1.Bone formation of alveolar augmentation with hydroxylapatite, bone morphogenetic protein and fibrin sealant composite

AIM: To investigate new bone formation of alveolar augmentation with hydroxylapatite(HA)+bone morphogenetic protein(BMP)+fibrin sealant FS(HA+BMP+FS) composite.METHODS:Bone morphogenetic protein was extracted from bovine cortical bone. Hydroxylapatite particles were combined with fibrin sealant and BMP, which is of high efficient osteoinduction, forming a new complex material. The alveolar augmentation was made with this composite. The samples were investigated 3, 6,12 and 24 weeks postoperatively, with gross observation, microscope, calculation of new bone formation, tetracycline fluorescent microscope, scanning electron microscope, backscattered electron image and X-ray energy analysis.RESULTS: 1. In experimental group, weave bone had been seen since 3 weeks postoperatively and more new bone 6 weeks postoperatively; remolded bones united and distributed to all spaces of HA particles 12~24 weeks postoperatively. In control group, no new bone appeared 3 weeks postoperatively, only some HA particles contacted base bone 6 weeks postoperatively, a little of new bone grew into HA spaces 12~24 weeks postoperatively. 2. New bone directly incorporated with HA. The quantity of calcium and phosphate are similar to HA and base bone. CONCLUSION:This composite in alveolar augmentation obviously promotes incorporation HA with base bone, speeds formation of new bone and adds amount of new bone, reconstructing a bony alveolar ridge.
2.Surgical treatment of carotid body tumor

Objective To explore the surgical treatment of carotid body tumor(CBT). Methods The clinical data of 78 patients with CBT were retrospectively analysed. Results Seventy eight patients had 88 tumors (10 patients had bilateral CBT),of which 83 were resected. The operations included:CBT enucleation (28 tumors);CBT resection along with the external carotid artery resection(12 tumors); CBT enbloc resection with partial carotid artery(CA) resection and interposition grafting(3 tumors) or end to end anastomosis (1 tumor), and external to internal CA anastomosis (1 tumor); CBT enbloc resection with CA reconstruction(30 tumors), and CBT resection and CA ligation (8 tumors). The operative mortality was 3.8%(3/78). 65 tumors were followed up for 1~21 years,and one patient died of pulmonary metastasis seven years after the surgery. In the follow up period, 1 patient had a local recurrence two years after the surgery,and reoperation was performed,the patient did well 8 years after the reoperation. One patient with family CBT had an opposite lesion 10 years after the first lesion resection. Conclusions CBT may be bilateral,family occurrence,malignant changes and endocrine secretion charateristics. For small CBT, enucleation can be done first.If CA is involved, external CA remove along with CBT and end to end anastomosis may be a better choice . For tumors invading the internal CA, it is better to resect the lesion with reconstruction of the CA with, an autogenous internal jugular vein graft or prosthesis. For large CBT or CBT adhensed to nearby tissues seriously, end to end anastomosis can be carried out to internal CA at a transversely cutting surface of CBT.
4.A clinical study on the treament of Sjogren's syndrome with electrical stimulation

objective: To investigate the effect of electrical stimulation on the treament of Sjogren's syndrom(SS) . Methods: Ten SS patients were treated with electrical stimulation at less than 6 V and 9 mA of the current for 6 min each time and three times each day, the treatment was countinued for 20 days. The symptom of dry mouth was classified into 0,1,2,3 and 4 according to a treatment emergent symptom scale (TESS). Unstimulated and stimulated whole saliva flow rates on chewing medical paraffin were determined before and after treatment.Results: The symptom of dry mouth was markedly improved in two SS patients, moderately in 2 and slightly in 5. No improvement was found in 1 case. Before and 1, 2 and 3 weeks after treatment unstimulated whole saliva flow rate(ml/min) was 0.14?0.35, 0.14?0.38, 0.16 ?0.35 and 0.22?0.42; stimulated 0.58?0.40, 0.61?0.41, 0.78?0.37 and 0.90?0.39, respectively.Conclusion: Electrical stimulation can markedly improve the symptom of dry mouth in the patients with SS.

Objective To make sure whether or not Bcrp1 is the marker of cervical cancer stem cells or not by studying the invasive ability and formation of tumors of Bcrp1 + phenotype HeLa cells.Methods The tumor cell migration and invasion assay were used by boyden chamber to identify the invasive ability of Bcrp1 + phenotype HeLa cells.The formation of tumors in vivo experiments were completed,in which the two groups of cells with different concentrations were inoculated in non obese diabetes-severe combined immunodeficiency disease ( NOD/SCID ) mice ( 1 × 104,1 × 105,1 × 106/ml ) and the differences of time,rate and volume in the formation of tumors between two groups were observed.Results ( 1 ) In the invasion assay,the amount of cells that invaded through the artificial basement membrane in Bcrp1 + group were 99 ± 14,which was significantly greater than those in Bcrp1- group ( 57 ± 13,P < 0.05 ) ; the length of the Bcrp1 + group was ( 366 ± 52 ) μm,which was significantly greater than the Bcrp1 - group ( 301 ± 54) μm ( P < 0.05 ).( 2 ) Following transplantation of 1 × 104 cells,only the Bcrp1 + cells formed tumors in NOD/SCID mice.When 1 × 105 or 1 × 106 cells were transplanted,the tumor incidence and the tumor mass were greater in the Bcrp1 + groups than those in the Bcrp1 - groups ( P < 0.05 ).Conclusion Bcrp1 + HeLa cell have the greater capacity of invasive and the tumorigenicity,which may contain cancer stem cells.
7.Effects of Jiawei Wuzi Yanzong Pill and Its Components on Central Cholinergic System in Mice with Learning and Memory Disorders Induced by Scopolamine

