Objective To retrospectively analyze the expression of SARS-CoV (IgG) in the serum of hospitalized children who were not suffering from SARS, and retrospectively evaluate the difference of generation of the antibody between the children and adults. Methods Serum samples were collected from 85 hospitalized children, 500 health-check adults, 976 hospitalized adults, and 156 adults recovered from severe acute respiratory syndrome (SARS), and SARS-CoV (IgG) in the specimens was determined by ELISA and biologic chip. Results The level of serum SARS-CoV (IgG) in childhood patients was significantly higher than that in adult patients (17.6% vs 6.3%, P

0.05). Conclusion:There is bigger difference process of tumor apoptosis between different cell types in the same topography site. Survivin may play adjuvant roles and Bcl-2 may play main roles in the inhibition of cell apoptosis in SCLC while the case is just reversed in NSCLC,it suggests that there were different apoptosis inducing factor and different apoptosis pathway due to different cell type in NSCLC and SCLC.

Objective To investigate the effect of high viral loads(HBV DNA concentration in blood ＞2.0 copy/ml)on the vertical transmission of hepatitis B virus in mothers with HBV DNA positivity.Method Forty pregnant women with HBV DNA positivity were divided randomly.double-blindly into 2 groups:at 28 weeks of pregnancy,one group received oral lamivudine(100 mg/d)and the other received oral placebo.The serum HBV DNA loads were tested at 28 and 40 weeks'gestation in mothers,and serum HBV DNA,HBsAg,HBeAg and anti-HBs were examined in infants at 12 month follow up.Result Thirtynine infants finished(one twins)the follow up,and 2 infants lost(5%).Among them 4 infants were confirmed to be HBV infection(10%,4/39),2 in the treatment group(10%,2/20)and 2 in the control group(11%,2/19)(P＞0.05).The serum HBV DNA levels of 40 weeks'gestation in the treatment group,compared with the levels of 28 weeks'gestation in the treatment group and 40 weeks'gestation in the control group,showed a significant decline(P＜0.01).The HBV DNA levels of the mothers whose infants were infected,were(3.1±3.4) copy/ml,(3.1±3.2) copy/ml during 28 and 40 weeks'gestation,and for mothers whose infants were non-infected,the levels were(3.4±2.2) copy/ml,(2.6±1.5) copy/ml respectively(P＞0.05).The mean values of anti-HBs of 18 infants in the treatment group showed no significant difference as compared to 17 infants in the control group,(594±416)U/L vs (458±398) U/L (P＞0.05).Conclusion The pregnant women's HBV DNA loads could be obviously decreased from high viral loads (HBV DNA concentrations in blood＞2.0 copy/ml)after they take lamivudine from 36 weeks'gestation.But it might not reduce the maternal-fetal vertical transmission of HBV infection.

This study was aimed to discuss the distribution and protein expression level ofμ-opioid receptor (MOR) in hippocampus of premenstrual syndrome (PMS) liver-qi stagnation rat model, in order to initially reveal the action mechanism of PMS liver-qi stagnation and intervention effect ofShu-Yu (SY) capsule. Chronic restraint stress method was used to copy PMS liver-qi stagnation rat model.SY capsule ofTiao-Gan prescription was given as intervention. Immunofluorescence (IF) and western blot (WB) technique were used to detect MOR in hippocampal CA1 and CA3 brain area of rats from each group. The results showed that compared with the normal group, the hippocampus MOR distribution arrangement was messy with increased protein concentration in the model group (P< 0.01). After drug intervention, the MOR protein level returned to normal level. It was concluded that the pathogenesis of PMS liver-qi stagnation may be associated with high expression of MOR in hippocampus CA1 and CA3 region of rats. SY capsule can effectively correct and restore it to nearly normal level. It may be one of the central mechanisms in SY capsule treatment of PMS liver-qi stagnation.

