1.Influence of circadian genes hClock and hBmal1 on the chemosensitivity of gastric cancer

Objective:To study the influernce of circadian genes hClock and hBmal1 on the chemosensitivity of SGC-7901 cells. Methods: SGC-7901 cells were cultivated under continuous darkness in vitro.The expression levels of the two main circadian genes hClock and hBmal1 at the different time were determined by real-time polymerase chain reaction(PCR). Docetaxel was administered at the peak and nadir time point respectively. The inhibition of SGC-7901 cell proliferation was measured using a CCK-8 kit. Result:The expression of circadian genes hClock and hBmal1 varied at different times, as shown by real-time PCR. The expression of circadian genes hClock and hBmal1 showed Phase oseillation. The maximum expression of hClock and hBmal1 mRNA was at 20:00. whereas their minimum expression was at 08:00. The inhibition ratio of docetaxel to SGC-7901 cells at the maximum expression of hClock and hBmal1 genes was lower than that at the minimum expression. Conclusion:Circadian Genes hClock and hBmal1 can reduce the drug sensitivity of SGC-7901 cell line to docetaxel in vitro.
2.Peroxisome proliferator activated receptor β agonist, GW501516,ameliorates insulin resistance in glucose intolerant mouse model

AIM The effects of GW501516, a peroxisome proliferator-activated receptor β (PPARβ) agonist, in long term diet induced obesity (DIO, high fat and maltose diet for 4 months) mice were evaluated, and the efficacy of GW501516 against insulin resistance and the involved mechanism was investigated. METHODSMice were divided into 3 groups: normal control, DIO model and DIO model+GW501516. GW501516 (10 mg·kg-1·d-1) was administered by ig once a day for 14 d. During the treatment, body weight and food intake were monitored every other day. The oral glucose tolerance test, and the serum biochemical parameters including the serum triglyceride, total cholesterol and high density lipoprotein cholesterol (HDL-C) levels were measured according to the specifications. To confirm the GW501516-mediated PPARβ activation, the mRNA levels of downstream genes related to glucose, lipid metabolism and energy expenditure was measured. RESULTS GW501516 treatment effectively improved the glucose intolerance, increased the area under the glucose curves[DIO model, (32.4±4.6) mmol·h·L-1 compared with DIO model+GW501516, (23.4±2.5) mmol·h·L-1, n=7-8, P<0.05], normalized the fasted blood glucose, and increased serum HDL-C level, besides, histological analysis revealed the decreased hepatic lipid accumulation and hypertrophy of hepatocyte in DIO mice. Moreover, RT-PCR results indicated that carnitine palmitoyltransferase 1b, uncoupling protein 2, uncoupling protein 3 and glucose transport protein 4 were all upregulated. CONCLUSIONGW501516 significantly ameliorates glucose intolerance, decreases fasted blood glucose and hepatic steatosis, which might be related to ① the enhancement of fatty acid oxidation and energy uncoupling in muscle, and ②the improvement of insulin-stimulated glucose transportation in skeletal muscle in the long term DIO mice.
3.The expression and clinical significance of mineral dust-induced gene in malignant and tuberculosis pleural effusion

Objective To investigate the expression and clinical significance of mineral dust-induced gene(MDIG) in malignant pleural effusions (MPE) and tuberculosis pleural effusions (TBPE).Methods Fifty-four patients with MPE (MPE group) and 50 patients with TBPE (TBPE group) were collected.The MDIG protein was detected by enzyme-linked immunosorbent assay and the relative quantification of MDIG mRNA was measured by real-time quantitation polymerase chain reaction.The cutoff value,sensitivity and specificity of the MDIG protein and MDIG mRNA to diagnose the MPE were evaluated by receiver operating characteristic (ROC) curve analysis.By Spearman correlation analysis,the correlation of MDIG protein and MDIG mRNA was evaluated.Results The MDIG protein in MPE group was significantly higher than that in TBPE group [(304.38 ± 228.47) ng/L vs.(44.43 ± 40.57) ng/L],and there was statistical difference (P < 0.01).The MDIG mRNA in MPE group was significantly higher than that in TBPE group (6.27 ± 3.54 vs.1.82 ± 0.64),and there was statistical difference (P < 0.01).With a cutoff point of 114.23 ng/L,MDIG protein had a sensitivity of 77.8% and a specificity of 94.0% for differential diagnosis.With a cutoff point of 2.75,MDIG mRNA had a sensitivity of 88.9% and a specificity of 92.0% for differential diagnosis.There was a positive correction between MDIG protein and MDIG mRNA (r =0.915,P < 0.01).Conclusions The MDIG protein and MDIG mRNA are highly expressed in MPE with a good sensitivity and specificity.MDIG protein and MDIG mRNA maybe a good clinical indicator in the diagnosis of malignant pleural effusions.
4.Liposoluble constituents from two Varieties of Chrysanthemum morifolium Ramat in Kaifeng City

