Aberrant promoter methylation in special genes such as tumor suppressor genes is early eventin carcinogenesis of lung cancer. Aberrant methylation, resulting in corresponding mRNA silence or overexpression , closely relates to carcinogenesis, development, treatment response and prognosis of lung cancer and it may become a potential biomarker. Methylation of special genes may provide evidences for individual treatment of lung cancers.
2.Changes of the CD4+ CD25+regulatory T cells in infant with sepsis

Objective To study the change of CD4+ CD25+ Foxp3high regulatory T cells (Treg cells) and the molecules associated with Treg cells in different immune status in infant with sepsis, and to further clarity the pathogenesis of disturbed immune function in infant with sepsis. Method Totally 36 sepsis infants admitted in In-tensive Care Unit of Shenzhen Children' s Hospital from May 2007 to November 2007 and 16 age-matched healthy infants were collected for prospective study, after excluding autoimmune disease, immunodeficiency, inherited metabolic disorders, tumor, and drug-treatment that could affect immune function during lately 6 months. The study was approved by Ethics Committee of Shenzhen Children's Hospital. The 36 infants with sepsis were divided into two groups according to expression levels of HLA-DR in CD14-positive cells: DR-H group was defined as patients with HLA-DR > 30%, while DR-L group was defined as patients with HLA-DR < 30%. Expression levels of HLA-DR in CD14-positive cells and the proportion of Treg cells were analyzed by flow cytometry. Real-time PCR were used to evaluate the mRNA levels of Foxp3, CTLA-4,GITR, and IL-10 in CD4-posidve ceils. Statistical analysis was performed by one-way Anova. There was statistical difference with P < 0.05. Results The proportion of Treg cells in DR-L group was found to be significantly higher than that in healthy control or DR-H group (P <0.05).Compared with healthy control group or DR-H group, transcriptional levels of Foxp3, CTLA-4 and IL-10were significantly increased in DR-L group (P <0.05). The levels of GITR mRNA in DR-L group were detected to be higher than those in DR-H group (P < 0.05). Conclusions Aberrant increased proportion of Treg cells may be associated with suppressed immune status in infant with sepsis.
3.Impact of psychological intervention to compliance and quality of life for cancer patients and their primary caregivers

Objective To investigate the impact of compliance and quality of life of psychological intervention for cancer patients and their primary caregivers,as well as the correlation between the psychological issues of patients and their primary caregivers.Methods The enrolled patients were randomly divided into intervention group and control group.The patients in intervention group were given to standardize anti-tumor therapy,while the patients and their primary caregivers were given psychological intervention once a week.The patients in the control group only received standard anti-tumor therapy.By TDL determination of quality of life,anxiety rating scale (SAS) and self-rating depression scale (SDS),in front of the psychological intervention after 8 weeks of intervention,the two groups of patients and their primary caregivers were questionnaired,and recorded the completion of the treatment plan.By SPSS 12.0 software,the statistics were completed.Results 51 cases in intervention group and 38 cases in control group were able to complete the number of people expected to treat there was a statistically significant (P < 0.05).TDL determination and quality of life scores in intervention group patients and their primary caregivers were significantly higher (P < 0.05).SAS and SDS score in intervention group patients and their primary caregivers were significantly lower than control group (P < 0.05).Conclusion The effective psychological intervention to cancer patients and their primary caregivers during the treatment of patients could improve the compliance of cancer patients,the quality of life of cancer patients and their primary caregivers.The psychological problems between the patients and primary caregivers are positive correlation.
4.Three different strategies of urine drainage following hypospadias surgery: clinical nursing and observation.

OBJECTIVETo study 3 different strategies of urine drainage following hypospadias urethroplasty, the clinical nursing in their application, and their effects.
METHODSWe retrospectively analyzed the clinical data of 595 cases of hypospadias treated by urethroplasty. After surgery, 133 of the patients underwent urine drainage by suprapubic cystostomy (group A), 202 by urethral stent- tube indwelling (group B), and 260 by early initiative micturition with the urethral stent-tube (group C). All the patients received routine postoperative nursing care required for hypospadias repair.
RESULTSOperations were successfully completed in all the cases. Group C showed a remarkably shorter hospital stay and lower incidence rates of urinary fistula and urethral stricture than groups A and B (P<0.05), but there were no significant differences in the three indexes between A and B (P<0.05).
CONCLUSIONFor urine drainage following hypospadias repair, early initiative micturition with the urethral stent-tube can significantly reduce postoperative complications, decrease difficulties and workload of nursing care, and shorten the hospital stay of the patient.

