1.Preparation of porous biodegradable chitosan/berberine hydrochloride composite xerogel and its antibacterial and hemostatic properties

BACKGROUND:Chitosan (CS) holding a good biocompatibility, biodegradability, antibacterial property and hemostasis effect cannot only promote wound healing and tissue repair, but also exhibit sustained-control release when carrying berberine hydrochloride (Bh). Therefore it functions as an excellent antibacterial and hemostatic material. OBJECTIVE:To prepare the porous CS/Bh composite xerogel and observe its antibacterial and hemostatic properties. METHODS:The porous CS/Bh composite xerogel was prepared using freeze-drying method, and its microstructure was characterized by scanning electron microscope and Fourier transform infrared spectrometer. The biocompatibility was detected through MTT assay and the in vitro antibacterial and hemostatic properties were observed through antibacterial test and blood clotting index. RESULTS AND COCNLUSION:The porosity of the CS/Bh composite xerogel was 83.1%-90.5%and almost degraded after 72 hours of immersion in PBS. The in vitro antibacterial and hemostatic properties of the composite xerogel were significantly better than those of CS, which were enhanced with the Bh content increasing. Additionally, the CS/Bh (10%) composite xerogel showed the best hemostatic and antibacterial abilities in vitro. To conclude, the CS/Bh composite xerogel possesses a good biocompatibility and strong hemostatic and antibacterial abilities, which is a promising postoperative antibacterial and hemostatic material.
2.Protective effects of hyperbaric oxygen in secondary spinal cord injury

Objective To investigate the mechanism underlying the protective effect of hyperbaric oxygen (HBO) in secondary spinal cord injury (SCI). Methods Models of acute SCI were established in 96 SpragueDawley rats using Allen's dropping weight technique. The rats were then divided into a HBO group, a high pressure nitrogen normal oxygen group, a normal pressure oxygen group and a normal pressure air group. The injured spinal cords were sampled for morphological studies at the 1 st, 3rd, 7th and 14th day after injury. Apoptotic cells were labeled using the TUNEL technique, and the expression of caspase-3 was detected. The neurological functionality of the spinal cord was assessed by open field locomotor evaluation ( the BBB score). Results The expression of caspase3 in the HBO group decreased significantly more than in the other groups after injury. The number of TUNEL-positive cells was significantly lower in the HBO group as well. Neurological function improved significantly after HBO therapy. Conclusions HBO can down-regulate the expression of caspase-3 and inhibit cell apoptosis in rats after SCI.The protective effect of HBO was related with the oxygen level.
3.THE EXPRESSION OF SSEA-1 AND OCT-4 ON HUMAN EMBRYONIC GERM CELLS

Objective To culture human embryonic germ cells in vitro and identify them by observing the expression of the surface markers SSEA-1 and Oct-4. Methods The gonadal ridges and mesenteries of human embryos at 5~9 weeks post-fertilization were isolated and then cultured in vitro.Flow cytometry and immunocytochemistry were applied to detect whether the cells cultured express SSEA-1 and Oct-4.Immunohistochemistry was also carried out to observe the expression of SSEA-1 on the PGCs which locate in the gonadal ridge. Results Both the cells and the cellular colonies cultured in vitro expressed SSEA-1 and Oct-4.The immunohistology study of gonadal ridges also showed that there were many PGCs which were SSEA-1 positive in it.Conclusion The cells we obtained through tissue culturing are undifferentiated human embryonic germ cells,which originate from hPGCs.
4.Clinical significance of anti-β2glycoprotein I antibodies in systemic lupus erythematosus

