2.Study on base insertion/deletion of CAG-rich region in insulin receptor substrate-1 5′-regulatory sequence

To study the relationship of the polymorphism of the insulin receptor substrate-1(IRS-1) gene 5′-flanking regulatory sequence and Type 2 diabetes,the IRS-1 gene 5′-flanking regulatory sequence was scanned by PCR-SSCP in 78 healthy control subjects and 76 Type 2 diabetic subjects. Applying PCR-denatured polyacrylamide gel electrophoresis and silver staining, the insertion/deletion polymorphism of the CAG-rich region was analyzed. The genome DNA of the normal and variant subjects was amplified with high-fidelity pfu DNA polymerase. The purified and digested target fragments were then subcloned into the pCAT Basic vector. Each allele was identified according to the mobility by the restrictive endonuclease digestion of the recombinant combined with denatured polyacrylamide gel electrophoresis and silver staining, and finally the constructive plasmids containing different alleles were analyzed by DNA sequencing. Firstly, we found several insertion/deletion variations in the CAG-rich region of IRS-1 gene. Secondly,7 genotypes and 6 alleles(T1~T6) in this site were detected.Moreover, T5 and T6 were only observed in Type 2 diabetic group.
3.Observation of clinical curative effect of the joint application of Vitapex paste and AH plus to cure the com-bined periodontic-endodontic lesion

Objective To observe the clinical curative effect of the joint application of Vitapex paste and AH plus,which cured the combined periodontic -endodontic lesion .Methods 72 patients of 113 teeth with the com-bined periodontic-endodontic lesion were chosen ,which were randomly divided into the two groups ,with all complete periodontal initial therapy and routine root canal preparation and dressing at the same time .The observation group included 36 cases of 58 teeth,when the first time to see a doctor ,injected Vitapex paste to the periapical tissue and periodontal tissue sparse area for 1 -2weeks.The sceond time,the tap root canal was closely filled withc AH plus paste and gutta-percha.The control group concludes 36 cases of 55 teeth,with the traditional method,the tap root canal was filled with AH plus paste and gutta -percha directly .Then observation of postoperative emergency response within 1 week,of periodontal clinical indicators (including the PD,BOP,PLI) in 1 month,3 months,6 months after periodontal initial therapy ,and of the long-term curative effect after 1 year.Results 1 week after root canal filling , there were 48 cases no reaction , 9 cases mild reaction and 1 case severe reaction in the observation group , while 47cases no reaction ,7 cases mild reaction and 1 case severe reaction in the control group .There was no statistical difference of evaluation of emergency response between the two groups (χ2 =0.002,P>0.05).Compared with the situations before treatment ,PD,BOP and PLI were all reduced significantly in 1,3 and 6 months after periodontal ther-apy (P<0.05).There was no significant differences of PD ,BOP and PLI between the two groups before treatment (t=0.050,0.019,0.004,all P>0.05),while 1 month (t=2.581,5.984,1.870,all P<0.05),3 months (t=4.806,7.640,1.034,all P<0.05) and 6 months (t=4.612,8.043,1.167,all P<0.05) later,there was statistical difference of decreased value between the two groups .The 1 year follow up showed that significant efficiency in the observation group was 46.55%,and total efficiency was 91.38%,while significant efficiency 27.27% and total effi-ciency 70.91%in the control group ,and there were statistical differences of significant efficiency and total efficiency between the two groups (χ2 =16.761,21.061,all P<0.05).Conclusion With the joint application of Vitapex paste and AH plus ,and two method curing the combined periodontic -endodontic lesion ,there was no evident differ-ence compared with the traditional short -term emergency response .By long-term observation ,found that this method could effectively promote the healing of periapical tissue and periodontal tissue ,and that curative effect is remarkable compared with the traditional method .
4.Observation on clinical effects of herbal cake-partitioned moxibustion for knee osteoarthritis

