess risk factors in NSCLC with N2 metastasis.Methods A total of 177 NSCLC patients (109 males,68 females; age:(60.1 ± 10.6) years) who underwent both 18F-FDG PET/CT scan and surgery were enrolled.They were divided into two groups.One was with N2 metastasis (N2+) and the other was without (non-N2+).The gender,age,location,size,primary tumor SUVmax,histopathologic type and differentiation grade between N2+ group and non-N2+ group were compared with x2 test and two-sample t test.After assignment of significant factors and groups,correlations between each factor and group were analyzed with uni-and multivariate analyses,and then the risk factors were identified finally with logistic regression analysis.Results Significant difference among factors of tumor size ((36.1±19.7) mm vs (49.3±24.4) mm),SUVmax(8.81±6.23 vs 11.21±4.43) and differentiation grades (high:3 vs 0; moderate:74 vs 11; poor:65 vs 24) were observed between non-N2+ group and N2+ group (t =-2.969,-2.633,x2 =6.143,all P<0.05).Besides,the univariate analysis showed significant correlations between tumor size,SUVmax,differentiation grade and N2 status (r=0.201,0.245,0.185,all P<0.05).However,multivariate logistic regression revealed that only SUVmax had a predictive value (β=0.546,P<0.01).The incidence of N2 metastasis went up along with the increase of SUVmax.There was no N2 metastasis in the patients with SUVmax ≤2.5,and the incidence rate of metastasis increased to 0.29 in those with SUVmax>7.5.Conclusion SUVmax may be an independent risk factor to predict N2 metastasis in NSCLC patients.
7.EXPRESSION OF Fos PROTEIN IN RAT BRAINS FOLLOWING RESTRAINT STRESS

Objective To investigate expression of Fos protein in rat brains following restraint stress. Methods The experimental rats were restrained in a small plastic tub for,1,3 and 6 hours,and were sacrificed at 30 min after removing restraint.Immunohistochemical ABC method was used to observe distribution of Fos protein-like immunoreactive(-LI)products in rats brain.Results Fos-LI neurons appeared in (1)Frontal brain:the cingulum,cortex(especially in third and fifth layer),lateral septal nucleus,central amygdaloid nucleus.(2)Diencephalon:the thalamic paraventricular nucleus,lateral geniculate body and medial genicular body,hypothalamic paraventricular nucleus,supraoptic nucleus,periventricular area of third ventricle,arcuate nucleus.(3)Brain stem:the superficial layer of superior colliculus,periaqueductal gray,cortical area of inferior colliculus,lateral parabrachial nucleus,locus coeruleus,A5 area,cochlear nuclei,medullary viceral zone in medulla oblongata.The expression of Fos-LI neurons peaked in rats restrained for 1h,at 3h,then began to decrease,at 6h,significantly decreased. Conclusion Fos-LI neurons appeared in many areas of brain induced with restraint stress.The number of Fos-LI neurons decreased following restraint time.
8.Prevention and treatment for acute left heart failure in patients undergoing maintaining hemodialysis

Objective To study prevention and treatment for patients during the intermittent period of maintaining hemodialysis complicated by acute left heart failure. Methods The treating process of 126 examples suffered from acute left heart failure during the intermittent period of 14 225 person-time maintaining hemodialysis in 143 patients were retrospectively analyzed. Results In this case group, 0.89% suffered from acute left heart failure during the intermittent period of hemodialysis and the successful rescuing rate was 88.9%. Conclusions Applying the treatment of emergency hemodialysis, quickly filtering body fluid, the curative effect is reliable and rapid. It is one of the best ways to cure patients suffered from acute left heart failure during the intermittent period of maintaining hemodialysis. Combined with other treatments, it will be able to improve the successful rate, and ensure the sufficiency of hemodialysis. United with other steps of reducing blood pressure, such as using angiotensin converting enzyme inhibitors, angiotensin II receptor blocker, and ? receptor blocker, it is an effective countermeasure to prevent patients suffering from acute left heart failure during the intermittent period of maintaining hemodialysis, and effectively reduces its appearance as well.
9.Clinical Study of Chinese Herb Formula Tangtongfang by Bathing the Affected Feet with Diabetic Peripheral Neuropathy

Objective To investigate the effects of Chinese herb formula Tangtongfang by bathing the affected feet of patients with diabetic peripheral neuropathy (DPN) in the pattern of Qi deficiency and blood stasis. Methods A parallel, randomized, controlled clinical research was performed. 70 diabetics in the inpatient and outpatient divisions of the Department of Endocrinology, Guanganmen Hospital, from January 2005 to October 2007 were investigated. They were divided randomly into two groups, treatment group and control group, 35 cases in each group. Before study, there were no statistic differences between the two groups, including gender, age, course of diabetes mellitus(DM), course of DPN, blood glucose (including fasting blood glucose and postprandial blood glucose), traditional Chinese medicine(TCM) symptom scores and nerve conduction velocity (NCV). The treatment group were treated with Tangtongfang bathing and the control group were treated with warm water bathing. Each group was also administered Vitamin B1 20 mg, t.i.d and Vitamin B6 20 mg, t.i.d respectively. All patients were keeping their routine treatment and diabetic care to control their blood glucose, such as diets, practices and drugs. Clinical symptoms and electrophysiological tests in diabetic peripheral neuropathy syndrome were investigated. The period of treatment was 8 weeks.Results After study, 9 cases retreated from this clinical trial totally, 4 cases in the treatment group whereas 5 in the control group. Among 4 common single symptoms, coolness feeling of the feet of diabetic patients was significantly improved (P<0.05) after bathed with Tangtongfang. The change of TCM symptom scores and tibialis NCV was significantly improved (P<0.001, P<0.05) in the treatment group. The improvement ratio was 45.2% and 61.3% in the treatment group, while was 10% and 3.3% in the control group. In each group there was no statistical difference between pre-and post-study with blood test, urine test, blood ALT, blood BUN and Cr, ECG examination (P>0.05). No adverse event was observed in each group.Conclusion Tangtongfang bathing improved several symptoms of patients suffering from DPN, especially the symptom of coolness feeling of the patients' feet. No adverse event was observed. This result showed that Tangtongfang bathing was worthy of using as a safe agent in treatment of DPN.
