1.Expressions of tumor angiogenesis related factors CD105 and Galectin-3 in laryngeal squamous cell carcinoma and biological assessment

[ABSTRACT]OBJECTIVETo investigate the expressions of Endoglin (CD105) and Galectin-3 protein in laryngeal squamous cell carcinoma (LSCC) as well as the relationship between their expressions and the clinicopathological factors of LSCC.METHODSThe expressions of CD105 and Galectin-3 protein were detected in 76 samples of LSCC and 25 normal laryngeal tissues (NLT) by immunohistochemical staining (S-P).RESULTS 1.The mean of Microvessel density (MVD) value marked by CD105 in LSCC was 10.33±2.29, which was significantly higher than that in NLT (1.20±1.04) (P<0.05). The expression of MVD marked by CD105 (CD105-MVD) was correlated with histological grading, T stage, clinical stage, lymph node metastasis, recurrence and prognosis in LSCC (P<0.05). 2.The positive expression rate of Galectin-3 protein in LSCC was 86.84%, which was significantly higher than that in NLT (36%)(P<0.05). The expression of Galectin-3 was correlated with T stage,clinical stage, lymph node metastasis, recurrence and prognosis in LSCC (P<0.05). 3.There was a positive correlation between CD105 and Galectin-3 protein. 4.Survival analysis indicated that the expressions of CD105 and Galectin-3, histological grading, lymph node metastasis,T stage and recurrence were independent factors for tumor prognosis in LSCC (P<0.05). CONCLUSIONThe expressions of CD105 and Galectin-3 protein have a positive correlation in LSCC. They may play important roles in the tumorigenesis, malignant progression and poor prognosis of LSCC. Combined detection of them may be great value in diagnosis and predicting prognosis of LSCC.
2.Quality standard study on Mori Cortex liquid extract.

A reasonable and practicable quality standard was developed for mori liquid extract from different sources by TLC, HPLC and fingerprint technology. In TLC method, the compounds were separated on polyamide film using glacial acetic acid-water (1: 3) as mobile phase at a UV wavelength of 365 nm. All qualified samples had the spots of the same color as the control herb and substance. The RP-HPLC method was used to determine the content of mulberroside A with mobile phase of methanol-water (25: 75) at a wave-length of 326 nm. The mulberroside A was in good linear with a regression equation of Y = 46.965X (r = 0.999 6) in the range of 4.6 - 228 mg x L(-1). In 14 batches of samples, the mulberroside A in 4 batches of them was less than 0.5 g x L(-1), and was more than 2.0 g x L(-1) in the other batches. It was suggested that the content limit of mulberroside A should be no less than 1.5 g x L(-1). The HPLC fingerprints were evaluated by the similarities. It has found that the similarities of different mori liquid extracts were very low and the chemical diversity of mori cortex was the major factor of similarity. Moreover, the process impact was minimal. Thus the fingerprint was not included in this quality standard.

Objective To investigate the effects of siRNA targeting CD 147 gene on the expression of CD147 in melanoma cell line A375, and on proliferation of these cells. Methods Previously prepared recombinant plasmid pSUPER/CD147 siRNA was used. In this study, the recombinant was transfected into the A375 cells. The mRNA expression of CD147 was measured by semi-quantitive RT-PCR, the proliferation of the cells by MTT assay. 4.Effect of admission requirement questionnaire on type 2 diabetes mellitus patients' satisfaction with hospitalization

