3.Phase Ⅱ rehabilitation/prevention of brain tumor: effect of the HSV-TK/ACV system mediated by cationic lipsome on the proliferative activity of human glioma cells

BACKGROUND: Gene therapy is a popular topic in domestic and overseas studies on biological therapy for brain tumor.OBJECTIVE: By using a newly constructed eukaryotic expression vector of pCR3-TK, the effect of the HSV-TK/ACV system on the proliferative activity of human glioma cells was investigated.DESIGN: Experimental study based on cells.SETTING: Department of neurosurgery and department of oncology in a university hospital.MATERIALS: The study was conducted at the National Key Laboratory of Veterinary Biotechnology of Harbin Veterinary Research Institute from January to April in 2004. The eukaryotic expression vector of pCR3-TK was constructed by the author. The TJ905 strain was a gift from professor Pu Pei-yu, who worked in the Neurology Institute of Tianjin city. The nontransfected cells and the cells transfected with pCR3-Uni vector were set as controls.METHODS: By using Lipofectamine(a cationic liposome), the pCR3-Uni vector and the recombinant pCR3-TK plasmid(inserted with HSV-TK gene)were transfected into the human glioma cell strain-TJ905. Then the positive clones were picked out and were given ACV(50 mg/L) . Totally 72 hours later, the cover slips were collected and silver staining for nucleolus organizer regions(AgNORs) was performed.MAIN OUTCOME MEASURES: After the ACV treatment and AgNORs staining, the numbers of silver-stained granules in TJ905 cells with or without transfections were counted respectively.RESULTS: In those cells transfected with HSV-TK gene, after ACV treatment, a significant decreasing in proliferative activity could be observed, and the average numbers of the silver-stained granules in cells transfected with pCR3-Uni or pCR3-TK were 14.33 and 6.67 respectively( P < 0.01).CONCLUSION: As an easy-to-operate method, AgNOR counting is helpful for the studies on the proliferative activity of cells and the investigations into the potential anti-tumor mechanism of the HSV-TK/ACV system.
4.Prognostic factors for bloodstream infection in adults

Objective To investigate prognostic factors for bloodstream infection in adult patients. Methods Clinical data of 131 adult patients with positive blood cultures during January 2002 to December 2003 in the Hospital were collected and 91 cases of them were retrospectively analyzed to understand their pathogen species and prognostic factors for it.Results Blood samples from 91 patients were cultured positive,53 cases(58.2%)with gram-negative bacteria mainly including Escherichia coli,Salmonella spp. and Klebsiella pneumoniae,28(30.8%)with gram-positive bacteria,mainly including Staphylococcus aureus and coagnlase-negative Staphylococci,eight(8.8%)with fungi and two(2.2%)with multiple infections.Case fatality ratio in this group of patients with septicemia was 30.8% during their hospitalization,and that in those with Pseudomonas aeruginosa,Staphylococcus aureus,Stenotrophomonas maltophilia and E.coli with extended-spectrum beta-lactamase was over 50%.Case fatality ratio was associated with severity of sepsis(OR=1.15)and inappropriately initial empirical treatment with antibiotics (OR=6.77).Conclusions Pathogen causing bloodstream infection in adults were mainly gram-negative bacteria and severity of infection and inappropriate initial antibiotics treatment could increase their fatality.
5.Effects on performance of high-speed dental handpieces subjected to autoclaving.

OBJECTIVETo investigate the effects on performances of high-speed dental handpieces subjected to autoclaving.
METHODSThe method of trial used to evaluate the effect of autoclaving included that ten chemical and ten biological indicators were arranged in each trial group being set by one sort of temperature and sterilization time. The range of temperature was from 100 degrees C to 134 degrees C and the sterilization time was from 1 to 45 minutes. The reliability of autoclaving was evaluated by the result of chemical indicator and the endospores of Bacillus stearothermophilus subjected to autoclaving. Ten newly imported and ten new domestic dental handpieces in two different brands were used to carry out this trial. The effects produced by autoclaving on performances of dental handpieces subjected to different type autoclaves were evaluated by some parameters.
RESULTSThe eligible outcome in killing test of the 221 sterilized dental handpieces proved that the sterilization effect of autoclaving on dental handpieces was reliable. The chemical indicators showed that under the condition of 121 degrees C it needed 20 minutes to reach sterilization standard in the inner package and only 15 minutes on the outer package, and under the condition of 134 degrees C it took 4 minutes to reach sterilization standard in the inner package while only 2 minutes on the outer package. This outcome proved that material with package was more time-consuming than that without package during autoclaving. Autoclaving has positive effects on the performance of dental handpieces, while the autoclave with less-time full sterilization cycle has less effect on it. The performance of dental handpieces sterilized by the autoclave with 6-minute full sterilization cycle might be restorable within certain times of sterilization cycle.
CONCLUSIONThe sterilization effect of autoclaving is reliable. In order to reduce the influence on the performance of dental handpieces subjected to autoclaving, it should be reasonable to use the autoclave by which a full sterilization cycle might take less time and no use of package.

