1.Haemostatic percutaneous injection therapy for renal trauma guided by contrast-enhanced ultrasonography

Objective To study haemostatic percutaneous injection therapy for the management of vascular damage in patients with renal injuries guided by contrast-enhanced ultrasonography(CEUS).Methods Which of 56 patients with renal trauma were diagnosis by CEUS,37 cases with grades Ⅱ-Ⅳ renal injuries were brought into our study.According to wound degree and accompanying active bleeding,they were divided into experiment group (percutaneous injection hemostatic treatment)and control group(conservative treatment).Results Thirty-seven renal trauma manifest low perfusion in lesions by CEUS,and the contrast agent could be seen overflow to renal pelvi and the location of capsule in 13 patients.The patients were divided into experiment group(17 cases)and control group(20 cases).The color of hematuria of 9 patients in experiment group became gradually light at 30 mins after treatment.and the color of 7 cases become normal,and hematuria of the only one was iterative appear.The color of hematuria of 9 patients in control group became gradually light in 24-72 hours,others' hematuria became gradually light in 5-14 days.The time of color of hematuria become light of the former was shorter than those of the latter(P<0.05).Reexamination by ultrasound and renal function and urine routine at 1,3 and 6 months after treatment,the results of all patients indicated normal.Conclusions Haemostatic percutaneous injection therapy for renal trauma guided by contrast-enhanced ultrasonography has very obvious hemostatic efficacy.Its advantages included may be used for effective,minimally invasive control of renal injuries(grades Ⅱ-Ⅳ),and can be a feasible management of active bleeding at bedside.
2.Expression and significance of Th17 cells from peripheral blood of patients with rheumatoid arthritis

Objective To study the expression and significance of Th17 cells from peripheral blood of patients with rheumatoid arthritis (RA). Methods Intracelluar flow cytomete detection of IL-17/IFN-γ and IL-17/IL-6 was established using anti-CD3/Anti-CD28/IL-23 as stimulators after isolation of untouched human CD4~+T cells from PBMC. There were three groups in the present study: ①healthy controls group; ② RA stable group; ③RA active group. Results The isolation of untouched human CD4~+T cells from PBMC was effective and its purity was over 90%. The percentage of intracelluar IL-17 in CD4~+ T cells from RA patients was increased significantly. Such percentage in active group (1.54±0.41) was higher than that of stable group (0.70±0.21, P<0.01) and both of them were higher than those of healthy controls (0.42±0.12, P<0.01). Under anti-CD3/Anti-CD28/IL-23 stimulation, the percentage of intracelluar IL-17 was also increased significantly(P<0.01). The porcentage of intracellular IFN-γ was similar to that of IL-17, while that of IL-6 was not significantly different. There is an correlation between IL-17 and IFN-γ or IL-6. Conclusion There is an abnormal expression of IL-17 and IFN-γ in human CD4~+T cells in RA patients, which is related to disease activity . Th17 cells may be used as a new marker for the assessment of RA activity.
3.Combined haemostatic percutaneous injection therapy for renal trauma under guidance of contrast-enhanced ultrasonography

Objective To study feasibility of combined haemostatic percutaneous injection therapy guided by contrast-enhanced ultrasonography (CEUS) in treatment of renal injuries. Methods Eighteen New Zealand rabbits were inflicted with kidney injury imitating grades Ⅲ-Ⅳ blunt injuries. The animals were randomly and equally divided into three groups, Group A ( treated with hemocoagulase),Group B ( treated with hemocoagulase and Alpha-cyanoacrylate) and Group C ( control group, given normal saline). The hemostatic time, hemostatic effect, and perirenal hematoma were observed. Results A perirenal hematoma was observed one hour after treatment. The perirenal fluid thickness was (0.200 ±0.012) cm in Group A, (0.050 ±0.002) cm in Group B and (0.400 ±0.009) cm in Group C, with statistical significance between two test groups and Group C (P < 0.05 ). At days 7 and 14 following treatment, lesion length and cross section was ( 1. 107 ±0. 143) cm and (0.433 ±0. 163) cm in Group A, (0.567 ±0.082) cm and (0. 160 ±0. 078) cm in Group B, and (0.980 ±0. 203) cm and (0.686 ± 0. 157) cm in Group C. There was statistical significance between the test groups (Groups A and B) and Group C (P<0. 01) at day 14. The lesion size in Group A was lager than that in Group B (P < 0.01 ). One month after treatment, a slight nephrohydrosis occurred in Group B. Conclusions Either injection of simple hemocoagulase or combined use of hemocoagulase and Alpha-cyanoacrylate guided by CEUS can attain positive hemostatic effect, but the latter one is more rapid and reliable.
4.Experiment research of contrast-enhanced ultrasonography in diagnosis of pancreatic laceration