Objective To investigate the effect of admission requirement questionnaire on improving diabetes mellitus patients' satisfaction with hospitalization.Methods 120 patients with type 2 diabetes mellitus from January to December in 2011 were randomly divided into observation group and control group,each with 60patients.The control group received conventional admission evaluation table and nursing care in department of Endocrinology,while the observation group was evaluated with admission requirement and received individualized care according to the results of admission requirement questionnaire.Two groups' satisfaction with hospitalization was compared on their discharge.Results Scores of satisfaction with health education,service quality and attitude,and nursing skill of the observation group were (4.33 ± 0.60),(4.50 ± 0.68 ) and (4.25 ±0.57),which were significantly higher than those of the control group [ (4.02 ±0.70),(4.18 ±0.54)and (4.03 ±0.61 )],and the difference were statistical significant (t =2.656,2.841,2.009,respectively; P<0.05).There was no statistically significant difference in their satisfaction with hospital environment between two groups [ (4.07 ±0.41) vs (3.92 ±0.53) ;t =1.74,P>0.05].Conclusions The application of admission requirement questionnaire can improve type 2 diabetes mellitus patients' satisfaction with hospitalization,thus worthy of clinical popularization.
5.Correlation study on 12p13 single nucleotide polymorphism rs12425791 and Chinese medical syndrome types in ischemic stroke patients of the Han nationality.

OBJECTIVETo explore the correlation between 12p13 single nucleotide polymorphism (SNP) rs12425791 and Chinese medical syndrome types of ischemic stroke patients of the Han nationality.
METHODSA case-control study was used. Recruited were 148 ischemic stroke patients of the Han nationality (67 patients of phlegm syndrome and 81 patients of blood stasis syndrome). Another 192 healthy subjects were recruited as the control group. The genotypes of rs12425791 were performed by TaqMan SNP genotyping assays to analyze the distribution of genes and the distribution frequency of alleles.
RESULTSThere was no statistical difference in the genotype and alleles of rs12425791 between the ischemic stroke patients of phlegm syndrome and the control group, or between the ischemic stroke patients of blood stasis syndrome and the control group (P > 0.05).
CONCLUSIONOur results did not support that 12p13 common variant rs12425791 was correlated with the pathogeneses of ischemic stroke patients of phlegm syndrome and ischemic stroke patients of blood stasis syndrome.

6.Study on treatment of irritable bowel syndrome: based on relationship between heart and intestines.

7.Morphology and microleakage study of repairing subpulpal wall perforation with resinous inlay.

OBJECTIVEThe purpose of this study is to study the sealing ability and the furcal appearance of repairing subpulpal wall perforation with resinous inlay.
METHODSFifty newly extracted human molars were randomly divided into three experiment groups (group A, group B, group C, 15 teeth each) and one control group (5 teeth). In experiment groups, perforations were made perpendicularly to the center of the pulp chamber floor. Perforations of group A and B were repaired with resinous inlay and sealed by AH Plus sealer and luting glass-ionomer, respectively. Perforations of group C were directly repaired using light-cure composite resin. Perforations were not made in five teeth of control group. The furcal appearances were evaluated under stereomicroscope after repairing. Microleakage was measured by glucose oxidase detection.
RESULTSThe fineness rate of furcal appearances with resinous inlay repairing were 83.3%, while the fineness rate of furcal appearances with light-cure composite resin directly repairing were 46.7%. There were statistics difference between resinous inlay repairing and light-cure composite resin directly repairing (P<0.05). There were statistics difference among the daily microleakage of three experiment groups, group A CONCLUSIONUsing resinous inlay to repair the subpulpal wall perforation can improve the sealing effect and avoid material overextension. AH Plus can be used as perforation sealant because of its better sealing ability.

8.Study on the complications of radical vaginal hysterectomy for locally bulky cervical cancer patients

