1.IMMUNOPATHOLOGIC STUDY IN THYROTOXICOSIS WITH THYROIDITIS AND HASHIMOTOXICOSIS

A morphologic and immunohistochemical studies were performed on the thyroid tissue of 13 thyrotoxicosis with thyroiditis(TTOT) and 9 hashimotoxocosis(HSHT). We found that the inflmmatory injure in HSHT was stronger than that in TTOT, and also, there were (?)ore infiltrating cells and activated T cells in HSHT (?) that in TTOT. Nevertheless, in both HSHT and TTOT, most of the infiltrating mononclear cells were T lymphocytes, and the proportion of T cell subsets, B lymphocytes and macrophages were almost the same, suggesting there might. be similar immunopathogencsis in these two diseases.
2.EFFECTS OF ABERRANT HLA-DR ANTIGEN EXPRESSION ON THYROCYTES

Tru-cut specimens of thyroid tissue from 37 cases of autoimmune thyroiditis, 23 cases of hyperthyroidism and 13 cases of thyrotoxicosis with thyroiditis were analyzed by immunohistochemistry using monodonal antibody specific for HIA-DR antigen. Results showed that thyroid epithelial cells aberrant expressed DR antigen and the number of DR~+ thyrocytes was significantly increased in thyroid gland with intense lymphocytes infiltration and severe destruction of throid architecture. The data suggest that TSH, thyroglobulin antibodies, thyroid microsomes antibodics can bring some stimulation HLA-DR antigen expression in thyrocytes.
3.Research progress of pathogenesis of phenotype shift of gastric intestinal metaplasia

Gastric intestinal metaplasia (IM), a common clinical pathological diagnosis, is a premalignant condition of intestinal type gastric cancer. The pathogenesis is still unclear, which links with helicobacter pylori (Hp) infection, bile reflux, nitrite intake, vitamin deficiency and so on. In recent studies, the relevant genes involved with transdifferentiation of gastric epithelium to intestinal epithelium are CDX2, SOX2, PDX1, OCT1, SHH, Runx3, Tff, and so on. Overall, the phenotype shift of gastric intestinal metaplasia is a complex and long process, current treatments targeted to the cause of IM such as Hp infection, bile reflux only can prevent its development, but fail to reverse the clinical process, so further clarification to the pathogenesis is still needed to guide the clinical treatment.

Brain ischemia/hypoxia preconditioning(I/HPC) is one kind of endocardial protective phenomenon by organism against blood and oxygen deficit.Researches discovered various proteins such as hypoxic inducible factor-1(HIF-1),heat shock proteins(HSPs),erythropoietin(EPO),Bcl-2 family,calcitoningene related peptide(CGRP) and so on participated in brain I/HPC.This essay explains function and participating I/HPC mechanism of these proteins.
6.Effects of muscle-derived stem cell transplantation on denervated gastrocnemius atrophy in rats

BACKGROUND: Studies concerning muscle-derived stem cell transplantation mainly focused on treatment of muscular dystrophy. There are no studies on muscle-derived stem cells for treatment of denervated amyotrophy combined therapy. OBJECTIVE: To explore effects of muscle-derived stem cell transplantation on denervated gastrocnemius atrophy. DESIGN, TIME AND SETTING: The randomized controlled animal study was performed at the Shenyang Medical College from February 2007 to July 2008. MATERIALS: A total of 19 adult healthy male Wistar rats were supplied by Animal Experimental Center, Shenyang Medical College. METHODS: Triceps brachii was sterilely collected from 3 rats, and muscle-derived stem cells were isolated in vitro, and purified by adherence method. The tibial nerve of right posterior limb was cut in remaining 16 rats to establish animal models of denervated gastrocnemius. In the cell transplantation group, 0.2 mL DAPI-labeled muscle-derived stem cell suspension was infused into the denervated gastrocnemius. The same volume of saline was injected into the model control group, for 4 weeks. MAIN OUTCOME MEASURES: Computer biological signal collection processing system was used to measure recovery rate of threshold intensity of gastrocnemius and the optimal intensity recovery rate. Electronic balance was utilized to calculate residual rate of wet weight. Image analysis system was used to measure residual rate of cross section area. 7.The empirical pharmacodynamics study of compound orange peel bamboo shavings formula capsules in chemical constipation