Objective To approach the diagnostic value of contrast-enhanced ultrasonography(CEUS)for the detection of traumatic laceration of pancreas. Methods Sixty cases of pancreatic traumatic model were made in twelve healthy swines after the animals were anesthetized and laparotomized. Then the conventional ultrasonography(US) and CEUS were performed in each case to diagnose the traumatic region,immediately. The results were compared with surgical findings. Results Among sixty injuries,the detection rate of conventional ultrasonography was 66. 7%,the detection rate of CEUS was 88.3%. Conclusions CEUS shows higher detection rate than conventional US in diagnosing pancreatic laceration,and it also can improve the diagnostic value of ultrasound for the detection of pancreatic laceration.
5.Changes of sarco(endo)plasmic reticulum Ca2+-Mg2+-ATPase activity of cultured myoblast induced by cyclic stretch.

OBJECTIVETo evaluate the effect of stretch on sarco(endo)plasmic reticulum Ca2+-Mg2+ ATPases activity and mRNA level and study the remodeling reaction of muscle in a variety of mechanical environments.
METHODSMyoblast from maxillofacial skeletal muscle of one-week-old male Sprague-Dawley rat was cultured and stretched cyclicly using a four-point bend device. Inorganic Phosphorus test was used to compare the activity of Ca2+-Mg2+ ATPases of myoblast before and after stretch. RT-PCR was also used to observe the Ca2+-Mg2+ ATPases mRNA level.
RESULTSThe activity of Ca2+-Mg2+ ATPases of myoblast down regulated significantly in 4 hours. During the period of 8 hours to 24 hours, up-regulation followed then returned to control level at the 48 hour point. RT-PCR showed that Ca2+-Mg2+ ATPases mRNA level were elevated by stretch, particularly at 2 hour and 48 hour point.
CONCLUSIONThe results suggested a transcriptional control of Ca2+-Mg2+ ATPases activity was involved in the muscle remodeling process induced by stretch.

6.Clinical evaluation of liver stiffness by acoustic radiation force impulse imaging in patient with ischemic type biliary lesion after orthotopic liver transplantation

Objective To study the clinical value of acoustic radiation force impulse imaging (ARFI) in patients with ischemic type biliary lesion (ITBL) after orthotopic liver transplantation. Methods Between July 2012 to January 2013, forty-one patients in General Hospital of the People′s Liberation Army were enrolled in this study who were admitted for post-transplantation follow-up. Among them, 15 patients were diagnosed as ITBL by ultrasonography. Besides conventional ultrasonography, ARFI was used to detect the shear wave velocity (SWV) of liver tissue at depth of 4 cm and 5 cm respectively. Simultaneously liver function test was performed. Independent-samples t test was performed to compare the difference of SWV between ITBL and non-ITBL group at the same depth. Paired-sample t test was performed to compare the difference of SWV for the same ITBL patient. Pearson correlation analysis was used in analyzing the relation between SWV and liver function. Results The average SWV in depth of 4 cm was (1.561±0.425) m/s and (1.121±0.160) m/s in ITBL and non-ITBL group, respectively. Signiifcant differences were found among the ITBL and non-ITBL groups (t=-3.173, P=0.01). The average SWV in depth of 5 cm was (1.608±0.545) m/s and (1.175±0.173) m/s in ITBL and non-ITBL group, respectively. Signiifcant differences were found among the ITBL and non-ITBL groups (t=-2.454, P=0.034). There was no signiifcant difference between SWV measurements at different depth for the same ITBL patient. For all patients, SWV at different depths were both strongly correlated with alkaline phosphatase (r=0.656, 0.667, respectively;both P=0.000) andγ-glutamyl transpeptidase (r=0.482, P=0.007;r=0.508, P=0.004). Conclusion The liver stiffness measurement is valuable for the clinical evaluation of post-transplantation ITBL.
7.Effects of programmed cell death on human dental follicle cells and changes of programmed cell death under different hydrostatic pressures.

