Objective To investigate multiparametric immunophenotypic features in patients with acute myelocytic leukemia(AML)-M_2 bearing AML-1/ETO gene rearrangements and its predicting value.Methods A multiparametric flow cytometry was used in the study of phenotypic characterization of the subtype of AML.Immunophenotype of 30 patients with AML(M_2/ETO~+)was analyzed by fluorescence in situ hybridization(FISH).The results were compared with 36 patients of AML-M_2 with AML-1/ETO~- (M_2/ETO~-)and 34 acute promyelocytic leukemia(APL)patients.Results There were a population. 15.89%-68.53% the blast cell and a population of more differentiated and heterogeneous myeloid cells in the marrow of 30 patients with M_2/ETO~+.The blast cells had a myeloid phenotype(CD_(33),CD_(13)and MPO) and showed a characteristic pattern of antigen expression.The fluorescent intensity of CD_(33)in patients with M_2/ETO~+ was less than in patients with M_2/ETO~-and APL [ mean fluorescent intensity(MFI):98?75 v. 244?184 and 845?523,both P
2.Effect of Respiratory Training on Motor Function in Acute Stroke Patients

Objective To investigate the effects of respiratory training on motor function in patients with acute stroke. Methods From 2012 to 2014, 80 patients with acute stroke were randomly assigned into treatment group and control group equally. The control group re-ceived routine rehabilitation training, while the treatment group received respiratory training in addition. All the patients were assessed with Fugl-Meyer Assessment (FMA) and modified Bathel Index (MBI) before and eight weeks after treatment. Results The scores of FMA and MBI improved more in the treatment group than in the control group (t>3.938, P<0.001) after treatment. Conclusion Respiratory training may promote the recovery of motor function in acute stroke patients.
3.Correlation between Serum Brain Natriuretlc Peptide and Cardiac Function in Children with Pneumonia

Objective To explore the correlation between serum brain natriuretlc peptide(BNP) and cardiac function in children with pneumonia.Methods Fifty-two children with pneumonia were seleated.Among them 20 were complicated with congestive heart failure(CHF),including 9 boys and 11 girls,their ages ranging from 4 months to 4 years old;and 32 cases non-complicated with CHF,including 17 boys and 15 girls,their ages ranging from 5 months to 6 years old.Besides,there were 30 healthy children as control group,including 16 boys and 14 girls,their ages ranging from 6 months to 5 years old.For all the children,their serum BNP was measured by using ELISA met-hod,and CK-MB was measured by automatic biochemistry analyzer.In the sametime,their cardial function was measured by echocardiography.Results In the children with pneumonina complicated with CHF,the levels of serum BNP[(182.45?57.32) ng/L] and CK-MB [(48.19?16.08) IU/L] were higher than those of healthy control group[(35.12?13.84) ng/L,(11.48?4.29) IU/L Pa0.05).The levels of left ventricular ejection fraction(LVEF),left ventricle shortening fraction(FS),peak flow velocity of arteriae arota(PFVA),peak flow velocity of pulmonary artery(PFVP) in the children with pneumonia complicated with CHF were lower than those of healthy control group(Pa0.05).There was a positive correlation between the levels of serum BNP and CK-MB(r=0.28 P0.05).Conclusion The levels of serum BNP may be an important index in diagnosis of infant pneumonia complicated with CHF.
4.Experimental studies on the protective effect of defibrase againstreperfusion injury after cerebral ischemia

Aim To ascertain whether defibrase has the protective effect against reperfusion injury after cerebral ischemia.Methods 70 renovascular hypertensive rats(RHR) were randomly divided into defibrase group, control group and sham-operated group.Reversible middle cerebral artery occlusion(MCAO) models were produced by the modified. Longa's method,and reperfusion was begun 2 hours after occlusion.Rats in the defibrase group were given defibrase 10 U?kg-1 body weight via femonal intraveneous injection, and in the control group with the same amount of saline. The brain pieces were processed by TTC and HE staining and the infarct size,brain microvessels damage and secondary bleeding were compared between the two groups. Results The volume of infarction in the defibrase group was obviously smaller than in the control group, the damage of brain microvessels was less severe, and the bleeding lesions under optical microscope were less than in the control group. Conclusion Defibrase has protective effect against reperfusion injury post cerebral ischemia.
5.Coronary artery disease in Chinese adults with type 2 diabetes mellitus estimated by electron beam computed tomography and electron beam computed tomography angiography

Objective To assess the prevalence and risk of coronary artery disease (CAD) in Chinese adults with type 2 diabetes mellitus (T2DM) using electron beam computed tomography (EBCT) and EBCT angiography (EBCTA). Methods: Ninety-four cases were enrolled in this study including diabetes (n=28), impaired glucose tolerance (IGT, n=30), coronary heart disease (CHD, n=11), and control (n=25). Cardiac EBCT plain scanning and EBCTA were performed on all of these subjects to evaluate coronary artery calcification (CAC) scores, and number of segments of stenosed coronary arteries. Both CAC and/or coronary artery stenosis were defined as patients with coronary artery lesions (CAL). Results CAC scores were not different with the control, diabetes, IGT, or CHD (P>0.05)groups. Compared to control (0.520±1.295), more stenosed coronary arteries segments (P<0.05) were detected in diabetes (2.964±1.915), IGT (2.200±2.024), and CHD (2.273±1.679). Number of stenosed artery segments were correlated with age (r=0.215, P=0.019),postprandial glucose (r=0.224, P=0.015), total cholesterol (r=0.323, P=0.000), and duration of diabetes (r=0.208, P=0.004). The incidences of CAL in diabetes (96.43%), IGT (93.33%), and CHD (90.91%) was substantially higher than that in normal control (56.00%, P<0.01).The odds ratio of CAL associated with having diabetes was estimated to be 7.514 (95% CI: 1.885-63.778). Conclusions Coronary artery lesions are prevalent in Chinese adults with type 2 diabetes, implying a high CAD risk. EBCTA holds potential in depicting the details of CAL and can be used to track the progression of CAD in diabetes patients.
6.Determination of Anthraquinones in Xinshenyan Capsules by HPLC