6.Analysis of Genotypes and Drug Resistance of Extended-Spectrum-Lactamases-Producing Strain in Pat-hogenic Gram-Negative Rod in Infection of Newborn in Guangzhou

Objective To investigate the distribution,drug resistance characteristics and genotypes of extended-spectrum-lactamases (ESBLs)-producing strain in pathogenic gram-negative rod in infection of newborn in Guangzhou.Methods The standard was performed by the production for ESBLs by phenotypic screening and confirmatory test provided by the National Committee for Clinical Laboratory Standards in 2001.The method of polymerase chain reaction(PCR) amplification was perbonmed and DNA sequences were analyzed by ESBLs gene sequencing.Results Total of 71 un-repicated and consecutive Gram-negative bacilli were isolated from 13 hospitals in Guangzhou,and the prevalence of ESBLs-producing clinical Gram-negative isolates was 59.2%(42/71).The PCR results showed that most pathogenic bacilli which infected newborn could be separated two or more genes of ESBLs.The type of TEM,SHV,CTX-M1,CTX-M9,OXA was 35.6%, 26.7% ,10.9%,24.8%,2.0%,respectively.The result of drug resistance monitoring showed that pathogenic gram-negative bacillui which infected newborn were Escherichia coli and Klebsiella pneumonia mostly.Most parts of them were drug fast and even multidrug resistant to the commonly used antibiotics.The sensitive drugs were lmipenem(the rate of sensitivty 100%),cefoperazone/sulbactam(87.3%),piperacillin/tazbatam(85.3%),ceftazidime(82%),aztreonam(82%),cefepime(81.8%).Conclusions In Guangzhou,the incidence rate of ESBLs-producing strain are very high inpathogenic bacilli which infected in newborn and is multidrug resistance.The genetypes of produced ESBLs are TEM,SHV,CTX-M1,CTX-M9,OXA.
7.Effect of traditional Chinese medicines with different properties on thermoregulation and temperature-sensitive transient receptor potentialion channel protein of rats with yeast-induced fever.

OBJECTIVETo compare the intervention effects of four traditional Chinese medicines (TCMs) with typical cold or hot property on body temperature and temperature-sensitive transient receptor potential ion channel proteins (TRPs) of rats with yeast-induced fever.
METHODThe pyrexia model was induced by injecting yeast suspension subcutaneously. Totally 108 male SD rats were randomly divided into the normal group, the model group, the Rhei Radix et Rhizoma treated group, the Coptidis Rhizoma treated group, the Euodiae Fructus treated group, and the Alpiniae Officinarum Rhizoma treated group, with 18 rats in each group. At the 4 h, 8 h and 12 h after injection of yeast, the rats were sacrificed to collect their hypothalamus and dorsal root ganglion. The expressions of TRPV1 and TRPM8 were detected by immunohistochemistry and Western blot method.
RESULTCompared with the normal group, after injection of yeast, the temperature of rats in the model group notably increased, and reached the peak at 8 h (P < 0.01). The TRPV1 level in hypothalamus and dorsal root ganglia (DRG) of the model group significantly increased, whereas the TRPM8 level significantly reduced. Compared with the model group, the Rhei Radix et Rhizoma group and the Coptidis Rhizoma group showed significant decrease in the high body temperature of rats caused by yeast, down-regulation in the expression of TRPV1, and up-regulation in the expression of TRPM8 (P < 0.05 or P < 0.01). Euodiae Fructus and Alpiniae Officinarum Rhizoma had no significant effect on either temperature or TRPs of fever rats.
CONCLUSIONRhei Radix et Rhizoma and Coptidis Rhizoma, both are TCMs with cold property, can reduce the temperature of fever rats induced by yeast, which may be related to their effective regulation of TRPV1 and TRPM8 in hypothalamus and DRG, while Euodiae Fructus and Alpiniae Officinarum Rhizoma had no relevant effect.