Objective To investigate the clinical application of anti-β2 glyeoprotein Ⅰ antibodies (IgG, IgM, IgA)in systemic lupus erythematosus(SLE). Methods The anti-β2-GP Ⅰ antibodies and anti-cardiolipin antibodies(ACL) level were measured by ELISA in 100 SLE patients, 39 other rheumatoid arthri-tis patients and 30 healthy control people. Their clinical application was analyzed in SLE diagnose and thera-py. Results The level of anti-β2-GP Ⅰ (IgG, IgM, IgA)were significantly higher in SLE than that in healthy (P < 0. 01 ). Sensitivity, specificity, positive predictive value and negative predictive value were 17.2%, 95.7%, 85.0% and 44. 6%, respectively. There was a significant and positive correlation be-tween anti-β2-GP Ⅰ antibodies and ACL antibodies ( IgG, IgM, IgA) (r = 0.418, 0. 624, 0.518, 0. 583, P <0.01). In multivariate analysis the factors(anti-β2-GP Ⅰ antibodies, ACL antibodies, dsDAN, u1-RNP, Sm, SSA, SSB, Jo-1, Scl-70, P-protein, PT, APTT) associated with SLE disease activity index(SLEDAI) were anti-β2-GP Ⅰ (IgG) and dsDNA. Conclusion anti-β2-GP Ⅰ antibody has high specificity and positive predict value, also is associated with SLE's thrombosis. It has some values in the clinical application.
7.Pseudo-words reading of Chinese patients with mild Alzheimer's disease

Objective To analyze the language function impairment differences between the two steps from the form to sound processing and from the sound to meaning processing for pseudo-words reading in Alzheimer' s disease (AD).Methods The pseudo-words reading test consists two parts:pseudo-true words and pseudopseudo words.Each part had 10 words.All of the 20 words were presented randomly.Correct reading presents the processing of from form to sound,and correct judging presents the processing of from sound to meaning.Each correct respond get 1 score,otherwise get 0.Each part had a total score of 20.Results The pseudo-words reading test was performed to 41 mild AD participants with Mini-mental State Examination (MMSE) average scores of (21.06 ±2.86),and 135 normal controls with MMSE average scores of (28.08± 1.52).The two groups were matched in age,gender and education.For all of the mild AD group and normal control group,there was no significant difference between reading score and judging score in either pseudo-true words or pseudo-pseudo words (P>0.05).Comparing with mild AD group and normal control group,there was no significant difference in reading score in either pseudo-true words or pseudo-pseudo words (P>0.05).For judging score,there was significant difference in pseudotrue words (P<0.01) but not in pseudo-pseudo words (P>0.05).Conclusion Understanding real syllable is impaired in patients with mild AD.
8.Repressive effects of RNA interference technique on expression and function of glucocorticoid receptor in human macrophage cell line U937

Objective To establish a glucocorticoid receptor knockdown model of human macrophage cell line U937 with RNA interference technique. Methods Two RNAi recombinant plasmids (named pSilencer 3.1-GR 1 and pSilencer 3.1-GR 2) targeting to GR gene were constructed. After RNAi recombinant plasmids were transfected into human macrophage cell line U937, the expressions of GR mRNA and GR protein were evaluated with RT-PCR and Western blotting respectively. The transcriptional activation function of GR was evaluated through the detection of relative luciferase activity after dexamethasone treatment. Results Two RNAi recombinant expression plamids were constructed and identified by sequencing. pSilencer 3.1-GR 2 transfection could inhibit not only GR mRNA and protein expressions, but also transcriptional activation function of GR specially; pSilencer 3.1-GR 1 transfection had no significant changes as compared to normal control. Conclusion A glucocorticoid receptor knockdown model has been established successfully, which offers a new method for the further research of GR biological functions.
9.Roles of distinguish vascular dementia from Alzheimer's disease with memory and executive screening scale