OBJECTIVETooth eruption requires the presence of the dental follicle (DF) around the unerupted tooth. This study is to investigate programmed cell death on human dental follicle cells and changes of programmed cell death under different hydrostatic pressures: 0, 50 and 100 kPa.
METHODSHuman dental follicles from third mandibular molars were surgically removed from adolescents who need for orthodontics treatment after informed content, then trypsinized and cultured. Human dental follicle cells were divided into three groups according to different hydrostatic pressures: 0, 50 and 100 kPa and their programmed cell death were labeled by using TdT-medi-ated-dUTP nick and labeling (TUNEL).
RESULTSDental follicle cells cultured were elongate shape and exhibited fibroblastic characteristics. Compared with 0 kPa, programmed cell death cells on human dental follicle cells were increased 0.23% and 31.65% under 50 kPa and 100 kPa hydrostatic pressures respectively. 100 kPa group increased significantly (P < 0.01).
CONCLUSIONIt suggested that programmed cell death occured in human dental follicle cells cultured in vitro and was influenced by different hydrostatic pressures. Hydrostatic pressure may improve tooth erup-tion through dental follicle.

8.Analysis for expression and significance of interleukin-17, RORγt in CD4+ T cells from patients with ankylosing spondylitis

Objective To study the expression of IL-17, RORγt in CD4+T cells from patients with ankylosing spondylitis (AS). Methods The specimens of venous blood PBMC were collected from 28 patients with AS and 15 healthy subjects. Intracellular flow cytometry detection of IL-17 was established after isolation of human CD4+ T cells from PBMC. The expression level of IL-17, RORγt mRNA in CD4+ T cells was determined from 28 AS patients and 15 healthy controls by real-time fluorescence quantitative RT-PCR using Anti -CD3/Anti -CD28 as stimulators or not. Analysis of variance and Pearson correlation were selected. Results The isolation of human CD4+ T cells from PBMC was effective and its purity reached 90%. The percentage of intracellular IL-17 in CD4+ T cells from AS pati-ents in the AS active group was higher than that of the AS stable group and healthy control group (P<0.01). The expression level of IL-17, RORγt mRNA in CD4+ T cells was significantly higher in patients with AS than in controls. After stimulated with anti-CD3/ anti-CD28 stimulation, the percentage of IL-17, RORγt mRNA was increased significantly (P<0.01). The percentage of IL-17, RORγt mRNA in the 12 h group was higher than that of the 24 h group, while both of them were higher than those without stimulation (P<0.05). Conclusion There is an abnormal expression of IL-17, RORγt in human CD4+ T cells from AS patients. Our results indicate that the abnormal expression of IL-17 might play a role in the development and progression of AS.
9.Changes of the acetylcholine receptor on rat's superficial masseter muscles after functional mandibular advancement.

OBJECTIVEThe purpose of this study was to evaluate the maximum binding (Bmax) and affinity Kd value changes of acetylcholine receptor (n-AchR) on rat's superficial masseter muscles after functional mandibular advancement.
METHODSForty 5-week-old male SD rats were randomly divided into experimental and control groups. The mimic functional appliances were only used in experimental groups, and the rats of two groups were killed after 1, 3, 7, 14 days. Radio-ligand binding assay (RBA) was applied to determine the Bmax and Kd value of n-AchR on the superficial masseter muscles.
RESULTSThe Bmax of n-AchR in experimental groups was higher than that in the control groups at all time points, and the differences had statistical significance. The Kd value of n-AchR was higher in experimental group than that in control groups in 1, 3, 7 days, while Kd value in experimental group of n-AchR was lower than that in control groups in 14 days. The differences had statistical significance in 7 and 14 days, but the differences had no statistical significance in 1 and 3 days.
CONCLUSIONThe functional orthopedics can increase the Bmax and affinity of n-AchR on rapid growing rat's superficial masseter muscles.

10.The effects of growth hormone on rabbit's mandibular condylar chondrocytes proliferation and secretion in vitro.

OBJECTIVETo study the effect of growth hormone (GH) on the proliferation and type II collagen secretion of chondrocytes of mandibular condyle in rabbit in vitro.
METHODSFlow cytometry (FCM) and immunohistochemical technique were employed to observe the possible changes.
RESULTS(1) The exogenic GH can enhance the proliferation and synthesis of DNA of the chondrocytes of mandibular condyle in rabbit in vitro. The suitable concentration of GH is 10 microg/ml. The synthesis of DNA reaches the highest level after 12 hours, while the proliferation index (PI) hits the highest after 24 hours. (2) GH (10 microg/ml) can stimulate the secretion of type II collagen of the chondrocytes.
CONCLUSIONThe exogenic GH can enhance the proliferation, the synthesis of DNA and the secretion of type II collagen of the chondrocytes of mandibular condyle in rabbit in vitro.