5.Effects of acute cold exposure on pulmonary proinflammatory cytokine of rat.

OBJECTIVETo study the effects of acute cold exposure on the inflammation and pathologic injuries in pulmonary of rats, and explore the mechanism induced by cold stress.
METHODSForty male Wistar rats were randomly divided into five groups(n = 8): control group (23 ± 2) °C 2.5 h, -25°C 0.5 h group, -25°C 1 h group, -25°C 2 h group and -25°C 2.5 h group. Rats were exposed to cold at -25°C and no wind by keeping them in a low temperature chamber except control group. Rectal temperatures of the rats were measured before and after cold exposure. The morphological changes of pulmonary were observed by the optics microscope. The levels of tumer necrosis factor-α(TNF- α), interleukin-6 (IL-6) and interleukin-β (IL-1β) in lung tissue homogenate were measured by ELISA.
RESULTSCompared to the control group, body core temperatures of the -25°C 1 h group, -25°C2 h group and -25°C 2.5 h group were decreased significantly, and the D-values of rectal temperature were increased before and after cold exposure (P < 0.05). The infiltration of inflammatory cells and alveolar edema fluid appeared in the lung tissue of the -25°C 2.5 h group. The concentrations of tumor necrosis factor-α (TNF α), interleukin-6 (IL-6) and inter- leukin-1β (IL-1β) in lung tissue homogenate were increased significantly in -25°C l h group, -25°C 2 h group and -25C° 2.5 h group (P < 0. 05).
CONCLUSIONThe infiltration of inflammatory cells and the increase in proinflammatory cytokine from pulmonary may lead to the lung tissue injury after acute cold exposure.

6.Pharmacokinetic Effect of Aikeqing Granule by Different Medication Ways on Zidovudine in HAART of Rats.

OBJECTIVETo study pharmacokinetic effect of Aikeqing Granule (AG) by different medication ways on zidovudine (AZT) in highly active antiretroviral therapy ( HAART) of rats.
METHODSTotally 36 rats were administered with corresponding medications by gastrogavage, group I [HAART: AZT 31.5 mg/kg +3TC 31.5 mg/kg + Efavirenz (EFV) 63.0 mg/kg], group II (HAART+AG525 mg/kg), group III (HAART and AG 525 mg/kg after a 2-h interval). Drug concentrations of AZT were determined by high performance liquid chromatography-mass spectroscopy (HPLC-MS) before HAART, and at 0.5, 1, 2, 3, 4, 6, 8, 10, 12 h after HAART, respectively. Pharmacokinetic parameters [such as t1/2, Tmax, Cmax, AUCo-t, plasma clearance rate (CL)] were calculated by DAS2.0 Software.
RESULTSThe-equation of linear regression of AZT was good, with the precision, coefficient of recovery, and stability definitely confirmed. AUC in group II and III was larger than that of group I. There was no statistical difference in t1/2, Tmax, Cmax, AUC0-12 h, or AUC0-∞ among groups (P > 0.05).
CONCLUSIONAG combined HAART could enhance the Cmax of AZT.

7.Effect of sevoflurane exposure on mRNA expression of N-methyl-D-aspartate receptor subunit in neonatal rat hippocampi

Objective To investigate the effect of sevoflurane on mRNA expression of N -methyl -D -aspartate ( NMDA ) receptor subunit in neonatal rat hippocampi.Methods Twenty-four SD rats were devided into 2 groups( n=12 each) via a random number table:sevoflurane group and control group.The 3.0% sevoflurane and oxy-gen inhalated for 6 h respectively.Rats were sacrificed at 24 h after inhalation of sevoflurane or oxygen.Hippocampi were removed to idendtify the mRNA expression of NMDA subunit and the ratio of NR2 B/NR2 A through in situ hybridization.Results Compared with control group, NR2BmRNA and the ratio of NR2B/NR2A increased, and NR2 AmRNA decreased significantly in sevoflurane group ( P<0.05 ).Conclusion Neurotoxicity of sevoflurane in immature hippocampi might be attributed to regulation of mRNA expression of NMDA subunit in neonatal rat.
8.Current status of social anxiety and depression among primary school students in Urumqi, China.