Objective To investigate the value of differential diagnosis for subcortical ischemic vascular dementia(SIVD)and Alzheimer's disease(AD)with memory and executive screening(MES) scale. Methods Fifty SIVD and 50 AD patients treated in the outpatient and wards of the Department of Neurology,Fuxing Hospital,Capital Medical University from June 2012 to November 2013 were enrolled retrospectively. They were enrolled in either a SIVD group or an AD group. Fifty healthy subjects in Yuetan community of Fuxing hospital were selected as a control group at the same period. The age,sex and education level of the subjects in the 3 groups were matched. The Mini-Mental State Examination( MMSE) and MES were used to measure all subjects and were compared;receiver operating characteristic curves was used to analyze the radio of memory and executive part for SIVD and AD discrimination. Results (1)There was no significant difference in total MMSE score between the SIVD group and the AD group(20. 6 ± 5. 5 vs. 20. 6 ± 3. 3;P>0. 05).(2)There were significant differences in each MES indexes among the patients of the 3 groups(all P>0. 05). The 1,3,4 and 5 scores for the sentence memory in the SIVD group were significantly higher than those of the AD group. There were significant differences(all P<0. 01). The indicators of executive part:the continuous action,instruction and action contradictory stimulation,knock and un-knock with finger scores of the SIVD group were all lower than those of the AD group(5. 1 ± 3. 1 vs. 7. 3 ± 2. 1, 5. 2 ± 3. 0 vs. 7. 7 ± 1. 9,3. 5 ± 4. 4 vs. 6. 2 ± 4. 3;all P<0. 01). The MES-R of the SIVD group was higher than that of the AD group(1. 2 ± 0. 7 vs. 0. 5 ± 0. 2;P>0. 05).(3)When MES-R was≥0. 7,the sensitivity and specificity of differentiating SIVD were 76. 7% and 83. 3%,and the total score of MMSE for differentiating the sensitivity and specificity of SIVD were 40. 0% and 0. 7%. Conclusion The differences of MES reflecting sentence memory and executive part may better distinguish the patients with SIVD and AD.
10.Change of serum interleukin-23 levels and sacroiliac joint magnetic resonance imaging before and after treatment with recombinant human tumor necrosis factor-α receptor Ⅱ IgG Fc fusion protein for injection in axial spondyloarthritis patients

Objective We investigated interleukin (IL)-23 that might play a role in the pathogenesis of rheumatoid arthritis (RA) and whether it was correlated with disease activity and clinical manifestations in axial spondyloarthritis (SPA).In addition, the Spondyloarthritis Research Consortium of Canada (SPARCC) scores was used to examine whether recombinant human tumor necrosis factor-α receptor Ⅱ IgG Fc fusion protein for injection (rhTNFR:Fc) was effective for the reduction of magnetic resonance imaging (MRI)-proven sacroiliac joint (SIJ) inflammation.Methods The serum IL-23 levels of etanercept and conven-tional treatment groups were measured using enzyme-linked immunosorbent assay kits.At the same time, SIJ MRI SPARCC score levels were assessed by MRI, the change of clinical indicators of patients was observed.ANOVA, repeated measure data of ANOVA and Spearman's correlation analysis were used for statisical analysis.Results ① The basal serum IL-23 levels of the Etanercept group were (34.2±1.8) pg/ml and those of the conventional treatment group were (34.1 ±1.8) pg/ml (F=1 073.790, P=0.991), both were significantly higher than the healthy control group (18.1±0.8) pg/ml (P=0.005).After treatment, serum IL-23 levels of the rhTNFR: Fc treatment and conventional treatment group were (24.5 ±1.7) pg/ml and (25.2±1.7) pg/ml (F=232.488, P=0.242), (19.2±0.8) pg/ml and (21.6±1.3) pg/ml (F=114.135, P=0.025), (19.0±0.8) pg/ml and(19.4± 0.8) pg/ml (F=23.374, P=0.085) respectively.A significant decrease was observed in the two groups and the serum level of IL-23 of the rhTNFR:Fc group was lower than that of the conventional group at week 12.② SIJ MRI SPARCC scores of the rhTNFR:Fc group and the conventional drugs group were (20.1± 1.2) scores and(20.7±1.5) scores (F=2003.660, P=0.191), (12.5± 0.8) scores and (15.4±0.9) scores (F=1 680.430, P=0.004), (8.8±0.9) scores and (12.8± 0.9) scores (F=972.877, P=0.002), the scores of two group were significantly decreased after treatment.At week 12, 24 of the treatment, the rhTNFR:Fc group scores were lower than the conventional drugs group.③ The serum IL-23 levels, SLJ MRI SPARCC scores and clinical index (ESR, CRP, PGA, BASDAI, BASFI, BASMI and number of painful joints) were not correlated (P>0.05), the SIJ MRI SPARCC scores and clinical indicators were not correlated (P>0.05).Conclusion ① The serum IL-23 levels of the rhTNFR:Fc are higher than healthy controls.② rhTNFR:Fc treatment could significantly decrease IL-23 levels and improve the sacroiliac joint inflammation compared to conventional treatment.③ SIJ MRI is a good assessment method for the detection of SpA sacroiliac joint inflammation.