Objective:To observe the clinical effects of herbal cake-partitioned moxibustion in the treatment of knee osteoarthritis (KOA).
Methods:A total of 120 cases with KOA were randomly divided into two groups by the random digital table, 60 cases in each group. The herbal cake-partitioned moxibustion (HCPM) group was treated by herbal cake-partitioned moxibustion, once per day and five days per week, with ten sessions as one course. The Western medication (WM) group was treated by the oral administration of Diclofenac Sodium Sustained-release Tablets. The knee functions of the patients were assessed by Western Ontario and McMaster Universities osteoarthritis index (WOMAC) before and after the treatment. The clinical effects were observed in the two groups after four-week continuous treatment.
Results:The total effective rate was 86.7% in the HCPM group and 73.3% in the WM group. The difference in the total effective rate between the two groups was statistically significant (P<0.05). After treatment, WOMAC scores in the two groups were statistically different than those of the same group before the treatment (bothP<0.05); the difference between the two groups were statistically significant (P<0.05).
Conclusion:Herbal cake-partitioned moxibustion and oral administration of Diclofenac Sodium Sustained-release Tablets can improve the knee functions of KOA patients, but the therapeutic effect was better by herbal cake-partitioned moxibustion than by oral administration of Diclofenac Sodium Sustained-release Tablets.
5.Testicular tumors in children (report of 55 cases)

Objective To summarize the management of testicular tumors in children. Methods The clinical data of 55 children (age between 2 months to 12 years) with testicular tumors were retrospectively analyzed.Most of the children presented with painless scrotal mass.Of them 25 cases had yolk sac tumors,24 had teratomas,2 lymphomas,1 seminoma,2 dermoid cysts and 1 lymphangioma. Results Of the 55 cases 31(56%) were followed up with a mean time of 3 years.Among the 25 (68%) cases with yolk sac tumors 17 were followed up,of whom,9 were with stage Ⅰ and they underwent high amputation of spermatic cord with orchiectomy;one with stage Ⅳ and 3 with relaps of tumors who underwent chemotherapy and surgery were alive and well.Of the 14 cases with teratomas who were followed up,5 underwent enucleation of teratomas with retention of testiculi;and one with malignant teratoma underwent surgery.All of them are alive and free of tumor recurrence. Conclusions Children with testicular yolk sac tumors in stage I can be managed by orchiectomy alone,while those in stage Ⅱ-Ⅳ can be treated with a combination regimen of surgery and chemotherapy (PVB).Testicular-sparing enucleation surgery can be used in some benign testicular tumors.
6.MR Imaging of Short-term Expansion of Bone Marrow Haemopoiesis In Vivo

Objective To study MR imaging features of bone marrow after priming.Methods 11 cases with lymphoma or myeloma alleviated after treatment were administration of recombinant human granulocyte colony-stimulating factor(rhG-CSF)injection.Two regions of bone marrow in L3 and iliac were followed up with T1WI,and STIR fat saturation before treatment(0 week),and 1 week,2 weeks and 4 weeks after treatment respectively.MRI features were observed by three experienced doctors independently and the average results acted as the final result.In the meantime,CNR of marrow and fat were calculated and paired sample test.Statistical process was performed in SPSS 10.0 for Windows.Results In comparison with 0 week,the signal intensity of bone marrow was no changed in all cases one week after intensive chemotherapy,slight high in 5 cases 2 weeks later and obvious low signal intensity in all cases three weeks after priming with rhG-CSF.In comparison with 0 week,CNR was no any different significance at 1 week,2 weeks and 4 weeks.Bone marrow changes in lumber and iliac were parallel except one case at 2 weeks and one case at 4 weeks.Conclusion Obvious low signal intensity of bone marrow is found three weeks after priming with rhG-CSF.
7.Factors influence long-term effect of diabetes mellitus treated by islet transplantation