Objective To investigate the effects of Jiawei Wuzi Yanzong Pill and its components on central cholinergic system in the model mice with learning and memory disorders induced by scopolamine. Methods Mice were treated by Jiawei Wuzi Yanzong Pill and its components for 15 days and induced by ip scopolamine. After the experiment in Morris water maze, the content of acetylcholine and the activity of acetylcholinesterase from brains of mice were determined by colorimetry. Results Significant differences were observed between model group and naked group that the content of acetylcholine and the activity of acetylcholinesterase from brains of model mice were decreased (P 0.05). Conclusion Jiawei Wuzi Yanzong Pill and its components could improve the function of central cholinergic system by increasing the content of acetylcholine and the activity of acetylcholinesterase and possesses a certain preventive effects on mice with learning and memory disorders induced by scopolamine.
8.Identification and characterization of stem cells in an ovarian cancer cell line and examination their drug resistance

Objective To isolate side population (SP) cells from an established ovarian cancer (OC)cell line,characterize these cells,and examine their drug resistance. Methods SP and non-SP (NSP) cells were isolated by fluorescence-activated cell sorting (FACS),and cultured in differential conditions,then detected their SP ratio to compare their capability of differentiation and self-renewal. Moreover,SP and NSP cell tumorigenesis was examined by subcutaneous and intraperitoneal injection of these cells to nonobes ediabetic(NOD)-severe combined immundeficient(SCID)mice. Drug resistance to cisplatin was examined by cell counting kit-8 (CCK-8).Results SP cells could be isolated stablly and insistently. There was(4.81 ± 0.43)%of SP cells in the established OC cell line and(4.89 ± 0.33)%of SP cells after cultured the isolated SP cells in differentiation condition,and there was no significant different between these two quantities (P>0.05). However,after cultured the NSP cells,there was only (0.10 ± 0.03)%of SP cells which was significantly lower than that contained in the OC cell line(P<0.01). In the tumorigenesis assay 1.0 × 103 SP cells were injected subcutaneously and formed the xenografted tumors in 6 weeks(3/3),and 1.0×104 NSP cells were injected subcutaneously and did not form xenografted tumors in 12 weeks(0). The tumorigenic capability of SP cells was higher than that of NSP cells(P<0.01). Both the original and the xenografted tumors were low differentiated serous cystadenocarcinomas and expressed the ovarian serous cystadenocarcinomas CA125 marker after stained by HE and immunohistochemistry. Simultaneously,the SP cells were also capable to form tumors as shown by intraperitoneal injection. In the drug resistance assay shown that the 50% inhibitory concentration (IC50) of the SP and NSP cells were respectively(2.33 ± 0.14)μg/ml and(1.60 ± 0.04)μg/ml(P<0.05). After treated the unsorted OC cells with cisplatin,the quantity of SP cells increased to(40.10 ± 4.22)%and there was significant difference,when compared to the untreated cells which was(4.81±0.43)%(P<0.01). The SP cells survival rate was(58.7± 3.3)%when treated with cisplatin at its IC50 dose,and the rate decreased to(7.2±1.3)%(P<0.01)when verapamil was present. Conclusions The SP cells could be isolated from the established OC cell line. They had the capacities of self-renewal,differentiation,and tumorigenesis,and the new tumor demonstrated the original tumor′s phenotype. The SP cells also had stem cells′ biological characteristics and is resistant to cisplatin.
9.Quality standard for Fukang Capsules

AIM: To establish the quality standards for Fukang Capsules(Cortex Phellodendri Chinensis,Cortex Ailanthi,Fructus Schisandrae Chinensis,etc.). METHODS: Cortex Ailanthi,Fructus Schisandrae,Poria were identified by TLC,and the content of berberine hydrochloride was determined by TLC-scanning. RESULTS: Cortex Ailanthi,Fructus Schisandrae,Poria could be identified by TLC.Berberine hydrochloride showed a good linear relationship at a range of 25.32 ng-354.48 ng,r=0.993 28.The average recovery was 100.3%,and RSD was 2.04%. CONCLUSION: The method is accurate and can be used for the quality control of Fukang Capsules.
10.Study on Microsatellite Instability in Breast Cancer

Objective To investigate the incidence and clinicopathologic significance of MSI in breast caner.Methods 40 paired sporadic invasive breast cancer were collected.Genomic DNA was extracted from live sample.Twelve microsatellites on chromosomes 2p,3p,5q,6q,16q,17q were amplified for MSI,respectively,by polymerase chain reaction(PCR)with designed primers and detecting after polyacrylamide gel electrophoresis.Results MSI in 15 out of 40(35%)of the carcinomas were observed.There was no MSI in benign hyperplasia.MSI was mainly located at D3S1766,D2S2739 and TP53 in the breast cancer.The incidence rate of MSI in breast cancer is associated with the degree of carcinoma differentiation.Conclusions Microsatellite instability might play a role in the early stage during multistep breast carcinogenesis.MSI indicated poor histologic differentiation in breast carcinoma.D3S1766,D2S2739 and TP53 might be the sensitive sites to detect MSI in breast carcinoma transformation.