Objective To construct the RNA interference(RNAi)recombinant adenovirus vector targeting at human hypoxia-inducible transcription factor 1?(HIF-1?)and to evaluate its effect on human lung adenocarcinoma cell line SPCA-1.Methods The recombinant adenovirus Ad was constructed.HIF-1? inserted with HIF-1? RNAi fragment via AdEasy system.The virus was purifed by CsCl gradient centrifuge.The functional titer of recombinant adenovirus was measured by transfection test in HEK 293 cells.SPCA-1 cells were transducted with 2 multiplicity of infection(MOI)Ad.HIF1? in vitro,the expression rate of green fluorescence protein(GFP)was recorded by flow cytometry,HIF-1? mRNA and protein level was measured by Real-Time RT-PCR and flow cytometry.ResultsThe recombinant shuttle plasmid PAdTrack.HIF-1? and adenovirus plasmid Ad.HIF-1? were all correct shown by enzyme digestion confirmation.The plasmid pAd.HIF-1? was transducted into HEK293 cells,15%GFP expressionwere seen after 3 days.The final titers of recombinant adenovirus were 5.0?1010 TU/mL.SPCA-1 cells was transducted by Ad.HIF-1? in vitro for 48 h,GFP expression rate was 92%,HIF-1? mRNA and protein level decreased 89% and 87%,respectively.Conclusion RNAi adenovirus vector of human HIF-1? gene has been successfully constructed,which could facilitate the research onHIF-1? gene related gene therapy for lung cancer.

AIM: To investigate the anti-cancer effect of cytotoxic T lymphocytes (CTL) activated by sensitized dendritic cells (DCs). METHODS: Immature DCs were induced in vitro from peripheral blood monocytic cells (PBMC) and sensitized by adding tumor cells antigen extract. DCs were identified by their morphology and surface markers. MTT assay was used to evaluate the killing activity of CTL activated by sensitized DCs. The effects of specific CTL cells on inhibiting transplanted tumor HT-29 growth and on preventing HT-29 tumor generation were evaluated by injecting CTL into nude mice. RESULTS: After cultured for seven days, a large number of activated DCs were obtained with typical morphology, extensive stimulatory proliferation capacity and high CD80 (63.5%), CD83 (67.6%) and CD3/HLA-DR (83.2%) expressions. Objective To analyze the significance of global end-diastolic volume index (GEDVI) in acute kidney injury (AKI) after septic shock.Methods A retrospective analysis of 61 patients was performed.The patients were diagnosed of septic shock in emergency ward of Shenyang Military Hospital from 2012 March to 2013 May and were monitored by pulse indicator continuous cardiac output (PiCCO).The patients were divided into two groups:low GEDVI group (GEDVI ＜ 700 ml/m2,29 cases) and high GEDVI group (GEDVI≥700 ml/m2,32 cases) by evaluating GEDVI of 24 hour after PiCCO.Several physiologic and biochemical indexes were recorded,including the hemodynamic parameters at the beginning and the 24 h of PiCCO monitoring,Scr,BUN,lactic acid,incidence and mortality of AKI,baseline glomerular filtration rate,baseline Scr,APACHE Ⅱ scores,mortality during the period of emergency ward or within 28 d after the diagnosis.Results A total of 26 cases in high GEDVI group (81.3％) were attacked with AKI,while 16 cases in low GEDVI group (55.2％) were attacked with AKI,the incidence of AKI in high GEDVI group was significantly higher than that in the low GEDVI group.A COX regression analysis of mortality was performed between the patients staying at emergency ward and during 28 d after diagnosis.The results indicated that AKI and GEDVI had no relation with patients' death.Therefore,AKI and GEDVI could not be considered as the risk factors for the prognosis.Conclusions High GEDVI can significantly increase the incidence of AKI after septic shock,therefore high GEDVI should be avoided as much as possible in the course of clinical treatment.

Objective To investigate the correlation of bone mineral density (BMD) with plasma Klotho levels and its related factors in type 2 diabetic patients (T2DM) . Methods BMD was measured by Dual-energy X-ray absorptiometry (DEXA) in 159 T2DM patients. The patients were divided into three groups:normal bone mass, reduced bone mass and osteoporosis. The fasting plasma levels of Klotho were detected in these patients using enzyme linked immuno sorbent assay (ELISA), clinical and biochemical parameters also were tested, the difference and related factors were compared and analyzed in each group. Results Plasma Klotho levels were not significantly different among the three groups (4.95±0.48 vs 4.96±0.47 vs 4.91±0.49,P>0.05) . BMD at the first, second, third, fourth and total lumbar spine, femoral neck, trochanter and total body were not associated with plasma Klotho levels in these patients (P>0.05) . Age, diabetic duration, HDL-C and BMI were independent determinants for BMD in T2DM patients. Conclusions BMD might be not associated with plasma Klotho level in T2DM patients. But age,diabetic duration,HDL-C and BMI are associated with reduced BMD and osteoporosis in T2DM patients.