Objective To explore the effect of compound orange peel bamboo shavings formula capsules on constipation by examining on chemical constipation mice.Methods Constipation was induced by oral administration of diphenoxylate and ondansetron.Mice were fed with compound orange peel bamboo shavings formula capsule at the doses of 3.2, 1.6 and 0.8 g/kg, respectively.Results The middle dose and high dose of compound orange peel bamboo shavings formula capsule shortened the start time of defecation, and increased fecal pellets number and wet weight in the diphenoxylate-induced constipated mice or the ondansetron-induced constipated mice (P < 0.05).Conclusion Compound orange peel bamboo shavings formula capsule produces a laxative activity against diphenoxylate-induced constipated mice or ondansetroninduced constipated mice.
8.Clinical significance of level of CD4+CD25HiCD127Low regulatory T cells in the peripheral blood of patients with esophageal cancers

Objective To detect the levels of CD4+CD25HiCD127Low regulatory T cells (Treg) in the peripheral blood and its clinical significance in patients with esophageal cancer. Methods The levels of Treg in the peripheral blood were detected by three-color flow cytometry (FCM) in 80 patients with esophageal cancer and 20 healthy controls. Among the 80 patients, 30 patients were also further studied for preoperative and postoperative comparison after operation. The clinical and pathological data of each patient were collected and analyzed for the correlation with the Treg levels. Results The level of Treg in the peripheral blood of the control group was lower than that of the esophageal cancer patients [(3.36±1.14) % and (5.70±1.96) %, respectively], with significant difference (P <0.01). The levels of Treg in the peripheral blood was higher in the patients with metastasis of lymph node (n=40) than that in the patients without metastasis of lymph node (5.96±1.36) % and (4.23±1.18) %, respectively] (n=30), with significant difference (P <0.01). The levels of Treg in the peripheral blood of the patients were negatively correlated with their TNM classification. As the TNM classification advanced, the level of the Treg in the peripheral blood increased. Conclusion The levels of Treg in the peripheral blood of the patients with esophageal cancer are significantly higher than that of the healthy subjects, which is correlated with the clinical and pathological conditions. The development of esophageal cancer may relate with suppression of immune function.
9.Study on cell-penetrating peptides carrying foreign proteins into eyeball

Objective To investigate whether poly arginine as the carrier can carry foreign proteins to penetrate the cell membrane and even penetrate the eyeball barrier.Methods Poly-Args (R9) was used as a CPP in this study.R9-green fluorescent protein (GFP) and GFP were constructed.In vitro,human lens epithelial cells were treated with these two proteins.Then,MTT assay were used to detect whether the protein could affect the proliferation of the cells.Flow cytometry and laser confocal microscopy were used to detect the penetrability of CPPs on the cells.In vivo,eyes of mice were treated with protein in eye drops way for 7 days.Then total protein were extracted,ELISA were used to detect the penetrability of CPPs.Results The results of MTT,flow cytometry and laser confocal microscopy showed that CPPs could carry protein into cells in a dose dependent manner without affecting cell proliferation.In vivo,slit lamp showed that the mice eyeballs had no any abnormal after treated by GFP,R9-GFP,and ELISA results also showed that R9 could effectively get foreign protein into the eyeball.Conelusion R9 can carry foreign protein into the cell membrane and eyeball barrier.This study provides the basis for the eye medication and dosing mode improvement.
10.Esophageal teratoma complicated with rupture of esophagus:A case report and literature review

Objective:To explore the clinical appearance,diagnosis method and curative efficacy of one patient with esophageal teratoma complicated with rupture of esophagus in order to raise the awareness of the clinicians for the disease. Methods:The patient with chest pain after eating was admitted to hospital,and upper gastrointestinal radiography,chest CT and endoscopy were taken,which showed rupture of esophagus.Then an emergency thoracic exploration was performed.A too long cleft and much necrosis were identified,so the repair of esophagus was changed into excision and anastomosis. Results: The operation was successful,and the pathological findings showed the rupture of esophageal teratoma.Follow-up showed no complication till now.Conclusion:Esophageal teratoma complicated with esophageal rupture is rare.Clinicians should improve the understanding and reduce the misdiagnosis rate.