AIM: To analyze the liposoluble constituents from two Varieties of Chrysanthemum morifolium Ramat(Changhong and Lengyan) cultivated in Kaifeng. METHODS: Changhong and Lengyan were extracted by Soxhlet extraction,and GC-MS was used to analyze their components. RESULTS: Forty compounds were identified from Changhong,amounting to 90.09% of the total constituents.Thirty-five compounds were identified from Lengyan,amounting to 91.57% of the total constituents. CONCLUSION: There appears to be considerable difference in triterpenoids and sterols between the samples of Changong and Lengyan.
5.The application of quantitative transfusion of hemoglobin in neonates

Objective To establish the quantitative transfusion of hemoglobin method and explore the application of this method in neonatal transfusion.Methods This study selected 93 cases of anemic neonates who had no other underlying diseases from the Neonatal Department of Maternal and Child Health (MCH) in Dalian,and they were the principle of weight and age divided into two groups.The first group (control group:conventional method) was transfused and injected the red blood cells that were leukocytereduced,irradiated and washed twice by 0.15 U/kg under the normal reservation.The study measured the hemoglobin value before transfusion and within 24 h after being transfused,and then detected the hemoglobin,volume,and hematocrit of a small amount of blood during the preparation,and finally calculated the utilization rate of hemoglobin in neonates who were transfused.For the second group (observation group:quantitative erythrocyte injection of hemoglobin),it calculated in reverse how much hemoglobin the children needed to be supplemented,based on the doctor's expected hemoglobin values achieved after children being transfused and the utilization rate of hemoglobin obtained from the first group.Then,according to the calculation,it prepared a small amount of blood of the red blood cells that were leukocyte-reduced,irradiated and washed,made the quantitative injection of hemoglobin twice,and measured the hemoglobin value before transfusion and within 24 h after being transfused.At last,it analyzed statistically the results.Results Both two methods effectively improved the children's anemia (P < 0.05).There was no difference (P > 0.05) for two methods in improving the anemia of children,but the quantitative transfusion of hemoglobin (s =6.6,cv =4.6%) could basically reach the doctor's expected hemoglobin value,and it avoided the situations in the first method (s =14.45,vc =8.6%),like that after transfusion a minority of hemoglobin was too high or it did not reach complete amelioration of anemia of the children.Conclusions Both traditional transfusion method and quantitative transfusion of hemoglobin method can improve the anemia of the children,but the latter one is more suitable for the clinical needs and achieves the true quantitative transfusion.
6.Risk factors analysis of liver dysfunction in patients with colorectal cancer after chemotherapy

Objective:To investigate the risk factors, clinical features and prognosis of abnormal liver function after receiving oxaliplatin-containing chemotherapy regimen in patients with colorectal cancer, and to provide a relevant basis for clinical diagnosis and treatment.Methods:The clinical data of 108 colorectal cancer patients who received XELOX (oxaliplatin+capecitabine) or mFOLFOX6 (oxaliplatin+leucovorin+ 5-fluorouracil) chemotherapy regimen from October 2017 to May 2019 in the First Hospital of Shanxi Medical University were analyzed retrospectively. According to the liver function indexes after chemotherapy, the patients were divided into abnormal liver function group and normal liver function group. The observation indexes included alanine aminotransferase, aspartate aminotransferase, total bilirubin, direct bilirubin, alkaline phosphatase, γ-glutamyl transpeptidase. The clinical characteristics of liver dysfunction after oxaliplatin-containing chemotherapy were analyzed and the related factors that might lead to liver dysfunction were analyzed by using multivariate logistic regression analysis.Results:Among 108 patients receiving chemotherapy, there were 67 (62.0%) cases of abnormal liver function. The main grades of liver dysfunction were grade 1 and grade 2, including 49 cases of grade 1 (73.1%) and 16 cases of grade 2 (23.9%). After chemotherapy, the abnormal liver function usually began in 1-4 cycles, of which 22 cases were 1 cycle (32.8%), 17 cases were 2 cycles (25.4%), 20 cases were 3 cycles (29.8%), and 4 cases were 4 cycles (6.0%). Univariate analysis showed that the age <60 years old, chemotherapy cycle >6, the use of mFOLFOX6 regimen, unprotected hepatoprotective drugs were related to liver dysfunction ( χ2 values were 3.910,4.799, 12.861, 4.044; all P < 0.05). Multivariate logistic regression analysis showed that mFOLFOX6 regimen and unprotected hepatoprotective drugs were independent risk factors of abnormal liver function ( HR = 3.405, 95% CI 1.266-9.159, P = 0.015; HR = 2.348, 95% CI 1.012-5.477, P = 0.047). Conclusions:For patients with colorectal cancer who have a high risk of liver dysfunction after chemotherapy, it is recommended to prefer XELOX regimen among oxaliplatin-containing chemotherapy regimens and to take preventive liver protection treatment.
7.Clinical and CYP17A1 gene mutation analysis of 6 children with 17α-hydroxylase/17, 20-lyase deficiency