OBJECTIVETo investigate the current status of social anxiety and depression among the fourth-, fifth-, and sixth-grade students in Urumqi, China, and to provide data support for mental health education for primary school students in the future.
METHODSStratified cluster random sampling was performed to select 919 fourth-, fifth-, and sixth-grade students from four primary schools for the Han and minority ethnic groups in Urumqi as research subjects. Social Anxiety Scale for Children and Children's Depression Inventory were used to evaluate the current status of social anxiety and depression.
RESULTSAmong the 919 fourth-, fifth-, and sixth-grade students, the detection rate of social anxiety was 28.5% (262 students). The fourth-grade students had significantly lower scores on each subscale of social anxiety and total score of social anxiety than the fifth- and sixth-grade students (P<0.05). Girls had significantly higher score on the subscale of fear of negative evaluation and total score of social anxiety than boys (P<0.05). The Uyghur students had significantly higher scores on each subscale of social anxiety and total score of social anxiety than the Han students and the students of other minority ethnic groups (P<0.05). Among these fourth-, fifth-, and sixth-grade students, the detection rate of depression was 11.2% (103 students). The fourth-grade students had significantly lower scores on the subscales of negative self-esteem, ineffectiveness, and interpersonal problems and total score of depression than the fifth- and sixth-grade students (P<0.05). The Uyghur students had significantly higher scores on the subscales of anhedonia and negative mood and total score of depression than the Han students and the students of other minority ethnic groups (P<0.05).
CONCLUSIONSThere are significant differences in the status of social anxiety and depression among the primary school students with different grades, sexes or ethnic groups in Urumqi. The fifth- and sixth-grade students, girls, and Uyghur students tend to have more serious psychological problems, which should be taken seriously by parents, schools, and the society.
9.Intestinal absorption of pulchinenosides from Pulsatilla chinensis in rats.

HPLC-ELSD was applied to explore the absorption mechanism of pulchinenosides (B3, BD, B7, B10, B11) in rats. The experimental results showed that the absorption rate constant, Ka value (B3, BD) and Permeability coefficient, Peff value (B3, B7) displayed significant difference (P <0.05) in various intestinal segments, The Ka value and Peff value of PRS was different from each other with the highest absorption in duodenum (duodenum > jejunum > colon > ileum); The PRS displayed excessive satuation as the concentration increased over 0.05-2.5 g · L(-1). There were no obvious linear correlations between Peff values and concentrations in duodenum (0.6007 ≤ R2 ≤ 0.7727); Ka and Peff value declined when the PRS was perfused with P-glycoprotein promoter digoxin, on the other hand, inclined when perfused with P-glycoprotein inhibitor verapamil with significant difference among PRS B3, BD, B7, B11 (P <0.05). All the above results demonstrated that B3, BD, B7 were greatly influenced by absorption sites, duodenum was the main absorption site; PRS didn't entirely transported in a concentration dependent manner, and the transporter-protein involved the transportation, so the intestinal absorption of the five pulchinenosides was not entirely passive diffusion; and PRS might be the substrates of P-glycoprotein.

10.Current status of social anxiety among primary school students with myopia in Urumqi, China and risk factors for myopia.

OBJECTIVE:
To investigate the prevalence of social anxiety among the fourth-, fifth- and sixth-grade primary school students with myopia in Urumqi, China and the risk factors for social anxiety.
METHODS:
Stratified cluster random sampling was used to select 552 fourth-, fifth- and sixth-grade primary school students with myopia from four primary schools in Urumqi. A self-designed questionnaire, a social anxiety scale for children and a self-esteem scale were used to investigate the general demographic data and the current status of social anxiety and self-esteem. A multivariate logistic regression analysis was used to investigate the risk factors for social anxiety in primary school students with myopia.
RESULTS:
Of the 552 children, 173 (31.3%) were found to have social anxiety. The multivariate logistic regression analysis showed that a higher grade, female sex, autocratic family, high myopia, low self-esteem and wearing glasses for more than 2 years were risk factors for social anxiety (P<0.05).
CONCLUSIONS
There is a serious problem of social anxiety among the fourth-, fifth- and sixth-grade primary school students with myopia in Urumqi. The development of social anxiety is associated with age, sex, degree of myopia, time of wearing glasses, parental education style and self-esteem level.