8.The possibility of hepatitis B virus transmission through dental handpieces.

OBJECTIVETo discuss the possibility of hepatitis B virus (HBV) transmission through dental handpieces.
METHODSInvestigation was carried on methods for disinfecting and sterilizing dental handpieces and the condition of HBsAg contamination on dental handpieces before and after disinfection and sterilization by randomly sampling all special stomatological hospitals and dental clinics in a same city and 10 dental departments from the third, second and first class hospitals. The possibility of HBV transmission through dental handpieces was probed by investigating whether ducks can be infected by bath liquid of dental handpieces contaminated by DHBV, while in such bath liquid, DHBV can not be detected by serum dot hybridization.
RESULTSFrom 2001 to 2004, in methods to disposing dental handpieces, the use of autoclave was remarkably increased while of the disinfectant wipe, immersion and other methods was remarkably decreased. The positive rate of HBsAg from dental handpieces in practice was 1.65%. It was evident that the bath liquid of dental handpieces contaminated by DHBV can conduct infection in vivo test of duck, while DHBV can not be detected in such bath liquid by serum dot hybridization, it is proved that the negative result of HBsAg in non-sterilized dental handpieces can not eliminate the possibility of HBV transmission through dental handpieces.
CONCLUSIONThere might exist the possibility of HBV transmission through dental handpieces however, the autoclaves might kill the virus contaminating on dental handpieces.

9.ZHU Lian--the founder of Chinese acupuncture-moxibustion scientific research.

This article discussed ZHU Lian's contributions to acupuncture-moxibustion scientific research from three aspects: building the scientific thought of "new acupuncture-moxibustion", constructing the first domestic acupuncture-moxibustion institution and opening the door to modern acupuncture-moxibustion scientific research. ZHU Lian's visionary thought of "new acupuncture-moxibustion" has influenced the following researchers till now. She established the acupuncture-Moxibustion therapeutic institute affiliated to the Ministry of Health, set up the acupuncture-Moxibustion research platforms and teams and made research cooperation. She firstly carried out acupuncture-Moxibustion clinical and basic scientific research, which started the acupuncture-Moxi- bustion scientific research in China. ZHU Lian is the Pioneer of Chinese acupuncture-Moxibustion scientific research.

10.BAG-1, EGFR, and PARP-1 expressions in breast cancer and their clinical significance

To observe BAG-1, EGFR, and PARP-1 expressions in invasive breast cancer and its correlation with clini-cal pathological indicators, as well as to evaluate their clinical significance. Methods:The BAG-l, EGFR, and PARP-1 expressions in a tissue microarray of invasive breast cancer and peritumoral tissues were detected through immunohistochemical staining. The clinical and pathological significance of BAG-1, EGFR, and PARP-1 were evaluated. Results:The BAG-1, EGFR, and PARP-1 expression lev-els are higher in invasive breast cancer tissues than in peritumoral tissues (P<0.05). BAG-1 expression in invasive cancer tissues is not related to age, tumor site, lymph node metastases, and clinical TNM staging of patients, but is related to size, grade, ER, PR, and HER-2 expressions and molecular subtype (P<0.05). EGFR expression is related to size, clinical TNM staging, and molecular subtype (P<0.05). PARP-1 expression is related to grade, lymph node metastases, ER, and molecular subtype (P<0.05). BAG-1 expression is not significantly correlated with EGFR and PARP-1 in all cases, but BAG-1 and PARP-1 expressions are positively correlated in tri-ple-negative breast cancer tissues (P<0.05). Results of the univariate analysis revealed that the BAG-1 and PARP-1 expressions and the molecular subtypes are associated with the prognoses of breast cancer patients. Multivariate analysis revealed that BAG-1 and PARP-1 expressions are factors that are independent of the prognosis. Conclusion: BAG-1, EGFR, and PARP-1 overexpressions in human breast tissues suggest that BAG-1, EGFR, and PARP-1 are related to breast cancer development. BAG-1, EGFR, and PARP-1 are poten-tial biomarkers of breast cancer diagnosis and prognosis.