Objective:To analyze the clinical features and CYP17A1 gene mutation of 17α-hydroxylase/17, 20-lyase deficiency (17OHD). Methods:The clinical data, laboratory examination and genetic results of 6 children with 17OHD in the Department of Endocrinology, Children′s Hospital Affiliated to Capital Institute of Pediatrics from March 2014 to December 2019 were enrolled and analyzed retrospectively.Meanwhile, the clinical types of all congenital adrenocortical hyperplasia (CAH) patients were calculated and then the incidence of 17OHD was calculated.Results:The 6 cases were from 5 families, and the age at diagnosis was ranged from 1 year and 6 months to 15 years old, in which 2 cases were 46, XX and 4 cases were 46, XY.Their gender were all female.Three cases presented with hypertension (50.0%), 4 cases with hypokalemia (66.7%), and 1 case with labia mass (16.7%). The gonad developed into a testis in patients with 46, XY, and patients with 46, XX had ovarian hypoplasia.The laboratory tests revealed an decrease in the cortisol at 8 AM in all cases, ranging from 0.62 to 5.93 mg/L.Five cases displayed an increase in adrenocorticotropic hormone (ACTH) in the range of 84-271 ng/L, and 1 patient with normal ACTH (58 ng/L) had a peak cortisol of 1.75 mg/L after the ACTH challenge test.Elevated progesterone was detected in 6 patients with a normal 17 hydroxyprogesterone level.Further results proved low levels of testosterone and estradiol, and high levels of luteinizing hormone (LH), and follicle stimulating hormone (FSH). CT scan showed mild adrenal hyperplasia in all cases.Among 114 CAH patients during the same period, the incidence of 17OHD came second at 5.3%.The CYP17A1 gene mutation results indicated that 2 unrelated patients were homozygous mutation for p. Y329fs (c.985_987delTACinsAA), 2 siblings were compound heterozygous mutations for p. Y329fs and exon 1-7 deletion, 1 patient was compound heterozygous mutations of p. Y329fs and p. R416C (c.1246C>T), and 1 patient was homozygous mutations for p. L465P (c.1394T> C), which was first reported in China. Conclusions:17OHD is not rare in CAH.Female children with hypokalemia, hypertension, and hypogonadism can lead to diagnostic suspicion of 17OHD.The p. Y329fs mutation in Chinese 17OHD children is a hotspot.The p. L465P (c.1394T>C) mutation is a new mutation in China and it could enrich the mutant spectrum of CYP17A1 gene in China.
8.Studies on effects of calycosin-7-O-β-D-glucoside on prim-O-glucosylcimifugin and cimifugin in vivo pharmacokinetics.

