1.Literature-based analysis of conversion of components in fermentation process of Sojae Semen Praeparatum.

Based on the systematic retrieval and the reported components of Sojae Semen Nigrum and Sojae Semen Praeparatum, this study conducted in-depth analysis of conversion of components in the fermentation process, and discussed types and possible mec-hanisms of conversion of chemical components, so as to provide the basis for studying technology, medicinal ingredients and quality standards. According to the analysis, there is a certain degree of conversion of nutrients(like protein, sugar, lipid), bioactive substances(like isoflavones, saponins, γ-aminobutyric acid) and other substances(like nucleosides, melanoids, biamines, etc) in the process of fermentation.

2.Study on totai flavonoids of Epimedium assisted with soybean polysaccharide spray-drying powder.

In order to evaluate the characteristics of the spray drying of total flavonoids of Epimedium extracts assisted with soybean polysaccharide, a certain percentage of soybean polysaccharide or polyvidone were added to the total flavonoids of Epimedium extract to conduct the spray drying. The effect of soybean polysaccharides against the wall sticking effect of the spray drying was detected, as well as the powder property of total flavonoids of Epimedium spray drying powder and the dissolution in vitro behavior of the effective component. Compared with the total flavonoids of Epimedium spray drying powder, soybean polysaccharide revealed a significant anti-wall sticking effect. The spray drying power which had no notable change in the grain size made a increase in the fluidity, improvement in the moisture absorption and remarkable rise in the dissolution in vitro behavior. It was worth further studying the application of soybean polysaccharide in spray drying power of traditional Chinese medicine.

3.Study on preparation of salvianolic acid phospholipid compound.

To prepare salvianolic acid phospholipid compound. With the compound of salvianolic acids and soybean phospholipid as the index, mono-factor experiment and orthogonal design experiment were conducted to screen its technical parameters. According to the results, the optimal preparation conditions of salvianolic acid phospholipid compound were that THF were taken as the reaction solvent, the concentration time was 3 h, the reactant concentration was 5 g x L(-1), the mass ratio of salvianolic acids and phospholipid was 1: 1.5, and the reaction temperature was 40 degrees C. The oil/water partition coefficient of the prepared salvianolic acid phospholipid compound significant increased in water and buffers with different pH values. The results of phase analysis such as DSC, XRD and FTIR indicated that salvianolic acids existed in phospholipid in an amorphous state.

4.Study on preparation of intravenous submicron emulsions of Oleum Cinnamomi oil of Miao nationality herbal.

OBJECTIVETo study the prescription and preparation of intravenous submicron emulsion of Oleum Cinnamomi oil of Miao nationality herbal.
METHODUsing the high speed blender mixed round the Oleum Cinnamomi oil with the soybean phospholipids and Pluronic F68 as emulsifier, then using the high pressure homogenizer made the intravenous submicron emulsion of the Oleum Cinnamomi oil and investigate its grain path and distributing.
RESULTHaving been done by using hydroextractor 4,500 r min(-1) 15 minutes the submicron emulsion grain path has well proportioned distribution.
CONCLUSIONThe preparation technology is simple and has good stability, so it can be used as a method to make the intravenous submicron emulsion of the Oleum Cinnamomi oil of Miao nationality herbal.

5.Study on isoflavone active aglycone preparation by immobilized beta-glucosidase from Aspergillus niger.

With sodium alginate as a carrier and glutaraldehyde as the crosslinking agent, an improved immobilization method of beta-glucosidase for production of soybean genistein was developed. As compared with entrapment or entrapment-crosslinkage, crosslinkage-entrapment that beta-glucosidase was treated with glutaraldehyde and then entrapped in sodium alginate remained high loading efficiency and activity recovery, Effects of bead sizes, concentrations of alginate and glutaraldehyde as well, on the loading efficiency and activity recovery were assessed. When compared with the free enzyme, the optimum temperature, pH value and Km of the immobilized beta-glucosidase were respectively shifted from 50 degrees C to 40 degrees C, 4.5 to 4.0 and 2.57 microg/mL to 2.02 miocrog/mL. The stabilities of the immobilized beta-glucosidase were considerably better than that of the native enzyme. The immobilized beta-glucosidase was employed to genistein production, 84.94% of the activity and 56.04% of conversion were kept after consecutive use of 6 times.

7.Preparation and quality control of soybean isoflavone dropping pills.