Objective To establish an HPLC method for the determination of anthraquinones including rhein, emodin and chrysophanol in xinshenyan capsules. Methods Anthraquinones were determined by HPLC with Phenomenex-C18 column (250 mm×4. 6 mm, 5 μm) as the chromatographic column and methanol-1% acetic acid (70:30) as the mobile phase. The flow rate was 1. 0 mL·min-1 and the detection wavelength was set at 254 nm. Results The liner range of rhein, emodin and chrysophanol was 4. 96-24. 80 μg·mL-1(r=0. 999 6), 6. 58-32. 91 μg·mL-1(r=0. 999 9) ,and 15. 11-75. 55 μg·mL-1 (r=0. 999 9),respectively, and the average recovery was 100. 78%, 98. 13% and 99. 29%, respectively. Conclusion The method is simple and practical, the result is accurate and reliable and it can be used to determine the contents of rhein, emodin and chrysophanol in xinshenyan capsules.
7.Content Determination of Paeoniflorin and Icariin inQixue Shuangbu Oral Liquid by HPLC

Objective To improve the quality standard of Qixue Shuangbu Oral Liquid;To provide guarantee for the quality of oral liquids.Methods HPLC was used to simultaneously determine the contents of paeoniflorin and icariin in the oral liquid. The determination was performed on a Agiltnt TC-C18 (2) column (250 mm × 4.6 mm, 5μm) with the mobile phase of acetonitrile and 0.1% H3PO4 gradient mode. The flow rate was 1.0 mL/min;the column temperature was set at 25℃; the detection wavelength was 232 nm.Results The linear ranges of paeoniflorin and icariin were obtained between 0.542-8.677μg (r=0.999 9) and 0.185-2.963μg (r=0.999 9), with the average recoveries (n=6) of 99.29% (RSD=1.06%) and 98.91% (RSD=1.97%), respectively.Conclusion This method is accurate and feasible, which can be used conveniently in quality control ofQixue Shuangbu Oral Liquid.
8.Study on the Quality Standard for Naoxin Luotong Pills

Objective:To establish the quality standard for Naoxin Luotong pills. Methods:Astragali radix, bombyx batryticatus and scorpio were identified under a microscope. Peach kernel, astragali radix, paeoniae radix alba, rheum, paeonia suffruticosa and salvia miltiorrhiza were identified by TLC. The content of total ferulic acid was determined by HPLC with a Sapphire C18 column (250 mm × 4. 6 mm,5 μm). The mobile phase was acetonitrile-0. 1% phosphoric acid(12 ∶88)and the detection wavelength was 316 nm. The column temperature was 35℃ and the flow rate was 1. 0 ml·min-1 . Results:The microscopic characteristics of astragali radix, bombyx batryticatus and scorpio were obvious to be observed easily. The TLC spots of peach kernel, astragali radix, paeoniae radix al-ba, rheum, paeonia suffruticosa and salvia miltiorrhiza were clear and specific. Ferulic acid had a good linear relationship within the range of 5. 1-81. 6μg·ml-1(r=0. 999 9), and the average recovery was 103. 30% with RSD of 1. 96% (n= 9). Conclusion:The standard can be used for the quality control of Naoxin Luotong pills.

10.Relationship between endothelial microparticles and CD4+CD25+Foxp3+ regulatory T cells in patients with acute coronary syndrome

Objective:To explore the relationships between the endothelial microparticles (EMPs)in peripheral blood and CD4+ CD25 + Foxp3+ regulatory T cells and their cytokine levels in the patients with acute coronary syndrome (ACS),and to clarify the mechanism of EMPs in the pathogenesis of ACS by affecting the Treg cell differentiation and function.Methods:Twenty-three patients with stable angina (SAP)were allocated to SAP group,and 52 patients with ACS were allocated to ACS group.Twenty individuals with normal conventional coronary angiography results were recruited as control group.The levels of EMPs and the percents of CD4+ CD25 +Foxp3+ regulatory T cells in peripheral blood of the patients in various groups were measured by flow cytometry. The expressions of Foxp3 mRNA and the plasma levels of TGF-β1 were detected with Real-Time PCR and ELISA, respectively.Correlation analysis was performed between EMPs and regulatory T cells,Foxp3 mRNA expression level and TGF-β1 level.Results:Compared with control group and SAP group,the level of EMPs in peripheral blood of the patients in ACS group was increased significantly,but the percentage of regulatory T cells,the Foxp3 mRNA expression level and the plasma TGF-β1 level were decreased markedly (P <0.01).The correlation analysis showed that there were significantly negative correlation between EMPs and regulatory T cells,Foxp3 mRNA expression level and TGF-β1 level (r = - 0.452,P = 0.001;r = - 0.466,P = 0.001;r = - 0.555,P = 0.000 ). Conclusion:EMPs may involve in the process of occurrence and development of ACS and plaque instability by regulating the differentiation and function of CD4+ CD25 + Foxp3 + regulatory T cells.