Objective To analyze the complications of radical vaginal hysterectomy in patients with locally bulky cervical cancer.Methods All 258 patients of early cervical cancer (stage Ⅰ A2-Ⅱ A) selected in our hospital during the period of June 2011 to June 2016 undergoing radical vaginal hysterectomy and laparoscopic pelvic lymph node dissection were analyzed.All cases were divided into observation (locally bulky) and control (conventional) groups.Comparison analysis was performed for complications of the two groups and the influence of neoadjuvant chemotherapy on the complications of local bulky patients.Results The incidence of complications in the observation group was 26 cases (27.9%),higher than that in the control group of 28 cases (17.0%),the difference was statistically significant (P < 0.05).The incidence of bladder injury (5 cases vs 2 cases) in observation group was significantly higher than that in control group (P < 0.05).For patients with locally bulky early cervical cancer,the incidence rate of complications of the neoadjuvant chemotherapy group (15.8%) was significantly lower than the simple operation group (36.4%) (P < 0.05).The incidence rate of ureteral injury in neoadjuvant chemotherapy group (0 cases vs 6 cases) was significantly lower than that in simple operation group (P < 0.05).Conclusions Early cervical cancer patients with locally bulky (stage Ⅰ B2/ 1Ⅱ A2) had higher incidence of complications of radical vaginal hysterectomy,and neoadjuvant chemotherapy can reduce the incidence of surgical complications.
9.Expressions of angiogenesis-related factors: CD105, EphA2 and EphrinA1 in laryngeal squamous cell carcinoma and clinical implication

Objective To investigate the expressions of endoglin (CD105),erythropoietinproducing hepatocyte receptor A2 (EphA2) and its ligand ephrinA1 proteins in laryngeal squamous cell carcinoma (LSCC) and the relationship between their expressions and the clinicopathological factors of LSCC.Methods The expressions of CD105,EphA2 and EphrinA1 proteins were detected with immunohistochemical staining in LSCC in 76 cases and adjacent normal laryngeal tissues (ANLT) (S-P) in 25 cases.SPSS 17.0 software was used to analyze the data.Results The mean microvessel density (MVD) value marked by CD105 staining in LSCC was 10.33 ± 2.29,which was significantly higher than that in ANLT(1.20 ± 1.04,t =18.732,P < 0.05).The CD105-MVD was correlated with T stage,histological grading,clinical stage,lymph node metastasis,recurrence and prognosis in LSCC (F value was 5.34,4.79,5.36,t value was-2.70,2.56,all P < 0.05).The positive expression rates of EphA2 and EphrinA1 in LSCC were 78.95% (60/76),and 81.85% (62/76),which were respectively significantly higher than 40% (10/25) for EphA2 expression and 44% (11/25) for EphrinA1,expression in ANLT (x2 value was 13.41,13.26,both P < 0.05).EphA2 expression was correlated with histological grading,T stage,clinical stage,lymph node metastasis,recurrence and prognosis in LSCC (x2 value was 6.25,14.60,15.11,8.52,5.54,all P < 0.05).EphrinA1 expression was correlated with T stage,clinical stage,lymph node metastasis,recurrence and prognosis in LSCC (x2 value was 6.44,12.28,16.78,6.44,all P < 0.05).The expressions of CD105,EphA2 and EphrinA1 were positively correlated with each other r value was 0.72,0.74,0.64,all P < 0.05.Survival analysis indicated that the expressions of CD105 and EphA2,histological grading,lymph node metastasis,clinical stage and recurrence were independent factors for tumor prognosis in LSCC (P < 0.05).Conclusions The expressions of CD105,EphA2 and EphrinA1 protein were positively correlated with each other in LSCC.They may play important roles in the tumorigenesis,malignant progression and poor prognosis of LSCC.
10.Effects of conjugated linoleic acid on obese MSG mice with insulin resistance.

AIMTo study the effect of conjugated linoleic acid (CLA) on obese MSG mice with insulin resistance.
METHODSAbout four months old, obese MSG mice with insulin resistance were divided into control, CLA and rosiglitazone groups and drugs were administrated ig once a day. Body weights were recorded regularly, insulin and glucose tolerance were tested. In addition, serum insulin and TNF-alpha concentrations in serum and fat tissues were determined.
RESULTSCLA was shown to reduce the body weight and fat weight in MSG mice, but can not improve the abnormal insulin and glucose tolerance in these mice. Indeed, the serum insulin and TNF-alpha concentrations in the fat tissues of the group treated with CLA were higher than those in the models and the insulin sensitivity index was significantly lower than that in the model mice.
CONCLUSIONCLA can reduce the body weight of MSG mice, but can not improve the insulin resistance in these mice.