Study on the effects of Astragali Radix main active flavone calycosin-7-O-β-D-glucoside on Saposhnikoviae Radix main active ingredients prim-O-glucosylcimifugin and cimifugin, a UPLC-MS/MS method for simultaneous determination of prim-O-glucosylcimifugin and cimifugin in rat plasma was established, and the comparative pharmacokinetics of prim-O-glucosylcimifugin and cimifugin after oral administration of prim-O-glucosylcimifugin and calycosin-7-O-β-D-glucoside-prim-O-glucosylcimifugin to rats were carried out, which might be conductive in exploring the rationality of Astragali Radix - Saposhnikoviae Radix herb couple. Twelve male SD rats were divided into two groups. Prim-O-glucosylcimifugin and cimifugin in rat plasma of different time points after oral administration of prim-O-glucosylcimifugin and calycosin-7-O-β-D-glucoside - prim-O-glucosylcimifugin to rats were determinated. And the main pharmacokinetic parameters were investigated using DAS 3. 2. 4. The established method was rapid, accurate and sensitive for simultaneous determination of prim-O-glucosylcimifugin and cimifugin in rat plasma. The analysis was performed on a Waters Acquity BEH C18 column (2.1 mm x 100 mm, 1.7 μm) with the mixture of acetonitrile and 0.1% formic acid/water as mobile phase, and the gradient elution at a flow rate of 0.3 mL x min(-1). The analytes were detected by tandem mass spectrometry with the electrospray ionization (ESI) source and in the multiple reaction monitoring (MRM) mode. Compared with prim-O-glucosylcimifugin group, the AUC(0-t)., and AUC(0-∞) of p-O-glucosylcimifugin as well as the C(max) of cimifugin significantly increased (P < 0.05) in calycosin-7-O-β-D-glucoside-prim-O-glucosylcimifugin group. Calycosin-7-O-β-D-glucoside could enhance the absorption of prim-O-glucosylcimifugin and cimifugin and improve the bioavailability, explaining preliminarily the rationality of Astragali Radix-Saposhnikoviae Radix herb couple.

9.Relationship between serum ghrelin and nutrition status in patients with chronic obstructive pulmonary disease

Objective To investigate the correlation between the serum ghrelin levels and nutrition state in the patients with chronic obstructive pulmonary disease(COPD) ,as well as to explore the role of ghxelin in the nutri-tion metabolism. Methods Fifty-three COPD patients were observed, thirty-one of them were with malnutrition (group A), twenty-two COPD patients with normal nutrition status(group B), and twenty were healthy controls.Serum ghrelin, and nutritional parameters such as body mass index(BMI), ideal body weitht( % IBW), mid-upper arm cricumference(MAC), serum albumin(ALB), total lymphocyte counts(LYM) were determined. The correlation between ghrelin and nutritional parameters was analysed. Resuits The level of serum ghrelin in group A were sig-nificantly higher than those in group B and in healthy controls. And the serum ghrelin showed a negative correlation with BMI, % IBW, MAC, but there was no correlation between serum ghrelin level, ALB and LYM. Conclusion Ghrelin participated in the nutrition metabolism in patients with COPD, it would become much higher because of malnutrition in COPD.
10.The relationship of MMP-9, VEGF and PCNA expressions and their clinical significance in gastric adenocarcinoma

Objective To investigate abnormal protein expression of matrix metalloproteinase-9 (MMP-9), vascular endothelial growth factor (VEGF) and proliferating cell nuclear antigen (PCNA) in human gastric adenoearcinoma, and further reveal the clinical significance. Method The MMP-9, VEGF and PCNA proteins expression was determined by immunohistochemistry staining in 45 gastric adenocarcinoma tissues, 45 adjacent specimens and 10 normal gastric mucosa tissues via tissue arrays accordingly. The relationship of these protein expression with differentiation degree, development and progression of gastric adenocarcinoma were also analyzed. Results Positive rates of MMP-9, VEGF and PCNA in gastric adenocarcinoma, adjacent specimens and gastric normal mucosa were as follows: MMP-9, 82.2%(37/45), 64.4% (29/45), 30.0% (3/10) (P=0.019); VEGF, 73.3% (33/45), 62.2% (28/45), 30. 0% (3/10) (P=0.029); PCNA,84.4% (38/45), 71.1% (32/45), 10.0% (1/10) ,there were statistically significant difference (P = 0. 001). The positive rates of MMP-9, VEGF and PCNA in well-differentiated adenocarcinoma, moderately differentiated adenocarcinoma and poorly differentiated adenocarcinoma were as follows: MMP-9,70.0%(7/10), 80. 0% (8/10), 88.0%(22/25), there were statistically significant difference (P=0.015);VEGF, 50.0%(5/10), 60.0% (6/10), 88.0% (22/25), there were statistically significant difference (P =0.000);PCNA, 60.0% (6/10) ,90.0% (9/10) ,92.0% (23/25) ,the difference is significant statistically (P = 0.004). The expression of MMP-9, VEGF and PCNA showed positive relationship with each other by rank correlation analysis (P < 0. 05). Conclusion Tissue arrays technology is effective tool to analyze the expression of cancer related proteins in gastric adenocarcinoma. The expression of MMP-9, VEGF and PCNA proteins participates in the tumorigenesis and development process of gastric adenocarcinoma, and these can be used as indexes to evaluate prognosis in clinical.