OBJECTIVETo establish the optimized preparation procedure and study the method to determine the content for soybean isoflavone(SIF) Dropping Pills.
METHODThe preparation conditions, such as the proportion between SIF and PEGs, the temperature of mixture of SIF and PEGs, dropping distance, etc., were studied with Uniform Design and One-way ANOVA. SIF was identified by TLC and the content of SIF was determined by UV spectrometry at 262 nm detection wavelength.
RESULTThree batches of the prepared products meet the standards of the Chinese pharmacopoeia on dropping pills. SIF can be identified by TLC. Using UV spectrometry, the linear range of SIF was 0. 407 2 to 4. 072 g x mL(-1) and the correlation coefficient was 0. 999 8. In high, middle and low concentration, average recovery were 96. 54%, 97.27% and 97.21%, respectively (RSD were 1.3%, 0.78% and 0.71%).
CONCLUSIONThe preparation procedure is feasible, simple and suitable, the method established in this paper can be adopted for the quality control of SIF dropping pills, and the determination method is simple, relatively fast and accurate.

8.Effects of soy isoflavone on the reproductive system development and ER-beta expression in male rat offspring in different postnatal periods.

OBJECTIVETo investigate the effects of pre- and post-natal exposure to soy isoflavones (SIF) on the related indicators of the reproductive system development and ER-beta expression in male rat offspring.
METHODSPregnant SD rats were randomly divided into a corn oil group (control), 3 SIF groups (50, 200 and 400 mg/kg body weight), and a diethylstilbestrol (DES, 0.1 mg/ kg body weight) group to be treated respectively by intragastric gavage from pregnancy day 0. On postnatal day (PND) 21, the male offspring were treated in the same manner till sexual maturity, and on PND 49 and 90, 6 male rats from each group were killed for observation of the related indicators of the reproductive system development and the detection of the expression of ER-beta in the testis.
RESULTSThe mean daily food intake showed no evident differences among the SIF and DES groups, but the food efficiency ratio (FER) was significantly decreased in the 200 and 400 mg/kg SIF and 0.1 mg/kg DES groups (P < 0.05) at 49 and 90 PND. At the dose of > or = 50 mg/kg, SIF markedly reduced the body weight of the rats (P < 0.05), even more so at 90 PND than at 49 PND. The increased dose of SIF was correlated with the reduction of testis weight, sperm head count and daily sperm production, and so was it with the elevation of the ER-beta expression, both more obviously at 90 PND than at 49 PND.
CONCLUSIONPre- and post-natal exposure to SIF affects the reproductive system development of male rat offspring, and the elevation of ER-beta expression may be one of its mechanisms.

9.Effect of long term supplementation of mineral-fortified dephytinized soy milk powder on biomarkers of bone turnover in boys aged 12 to 14 years.

OBJECTIVESIn order to understand the effect of long term consumption of mineral-fortified dephytinized soy milk powder on skeletal health in children, biological effects of mineral-fortified dephytinized soy milk powder, dephytinized mineral-unfortified soy milk powder and fresh mineral-fortified cow milk were compared.
METHODSTotally, 90 boys aged 12 to 14 years were involved in the study and were given daily with mineral-fortified dephytinized soy milk powder, dephytinized soy milk powder and fresh fortified cow milk, respectively, for two months. The blood samples were collected from all the subjects for measuring activity of serum bone specific alkaline phosphatase and serum level of oesteocalcin, and the urine samples were collected for determining urine C-terminal telopeptides of type I collagen (CTx).
RESULTSAfter two-month intervention, serum level of oesteocalcin increased significantly in the all groups, (16.42 +/- 11.25) micro g/L for the group with fortified soy milk powder, (18.86 +/- 11.57) microg/L for the group with unfortified soy milk powder and (13.81 +/- 10.06) microg/L for the group with fortified cow milk, respectively. The activity of bone specific alkaline phosphatase significantly decreased in the all groups, by (0.28 +/- 0.40) micromol x s(-1) x L(-1) for the group with fortified soy milk powder, (0.26 +/- 0.35) micromol x s(-1) x L(-1) for the group with unfortified soy milk powder, and (0.17 +/- 0.28) micromol x s(-1) x L(-1) for the group with fortified cow milk, respectively. As well as, urine level of CTx decreased significantly, by (349.1 +/- 380.3) mg/L for the group with fortified soy milk powder, (260.4 +/- 412.4) mg/L for the group with unfortified soy milk powder, and (354.4 +/- 404.7) mg/L for the group with fortified cow milk, respectively.
CONCLUSIONSLong term consumption of mineral-fortified and dephytinized soy milk powder could improve bone mass retention and calcium nutrition status efficiently in children.