Islet transplantation is an effective method treating insulin-dependent diabetes mellitus, which is apt to supersede pancreas transplantation currently. Although human islet transplantation has already performed successfully, its long-term effect is still unsatisfying. Reasons causing loss of islet function is still not fully understood to date. Thus, deep research of factors influencing transplanted islet survival is of importance to improve long-term effect of islet transplantation. Patient selection and tissue matching, collection, preservation and transport of pancreatic organ, isolation and purification of islet cells, implement of islet transplantation, blood glucose control and immunosuppressive drug application during islet transplantation period, functional assessment after islet transplantation, etc. completion quality of each of these technical links is the factor influencing islet transplantation.
8.EFFECTS OF STATIC EXERCISE AND DYNAMIC EXERCISE ON SYSTOLIC TIME INTERVALS (STI) IN CHILDREN

The purpose of this study was to investigate STI changes resulting from static and dynamic exercises in boys, 9-10 years old, so as to reveal the effects of physical work of different types and loads on the heart function.Our research showed that the effects of static load and dynamic load on STI in children were different. After dynamic exercise all items of STI changed significantly and PEP/LVET decreased significantly. No PEP change was found after static exercises (50% and 80% max load). Static exercise of 50% max load resulted in no IVCT change, but static exercise of 80% max load resulted in very significant change of IVCT, and the longest recovery period was found afterwards. Thus, 80% static exercise would result in a heavy, transient heart load in children.Our research also showed that the changes of HR?systolic blood pressure?LVET after dynamic load and static load were significantly different indicating that oxygen consumption in heart muscles was higher during dynamic exercise and the effect of dynamic exercise on heart was stronger.
9.Cloning, Location and Differential Analysis of Transcription Factor Relish Gene from Anopheles stephensi under Different Feeding Conditions

Objective To clone, locate and differentially analyze the transcription factor Relish gene from Anopheles stephensi, and to examine its signals-modulating action on prophenoloxidase cascade and melanization of Plasmodium yoelii oocysts. Methods Relish cDNA of total mosquitoes was amplified by RT-PCR with degenerated primers. Target PCR product was purified,cloned,sequenced and identified. Special Relish gene was amplified with specific primers from hemocytes or midgut,respectively. Semi-quantitative analysis was made under different feeding conditions. Relish message ribonucleic acid was identified with hybridization in situ. Results One cDNA segment of Relish similar to An.gambiae was acquired from An. stephensi. The same Relish gene was also manifested in the hemocytes and midgut. Marked up-regulation expression of Relish was observed at 6,12,24 or 48 h of Plasmodium yoelii infection and at 12 and 24 h after sucking nitroquine-acetate sucrose solution,that was before inducible oocyst melanization. Relish was also expressed in the hemocytes and midguts by ISH. Conclusion Transcription factor Relish of An. stephensi might play a role in signal modulation of Plasmodium yoelii infection and oocyst melanization.
10.The risk factors of severe birth defects and mild birth defects of Guangdong province

Objective To find out and compare the risk factors of the severe birth defects and mild birth defects,and make it more effective to prevent the severe birth defects.Methods Fetus diagnosed with severe birth defects in Shunde district,Foshan during 2016 were selected as case group 1,those who diagnosed with moderate/mild birth defects were selected as case group 2,and those healthy infants whose mother's last menstrual period was at the same time with the case groups were selected as control group.Their mothers' relative information was collected.A case-control study was conducted to analyze the risk factors of the two case groups respectively.We applied Chi-square analysis and multivariate Logistic regressions to find out the independent risk factors.Results Chi-square analysis showed that maternal age above 35 years(χ2=19.69,P<0.01) and gravidity 3 times or more(χ2=10.06,P<0.01) were the risk factors of severe birth defects,and twins was the risk factor of mild birth defects(χ2=4.05,P<0.05).The multivariate Logistic regressions showed that maternal age above 35 years was the independent risk factor of severe birth defects(OR=2.75,P<0.001),and twins was the independent risk factor of mild birth defects(OR=2.22,P=0.05).Conclusion The women with age above 35 years have higher risk to pregnant a baby with severe birth defects and they are worthy of more attention during pre-pregnancy and antenatal examinations.
